        Immobilization of γ-glutamyltranspeptidase on Silylated Mesoporous TiO2 Whiskers

        The surface of mesoporous TiO2 whiskers (MTw)was chemically modified with 3-aminopropyltriethoxysilane(APTES). The point of zero charge (pzc) of MTw and themodified material (MTwA) was 5.3 and 6.8, respectively. MTw and MTwA were then used as carrier to immobilizeγ-glutamyltranspeptidase (GGT) from Bacillus subtilis (B. subtilis). Our results indicated that the loading capacity ofMTwA for GTT was significantly superior to that of MTw. Although the optimum temperature and thermal stability ofMTwA-GGT were slightly lower than those of MTw-GGT,its pH stability was greatly improved compared with eitherfree enzyme or MTw-GGT. The affinity constant (Km) ofMTwA-GGT to γ-glutamyl-p-nitroanilide (GpNA) was0.889 mM, higher than that of free enzyme but lower thanthat of MTw-GGT. In addition, MTwA-GGT displayed agood operational stability.

      • Predictive control for vehicular sensor networks based on round-trip time-delay prediction

        <P>With the rapid development of vehicular sensor networks (VSNs) technology, the potential use of networked real-time control and automation is enormous and appealing. However, closed-loop control VSNs via the Internet are very difficult to implement practically because of their stochastic nature. One of the biggest challenges in VSNs is how to deal with sensor networks time delay and data loss. The authors investigate the potential of using the predictive control scheme for VSNs based on round-trip time (RTT) delay prediction to overcome the VSNs transmission delay and data loss. The predicted RTT delay is taken as the sampling interval reference for variable-period sampling approach. Modelling the time delay of networked control systems as a non-linear time series, the least mean square (LMS) filter algorithm is adopted to predict online the time delay induced in VSNs. The simulation results show that the LMS algorithm can achieve ideal efficacy if time delay has no variety abnormally. The predictive compensation strategy is proposed to reduce the detrimental effect of stochastic time delays induced by communication networks on control performance. The results of offline simulations via the Internet illustrate that the predictive control scheme based on RTT delay prediction has the potential to overcome the VSNs time delay and data loss.</P>

        Association Analyses with Carcass Traits in the Porcine KIAA1717 and HUMMLC2B Genes

        By screening a subtracted cDNA library constructed with mRNA obtained from the longissimus dorsi muscles of F1 hybrids Landrace${\times}$Yorkshire and their Yorkshire female parents, we isolated two partial sequences coding for the H3-K4-specific methyltransferase (KIAA1717) and skeletal muscle myosin regulatory light chain (HUMMLC2B) genes. In the present work we investigated two SNPs, one (C1354T) at the 3' untranslated region (UTR) of KIAA1717 and one (A345G) at the SINE (PRE-1) element of HUMMLC2B, in a resource population derived from crossing Chinese Meishan and Large White pig. The selected pigs were genotyped by means of a PCR-RFLP protocol. Significant associations were observed for the KIAA1717 C1354T polymorphic site with thorax-waist backfat thickness (p<0.05), buttock backfat thickness (p<0.05), average backfat thickness (p<0.05), loin eye height (p<0.05), loin eye area (p<0.05), carcass length to 1$^{st}$ spondyle (p<0.01) and carcass length to 1st rib (p<0.01). HUMMLC2B A345G were significantly associated with loin eye width (p<0.05), loin eye area (p<0.05). Further studies are needed to confirm these preliminary results.

        Isolation, Expression Pattern, Polymorphism and Association Analysis of Porcine TIAF1 Gene

        TIAF1 is a TGF-${\beta}$1-induced anti-apoptotic factor that plays a critical role in blocking TNF (tumor necrosis factor) cytotoxicity in mouse fibroblasts and participates in TGF-${\beta}$-mediated growth regulation. In this study, we obtained the full-length cDNA sequence of the porcine TIAF1 gene. Real-time PCR further revealed that the TIAF1 gene was expressed at the highest level in liver and kidney with prominent expressions detected in uterus, and lower levels detected in heart, spleen, lung, stomach, small intestine, skeletal muscle and fat of Large White pigs. Sequence analysis indicated that a 6 base-pair deletion mutation existed in the exon of the TIAF1 gene between Meishan and Large White pigs. This mutation induced deletion of Gln and Val amino acids. PCR-RFLP was used to detect the polymorphism in 394 pigs of a "Large White${\times}$Meishan" $F_{2}$ resource population and four purebred pig populations. The frequencies of the A allele (with a 6 bp deletion) were dominant in Chinese Meishan and Bamei pigs, and the frequencies of the B allele (no 6 bp deletion) were dominant in Large White and Landrace pigs. Association analyses revealed that the deletion mutation had highly significant associations (p<0.01) with meat marbling score of the thorax-waist longissimus dorsi (LD) muscle (MM1) and intramuscular fat percentage (IMF), and significant associations (p<0.05) with carcass length (CL). The results presented here supply evidence that the 6 bp deletion mutation in the TIAF1 gene affects porcine meat quality and provides useful information for further porcine breeding.

        Age-dependent Changes of Differential Gene Expression Profile in Backfat Tissue between Hybrids and Parents in Pigs

        Large White, an introduced European pig breed, and Meishan, a Chinese indigenous pig breed, were hybridized directly and reciprocally and a total of 260 pigs, including purebreds, Large White and Meishan, and their hybrids, White${\times}$Meishan (LM) and Meishan${\times}$Large White (ML) pigs, were bred in our laboratory. The mRNA differential display PCR (DD-PCR) was used to detect the age-dependent changes of differential gene expression in backfat tissue between hybrids and parents. Some measures were taken to reduce the false positives in our experiment. Among the total of 2,686 bands obtained, 1,952 bands (about 72.67%) were reproducible and eight patterns (fifteen kinds) of gene expression were observed. The percentage of differentially expressed genes between hybrids and parents is 56.86% at the age of four months and 57.71% at the age of six months. This indicated that the differences of gene expression between hybrids and their parents were very obvious. U-test was used to compare the patterns of gene expression between the age of four and six months, and results showed that bands occurring in only one hybrid and bands displayed in one hybrid and one parent were significantly different at p<0.05, and bands visualized in only two hybrids were significantly different at p<0.01. These indicated that differential gene expression between hybrids and parents changed at different ages.

        Investigation of Gene and Microsatellite Heterozygosities Correlated to Growth Rate in the Chinese Meishan Pig

        A total of 130 Chinese Meishan piglets were scored for their genotypes at five gene loci and five microsatellite loci. The average gene heterozygosity was 0.3338 and the average microsatellite heterozygosity was 0.2954, and the heterozygosity of the overall ten loci was 0.3146. The data of birth weight (BW) and body weight on day 35 (W35) were collected, average daily gain (ADG) for each individual was computed as the slope from the regression of weight on age. There was no significant correlation between individual heterozygosity and birth weight (p>0.05). Significant regressions were observed between ADG and the heterozygosity of loci (p<0.01). Similar results were observed in W35 and loci heterozygosity (p<0.01). Heterozygosity at these ten loci explained 43.62% of the total variation in ADG and 45.48% in W35. Significant correlations existed not only in the function of gene loci but also in neutral microsatellite loci, so it indicated that associative overdominance affected piglet growth significantly.

      • Study in Mechanism of Hydrogen Retention by C - SiC Films with IR

        C-SiC films with different content of SiC on stainless steel substrate were prepared with ion beam mixing. It was found that hydrogen concentrations in C-SiC coatings was higher than that in stainless steel after H^+ ion implantation followed by thermal annealing. Infrared (IR) transmission measurement was used to study the mechanism of hydrogen retention by C-SiC films. The vibrational features in the range between 400 and 3200 ㎝-¹ in IR transmission spectra show the Si-CH₃, Si-CH₂, Si-H, CH₂, and CH₃ bonds, which are responsible for retaining hydrogen.

        Oviduct-specific Glycoprotein 1 Locus is Associated with Litter Size and Weight of Ovaries in Pigs

        Oviduct-specific glycoprotein 1 (OVGP1) is implicated in playing a role in fertilization and early embryo development. In this study, we have obtained the sequence of intron 9 of OVGP1 gene in swine. Comparative sequencing of Meishan (a native Chinese breed) and Large White pig breeds revealed an A/T substitution at position 943. A PCR-EcoRI-RFLP assay was developed to detect this mutation. Polymorphism analysis in Qingping animals showed that pigs with BB genotype had lower number of piglets born alive (NBA) in multiple parities than pigs with AA (p<0.05) and AB genotype (p<0.01). In Large $White{\times}Meishan$ ($LW{\times}M$) $F_2$ offspring, the weight of both ovaries (OW) of the BB genotype was significantly lighter than that of AB (p = 0.05) and AA (p<0.01) genotypes. Analysis of the data also revealed that the mutation locus affected these two traits mostly by additive effects. These studies indicated that the polymorphism was associated with NBA and OW in two distinct populations and further investigations in more purebreds or crossbreds are needed to confirm these results.

        Investigation on the Weld Damage Behavior of Steel Beam-to-column Connection

        A large number of steel moment frames fractured at the welds of beam-to-column connections in earthquake. As the weld damage behavior is a crucial factor for aseismic performance of steel frame connections, 20 local welded connections representing beam-to-column connections were tested under monotonic and cyclic loads to study the weld damage behavior in this paper. The failure modes and deformation curves were recorded; the effects of material strength, load types, and beam geometries on the connection damage behavior were analyzed. Three damage evolution models were calibrated, the relation between weld damage and macro connection performance was established. The results indicate weld cracking and plate buckling are the main causes of the connection damage; the range of cyclic loads is the key factor affecting the weld damage level; the plastic strain based damage model illustrate the damage process with good accuracy. This study provide technical basis for the damage evaluation and fracture prevention of the welded steel beam-to-column connections.

      • Enhanced strength and ductility in particulate-reinforced aluminum matrix composites fabricated by flake powder metallurgy

        Reinforcement agglomeration always leads to severe stress concentration and porosity, which is detrimental to the deformation ability and mechanical properties of particulate-reinforced metal matrix composites. In this study, uniform distribution of 32vol%B<SUB>4</SUB>C has been achieved in B<SUB>4</SUB>C/Al composite by means of flake powder metallurgy (Flake PM), in which flake Al powder is used as the starting material. The flake Al powder exhibits higher apparent volume than spherical powders of the same mass, and thus can provide more space to accommodate the B<SUB>4</SUB>C particles. Therefore, compared with conventional PM, Flake PM can lead to more uniform distribution of B<SUB>4</SUB>C particles in the composite powder as well as in the consolidated composite. Meanwhile, the flake Al powder has a nano skin of Al<SUB>2</SUB>O<SUB>3</SUB>, which could be fractured and dispersed inside the fine matrix grains during consolidation, and were found to induce a higher normalized strain hardening rate for the composite during deformation. As a result, the Flake PM 32vol%B<SUB>4</SUB>C/Al composite exhibits an ultimate tensile strength of 305MPa and a uniform elongation of 6.6%, 63% stronger and 13% more ductile than its counterpart fabricated by conventional PM.
