      • DMNQ S-64 Induces Apoptosis via Caspase Activation and Cyclooxygenase-2 Inhibition in Human Nonsmall Lung Cancer Cells

        <P>Shikonin has been reported to induce apoptosis and inhibit angiogenesis in vivo and in vitro. 6-(1-propoxyiminoalkyl)-5,8-dimethoxyoxy 1,4-naphtoquinone S-64 (DMNQ S-64) was synthesized as a shikonin derivative. In this article, the underlying apoptotic mechanism of DMNQ S-64 was examined. DMNQ S-64 exerted cytotoxicity against A549 lung carcinoma cells with IC(50) of 27.3 microM. Apoptotic bodies were observed in DMNQ S-64-treated A549 cells by 4'-6-diamidino-2-phenylindole (DAPI) staining assay. DMNQ S-64 also increased sub-G1 DNA portion in a concentration-dependent manner by flow cytometric analysis. Western blotting has revealed that DMNQ S-64 effectively activates the expression of caspase 8, 9, and 3, cleaves poly (ADP-ribose) polymerase, and increases the ratio of Bax/Bcl-2. Furthermore, cytochrome c was released in a concentration-dependent manner by DMNQ S-64. Similarly, DMNQ S-64 significantly increased caspase 3 activity by enzyme-linked immunosorbent assay (ELISA). It also significantly inhibited the level of prostaglandin E2 (PGE(2)) by ELISA and downregulated the expression of cyclooxygenase-2 (COX-2) in a concentration-dependent manner. Taken together, DMNQ S-64 may exhibit cytotoxicity against A549 cells via caspase activation and COX-2 inhibition.</P>

      • Caspase-3 activation as a key factor for HBx-transformed cell death

        <P>Abstract. </P><P><I>Objectives</I>: Nuclear factor-kappa B (NF-&kgr;B) activation has been associated with the tumorigenic growth of hepatitis B virus X protein (HBx)-transformed cells. This study was aimed to find a key target for treatment of HBx-mediated cancers. <I>Materials and methods</I>: NF-&kgr;B activation, endoplasmic reticulum-stress (ER-stress), caspase-3 activation, and cell proliferation were evaluated after Chang/HBx cells permanently expressing HBx viral protein were treated with inhibitors of NF-&kgr;B, proteasome and DNA topoisomerase. <I>Results</I>: Inhibition of NF-&kgr;B transcriptional activity by transient transfection with mutant plasmids encoding Akt1 and glycogen synthase kinase-3&bgr; (GSK-3&bgr;), or by treatment with chemical inhibitors, wortmannin and LY294002, showed little effect on the survival of Chang/HBx cells. Furthermore, I&kgr;Bα (S32/36A) mutant plasmid or other NF-&kgr;B inhibitors, 1-pyrrolidinecarbonidithioic acid and sulphasalazine, were also shown to have little effect on the cell proliferation. By contrast, proteasome inhibitor-1 (Pro1) and MG132 enhanced the HBx-induced ER-stress response and the subsequent activation of caspase-12, -9 and -3 and reduced cell proliferation. Camptothecin (CPT), however, triggered activation of caspase-3 without induction of caspase-12, and reduced cell proliferation. In addition, CPT-induced cell death was reversed by pre-treatment with z-DEVD, a caspase-3-specific inhibitor. <I>Conclusions</I>: Detailed exploitation of the regulators of caspase-3 activation could open the gate for finding an efficient target for development of anticancer therapeutics against HBx-transformed hepatocellular carcinoma.</P>

      • Fabrication of hollow porous PLGA microspheres using sucrose for controlled dual delivery of dexamethasone and BMP2

        <P>Hollow porous poly(D,L-lactide-co-glycolide) microspheres (HP-PLGA-MS) were manufactured using the water-in-oil-in-water double emulsion solvent evaporation method with sucrose as a novel porogen. Dexamethasone (DEX) was dissolved in the oil phase and loaded during the preparation of the HP-PLGA-MS. Bone morphogenetic protein-2 (BMP2) with type I collagen (Coll) was incorporated into HP-PLGA-MS using a simple immersion method. The release rate of BMP2 could be controlled by the amount of Coll. DEX and BMP2 showed a sustained release profile. These results showed that the HP-PLGA-MS could be fabricated successfully using sucrose and showed great potential as a controlled dual drug delivery system.         Bi-S 쾌삭강의 칩생성특성

        In this study, the characteristics of chip formation of the cold drawn Bi-S free machining steels were assessed. And for comparison, those of the cold drawn Pb-S free machining steel, the hot rolled low carbon steel which has MnS as free machining inclusions and the conventional steels were also investigated. During chip formation, the cold drawn free machining steels show relatively little change in thickness and width of chip compare to those of the conventional carbon steels. And a single parameter which indicates the degree of deformation during chip formation, 'chip cross-section area ratio' is introduced. The chip cross-section area ratio is defined as chip cross-section area is divided by undeformed chip cross-section area. The variational patterns of the chip cross-section area ratio of the materials cut are similar to those of the shear strain values. The shear stress, however, seems to be dependent on the carbon content of the materials. The cold drawn Bi-S and Pb-S steels show nearly the same chip forming behaviors and the energy consumed during chip formation is almost same. A low carbon steel without free machining aids shows poor chip breakability due to its high ductility. By introducing a small amount of free machining inclusions such as MnS, Bi, Pb or merely increasing carbon content the chip breakability improves significantly.

        Whole genome de novo sequencing and genome annotation of the world popular cultivated edible mushroom, Lentinula edodes

        <P>Lentinula edodes, the popular shiitake mushroom, is one of the most important cultivated edible mushrooms. It is used as a food and for medicinal purposes. Here, we present the 46.1 Mb draft genome of L. edodes, comprising 13,028 predicted gene models. The genome assembly consists of 31 scaffolds. Gene annotation provides key information about various signaling pathways and secondary metabolites. This genomic information should help establish the molecular genetic markers for MAS/MAB and increase our understanding of the genome structure and function. (C) 2016 Elsevier B.V.       • 중·고등학교 여학생의 월경 전후기 불편감에 따른 월경태도 및 성 역할 고정관념의 관계

        Purpose: The purpose of this study was to provide the basis for program helping adolescent girls to form correct attitude toward menstruation and gender role stereotype by identifying the relationship among menstrual distress, attitudes toward menstruation and gender role stereotype. Method: The 392 participants of this study were selected among students of 5 middle schools and 3 high schools located in Seoul and Gyeong-gi province. Data were collected from 16 September to 23 September, 2013. The students were asked to answer the questionnaires consisting of MDQ(Menstrual Distress Questionnaire), KGES-A(Korean Gender Egalitarianism Scales for Adolescents), and menstrual attitude measurement scale developed by Min in 2003. Descriptive statistics, independent t-test and One way ANOVA were used to analyze the data with SPSS 21.0. Results: The results of this study were as follows: 1. High school students were more vulnerable to pre and post menstrual discomfort and tend to accept menstruation statically than middle school students(p=.009). 2. Students who did not receive regular sexuality education had private attitudes toward menstruation(p=.002), were emotionally more sensitive(p=.002) than the other group. 3. Students, who had higher menstrual discomfort, were more emotionally sensitive toward menstruation(p<.001), had variety of menstrual symptoms(p<.001), actively managing menstrual symptoms(p<.001), and accepted menstruation statically(p=.002) than the lower group. 4. There was no statistically significant relationship between gender role stereotype and pre and post menstrual discomfort. Conclusion: It was suggested that effective education program should be developed and implemented periodically in order to help adolescent school girls to understand pre and post menstrual discomfort and manage menstrual symptoms by themselves. Also, professional and systematic studies should follow up with larger population and more specific general characteristics.

        1. 고온, Hydrogen Attack, 펄라이트 분해(메탄반응) 2. 저온, Hydrogen Embrittlement, Delayed 균열 3. 응력/부식, Hydrogen Stress Corrosion, S/CI 집적동반 수소취성을 대하며 첫 번째 결정은 이것을 손상분류체계 중 어디에 속하게 할 것인가 이었는데, 비교적 큰 갈등 없이 사용상>피로/부식/고온 분류 중 부식에 넣는다. 고전적인 부식은 틈새/갈바닉/응력-부식/입계예민화/피트/마식/염기성부식의 7개 분야로 나눌 수 있는데, 여기에 특별히 “수소취성” 부식을 여덟 번째로 더하는 것이다. 아주 잦지는 않지만 그래도 종종 손상진단 사건들에서 수소취성 관련 의심 증거들이 나타난다. 그런데 사실 손상진단 현장에서 자신 있게 수소취성을 원인으로 내세우는 것은 실험적인 증거가 미약하여 쉽지 않다. 이것은 원자번호 1번인 작은 수소(분자 또는 이온형태)가 금속과 어떤 작용을 하는 가에 대한 분석이 어렵기 때문이다. 본 연구는 수소취성을 종합 정리해 보고자한 것이다. 우선 온도에 따른 반응기구별 분류를 하였는데, 잘 밝혀보지 않은 것은 부식관련 기구로 마지막 분류체계에 넣었다. 이와 같은 분류체계가 수소취성의 “The best”일지는 확신할 수 없다. 하지만 현장에서 종종 의심되는 수소취성의 손상사건들을 인단 정형화된 분류 틀에 넣어 진단 논리를 단순화할 필요가 크다.

      • 뇌막염의 임상적 연구

        Total 62 meningitis cases since 1958 to 1971 are analyzed. T.B. origin are 13 cases,. bacterial 4, syphilitic 3, toxic 1, and unknown origin 41 cases. Age is mainly 2nd and 3rd decades. Male female ratio is approximately equal. Duration of the disease is mainly 1 month, esp. within 2 weeks. The main symptoms are headache, fever, vomiting, stupor, nausea, and dizziness and main signs are stiff neck, Kernig sign, weak deep reflexes, Babinski sign, and semi-coma. The cerebrospinal fluid showed often confusing picture for T. B. meningitis. Medications for both T. B. and bacterial meningitis together resulted in about 67. 7% improvement (probably cure) and the worsening or expiring cases are those who were too late for treatment.
