        Anti-inflammatory Effects of Quercetin and Vitexin on Activated Human Peripheral Blood Neutrophils - The effects of quercetin and vitexin on human neutrophils -

        Objectives: Polymorphonuclear neutrophils (PMNs) constitute the first line of defense against invading microbial pathogens. Early events in inflammation involve the recruitment of neutrophils to the site of injury or damage where changes in intracellular calcium can cause the activation of pro-inflammatory mediators from neutrophils including superoxide generation, degranulation and release of myeloperoxidase (MPO), productions of interleukin (IL)-8 and tumor necrosis factor ${\alpha}$ ($TNF-{\alpha}$), and adhesion to the vascular endothelium. To address the anti-inflammatory role of flavonoids, in the present study, we investigated the effects of the flavonoids quercetin and vitexin on the stimulus-induced nitric oxide (NO), $TNF-{\alpha}$, and MPO productions in human neutrophils. Methods: Human peripheral blood neutrophils were isolated, and their viabilities were determined by using the Trypan Blue exclusion test. The polymorphonuclear leukocyte (PMNL) preparations contained more than 98% neutrophils as determined by morphological examination with Giemsa staining. The viabilities of cultured neutrophils with various concentrations of quercetin and vitexin ($1-100{\mu}M$) were studied using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assays. Neutrophils were cultured in complete Roswell Park Memorial Institute (RPMI) medium, pre-incubated with or without quercetin and vitexin ($25{\mu}M$) for 45 min, and stimulated with phorbol 12-myristate 13-acetate (PMA) ($10^{-7}M$). NO production was carried out through nitrite determination by using the Griess method. Also, the $TNF-{\alpha}$ and the MPO productions were measured using enzyme-linked immunosorbent assay (ELISA) kits and MPO assay kits. Results: Neutrophil viability was not affected up to a concentration of $100{\mu}M$ of quercetin or vitexin. Both quercetin and vitexin significantly inhibited $TNF-{\alpha}$, NO, and MPO productions in human neutrophils (P < 0.001). Conclusion:The present study showed that both quercetin and vitexin had significant anti-inflammatory effects. Thus, treatment with either quercetin or vitexin may be considered as a therapeutic strategy for treating patients with neutrophil-mediated inflammatory diseases.

Immediately after stimulation, neutrophilic enzymes are activated and produce toxic mediators such as pro-inflammatory cytokines, nitric oxide (NO) and myeloperoxidase (MPO). These mediators can be toxic not only to infectious agents but also to host tissues. Because flavonoids exhibit antioxidant and anti-inflammatory effects, they are subjects of interest for pharmacological modulation of inflammation. In the present study, the effects of rutin on stimulus-induced NO and tumor necrosis factor $(TNF)-{\alpha}$ productions and MPO activity in human neutrophils were investigated. Methods: Human peripheral blood neutrophils were isolated using Ficoll-Hypaque density gradient centrifugation coupled with dextran T500 sedimentation. The cell preparations containing > 98% granulocytes were determined by morphological examination through Giemsa staining. Neutrophils were cultured in complete Roswell Park Memorial Institute (RPMI) medium, pre-incubated with or without rutin ($25{\mu}M$) for 45 minutes, and stimulated with phorbol 12-myristate 13-acetate (PMA). Then, the $TNF-{\alpha}$, NO and MPO productions were analyzed using enzyme-linked immunosorbent assay (ELISA), Griess Reagent, and MPO assay kits, respectively. Also, the viability of human neutrophils was assessed using tetrazolium salt 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), and neutrophils were treated with various concentrations of rutin ($1-100{\mu}M$), after which MTT was appended and incubated at $37^{\circ}C$ for 4 hour. Results: Rutin at concentrations up to $100{\mu}M$ did not affect neutrophil viability during the 4-hour incubation period. Rutin significantly decreased the NO and $TNF-{\alpha}$ productions in human peripheral blood neutrophils compared to PMA-control cells (P < 0.001). Also, MPO activity was significantly reduced by rutin (P < 0.001). Conclusion: In this in vitro study, rutin had an anti-inflammatory effect due to its inhibiting NO and $TNF-{\alpha}$ productions, as well as MPO activity, in activated human neutrophils. Treatment with rutin may be considered as a therapeutic strategy for neutrophil-mediated inflammatory/autoimmune diseases.

      • Demographic Survey of Four Thousand Patients with 10 Common Cancers in North Eastern Iran over the Past Three Decades

        Background: Cancer is a major cause of mortality in developing countries and correct and valid information about the epidemiology of this disease is the first step in the planning of health care in each region. The aim of this study was to determine the relative frequency, mean age and sex ratio of the most 10 common non-skin cancers in the world and Iran, among patients referred to an oncology clinic. Materials and Methods: This descriptive study was conducted in Mashhad, north east of Iran. The data obtained from the records of patients referred to the private oncology center between the years of 1985-2012". According to the latest report of GLOBOCAN study commonest malignancies included were lung, breast, colorectal, prostate, stomach, liver, cervix, esophageal, bladder cancers and Non-Hodgkin lymphoma. Results: A total of 4,606 cases were analyzed. The mean age was $55.5{\pm}13.8years$ (male: $59.5{\pm}13.9$, female: $52.6{\pm}12.9$). Overall, breast cancer (1,264 cases, relative frequency of 27.4%) was the most prevalent cancer; however the mean ages of diagnosis were not significantly different between 5-year time period divisions (p=0.290). The most common cancer in men was esophageal cancer (26.3%).The lowest mean age was related to women diagnosed with breast cancer ($48.5{\pm}11.8$) and men with non-Hodgkins lymphoma ($48.4{\pm}17.8$). There were statistically significant differences between the mean age of men and women with gastric (p=0.003) and esophageal cancers (p<0.001). Male to female sex ratios in our study for bladder, lung and stomach cancers were 6.57, 2.60 and 2.50 respectively. Conclusions: The results showed that breast cancer tends to be found in younger female patients and bladder cancer appears more often in men. Screening in target population in addition to early diagnosis may reduce death and disability.

Conclusion: The present study showed that both quercetin and vitexin had significant anti-inflammatory effects. Thus, treatment with either quercetin or vitexin may be considered as a therapeutic strategy for treating patients with neutrophil-mediated inflammatory diseases.

        Flocculation properties of a natural polyampholyte

        Polyelectrolytes are commonly used as flocculants in drinking water treatment. However the growing concerns about their toxicity have motivated the search for biocompatible flocculants. Here, we show that gelatin, a natural amphoteric polyelectrolyte, can be effectively adsorbed on clay surfaces and can potentially be a suitable substitute for existing flocculants. The adsorption of gelatin from its aqueous solution onto the mineral clay surfaces at different conditions was systematically investigated using the design of experiments methodology. The gelatin adsorption was found to vary considerately with pH variation showed a maximum adsorption at its isoelectric point. The amount of adsorbed gelation increased with increasing pH from 3 to 5, attained a maximum at pH 5 and then decreased with increasing pH from 5 to 11. Similarly, the amount of adsorbed gelatin showed decreasing trends around salt concentration of 0.05 M and temperature 35°C. On the other hand, the adsorption was continuously increased with time and polymer concentration in the range of 0.1-0.9 mg/dL. Finally, the jar tests confirmed the ability of gelatin for using a natural flocculant for water treatment.

        Developing a wireless sensor network based on a proposed algorithm for healthcare purposes

        This letter describes a developed wireless sensor network based on a proposed algorithm for monitoring the environmentalparameters in healthcare intentions. This proposed algorithm contains a frame with diff erent packets that are implemented onthe developed wireless sensor network. The developed wireless sensor network consists of one central node as well as foursensor node that has been equipped with various sensors such as temperature, humidity, CO, CO 2 , and passive infrared sensor. In order to test the presented algorithm and the developed wireless sensor network, the sensor nodes are situated in fourdiff erent rooms in a hospital for recording essential parameters of the environment while the central node is put in the nursestation for warning to nurses. The obtained result of the proposed sensor nodes in comparison to gold standards shows rootmean square error 1.1%, 0 .35 ◦C , 0.98% for humidity, temperature and gas, respectively. Also, the obtained results illustratethat the system gives accurate feedback from environmental temperature, humidity, and CO, and CO 2 to the nurse station inorder to increases the possibility of a healthy environment condition for patients.

        Modeling of the solubility of H2S in [bmim][PF6] by molecular dynamics simulation, GA-ANFIS and empirical approaches

        Predicting the solubility of acid gases in ionic liquids (ILs), has lately appeared as advantageous for natural gas purifying, which is equipped by powerful models considering technical and economic aspects. Important issue in the assessment of ILs for potential utilization in gas sweetening process is estimating the H2S solubility at various temperatures and pressures Experimental measurements are costly and take considerable time and effort. As a result, proposing methods for predicting the behavior of this system over a wide range of conditions is vital. In this regard, molecular dynamics simulation (MD) technique as well as artificial intelligence knowledge of hybrid genetic algorithmadaptive neuro fuzzy inference system (GA-ANFIS) and an empirical polynomial regression (PR) model were employed to estimate the solubility of H2S in [bmim][PF6] IL. Pressure and temperature are considered as the independent input variables and H2S solubility as the dependent output variable. The results of this study reveal that the simple fourthorder PR model and GA-ANFIS have the highest accuracy. As a result of the simplicity and accuracy of PR model, it can be used without any prior knowledge about MD and artificial intelligence (AI). According to the accuracy and precision of model proved by the obtained result, the solubility of H2S in ILs has been estimated. The results show that the PR method is more trustworthy than other models.
