The conversion of rice-fields to uplands is increased in last 8 years due to the government s policy. To obtain basic informations on plant-parasitic nematodes in converted uplands, we surveyed nematodes associated with crops cultured on the ridge of the rice-fields. The crops mostly cultivated at the ridge of rice-field were soybean (66%), red bean (21%), and perilla (13%). Ten important plant-parasitic nematodes species were identified: Helicotylenchus sp., Heterodera glycines, Meloidogyne arenaria, M. javanica, M. hapla, Paratylenchus projectus, Pratylenchus coffeae, P. neglectus, Rotylenchus incultus, Trichodorus sp. Tylenchorhynchus crassicaudatus, and T. claytoni. Among them, Rotylenchus incultus was the most common plant-parasitic nematode in all three crops. The ridge is a border of rice field, thus plant-parasitic nematodes infested on ridges should be cautioned for future cropping.
밀양근교에서 채집한 야생 동충하초 계통의 PCR 산물에 근거한 계통 유전학적 연구

The phylogenetic relationships among thirty-two strains (P1∼P32; including Cordyceps sp., Paecilomyces sp., Beauveria sp., Aranthomyces sp., Isaria sp. and Himenostilbe sp.) in Miryang region located in the southern part of Korea, were investigated based on internal transcribed spacer (ITS) sequences of ribosomal DNA. A fragment of ITS region was amplified by polymerase chain reaction (PCR) using the specific primer pairs ITS1 and ITS4. After obtained same size of PCR products from various strains, we cloned them into a pGEM-T easy vector to determine their sequences. BLAST analyses of the nucleotide sequence ITS1, 5.8S and ITS2 gene fragments revealed the identity and their phylogenetic relationship. Among 32 strains isolated from Miryang region, Cordyceps militaris was shared 100% sequences with Genbank (AY49191, EU825999, AY491992), while some species are not shared perfectly with reported sequences. For example, strain P17 (P. tenuipes in Ulju-gun Gaji Mountain) has some differences among the other strains of P. tenuipes (Miryang-si Jocheon-eup, Miryang-si Gaji Mountain) and those of gene bank. We conclude that ITS analyses with strains in the suburbs of Miryang in this study can be effectively used as a tool for classification, evaluation and collection of the natural eco-type genetic resources.
Molecular Biological Analysis for the Identification of Meloidogyne Species on Strawberry in Korea

This study is the identification of root-knot nematode (RKN), Meloidogyne hapla, from strawberry in Korea using molecular analyses. Strawberry plants showed localized stunting and galled roots. Molecular analyses of COⅡ/lrRNA, 28S rDNA D2-D3 segment and internal transcribed spacer (ITS) region were employed for the identification of Meloidogyne spp. Polymerase Chain Reaction (PCR) amplification of COⅡ/lrRNA region produced a single fragment ca 528bp. Restriction digestion of the amplified PCR products with Dra1 enzyme produced two fragments at 200 and 250bp indication M. hapla. 28S rDNA D2-D3 segment and ITS were cloned and sequenced. 28S rDNA D2-D3 segment and ITS region produced a single fragment of 1004bp and 560bp, respectively. In BLAST search in Genbank, all sequences accord with other known M. hapla. As a result, all of the RKN samples were M. hapla. This study suggests that the dominant species of RKN on strawberry is M. hapla in Korea.

<P><B>Abstract</B></P> <P>We conducted biochemical and physiological experiments to investigate the mode of action of tiafenacil (Terrad'or™), a new protoporphyrinogen IX oxidase (PPO)-inhibiting pyrimidinedione herbicide. Analysis of the half-maximal inhibitory concentration (IC<SUB>50</SUB>) against recombinant PPO enzymes from various plant species, including amaranth (<I>Amaranthus tuberculatus</I>), soybean (<I>Glycine</I> max), arabidopsis (<I>Arabidopsis thaliana</I>), and rapeseed (<I>Brassica napus</I>), showed that tiafenacil had an IC<SUB>50</SUB> of 22 to 28 nM, similar to the pyrimidinedione herbicides butafenacil and saflufenacil and the <I>N</I>-phenylphthalimide herbicide flumioxazin. By contrast, tiafenacil exhibited 3- to 134-fold lower IC<SUB>50</SUB> values than the diphenyl ether herbicides fomesafen, oxyfluorfen, and acifluorfen. Tiafenacil is non-selective and is herbicidal to both dicots and monocots, such as the weeds velvetleaf (<I>Abutilon theophrasti</I>), amaranth, and barnyardgrass (<I>Echinochloa crus-galli</I>) as well as the crops soybean, rapeseed, rice (<I>Oryza sativa</I>), and maize (<I>Zea mays</I>) at concentrations ranging from 1 to 50 μM. Treatment of plant tissue with tiafenacil in darkness resulted in the accumulation of protoporphyrin IX. Subsequent exposure to light increased the content of malondialdehyde and significantly decreased the Fv/Fm values of chlorophyll fluorescence. Characterization of White Soybean Cyst Nematode, Heterodera sojae

We reported a new white soybean cyst nematode, Heterodera sojae in 2016. This species is closely related to H. glycineswhich has the same host plant. Morphological observations of the cyst, female, male and second-stage juveniles indicatedthat this species is differed from those of H. glycines. Cysts of H. sojae appeared more round, shining and darker thanthat of H. glycines. The distribution of this new cyst nematode in soybean field in Korea is unknown yet. In 2016, 270soil samples were collected from soybean fields and examined the existence of H. glycines and H. sojae. Total of 111samples (41.1%) contained cysts. Among them 77% were H. glycines and 23% were H. sojae. Our results suggest thatextensive researches are required for white soybean cyst nematode.
노인의 디지털리터러시와 성공적 노화간의 관계에서 인지유연성의 매개효과

노인의 디지털리터러시는 사회적 관계 확장과 삶 만족에 영향을 미친다. 본 연구는 노인의 디지털리터러시와 성공적 노화와의 관계에서 인지적 유연성의 매개효과를 살펴보고자 한다. 연구 결과는 다음과 같다. 첫째, 디지털리터러시는 성공적 노화에 유의한 영향을 미쳤다. 둘째, 노인의 디지털리터러시는 인지적 유연성에 유의한 영향을 미쳤다. 셋째, 인지적 유연성은 성공적 노화에 유의한 영향을 미쳤다. 넷째, 디지털리터러시와 성공적 노화간의 관계에서 인지적 유연성은 두 변인의 관계를 매개하고 있었다. 노인의 디지털리터러시는 성공적 노화에 직접적인 관계가 있으며, 인지적 유연성이 높으면 성공적 노화를 보일 가능성이 높아진다고 해석된다. 연구 결과 노인의 디지털리터러시는 성공적 노화에 영향을 주며 노년기의 변화와 위기에 적응력을 높이는 인지적 유연성의 중요성을 확인할 수 있었다. The digital literacy of the elderly affects the expansion of social relationships and life satisfaction. The purpose of this study is to examine the mediating effect of cognitive flexibility on the relationship between digital literacy and successful aging in the elderly. The research results are as follows. First, digital literacy had a significant effect on successful aging. Second, the digital literacy of the elderly had a significant effect on cognitive flexibility. Third, cognitive flexibility had a significant effect on successful aging. Fourth, in the relationship between digital literacy and successful aging, cognitive flexibility mediated the relationship between two variables. The digital literacy of the elderly is directly related to successful aging, and it is interpreted that high cognitive flexibility increases the likelihood of successful aging. As a result of the study, it was confirmed that digital literacy in the elderly affects successful aging and the importance of cognitive flexibility to increase adaptability to changes and crises of old age.
Current Researches and Prospects of Soybean Cyst Nematode

Plant-parasitic nematodes causes yield losses of more than $100 million per year in the world. Among them, soybeancyst nematode (Heterodera glycines Ichinohe) causes the greatest yield loss to soybean compared to any other pest worldwide.SCN causes yield reductions by feeding on plant nutrients, retarding root growth and inhibiting Bradyrhizobium japonicum(Kirchner) Buchanan nodulation. The primary methods for controlling SCN include planting resistant cultivars and rotationwith nonhost crop. In order to control SCN, we should be breeding resistant soybean cultivars which has resistant genefor this pest. The research that can minimize the damage of the soybean yield is still insufficient, comprehensive andsystematic research is urgently required. Current researches in our lab are discussed.
