The therapeutic effects of the leaves of Couroupita guianensis, a large tropical tree in the family of Lecythidaceae improving testosterone-induced Benign Prostatic Hyperplasia (BPH) were tested in vitro and in vivo. In BPH rats induced by castration and testosterone treatment, the prostate index was improved in groups administered with the extracts of C. guianensis extracted with 50%-, 100%-ethanol or boiling water, which was comparable with positive control, finasteride. The extract C. guianensis leaves showed significant inhibition on the expressions of type 2 5-alpha reductase (5αR) in RWPE-1 human prostatic epithelial cells, and effectively attenuated the expressions of androgen receptor, type 2 5αR and proliferating cell nuclear antigen in LNCap human prostatic adenocarcinoma cells. The leaves of C. guianensis that exerted evident suppression on BPH-related biomarkers in vitro and improvement of prostate index in vivo has a potential therapeutic use for the treatment of BPH.

Natural products with antioxidant activity have received a great attention for photoaging therapy because this disease is well known as premature skin changes after continuous and long-term repeated exposure to chronic ultraviolet (UV)A and UVB. To investigate therapeutic effects of methanol extracts of Dipterocarpus tuberculatus Roxb. (MED) against UV-induced photoaging, alterations on antioxidant activity, anti-apoptotic effects, ECM modulation, skin appearances and anti-inflammatory response were assessed in normal human dermal fibroblasts (NHDFs) cells and nude mice treated with MED. MED showed the high level of tannin content (879.27 mg/g) and high free radical scavenging activity to DPPH (IC50 = 7.06 μg/ml). UV-induced alterations on NO concentration, SOD activity and expression, and Nrf2 phosphorylation were remarkably recovered by MED treated NHDF cells. The number of apoptotic cells, Bax/Bcl-2 ratio and cleaved Cas-3/Cas-3 ratio were significantly decreased in UV+MED treated group, while G2/M phase arrest was increased in the same group. Also, similar recoveries after MED treatment were detected on b-galactosidase, MMP-2/9 expression and intracellular elastase activity. MED treatment induced the suppressing COX-2 induced iNOS mediated pathway, expression of inflammatory cytokines and inflammasome activation in UV-radiated NHDF cells. Furthermore, anti-photoaging effects observed in NHDF cells were measured in UV+MED treated nude mice although there are differences in the level of change. The skin phenotypes including wrinkle score, hydration, TEWL, melanin index and erythema index as well as histopathological structure were significantly improved UV+MED treated mice. Thus, these results suggest that MED has therapeutic effects against photoaging induced by UV, and that it presents the potential for development as a treatment for photoaging in the future.
