      • 만성 간염 환자에 대한 Malotilate(Kantecs^�)의 임상적 연구

        Authors carried out the clinical study on Malotilate(Diisopropyl 1,3-dithio-2-ylidenemalonate) in patient with chronic liver diseases and in those with elevated SGOT and SGPT over normal range. We selected three patients with chronic liver diseases and six patients with elevated SGOT and SGPT levels, aged from 26 to 61 years. The daily dose was 3 tablets, each tablet containing 200mg of Malotilate(Kantecs). The clinical and laboratory evaluation were made before and after 8 weeks’ treatment. The results were as followings; 1. Serum total protein and albumin were significantly increased from 6.53±0.055gm/dL to 6.69±0.060gm/dL and from 3.43>0.065gm/dL to 3.71±0.096gm/dL respectively after 8 weeks’ administration of Malotilate(p<0.05, p<0.025). 2. Total cholesterol was significantly increased from 161.3±6.69mg/dL to 184.2±6.97mg/dL after 8 weeks’ administration of Malotilate(p<0.025). 3. The values of SGO, SGPT, and ν-GTP were significantly decreased after 8 weeks’ administration of Malotilate(p<0.05). 4. BUN,S-glucose, total bilirubin and alkaline phosphatase were not significantly altered after Malotilate administration. 5. No severe adverse side effects were found during 8 weeks‘ administration of Malotilate. Accordingly, it could said that Malotilate(Kantecs) was effective in the treatment of chronic hepatic parenchymal diseases without causing adverse effects.

      • 마이크로 컴퓨터를 이용한 맨-머시인 시스템에 의한 최적제어 문제의 해법

        This paper discribes the man machine system based on micro-computer for solving optimal control problems which are basically different in many point from the ordinary solving methods by use of big computer. The proposed man machine system is consist of a simulator related to a given differential equation system. CRT display unit showing instantaneous states of the simulator, micro-computer system for calculating the performance index value function, and a man who plays a role as a part of the overall system. In this man machine system, there are two feedback loops which are the conditional reflection loop and the intelligent feedback loop. By using this man machine system, some experiments were tested to solve optimal control problem of one dimensional heat conduction process. As a result, it was showed that the solutions by man machine system are reasonable ones when that are compared with the solutions by numerical calculation.

        Information has become a strategic tool with progress of sports science and sports technology. Demand on sports information is spreading to leisure, recreation, sociology, science of nutrition, psychology, sports medicine, sports therapy, prophylactic medicine, alternative medicine, sports industry and sports nature etc. if on line data base can be manipulated freely then acceleration and quality improvement of the study in this field can be effected and it is possible to gain new insight for receiving latest information But next problem is cost for use. For this reason this author summarized quick and effective on lline search points as follows: 1. Data base should be chosen with clear purpose for its use. Conduct prior investigation on contents and fee system of data base. With the same data base fee system may be differed depending on distributor therefore it is necessary to try different distributors at first as much as possible. 2. Prior to starting on line search prepare search method. If on line is started and then search method is checked than quickly telephone charge will go up. Search method should be prepared in advance by thinking sufficiently of what theme is to be searched. 3. Look for good advisor: lf there is no advisor nearby then the best thing is counselling with the person in charge for the distributor. If the result of search is stored at floppy disk by "downloading" then even after communication line is cut off it will be possible to print out anytime. 4. In this study the author explained about on-line search of lterature data line but recently it is possible to search overseas sports news or information on rate of victory or defeat of prfessional footbali etc. in real time. In Japan it is possible to search feature articles of Nihon Kesai or Asahi (except culture and sports column) by on-line. However in our country there are not many data bases in the field of sports and physical education which can be on-line searched. Our country belongs to up per level in the world in terms of study of sports science and technology and we are attracting attention from countries in Asia and from different parts of the world and the task facing us now is developing independent and unique data base for our country.

        As Follow suggested : The purpose of this study was to consider tendency of modern sports information. today Modernst information system had four areas such as collection, clearance, communication and application So, most literuture of America and Europe was collected, cleared, communicated and apilcated in international sports data base. If we utillited communication circuit properly, we could use sports information beneficently at the laboratory and office. In the future and now, one was that special sports institution needed to obtain Education about sports imformation such as information of specialization, and information of knowledge on both soft phase and hard phase of new media. Although korea used date base of foriegn country efficiently, korea had to make effort to create its own information system and wish to avoid friction of economy and fiction of information.

      • The Identification of Apoptosis-related Residues in Human Thymosin β-10 by Mutational Analysis and Computational Modeling

        Thymosin β-10 (TB10) is an actin monomer-sequestering peptide that consists of 43 amino acid residues and that tends to form α-helical structures. Previously, we showed that the overexpression of TB10 dramatically increases the frequency of apoptosis in human ovarian cancer cells. To identify the critical residues responsible for TB10-mediated apoptosis, we used a series of computational methods. First, a three-dimensional structure of human TB10 was constructed using the homology modeling method with the calf thymosin β-9 NMR structure as a template. Although the sequences of both of these structures are almost identical, 200-ps molecular dynamics simulation results showed that their secondary structures differ. Analyses of molecular dynamics snapshot structures suggested that the TB10 structure is conformationally more complicated than the TB9 structure. The conserved ^(17)LKKTET^(22) motif region of TB10 was tested by Ala and Ser scanning mutagenesis using computational and biochemical methods, and 12 mutants were transfected into cancer cell lines and tested for their effects on growth arrest. Of the 12 mutants examined, only the Thr ^(20) to Ser^(20) mutation showed reduced growth arrest. These results strongly suggest that Thr^(20) is specifically required for actin sequestration by TB10 in ovarian cancer cells. These results may provide useful information for the development of a new ovarian cancer therapy.

        LKB1/STK11 Tumor Suppressor Reduces Angiogenesis by Directly Interacting with VEGFR2 in Tumorigenesis

        Cervical tumors represent a prevalent form of cancer affecting women worldwide; current treatment options involve surgery, radiotherapy, and chemotherapy. Angiogenesis, the process of new blood vessel formation, is a crucial factor in cervical tumor growth. The molecular mechanisms underlying the effects of the liver kinase B1 (LKB1/STK11) tumor suppressor protein on tumor angiogenesis have not been elucidated. Therefore, we investigated the role of LKB1 in cervical tumor angiogenesis both in vitro and in vivo in this study. Our results demonstrated that LKB1 inhibited cervical tumor angiogenesis by suppressing the expression of angiogenesis-related factors such as vascular endothelial growth factor (VEGF) and hypoxia inducible factor-1α. LKB1 directly affected both carcinoma and vascular endothelial cells, resulting in a significant reduction in tumor growth and angiogenesis. Furthermore, LKB1 was found to bind to VEGF receptor 2 (VEGFR-2) and target the VEGFR-2-mediated protein kinase B/ mechanistic target of rapamycin signaling pathway in endothelial cells, thereby reducing cervical tumor growth and angiogenesis. Our study provides new insights into the molecular mechanisms underlying the anti-tumor and anti-angiogenic effects of LKB1 in cervical cancer. These findings will help develop new therapeutic strategies for cervical cancer.

        Novel Anti-Angiogenic and Anti-Tumour Activities of the N-Terminal Domain of NOEY2 via Binding to VEGFR-2 in Ovarian Cancer

        The imprinted tumour suppressor NOEY2 is downregulated in various cancer types, including ovarian cancers. Recent data suggest that NOEY2 plays an essential role in regulating the cell cycle, angiogenesis and autophagy in tumorigenesis. However, its detailed molecular function and mechanisms in ovarian tumours remain unclear. In this report, we initially demonstrated the inhibitory effect of NOEY2 on tumour growth by utilising a xenograft tumour model. NOEY2 attenuated the cell growth approximately fourfold and significantly reduced tumour vascularity. NOEY2 inhibited the phosphorylation of the signalling components downstream of phosphatidylinositol-3’-kinase (PI3K), including phosphoinositide-dependent protein kinase 1 (PDK-1), tuberous sclerosis complex 2 (TSC-2) and p70 ribosomal protein S6 kinase (p70S6K), during ovarian tumour progression via direct binding to vascular endothelial growth factor receptor-2 (VEGFR-2). Particularly, the N-terminal domain of NOEY2 (NOEY2-N) had a potent anti-angiogenic activity and dramatically downregulated VEGF and hypoxia-inducible factor-1α (HIF-1α), key regulators of angiogenesis. Since no X-ray or nuclear magnetic resonance structures is available for NOEY2, we constructed the threedimensional structure of this protein via molecular modelling methods, such as homology modelling and molecular dynamic simulations. Thereby, Lys15 and Arg16 appeared as key residues in the N-terminal domain. We also found that NOEY2-N acts as a potent inhibitor of tumorigenesis and angiogenesis. These findings provide convincing evidence that NOEY2-N regulates endothelial cell function and angiogenesis by interrupting the VEGFR-2/PDK-1/GSK-3β signal transduction and thus strongly suggest that NOEY2-N might serve as a novel anti-tumour and anti-angiogenic agent against many diseases, including ovarian cancer.

        Knockdown of LKB1 Sensitizes Endometrial Cancer Cells via AMPK Activation

        Metformin is an anti-diabetic drug and has anticancer effects on various cancers. Several studies have suggested that metformin reduces cell proliferation and stimulates cell-cycle arrest and apoptosis. However, the definitive molecular mechanism of metformin in the pathophysiological signaling in endometrial tumorigenesis and metastasis is not clearly understood. In this study, we examined the effects of metformin on the cell viability and apoptosis of human cervical HeLa and endometrial HEC-1-A and KLE cancer cells. Metformin suppressed cell growth in a dose-dependent manner and dramatically evoked apoptosis in HeLa cervical cancer cells, while apoptotic cell death and growth inhibition were not observed in endometrial (HEC-1-A, KLE) cell lines. Accordingly, the p27 and p21 promoter activities were enhanced while Bcl-2 and IL-6 activities were significantly reduced by metformin treatment. Metformin diminished the phosphorylation of mTOR, p70S6K and 4E-BP1 by accelerating adenosine monophosphatea-ctivated kinase (AMPK) in HeLa cancer cells, but it did not affect other cell lines. To determine why the anti-proliferative effects are observed only in HeLa cells, we examined the expression level of liver kinase B1 (LKB1) since metformin and LKB1 share the same signalling system, and we found that the LKB1 gene is not expressed only in HeLa cancer cells. Consistently, the overexpression of LKB1 in HeLa cancer cells prevented metformin-triggered apoptosis while LKB1 knockdown significantly increased apoptosis in HEC-1-A and KLE cancer cells. Taken together, these findings indicate an underlying biological/physiological molecular function specifically for metformin-triggered apoptosis dependent on the presence of the LKB1 gene in tumorigenesis.

        Snail Promotes Cancer Cell Proliferation via Its Interaction with the BIRC3

        Snail is implicated in tumour growth and metastasis and is up-regulated in various human tumours. Although the role of Snails in epithelial-mesenchymal transition, which is particularly important in cancer metastasis, is well known, how they regulate tumour growth is poorly described. In this study, the possible molecular mechanisms of Snail in tumour growth were explored. Baculoviral inhibitor of apoptosis protein (IAP) repeat-containing protein 3 (BIRC3), a co-activator of cell proliferation during tumourigenesis, was identified as a Snail-binding protein via a yeast two-hybrid system. Since BIRC3 is important for cell survival, the effect of BIRC3 binding partner Snail on cell survival was investigated in ovarian cancer cell lines. Results revealed that Bax expression was activated, while the expression levels of anti-apoptotic proteins were markedly decreased by small interfering RNA (siRNA) specific for Snail (siSnail). siSnail, the binding partner of siBIRC3, activated the tumour suppressor function of p53 by promoting p53 protein stability. Conversely, BIRC3 could interact with Snail, for this reason, the possibility of BIRC3 involvement in EMT was investigated. BIRC3 overexpression resulted in a decreased expression of the epithelial marker and an increased expression of the mesenchymal markers. siSnail or siBIRC3 reduced the mRNA levels of matrix metalloproteinase (MMP)-2 and MMP-9. These results provide evidence that Snail promotes cell proliferation by interacting with BIRC3 and that BIRC3 might be involved in EMT via binding to Snail in ovarian cancer cells. Therefore, our results suggested the novel relevance of BIRC3, the binding partner of Snail, in ovarian cancer development.
