S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

Polygonum cuspidatum root has been traditionally used for the treatment of dental diseases in Korea. The purpose of this study was to evaluate effects of P. cuspidatum root on the development of dental caries, especially its effects against bacterial viability and caries-inducing factors of Strptococcus mutans and Strptococcus sobrinus. Among methanol extract of P. cuspidatum root and its fraction tested, ethyl acetate fraction, composed of polydatin, resveratrol, anthraglycoside B, and emodin, showed inhibitory effects on glycolytic acid production and glucosyltransferase activity of S. mutans and S. sobrinus in addition to antibacterial activities.

<P>Hes6 is a member of the hairy-enhancer of split homolog (Hes) family of transcription factors and interacts with other Hes family genes. During development, Hes genes are expressed in neural stem cells and progenitor cells. However, the role of Hes6 in adult hippocampal neurogenesis remains unclear. We therefore investigated the effects of Hes6 on adult hippocampal neurogenesis, by comparing Hes6 knockout and wild-type mice. To this end, we immunostained for markers of neural stem cells and progenitor cells (nestin), proliferating cells (Ki67), post-mitotic neuroblasts and immature neurons (doublecortin, DCX), mature neuronal cells (NeuN), and astrocyte (S100 beta). We also injected 5-bromo-2'-deoxyuridine (BrdU) to trace the fate of mitotic cells. Nestin- and Ki67-positive proliferating cells did now show any significant differences between wild and knockout groups. Hes6 knockout negatively affects neuroblast differentiation based on DCX immunohistochemistry. On the contrary, the ratio of the BrdU and NeuN double-positive cells did not show any significance, even though it was slightly higher in the knockout group. These results suggest that Hes6 is involved in the regulation of neuroblast differentiation during adult neurogenesis, but does not influence integration into mature neurons.</P>

<P>Abstract</P><P><B><I>Background/Aims:</I></B> Lymphedema is a clinically incurable disease that occurs commonly after lymph node dissection and/or irradiation. Several studies have recently demonstrated that extracorporeal shock wave therapy (ESWT) could promote lymphangiogenesis associated with expression of vascular endothelial growth factor (VEGF)-C. This research concerned primarily the synergistic effect of ESWT combined with VEGF-C incorporated hydrogel (VEGF-C hydrogel) combination therapy for promoting lymphangiogenesis and ultimately alleviating lymphedema. <B><I>Methods:</I></B> The VEGF-C hydrogel was applied to the injury site in a mouse model of lymphedema and then regularly underwent ESWT (0.05 mJ/mm<SUP>2</SUP>, 500 shots) every 3 days for 4 weeks. <B><I>Results:</I></B> Four weeks after the treatment, mice treated with VEGF-C hydrogel and ESWT showed signs of the greatest decrease in edema/collagenous deposits when compared with the other experimental group. LYVE-1-positive vessels also revealed that the VEGF-C/ESWT group had significantly induced the growth of new lymphatic vessels compared to the other groups. Western blot analysis showed that expression of VEGF-C (1.24-fold) and VEGF receptor-3 (1.41-fold) was significantly increased in the VEGF-C/ESWT group compared to the normal group. <B><I>Conclusion:</I></B> These results suggested that VEGF-C and ESWT had a synergistic effect and were very effective in alleviating the symptoms of lymphedema and promoting lymphangiogenesis.</P><P>Copyright © 2012 S. There are two types of beam theories: the first one is the traditional Euler-Bernoulli theory while the second one is Timoshenko beam theory that considers additional effects of shear deformations and rotary inertia. In this investigation, the both theories are utilized to evaluate elastic constants. leading to direct comparisons of the two theories. Also, for a numerical analysis the commercial FEM program. ANSYS 5.3 is employed to conduct modelling of test specimens for a modal analysis, enhancing the understanding of the nature of the FEM analysis and allowing comparisons with experimental results.
A Competitiveness Evaluation of the Wine Industry at Yalu River Valley in China

The wine consumption market in China has been growing rapidly in recent years. The ‘Yalu River Valley’ strategy to promote the local wine industry was introduced in 2013 by the Tonghua City government. However, compared to large wine producing areas, wine produced in the Yalu River Valley has a small share of the market. The purpose of this study is to evaluate the competitiveness of the Yalu River Valley wine industry in China. This study used multicriteria decision making technique analytic hierarchy process methodology to identify each factor’s importance degree with three hierarchies and 31 critical factors. The survey was conducted with the help of 48 relevant experts. The results show that production factor condition is the most important category in the priority list, followed by domestic demand condition, structure and rivalry and related industry. This study would be helpful to give a strategy to policy makers for developing small wineries in China.

In this study, binanoenzyme (BNE) was synthesized using FateDH and Cyt-C in same bionano-interface;by formation of formate dehydrogenase-co-cytochrome C by using ANADOLUCA method. Thesynthesized novel binanoenzyme was embedded into HEMA based cryogel column and the conversionof CO2 to formic acid was investigated by using this cryogel column. To maintain the conversion, CO2saturated water was passed through the cryogel column and the converted formic acid was analyzedusing HPLC. The parameters which has an effect on conversion such as pH, sample volume and CO2concentration were analyzed. The results showed that the conversion process was succeeded effectively.
