        Retinoic acid induces expression of Ig germ line α transcript, an IgA isotype switching indicative, through retinoic acid receptor

        Retinoic acid (RA) is considered to possess an activity of IgA isotype switching. Thus far, TGF-β1 is known to be the most powerful IgA isotype switch factor. To elucidate the molecular mechanisms underlying the Ig germ line (GL) α transcriptional regulation by RA, we constructed three different sizes of mouse GLα promoter reporters; short-GLα(-130/+14), middle-GLα(-448/+72) and long-GLα(-3028/+72). Based on luciferase assay, RA increased the activity of all three GLα promoter reporters by approximately 2-fold and the effect was further enhanced by TGF-β1. Overexpression of Smad3/4 increased TGF-β1-induced GLα promoter activities but had no effect on RA-induced GLα promoter activities. In order to analyze the characteristics of the RA-inducible GLα promoter region,we also constructed two mutant reporters: Smad3 binding elements (SBEs)-substituted short-GLα (short-GLα mSBE)and Runx3 binding elements (RBEs)-substituted short-GLα(short-GLα mRBE) promoter reporters. Promoter activities of the two mutant reporters to RA were comparable to that of wild type reporter, while those of the two mutant reporters to TGF-β1 were markedly diminished as compared to that of WT short-GLα. Finally, RA-induced GLα transcription was virtually disappeared by LE540, an antagonist of RA receptor (RAR). Taken together, these results suggest that RA induces GLα transcription mainly through RAR pathway, where neither Smad3/4 nor Runx3 is involved.

        류마티스관절염 환자의 인슐린 저항성 결정인자

        Objective: Rheumatoid arthritis (RA) is associated with an increased cardiovascular events. These may be related to insulin resistance (IR). We evaluated status of IR and analyzed the relationship between IR and clinical and laboratory characteristics in patients with RA. Methods: We examined 52 RA patients (43 females) and 52 age and sex matched healthy controls. We measured Homeostasis model assessment (HOMA) IR, calculated according to fasting serum glucose and insulin. Results: In patients, age was 50.8±10.2 years old, disease duration was 42.1±30.9 months. In controls, HOMA IR was 0.62±0.30 and in patients, it was 1.28±0.50. This difference was highly significant (p<0.001). Patients with early RA (disease duration is less than 36 months) were 28, and established RA (more than 36 months) were 24. HOMA IR was significantly higher in patients with established RA (1.42±0.45) than those with early RA (1.16±0.45) (p=0.03), and significantly correlated with disease duration (r=0.36, p=0.01), BMI (r=0.36, p<0.001), total cumulative prednisolon dose (r=0.34, p=0.01). Disease duration and BMI were independent predictors for HOMA IR (p<0.01, p=0.03). Conclusion: In patients with RA, IR measured by HOMA IR was more significantly increased than that of healthy control and significantly correlated with disease duration, BMI, and total cumulative prednisolon dose; however, the determinants of HOMA IR in RA patients were disease duration and BMI.

        소득계층요인에 따른 자연녹지와 도시공원의 접근성 분석

        Natural greenspace and urban parks provide places for people to experience nature and engage in physical activity. For these reasons, greenspace and urban parks can be an index of environmental benefits. An equity mapping analysis was used to evaluate the area and the accessibility to the greenspace and urban parks by socioeconomic status. This study analyzed the accessibility of greenspace and urban parks in Seongnam-si, according to their locations using a Geographic Information System (GIS). In Seongnam-si, we found patterns of inequity in the distribution of urban parks and greenspace. This inequity can cause environmental inequity due to the lack of opportunity to use the greenspace and urban parks. The study shows how a GIS-based buffer analysis in conjunction with statistical analysis of socioeconomic data can be used to analyse the equity of access to community greenspace and urban parks. The results can be used to inform the local planning process and the GIS approach can be expanded into other local authority domains.

      • 젊은 남자 성인의 칼슘과 철의 영양상태에 관한 연구

        A survey was performed to 174 male young men who were living alone(40 persons) and shared living with friends(4143 persons) in Chungnam province. Their average age was 22.6 years old, height 171.8 ㎝, and weight 65.6 ㎏. Their daily calcium intake level was averaged 271.9±169.1 ㎎, which was 38.8 %RDA. Comparing calcium intake of each mealtime, breakfast, lunch, dinner, and snacks were composed of 18.6%, 30.0%, 34.0% and 17.4%, respectively. Their daily iron intake level was averaged 8.9±10.8 ㎎, which was 74.3 %RDA. Comparing iron intake of each mealtime, breakfast, lunch, dinner, and snacks were composed of 15.4%, 33.6%, 36.3% and 14.7%, respectively. These results suggested that deficiency of the two minerals in young male adults were severe and the nutrient qualities were also poor, no matter of their living alone or with accompanies. The deficiency was more profound in calcium than iron. And we observed iron nutriture differed significantly according to their living status, such as iron intake and its %RDA were significantly worse in single living subjects than the others. However calcium did not show significant difference. We concluded that shared living environment in young male adults could possibly improve their dietary mineral intake levels.

        류마티스관절염과 만성 호산구성 폐렴에 동반된 호산구성 흉막삼출

        We describe a 48-year-old man with family history of rheumatoid arthritis (RA) affected by chronic eosinophilic pneumonia (CEP) with severe peripheral eosinophilia. CEP might develop as a complication of longstanding active RA. The patient with 5 months history of seropositive RA and chronic respiratory symptoms, alveolar and blood eosinophilia, peripheral pulmonary infiltrates and pleural effusion on chest imaging. The lung may be involved as an extraarticular manifestation of RA. However, CEP is not recognized as a typical lung manifestation of RA, and the two diseases rarely coexist. The effusion was an eosinophil predominant exudates and was characterized by low pH, and glucose level and high lactic dehydrogenase. The patient responded rapidly to combination of steroids and disease modifying anti-rheumatic drugs.

      • 동천의 정화능력 평가

        This study was carried out to investigate the self-purification capacity of Dongcheon in Sunchon. According to water quality of Dongcheon, the mean concentrations of BOD and DO were 0.7∼1.6㎎/ℓ and 7.1∼8.0㎎/ℓ, which correspond to the grade Ⅱ of water quality standards. The ratio of BOD loading inflowed from Sunchon-city, Seocheon and Okchoen were measured 4.8%, 52.1% and 23.7%, respectively. The deoxygenation constant(k_1) and reaeration constant(k_2) of Dongchoen were estimated as 3.19day^(-1) and 2.181day^(-1). The results of DO distribution analysis by Streeter-Phelps equation showed that critical point was 12.1㎞ downstream, and then DO concentration was 6.94㎎/ℓ. In addition, the self-purification capacity assessment indicated that BOD loading to maintain over 5㎎/ℓ of DO should be less than 0.925ton/day.

      • 세균속간 시데로포아 생산능의 비교에 Chrome azurol S 한천배지 확산법의 응용

        연구배경 : 세균이 생산하는 시데로포아를 정량적으로 측정하기 위한 새로운 측정법인 chrome azurol S 한천배지 확산법(CASAD)이 저자들에 의해 개발되었다. 본 연구에서는 이 CASAD 법이 세균속간 시데로포아 생산을 비교하는 데에도 사용될 수 있는지를 알아보고자 하였다. 방법: Staphvlococcus aureus, Staphylococcus eptdermidis, Bacillus cereus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Vibrio vulnificus 균을 철을 제거한배지에 트렌스페린을 철공급원으로 공급한 배지에서 배양하면서 세균들의 증식은 파장 600 nm에서 흡광도를 측정하여 관찰하고 시데로포아 생산능은 CASAD법을 이용하여 관찰하였다. 트렌스페린으로부터 철을 획득하는 능력을비교하기 위하여 6 M urea를 함유하는 전기영동법을 사용하였다. 결과: 세균속간 시데로포아 생산능에는 큰 차이가 있었다. S. aureus, S. epidermidis, E. coli, P. aeruginosa는 B. cereus, B. subtilis, V. vulnificus체 비해 더 많은 양의 시데로포아를 생산할 수 있었고 트렌스페린으로부터 철을 더 효율적으로 획득할 수 있었으며 트렌스페린이 첨가된 배지에서 더 잘 자랄 수 있었다. 시데로포아 생산능과 트렌스페린으로부터 철을 획득하는 능력에는 밀접한 상관관계가 있었다. 결론: 본 연구를 통해 저자들은 CASAD법이 세균속간 시데로포아 생산능을 비교하는데에도 유용하게 사용될 수 있음을 알 수 있었다. Background: We recently developed a new chrome azurol S agar diffusion (CASAD) assay for the quantitative analysis of siderophore production. This study was designed to evaluate the usefulness of the CASAD assay for the comparison of the ability to produce siderophore among different genera of bacteria. Materials and Methods: Strains of Staphy lococctis aureus, Staphylococcus epidermidis, Bacillus cereus, Bacilhis subtilis, Escherichia coli, Pseudomonas aeruginosa, and Vibrio vulniftctis were used. The bacteria were cultured in deferrated heart infusion broth containing transferrin at 37℃ for 24 hours. During the cultures, Bacterial growth was measured by OD_(600), siderophore production was measured by CASAD assay, and iron-removal from transferrin was measured by 6 M urea-gel electrophoresis. Results: There was a big difference in the ability of the bacteria to produce siderophores. S. aureus, S. epidermidis, E. coli, and P. aeruginosa produced larger amounts of siderophores than B. cereus, B. subtilis, and V. vulnificus. S. aureus. S. epidermidis, E. coli, and P. aeruginosa utilized more efficiently transferrin-bound iron than B. cereus, B. subtilis, and V. vulnificus. The ability of the bacteria to produce siderophores was consistent with the ability of the bacteria to grow on transferrin-bound iron and to uptake iron from human transferrin. Conclusion: Through this study, we found that the CASAD assay is a useful universal method that can be applied for the intergeneric comparison of the ability to produce siderophores.
