보성산 유기농 녹차의 품질에 따른 카테킨 함량과 항산화능 비교 분석

유기농 녹차의 등급별 총페놀 함량과 항산화능은 각 추출 용매에 따른 수율을 확인하였을 때 물 추출에 비하여 수용성 유기용매의 추출 수율이 유의적으로 높았다. 유기용매를 이용한 추출수율은 물을 이용할 때 보다 총페놀 함량은 1.5에서 3.2배, 항산화능은 1.8에서 3.8배 정도 높게 나타났다. HPLC 분석을 통하여 총페놀 함량과 항산화능의 상관관계를 비교 시 총페놀 함량이 증가함에 따라 항산화능이 1차 선형관계로 증가하는 경향을 보였다. HPLC분석을 통하여 유기농 녹차에 함유되어 있는 다양한 카테킨의 함량이 채엽시기에 따라 변화가 나타났다. 유기농 녹차의 카테킨 성분 중 가장 많이 존재하는 EGCG가 건조중량당 5.8-7.7%의 함량을 보였고, caffeine은 1.7-2.9%의 함량을 보였다. 유기농 녹차의 카테킨 함량은 일반적으로 채엽 시기가 늦어짐에 따라서 점차 감소하였다. 유기농 녹차의 등급에 따른 항산화능은 최상급인 우전에서 가장 높았으며 최하등급인 엽차에서 가장 낮게 나타났다. HPLC 분석에 의한 유기농 녹차의 등급에 따른 총 카테킨 함량(㎎/g)은 우전(155.4), 세작(147.7), 중작(143.2), 엽차(135.1), 대작(130.5)의 순서로 감소하였다. 이를 통해 녹차의 채엽 시기가 유기농 녹차의 카테킨 성분 및 항산화능에도 영향을 미치는 것을 확인하였다. 전반적으로 상급의 유기농 녹차 일수록 총페놀 및 카테킨 함량이 많으며 또한 더 높은 항산화능을 가지는 것으로 나타났다. The objective of this study was to evaluate the effect of various solvents on extraction of bioactive phenolics and to analyze the antioxidant capacity and contents of individual catechins in various grades of green teas organically grown in Boseong, Korea. The organic green teas, based on their harvest seasons, were categorized into five grades such as Woo-Jeon, Se-Jak, Jung-Jak, Dae-Jak, and coarse tea. Solvents used to extract phenolics from these teas included water at 23℃ and 70℃ as well as 80% (v/v) aqueous methanol and ethanol. In general, aqueous organic solvents of methanol and ethanol led to higher extraction yields of phenolics than water at 23℃ and 70℃. Total phenolics and antioxidant capacity of the teas extracted with the aqueous organic solvents were approximately 1.5 to 3.2 and 1.8 to 3.8 times higher than those with water at 23℃ and 70℃, respectively. Coarse tea, the lowest grade of green tea, showed approximately 30-60% lower total phenolics and antioxidant capacity compared with the higher grade ones. Reversed-phase HPLC analysis was performed quantitatively to identify individual catechins, gallic acid, and caffeine in teas extracted with 80% (v/v) aqueous methanol. Based on their dry weights, the organic green teas contained about 1.7 to 2.9% of caffeine. Content (㎎/g dry weight) of tea catechins decreased in the following order: Woo-Jeon (155.4) > Se-Jak (147.7) > Jung-Jak (143.2) > coarse tea (135.1) > Dae-Jak (130.5). (-)-Epigall℃atechin gallate was the most abundant among the catechins analyzed. The highest grade of green tea, Woo-Jeon, had the highest amount of (-)-epigall℃atechin gallate at 77.4 ㎎/g dry weight. Overall, the higher grade of organic green teas tended to have the higher level of antioxidant capacity and catechins.
Vaginal wall endometriosis following total hysterectomy: a case report

Background: Endometriosis is defined as the endometrial glands and stroma that occur outside the uterine cavity. Endometriosis usually occurs within the pelvis such as the ovaries, cul-de-sac, broad ligaments, and uterosacral ligaments, but rarely, endometriosis may occur in the bladder, ureter, thorax, and skin. Patients who undergo hysterectomy with endometriosis may have a recurrence in the pelvic cavity or vaginal stump. However, we report a case of endometriosis of the vaginal wall after hysterectomy in a patient without endometriosis. Case: A 43-year-old female presented with cyclic vaginal bleeding. The patient had undergone a total hysterectomy 15 months prior due to symptomatic multiple leiomyomas. At the time of surgery, there was no evidence of endometriosis other than uterine myomas. Pelvic examination showed no hemorrhage or inflammation in the vaginal cuff, but small spotting lesions were observed in the left vaginal wall area. Excision of vaginal wall lesion was performed. The pathological diagnosis of the excised tissue was endometriosis. The patient has no abnormal findings on postoperative follow-up examination. Conclusion: In rare cases, endometriosis may recur in the vaginal stump after hysterectomy. However, vaginal wall endometriosis following hysterectomy is very rare. Regarding the menstrual cycle, periodic symptomatic lesions should be carefully observed with the possibility of endometriosis.

<P><B>ABSTRACT</B></P><P>Primary diffuse large B-cell lymphoma of the central nervous system (CNS-DLBCL) is an aggressive disease with a poor prognosis. The status of the tumor immune microenvironment in CNS-DLBCL remains unclear. We investigated the prognostic implications of tumor-associated macrophages (TAMs), regulatory T-cells (Tregs), and indoleamine 2,3-dioxygenase (IDO)<SUP>+</SUP> cells in primary CNS-DLBCL (n = 114) by immunohistochemical analysis. The numbers of tumor-infiltrating immune cells, including CD68<SUP>+</SUP> TAMs, CD163<SUP>+</SUP> or CD204<SUP>+</SUP> M2 macrophages, FOXP3<SUP>+</SUP> Tregs, and IDO<SUP>+</SUP> cells were all significantly lower in CNS-DLBCL versus systemic DLBCL (n = 165; all P < 0.001), but with little difference in the ratio of CD163<SUP>+</SUP>/CD68<SUP>+</SUP> or CD204<SUP>+</SUP>/CD68<SUP>+</SUP> cells. An increase in CD68<SUP>+</SUP> cell numbers was significantly associated with prolonged progression-free survival (PFS) and overall survival in patients with CNS-DLBCL (P = 0.004 and 0.021, respectively). In contrast, an increase in CD204<SUP>+</SUP> cell numbers or a higher ratio of CD204<SUP>+</SUP>/CD68<SUP>+</SUP> cells was related to a shorter PFS (P = 0.020 and 0.063, respectively). An increase in IDO<SUP>+</SUP> cell numbers was associated with a significantly longer PFS (P = 0.019). In combination, the status of low IDO<SUP>+</SUP> cell numbers combined with low CD68<SUP>+</SUP> cell numbers, high CD204<SUP>+</SUP> cell numbers, or a high CD204<SUP>+</SUP>/CD68<SUP>+</SUP> cell ratio all predicted poor PFS in multivariate analyses. This study showed that an increase in CD204<SUP>+</SUP> cell numbers, suggestive of M2 macrophages, was associated with poor clinical outcome in CNS-DLBCL, whereas increased CD68<SUP>+</SUP> or IDO<SUP>+</SUP> cell numbers were related to a favorable prognosis. The analysis of tumor-infiltrating immune cells could help in predicting the prognosis of CNS-DLBCL patients and determining therapeutic strategies targeting tumor microenvironment.</P>

<P>Given the growing number of cancer patients and the resulting increase in the administration of chemotherapeutic agents, convenient and effective methods for measuring the symptoms and quality of life associated with the hand-foot syndrome (HFS) are needed. Therefore, the aim of this study was to develop and validate the Korean version of the hand-foot skin reaction and quality of life questionnaire (HF-QoL-K), comprising a 20-item symptom domain and an 18-item daily activity domain. After we developed the HF-QoL-K, 209 Korean patients with gynecologic cancer who were undergoing chemotherapeutic agents relating the HFS were asked to fill in the questionnaire. The content validity, internal consistency reliability, and test-retest reliability were evaluated. The internal validity index, Cronbach’s alpha coefficient, and intra-class correlation coefficient of the HF-QoL-K were 0.90, 0.958, and 0.825 (95% confidence interval [CI], 0.774–0.865), respectively. The scatter plot (Pearson correlation coefficient, 0.826) and the Bland-Altman plot for test-retest reliability were also acceptable. The HF-QoL-K instrument is a valid and reliable questionnaire for the measurement of the symptoms and quality of life in Korean cancer patients suffering HFS.</P>

Background: Dipeptidyl peptidase-4 (DPP-4) inhibitor has been reported to have kidney-protective benefits. To elucidate how antidia-betic agents prevent diabetic kidney disease progression, it is important to investigate their effect on the kidney environment in type 2 diabetes mellitus (DM) patients. Herein, we investigated the expression pattern of urinary exosome-derived microRNA (miRNA) in patients taking a combination of DPP-4 inhibitor and metformin (DPP-4 inhibitor group) and compared them with patients taking a combination of sulfonylurea and metformin (sulfonylurea group). Methods: This was a prospective study involving 57 patients with type 2 DM (DPP-4 inhibitor group, n = 34; sulfonylurea group, n = 23) and healthy volunteers (n = 7). We measured urinary exosomal miRNA using the NanoString nCounter miRNA array (NanoString Technologies) across the three groups (n = 4 per each group) and validated findings using real-time polymerase chain reaction. Results: Twenty-one differentially expressed candidate miRNAs were identified, and six (let-7c-5p, miR-23a-3p, miR-26a-3p, miR-30d, miR-205, and miR-200a) were selected for validation. Validation showed no significant difference in miRNA expression between the DPP-4 inhibitor and sulfonylurea groups. Only miR-23a-3p was significantly overexpressed in the diabetes group compared with the control group (DPP-4 inhibitor vs. control, p = 0.01; sulfonylurea vs. control, p = 0.007). This trend was consistent even after adjusting for age, sex, and body mass index. Conclusion: There was no significant difference in urine exosome miRNA expression between diabetic participants taking DPP-4 in-hibitor and those taking sulfonylurea. The miR-23a levels were higher in diabetic participants than in nondiabetic controls.

Previously, we demonstrated that the erythropoietin receptor (EpoR) is present on fibroblasts, where it regulates focal contact. Here, we assessed whether this action of EpoR is involved in the reduced cell adhesion observed in colonocytes exposed to Clostridium difficile toxin A. EpoR was present and functionally active in cells of the human colonic epithelial cell line HT29 and epithelial cells of human colon tissues. Toxin A significantly decreased activating phosphorylations of EpoR and its downstream signaling molecules JAK-2 (Janus kinase 2) and STAT5 (signal transducer and activator of transcription 5). In vitro kinase assays confirmed that toxin A inhibited JAK 2 kinase activity. Pharmacological inhibition of JAK2 (with AG490) abrogated activating phosphorylations of EpoR and also decreased focal contacts in association with inactivation of paxillin, an essential focal adhesion molecule. In addition, AG490 treatment significantly decreased expression of occludin (a tight junction molecule) and tight junction levels. Taken together, these data suggest that inhibition of JAK2 by toxin A in colonocytes causes inactivation of EpoR, thereby enhancing the inhibition of focal contact formation and loss of tight junctions known to be associated with the enzymatic activity of toxin A.

The β-1,4-endoglucanase gene from Actinomyces sp. 40 was cloned into Escherichia coli DH5α with pUC19. Chromosomal DNA from Actinomyces sp. 40 was cleaved with the restriction enzyme Sau3Al and ligated into pUC19 for the transformation of Escherichia coli DH5α. Positive clones of β-1,4-endoglucanase gene were detected as the clear zones on a medium supplemented with carboxymethylcellulose (CMC). This transformant possessed a single plasmid, designated pDS1, which contained the vector DNA and a 3.5 kilobase (kb) Sau3Al insertion fragment encoding endoglucanase. The size of the cloned fragment was reduced to 2.0 kb. The endoglucanase activity produced by the E. coli DH5a (pDS6) was higher than that of Actinomyces sp. 40 strain. The optimum pH and temperature of the cloned enzyme were pH 4.0∼5.0 and 55℃, respectively. The cloned enzyme was stable at 55℃ or below and in buffer ranging from pH 4.0 to 7.0. The enzyme degraded CMC but did not degrade xylan, cellobiose, and methyl-umbelliferylcellobiopyranoside (MUC).
