Low-Dose Gamma Irradiation as Means of Isolating Carotenoid-Hyperproducing Yeast Mutant

In order to isolate carotenoid-hyperproducing yeast, low-dose gamma irradiation was used as means of mutagenesis. Phaffia rhodozyma was treated by gamma irradiation of less than 10 kGy, which is considered to be a wholesome irradiation condition established by the Food and Drug Administration. Through repeated rounds of gamma irradiation and visual screening, mutant 3A4-8 was obtained. It produced a 3,824 ㎍ carotenoid/g yeast 59% higher content than 2,265 ㎍/g yeast of the unirradiated one. This result indicates that low-dose gamma irradiation could be used as means of mutagenesis to obtain carotenoid-hyperproducing strain of Phaffia rhodozymam, since only carotenoid-hyperproducing yeast survived gamma irradiation by scavenging oxygen radicals generated by radiolysis of water.
The first two-dimensional reference map of the fission yeast, Schizosaccharomyces pombe proteins

<P>Cytosolic proteins of Schizosaccharomyces pombe were separated by two-dimensional (2-D) gel electrophoresis, to construct the first 2-D reference map. In the pI range 4–7, more than 500 spots were detected by silver staining, and 70 different proteins corresponding to 111 spots were identified by matrix-assisted laser desorption/ionization-time of flight mass spectrometry and tandem mass spectrometry, where necessary. In the pI range 6–9, approximately 330 spots were detected, and 31 proteins corresponding to 38 spots were identified by mass spectrometry. More than 50% of the identified proteins were involved in amino acid, carbohydrate or nucleotide metabolism, and energy production. A second large group of identified proteins comprises heat shock and other stress related proteins and chaperones.</P>

Background: Ginseng has been used as a traditional medicine for treatment of many diseases and for general health maintenance. Previously, we showed that ginseng did not demonstrate estrogenic property in ovariectomized mouse model. However, it is still possible that disruption of steroidogenesis leading to indirect hormonal activity. Methods: The hormonal activities were examined in compliance with OECD guidelines for detecting endocrine disrupting chemicals: test guideline (TG) No. 456 (an in vitro assay method for detecting steroidogenesis property) and TG No. 440 (an in vivo short-term screening method for chemicals with uterotrophic property). Results: Korean Red Ginseng (KRG) and ginsenosides Rb1, Rg1, and Rg3 did not interfere with estrogen and testosterone hormone synthesis as examined in H295 cells according to TG 456. KRG treatment to ovariectomized mice did not show a significant change in uterine weight. In addition, serum estrogen and testosterone levels were not change by KRG intake. Conclusion: These results clearly demonstrate that there is no steroidogenic activity associated with KRG and no disruption of the hypothalamic-pituitary-gonadal axis by KRG. Additional tests will be performed in pursuit of cellular molecular targets of ginseng to manifest mode of action.

We investigate whether the National Health Insurances‘ policy of expanding coverage for the uncovered medical services leads to a balloon effect since the uncovered medical services costs increased even after the benefit coverage expansion policy were implemented. We divided the increase in the uncovered medical services costs into the three items: a) natural growth from population, income and price level, b) perverse incentive effect from private health insurance market, and c) the new uncovered medical services from technology advance. As a result of estimating the increase rate for each item, the annual average rate of natural growth is estimated at 3%, the annual average growth rate of perverse incentive is 4.5%, and the annual average increase rate of new uncovered medical services is 0.5%. Our results imply that the main factor driving the increase in uncovered medical services costs is perverse incentive effect from private health insurance market, as providers and subscribers frequently used unnecessary medical services. In conclusion, it is necessary to reinforce the management of uncovered medical services costs and reorganize the product structure of private health insurance market, in order to strengthen public health insurance coverage and reduce the burden of medical expenses on the people.

저장조건이 딸기 "미녀봉"의 저장 중 품질 및 미생물학적 변화에 미치는 영향을 연구하기 위하여 저장 중 중량감소율, 부패율, pH 및 적정산도, 총균수 및 곰팡이와 효모 수의 변화가 측정되었다. 딸기 시료는 LDPE로 포장하여 4℃ 와 20℃에서 각각 저장하였다. LDPE 포장은 대조구에 비해 중량감소율 및 부패율을 감소시켰다. 또한 포장방법은 저장중 미생물학적 변화에 영향을 끼쳤는데 저장온도와 상관없이 대조구와 비교해서 효모와 곰팡이뿐만 아니라 총균수의 감소를 야기시켰다. 본 연구결과는 딸기 "미녀봉"의 유통기한을 고려할 때 4℃에서 LDPE 포장으로 저장하는 것이 바람직하다는 것을 나타냈다. To examine the effect of storage condition on the quality and microbiological change fo strawberry “Minyubong”, the rate of weight loss, decay rate, pH and litratable acidity, and microbial (total bacterial count, mold and yeast) change were determines during storage. Strawberry fruits were packaged with low-density polyethyiene (LDPE). Strawberries were then stored at 4℃ and 20℃, respectively. LDPE package was effective on the decrease of decay rate of strawberry as well as the rate of weight loss, compared with the non-packaged. Microbial changes of strawberry stored at 4℃ and 20℃ were monitored during storage. Packaging affected the microbial change, resulting in retarding the growth of total bacteria as well as mold and yeast, compared with the non-packaged, regardless of storage temperature. These results indicate that storage of strawberry fruits wrapped with LDPE at 4℃ should be recommended in terms of quality as well as its shelf-life.

Sodium glucose cotransporter 2 (SGLT2) inhibitor has been recently reported of diabetic ketoacidosis due to accumulation of ketone bodies in patients with severe dehydration caused from such like diarrhea even though the patient had normal glucose level. This is a case of ketoacidosis in normal glucose level as production of ketone bodies is stimulated in liver with increased secretion of glucagon by stimulation of α cells in pancreas due to increase of lipolysis caused from reducing insulin and by SGLT2 inhibitor among patients who are under concurrent insulin and SGLT2 inhibitor. Thus, insulin dosage reduction requires caution in order to control blood glucose level on combined treatment of SGLT2 inhibitor in a patient who is administering insulin because the patient may be caused ketoacidosis in normal blood glucose level.

Aims: Hepatocellular carcinoma (HCC) is one of the adverse clinical outcomes of alcoholic liver disease, but the risk is relatively low compared to those of chronic viral hepatitis. Alcohol increases the risk of colorectal adenoma and adenocarcinoma. In this study, we evaluated the relationship between detection of colorectal neoplasm and subsequent risk of HCC in alcoholic liver disease patients. Methods: This retrospective cohort study enrolled all consecutive patients with alcoholic liver disease who underwent colonoscopic screening in our institute. The risk of HCC development was compared by Kaplan-Meier analysis with log-rank test. Cox proportional hazard analysis was used to identify independent risk predictors for HCC. Results: A total of 1,294 patients were included. The median duration of follow-up was 62.3 months (range 12.1-163.9). Colonoscopic screening revealed hyperplasia, low-grade dysplasia, high-grade dysplasia and adenocarcinoma in 10.7, 37.2, 0.9 and 2.2% of patients, respectively. The cumulative incidence of HCC was 0.8%, 2.1% and 3.1% at 3, 5, 10 years, respectively after initial colonoscopic screening. Univariate analysis showed that old age, low albumin, high AFP, prolonged prothrombin time, daily alcohol consumption amount, smoking and history of colorectal adenocarcinoma was associated with significant increased risk for HCC. Cox analysis showed that low albumin, high AFP, prolonged prothrombin time and history of colorectal adenocarcinoma were independent HCC risk predictors with hazard ratio of 0.39, 2.33, 5.59 and 3.73, respectively. Conclusions: Detection of colorectal adenocarcinoma increased the risk of subsequent development of HCC in patients with alcoholic liver disease.
