      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

      • A long-term tunnel settlement prediction model based on BO-GPBE with SHM data

        The new metro crossing the existing metro will cause the settlement or floating of the existing structures, which will have safety problems for the operation of the existing metro and the construction of the new metro. Therefore, it is necessary to monitor and predict the settlement of the existing metro caused by the construction of the new metro in real time. Considering the complexity and uncertainty of metro settlement, a Gaussian Prior Bayesian Emulator (GPBE) probability prediction model based on Bayesian optimization (BO) is proposed, that is, BO-GPBE. Firstly, the settlement monitoring data are analyzed to get the influence of the new metro on the settlement of the existing metro. Then, five different acquisition functions, that is, expected improvement (EI), expected improvement per second (EIPS), expected improvement per second plus (EIPSP), lower confidence bound (LCB), probability of improvement (PI) are selected to construct BO model, and then BO-GPBE model is established. Finally, three years settlement monitoring data were collected by structural health monitoring (SHM) system installed on Nanjing Metro Line 10 are employed to demonstrate the effectiveness of BO-GPBE for forecasting the settlement.

      • Interaction of Coping Styles and Psychological Stress on Anxious and Depressive Symptoms in Chinese Breast Cancer Patients

        Purpose: This study aimed to assess possible interactive effects of coping styles and psychological stress on depression and anxiety symptoms in Chinese women shortly after diagnosis of breast cancer. Methods: Four hundred and one patients with breast cancer were face-to-face interviewed by trained research staff according to a standardized questionnaire including information on socio-demographic characteristics, psychological stress, coping styles, and anxiety and depressive symptoms. Interactive effects were assessed by hierarchical multiple regression analyses. Results: There were significant associations of the four domains of psychological stress with anxiety and depressive symptoms except for the relationship between "worrying about health being harmed" and depressive symptoms. "Abreaction coping behavior" and "escaping coping behavior" significantly increased the level of both anxiety and depressive symptoms; whereas an "active coping style" reswulted in significant decrease. The interaction of "active coping behavior" with "worrying about health being harmed" significantly increased the risk of the anxiety symptoms, while adopting "self-relaxing coping behavior" was associated with significant decrease. The interaction of "worry about daily life and social relationship being restricted" with "escaping coping behavior" significantly increased the risk of the depressive symptoms. Conclusions: The results of this study suggest that certain coping styles might moderate the association of psychological stress with anxiety and depressive symptoms in Chinese women with breast cancer.

        Uremic pruritus: pathophysiology, clinical presentation, and treatments

        Uremic pruritus is one of the most common and bothersome symptoms in patients with end-stage renal disease. Most patients withuremic pruritus experience a prolonged and relapsing course and significant impairments of quality of life. The pathophysiology of uremicpruritus is not completely understood. A complex interplay among cutaneous biology and the nervous and immune systems hasbeen implicated, with the involvement of various inflammatory mediators, neurotransmitters, and opioids. Uremic pruritus treatmentoutcomes are often unsatisfactory. Clinical trials have mostly been small in scale and have reported inconsistent results. Recent evidenceshows that gabapentinoids, nalfurafine, and difelikefalin are effective for relieving uremic pruritus in hemodialysis patients.This review provides an overview of the epidemiology and proposed mechanisms of uremic pruritus, then highlights the manifestationsof and clinical approach to uremic pruritus. Current evidence regarding treatment options, including topical treatments, treatmentof underlying disease, phototherapy, and systemic treatments, is also outlined. With a better understanding of uremic pruritus,more therapeutic options can be expected in the near future.

        Effects of Temperature and Ethylene Response Inhibitors on Growth and Flowering of Passion Fruit

        This study examined the effects of different day/night temperature regimes or silver ion on growth and flowering of passion fruit 'Tai-nung No.1'. Low temperature treatment ($20/15^{\circ}C$) caused passion fruit cultivar 'Tai-nung No.1' to fail to flower. Flowering induction occurred within a temperature range of $20-30^{\circ}C$, with no significant difference in the days to first flower bud and the total number of flower buds between plants grown at $30/25^{\circ}C$ and $25/20^{\circ}C$. However, plants grown at $30/25^{\circ}C$ exhibited their first flower buds set on the higher nodes and had higher abortion rates of flower buds than those at $25/20^{\circ}C$. Plants grown at $30/25^{\circ}C$ had the most rapid growth and the shortest plastochron. We also evaluated the effect of the ethylene response inhibitors silver nitrate ($AgNO_3$) and silver thiosulfate (STS) on growth and flowering of potted passion fruit 'Tai-nung No.1', when they were exposed to low temperature conditions ($20/15^{\circ}C$) following chemical treatments ($AgNO_3$ or STS, at 0.5 or 1.0 mM). $AgNO_3$ and STS treatments induced flower formation and initial flower bud formation within approximately two weeks at $20/15^{\circ}C$ whereas non-treated control plants exhibited no flower formation. ACC content and activity of ACC oxidase in the leaves of passion fruit 'Tai-nung No.1'exposed to low temperature conditions ($20/15^{\circ}C$) were significantly inhibited by the ethylene inhibitor treatments. These results indicate that ethylene, which is produced under low temperature conditions, plays an important role in inhibiting flower formation in passion fruit.

        Operable Breast Cancer of the Inner Hemisphere Is Associated with Poor Survival

        Purpose: This study investigated the clinicopathological featuresof operable breast cancer lesions located in different hemispheresof the breast and determined related survival outcomes. Methods: Data from 5,330 patients with invasive ductal carcinomawere retrospectively analyzed based on tumor location. Results:The median follow-up time was 68 months (range, 18–176months). Patients with breast cancer located in the outer hemisphereof the breast had lesions with more advanced nodalstages and more frequently received adjuvant chemotherapythan patients with breast cancer in the inner hemisphere. The5-year disease-free survival (DFS) rates of patients with tumorslocated in outer versus inner hemispheres were 81.5% and77.0%, respectively (p=0.004); the overall survival (OS) rateswere 90.7% and 88.8%, respectively (p<0.001). The associationbetween tumor location and the 5-year DFS rate was most apparentin node-positive patients (73.1% vs. 65.8% for outer vs. inner hemisphere lesions, p<0.001) and in patients with primarytumors greater than 2 cm in diameter (78.2% vs. 72.3%,p=0.002). Multivariate analysis showed that tumor location wasan independent predictor of DFS (hazard ratio [HR], 1.23;p=0.002) and OS (HR, 1.28; p=0.006). There were no significantdifferences in 5-year DFS or OS rates between patients with outerversus inner hemisphere tumors when internal mammary nodeirradiation was performed. Conclusion: This study demonstratedthat tumor location was an independent prognostic factor foroperable breast cancer. Internal mammary node irradiation isrecommended for patients with breast cancer of the inner hemisphereand positive axillary lymph nodes or large primary tumors

      • ZD1839 and Cisplatin Alone or in Combination for Treatment of a Nasopharyngeal Carcinoma Cell Line and Xenografts

        This study evaluated the effects of ZD1839, an orally active, selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, on nasopharyngeal carcinoma (NPC) both in vitro and in vivo. Influence of ZD1839 alone or combined with cisplatin on the NPC cell line CNE2 was detected by MTT assay with flow cytometry assessment of cell cycle distribution and apoptosis rates. Nude mice NPC xenografts were also used to evaluate the effects of ZD1839 alone or combined with cisplatin. The Student's t test evaluated statistical significance. ZD1839 alone or combined with cisplatin inhibited CNE2 cell line proliferation. ZD1839 induced CNE2 cell cycle arrest in the G1 phase, and higher concentrations induced apoptosis. Xenograft tumors were significantly smaller when treated with 200 mg/kg ZD1839, cisplatin, or cisplatin combined with 100 mg/kg ZD1839 than untreated controls. ZD1839 (200 mg/kg) alone showed good tumor inhibition effects, reduction of tumor weights, and smaller tumor volume without loss of body weight. ZD1839 (200 mg/kg) might provide a good and effective therapeutic reagent for NPC.

        Using Chlorophyll Fluorescence and Vegetation Indices to Predict the Timing of Nitrogen Demand in Pentas lanceolata

        The objective of this study was to predict the timing of nitrogen (N) demand through analyzing chlorophyll fluorescence (ChlF), soil-plant analysis development (SPAD), and normalized difference vegetation index (NDVI), which are positively correlated with foliar N concentration in star cluster (Pentas lanceolata). The plants were grown in potting soil under optimal conditions for 30 d, followed by weekly irrigation with five concentrations (0, 4, 8, 16, and 24 mM) of N for an additional 30 d. These five N application levels corresponded to leaf N concentrations of 2.62, 3.48, 4.00, 4.23, and 4.69%, respectively. We measured 13 morphological and physiological parameters, as well as the responses of these parameters to various N-fertilizer treatments. The general increases in Dickson's quality index (DQI), above-ground dry weight (DW), total DW, flowering rate, ${\Delta}F/Fm$', and qP in response to treatment with 0 to 8 mM N were similar to those of SPAD, NDVI, and Fv/Fm. Consistent and strong correlations ($R^2$= 0.60 to 0.85) were observed between leaf N concentration (%) and SPAD, NDVI, ${\Delta}F/Fm$', and above-ground DW. Validation of leaf S PAD, NDVI, and ${\Delta}F/Fm$' revealed that these vegetation indices are accurate predictors of leaf N concentration that can be used for non-destructive estimation of the proper timing for N-solution irrigation of P. lanceolata. Moreover, irrigation with 8 mM N-fertilizer i s recommended w hen leaf N concentration, SPAD, NVDI, and ${\Delta}F/Fm$' ratios are reduced from their saturation values of 4.00, 50.68, 0.64, and 0.137%, respectively.

        Genetic diversity and population structure of the amylolytic yeast Saccharomycopsis fibuligera associated with Baijiu fermentation in China

        The amylolytic yeast Saccharomycopsis fibuligera is a predominant species in starters and the early fermentation stage of Chinese liquor (Baijiu). However, the genetic diversity of the species remains largely unknown. Here we sequenced the genomes of 97 S. fibuligera strains from different Chinese Baijiu companies. The genetic diversity and population structure of the strains were analyzed based on 1,133 orthologous genes and the whole genome single nucleotide polymorphisms (SNPs). Four main lineages were recognized. One lineage contains 60 Chinese strains which are exclusively homozygous with relatively small genome sizes (18.55–18.72 Mb) and low sequence diversity. The strains clustered in the other three lineages are heterozygous with larger genomes (21.85–23.72 Mb) and higher sequence diversity. The genomes of the homozygous strains showed nearly 100% coverage with the genome of the reference strain KPH12 and the sub-genome A of the hybrid strain KJJ81 at the above 98% sequence identity level. The genomes of the heterozygous strains showed nearly 80% coverage with both the sub-genome A and the whole genome of KJJ81, suggesting that the Chinese heterozygous strains are also hybrids with nearly 20% genomes from an unidentified source. Eighty-three genes were found to show significant copy number variation between different lineages. However, remarkable lineage specific variations in glucoamylase and α-amylase activities and growth profiles in different carbon sources and under different environmental conditions were not observed, though strains exhibiting relatively high glucoamylase activity were mainly found from the homozygous lineage.
