OSI 환경하에서의 효율적인 네트워크 관리에 관한 연구

본 논문은 복잡해지는 네트워크를 관리하기 위해서 OSI 환경하에서 보다 효율적인 네트워크 관리 방안에 관해 연구하였다. 먼저 네트워크 관리의 기본적인 이론을 기술하였고, 기존 네트워크 관리 시스템을 검토하였다. 또한 OSI 환경하에 따른 네트워크 동작 관리의 자동화 모델과 성능 관리의 효율적 방법을 제시하고 표준화에 따른 네트워크 관리 구조의 개념적 모델에서 효과적인 관리를 위한 전략적인 방안을 제안하였다.

Background: The Ministry of Environment, Korea recognizes Cotoneaster wilsonii Nakai as a second-class endangered plant. It is a native species that grows in Ulleung-do, Korea. To our knowledge, the bioactivity of this plant has not yet been reported. Therefore, in this study, we have reported the bioactivity of C. wilsonii Nakai. Methods and Results: The anti-oxidant activities of C. wilsonii methanolic extract were investigated in vitro. The anti-oxidant activity was evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical assay, and the total phenolic and flavonoid content were measured. The leaf methanolic extract had the highest DPPH radical scavenging activity (IC50; 15.74 ㎍/㎖), and it also had the highest total phenolic and flavonoid content (220.95 ㎎·GAE/g, and 36.46 ㎎·QE/g extract respectively). Through high performance liquid chromatography (HPLC) analysis, chlorogenic acid was found to be the predominant among all phenolic compounds, showing a concentration of 84.24 ㎎/g extract. More than 60% decrease in nitric oxide (NO) production was found in the leaf methanolic extract. Conclusions: To the best of our knowldege, this is the first report of the bioactivities of C. wilsonii. The results demonstrate that the leaf methanolic extract of C. wilsonii shows potent anti-oxidant, and anti-inflammatory activities.

Background: Previously, studies have observed that the leaf extract of Cotoneaster wilsonii Nakai has potent antioxidant and anti-inflammatory activites. Therefore, further research was conducted to separate the active antioxidant and anti-inflammatory compounds in the leaf of the C. wilsonii. Methods and Results: The anti-oxidant effects were evaluated by analyzing the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, estimating the totla phenolic content (TPC) and total flavonoid content (TFC), and using reducing power assay method. The ethyl acetate (EtOAc) fraction demonstrated the highest DPPH radical scavenging activity (IC<SUB>50</SUB>; 9.69 ㎍/㎖), and the highest TPC and TFC (345.98 ㎎·GAE/g, and 74.23 ㎎·QE/g). Moreover, it indicated the highest effects on nitric oxide production. Therefore, the active compound was separated using the EtOAc fraction, determined on the basis of spectral data, and identified as (+)-catechin. Conclusions: The first compound separated was made from the leaf of C. wilsonii was (+)-catechin, which produced potent anti-oxidant and anti-inflammatory effects.

신장이식 공여자에서 <sup>99m</sup>Tc-DTPA를 이용한 Glomerular Filtration Rate 측정과 추정사구체여과율의 비교분석

신사구체여과율(glomerular filtration rate, GFR)은 신장 질환의 진단, 치료 및 추적 관찰에 중요한 지표이며, 건강인에서도 약제사용, 신장이식 공여자의 신장 평가 등에 사용된다. GFR 검사의 표준방법은 외인성 표지자인 이눌린(inulin)을 연속 주입하여 측정하는 방법이나, 시간이 많이 소요되고 검사 방법이 복잡하기 때문에, 혈청 크레아티닌 농도로 계산한 추정사구체여과율(estimated glomerular filtration rate, eGFR)을 대신 사용한다. 그러나 크레아티닌은 연령, 성별, 근육량 등에 영향을 받는 것으로 알려져 있으며, 현재 많이 사용되는 추정사구체여과율 공식에는 성인의 경우 Cockroft-Gault 공식, Modification of Diet in Renal Disease(MDRD) 공식, Chronic kidney Disease Epidemiology Collaboration (CKD-EPI) 공식 등이 있고, 소아의 경우 Schwartz 공식을 사용하고 있다. <sup>99m</sup>TC diethylenetriaminepentaacetic acid(<sup>99m</sup>Tc-DTPA)를 사용한 신사구체여과율(glomerular filtration rate, GFR) 측정은 이눌린을 대체 할 수 있어 현재 사용되고 있다. 이에 <sup>99m</sup>Tc-DTPA를 이용하여 측정한 신사구체 여과율과 CKD-EPI 공식을 이용한 추정사구체여과율을 비교하여 보았다. 서울 아산병원을 내원한 신장이식 공여자 200명(남성 96명, 여성 104명, 47.3세±12.7)을 대상으로 <sup>99m</sup>Tc-DTPA(0.5 mCi, 18.5 MBq)를 정맥투여 하고 획득된 plasma(Two-plasma-sample-method, TPSM)를 계측하여 신사구체여과율(glomerular filtration rate, GFR)을 측정하였다. 혈청 크레아티닌 농도를 근거로 chronic kidney disease epidemiology collaboration (CKD-EPI) 공식을 이용하여, 추정사구체여과율(estimated glomerular filtration rate, eGFR)을 도출하였다. 신장이식 공여자 200명(남성 96명, 여성 104명, 47.3세±12.7)을 대상으로 <sup>99m</sup>Tc-DTPA를 사용하여 측정된 신사구체여과율 평균값은 97.27±19.46 (GFR, ml/min/1.73m<sup>2</sup>)이고 CKD-EPI 공식을 이용한 추정사구체여과율 (estimated glomerular filtration rate, eGFR) 평균값은 96.84±17.74(CKD-EPI, ml/min/1.73m<sup>2</sup>), 혈청 크레아타닌의 농도는 0.84±0.39 (mg/dL)이다. 혈청 크레아티닌 근거 추정사구체여과율에 대한 <sup>99m</sup>Tc-DTPA 신사구체여과율의 회귀식은 Y= 0.5073X + 48.186, 상관계수는 0.698이었다.(P<0.01)혈청 크레아티닌 근거 추정사구체여과율에 대한 <sup>99m</sup>Tc-DTPA 신사구체여과율의 Difference(%)는 1.52±18.28 이었다. <sup>99m</sup>TC-DTPA를 이용해서 측정된 신사구체여과율과 혈청 크레아티닌 농도에 기반하여 도출된 추정사구체여과율에 대한 상관계수는 0.698로 중등도 정도의 상관성을 확인할 수 있었다. 이는 추정사구체여과율은 연령, 성별, 근육 양 등 신장 외적인 요소에 영향을 받고 신장질환자를 대상으로 만든 공식을 사용하는 원인에 기인하는 것으로 추정된다. 신장 질환의 진단, 치료 및 추적 관찰, 신장이식 공여자의 신장 평가 등에 사용되는 신사구체여과율 측정에 <sup>99m</sup>Tc-DTPA를 사용함으로써 신뢰성 있는 결과를 임상에 제공할 수 있다. Purpose Glomerular filtration rate(GFR) is an important indicator for the diagnosis, treatment, and follow-up of kidney disease and is also used by healthy individuals for drug use and evaluating kidney function in donors. The gold standard method of the GFR test is to measure by continuously injecting the inulin which is extrinsic marker, but it takes a long time and the test method is complicated. so, the method of measuring the serum concentration of creatinine is used. Estimated glomerular filtration rate (eGFR) is used instead. However, creatinine is known to be affected by age, gender, muscle mass, etc. eGFR formulas that are currently used include the Cockroft-Gault formula, the modification of diet in renal disease (MDRD) formula, and the chronic kidney disease epidemilogy collaboration (CKD-EPI) formula for adults. For children, the Schwartz formula is used. Measurement of GFR using <sup>51</sup>Cr-EDTA (diethylenetriamine tetraacetic acid), <sup>99m</sup>Tc-DTPA (diethylenetriamine pentaacetic acid) can replace inulin and is currently in use. Therefore, We compared the GFR measured using <sup>99m</sup>Tc-DTPA with the eGFR using CKD-EPI formula. Materials and Methods For 200 kidney transplant donors who visited Asan medical center.(96 males, 104 females, 47.3 years ± 12.7 years old) GFR was measured using plasma(Two-plasma-sample-method, TPSM) obtained by intravenous administration of <sup>99m</sup>Tc-DTPA(0.5mCi, 18.5 MBq). eGFR was derived using CKD-EPI formula based on serum creatinine concentration. Results GFR average measured using <sup>99m</sup>Tc-DTPA for 200 kidney transplant donors is 97.27±19.46(ml/min/1.73m<sup>2</sup>), and the eGFR average value using the CKD-EPI formula is 96.84±17.74(ml/min/1.73m<sup>2</sup>), The concentration of serum creatinine is 0.84±0.39(mg/dL). Regression formula of <sup>99m</sup>Tc-DTPA GFR for serum creatinine-based eGFR was Y = 0.5073X + 48.186, and the correlation coefficient was 0.698 (P<0.01). Difference (%) was 1.52±18.28. Conclusion The correlation coefficient between the <sup>99m</sup>Tc-DTPA and the eGFR derived on serum creatinine concentration was confirmed to be moderate. This is estimated that eGFR is affected by external factors such as age, gender, and muscle mass and use of formulas made for kidney disease patients. By using <sup>99m</sup>Tc-DTPA, we can provide reliable GFR results, which is used for diagnosis, treatment and observation of kidney disease, and kidney evaluation of kidney transplant patients.
