        Statistical Optimization of Medium Components for Milk-Clotting Enzyme Production by Bacillus amyloliquefaciens D4 Using Wheat Bran-an Agro-Industry Waste

        In this paper, two statistical methods were applied to optimize medium components to improve the production of the milk-clotting enzyme by Bacillus amyloliquefaciens D4. First, wheat bran juice, skim milk powder, and Na2HPO4 were shown to have significant effects on D4 enzyme production using the Plackett?Burman experimental design. Subsequently, an optimal medium was obtained using the Box?Behnken method, which consisted of 3.31 g/l of skim milk powder, 5.0 g/l of sucrose, 0.1 g/l of FeSO4?7H2O, 0.1 g/l of MgSO4?7H2O, 0.1 g/l of MnSO4?2H2O, 0.1 g/l of ZnSO4?7H2O, 1.52 g/l of Na2HPO4, and 172.45 g/l of wheat bran juice. With this optimal medium, the milk-clotting enzyme production was remarkably enhanced. The milk-clotting enzyme activity reached 3,326.7 SU/ml after incubation of 48 h, which was 1.76-fold higher than that of the basic medium, showing that the Plackett?Burman design and Box?Behnken response surface method are effective to optimize medium components, and B. amyloliquefaciens D4 possessed a high rennet-producing capacity in the optimal medium.

        A large-area free-standing graphene oxide multilayer membrane with high stability for nanofiltration applications

        <P><B>Abstract</B></P> <P>A flexible and free-standing graphene oxide and nylon 6 (GO@nylon 6) multilayer nanofiltration membrane was prepared by a layer-by-layer assembly process. The combination of electrospinning and electrospraying technique was employed, which can facilely prepare large-area membrane with size of 20 × 30 cm. The mechanical stability of multilayer membrane has enhanced significantly due to the tightly locked structure achieved by nylon 6 nanofibers network. The novel GO@nylon 6–13 multilayer nanofiltration membrane demonstrated a pure water flux up to 11.15 L m<SUP>−2</SUP> h<SUP>−1</SUP> bar<SUP>−1</SUP>, while keeping high organic dye rejection rate (>95% for methylene blue, and >99% for methyl orange). The rejections rate of the Na<SUB>2</SUB>SO<SUB>4</SUB>, NaCl, CuSO<SUB>4</SUB>, and Pb(NO<SUB>3</SUB>)<SUB>2</SUB> were 56.5%, 27.6%, 36.7%, and 18.9%, respectively.       • A Multi-center Survey of HPV Knowledge and Attitudes Toward HPV Vaccination among Women, Government Officials, and Medical Personnel in China

        Objectives: To assess knowledge of HPV and attitudes towards HPV vaccination among the general female population, government officials, and healthcare providers in China to assist the development of an effective national HPV vaccination program. Methods: A cross-sectional epidemiologic survey was conducted across 21 urban and rural sites in China using a short questionnaire. 763 government officials, 760 healthcare providers, and 11,681 women aged 15-59 years were included in the final analysis. Data were analyzed using standard descriptive statistics and logistic regression. Results: Knowledge of HPV among the general female population was low; only 24% had heard of HPV. Less than 20% of healthcare providers recognized sexually na$\ddot{i}$ve women as the most appropriate population for HPV vaccination. There was high acceptance of the HPV vaccine for all categories of respondents. Only 6% of women were willing to pay more than US $300 for the vaccine. Conclusions: Aggressive education is necessary to increase knowledge of HPV and its vaccine. Further proof of vaccine safety and efficacy and government subsidies combined with increased awareness could facilitate development and implementation of HPV vaccination in China.

      • Susceptibility Loci Associations with Prostate Cancer Risk in Northern Chinese Men

        Background: KLK3 gene products, like human prostate-specific antigen (PSA), are important biomarkers in the clinical diagnosis of prostate cancer (PCa). G protein-coupled receptor RFX6, C2orf43 and FOXP4 signaling plays important roles in the development of PCa. However, associations of these genes with PCa in northern Chinese men remain to be detailed. This study aimed to investigate their impact on occurrence and level of malignancy. Methods: All subjects were from Beijing and Tianjin, including 266 cases with prostate cancer and 288 normal individuals as controls. We evaluated associations between clinical covariates (age at diagnosis, prostate specific antigen, Gleason score, tumor stage and aggressive) and 6 candidate PCa risk loci, genotyped by PCR- high resolution melting curve and sequencing methods. Results: Case-control analysis of allelic frequency of PCa associated with PCa showed that one of the 6 candidate risk loci, rs339331 in the RFX6 gene, was associated with reduced risk of prostate cancer (odds ratio (OR) = 0.73, 95% confidence interval (CI) =0.57-0.94, P = 0.013) in northern Chinese men. In addition, subjects with CX (CC+TC) genotypes had a decreased risk for prostrate cancer compared to those carrying the TT homozygote (OR =0.64, 95% CI = 0.45- 0.90, P = 0.008). The TT genotype of 13q22 (rs9600079, T) was associated with tumor stage (P=0.044, OR=2.34, 95% CI=0.94-5.87). Other SNPs were not significantly associated with clinical covariates in prostate cancer (P > 0.05). Conclusions. rs339331 in the RFX6 gene may be associated with prostate cancer as a susceptibility locus in northern Chinese men.

        Background Bladder cancer is the 10th common cancer worldwide. Osteopontin has been found to enhance cell proliferation, metastasis and invasion in various human tumors. Objective To investigate the roles of osteopontin in bladder cancer. Methods The RNA interference and overexpression of osteopontin were performed in bladder cancer cell lines (T24 and SCaBER). Cell proliferation and apoptosis were measured using CCK-8 assay and flow cytometry, respectively. Cell invasion was determined using transwell assay. Results Osteopontin was highly expressed in bladder cancer tissues in comparison with the adjacent normal tissues. Its high expression significantly correlated with high histologic grade, high TNM stage (III and IV) and poor prognosis. For T24 cells with osteopontin interference and SCaBER cells with osteopontin overexpression, cell proliferation was significantly inhibited (3.58-fold vs. 5.62-fold) and enhanced (7.81-fold vs. 5.29-fold), respectively. The apoptosis portion of T24 cells significantly increased from 4.48 to 10.75%, and that of SCaBER cells significantly declined from 7.33 to 4.01%. The invaded T24 and SCaBER cells significantly decreased to 52.0% and increased to 2.0-fold, respectively. Osteopontin overexpression enhanced the expression (1.54-fold and 2.39-fold; 2.33-fold and 2.05-fold) and activation (1.80-fold and 1.96-fold; 2.00-fold and 2.59-fold) of JAK1 and STAT1 in two cell lines of bladder cancer. Conclusion Osteopontin might enhance proliferation, inhibit apoptosis and accelerate invasion and thus promote the development and metastasis of bladder cancer, and osteopontin’s functions might be mediated by activating JAK1/STAT1 signaling pathway.

        Political Connection, M&A and Enterprise Performance: Evidence from China Listed Companies

        Based on the samples of the Chinese listed companies, this paper studies the influence of political connection on enterprises M&A. We show that the political networks of the listed corporations in China considerably affect their M&A activities. More and greater M&A appear on the companies with political connection. The government mostly influences the political connections on the enterprises M&A. Moreover, we try to find out how much political connection affect the results of the enterprises M&A. Our study can contribute to a new aspect of the political connection in literature, as well as its fiscal consequences in China.

        The resource scheduling imbalance is a multi-objective optimization problem in cloud computing environment, this paper introduced the particle swarm optimization algorithm in cloud computing, a simulated annealing ideas is proposed in view of the prematurity of the algorithm, on the premise of performance determination, the position of the particle is determined by the probability choice, which helps the particle to escape. In order to enhance the global searching ability of the particle, the algorithm is combined with the chaotic mechanism to improve the accuracy of the algorithm. The inertia weight is adjusted dynamically according to the current state of the particle, accordingly, at the same time to obtain the optimal solution to ensure the convergence. Analysis of the experimental results show that the improved algorithm has a significant improvement in the ability of optimization and convergence speed, compared with other algorithms, the benchmark functions comparison is better, the different resource task proportion spent the shortest time and load balancing is the highest.

        Transcriptome analysis of the pheromone glands in Noorda blitealis reveals a novel AOX group of the superfamily Pyraloidea

        Noorda blitealis (Lepidoptera: Pyraloidea: Crambidae) is a major defoliating pest of Moringa trees. Focusing on its mating and reproduction, here we sequenced and analyzed the transcriptome of its pheromone glands (PGs) with a combination of Illumina sequencing, bioinformatics and phylogenetics approaches, coupled with a genomebased analysis. Transcriptome sequencing led to the yields of approximately 162 million clean reads, which were assembled into 60,578 unigenes and 121,692 transcripts, respectively. From the transcriptome, totally 117 genes encoding eight pheromone biosynthesis enzymes and one pheromone degradation enzyme were identified, 90 of which had complete open reading frames. A comparative analysis between PGs and bodies (removing PGs) revealed a large number of differentially expressed genes, including 79 pheromone biosynthesis and degradation related genes. Of the identified genes, NbliDES12 belonging to the △11 desaturase group was likely to a strong candidate for the desaturation of sex pheromones in N. blitealis, as implied by phylogenetic analyses and expression profiles. Finally and most notably, through genome and transcriptome analyses we discovered, for the first time, a novel aldehyde oxidase 6 (AOX6) group of the superfamily Pyraloidea that have been slightly expanded by gene duplications. Moreover, each orthologous AOX group shared highly conserved gene structure. Together, this current study has characterized the genes associated with sex pheromone biosynthesis and degradation from the PG transcriptome of N. blitealis, and more importantly, identifies a novel AOX group of the Pyraloidea.

        Nutritional characteristics and health effects of regional cuisines in China

        Background and objectives: Cuisine is the characteristic manifestation of the regional culture of Chinese food, which was formed and developed along with the distribution of physical geography and environment, the development of productivity and economy, cultural environment, ethnic food diversity, religion, and psychological and physiological factors. With the rapid development of the economy, food industry, and catering industry, various local cuisines learn from each other and develop greatly. The health consciousness of Chinese residents has gradually increased, and the requirements for nutrition and health of cuisine have also become higher and higher. The nutritional characteristics of regional cuisines and their effects on human health are increasingly being studied by scientists. The development of cuisine is facing new opportunities and challenges. It is necessary to understand the nutrition and health characteristics of cuisine and to take measures to reform and innovate the development of cuisines. The primary purposes of this paper are to introduce the formation and development of cuisines, to analyze the nutritional characteristics and health effects of four most influential regional cuisines—Sichuan Cuisine, Jiangsu cuisine, Shandong cuisine, and Cantonese, and to propose suggestions on future reform and innovation of cuisines. Methods: An online search with the following keywords of Chinese language and English language (cuisine OR dietary OR food) and (Chinese OR China) and (health OR diseases OR nutrition) was performed between January 1980 and September 2019. Multiple databases were used including CNKI, WanFang, PubMed, Health and Medical Collection, LWW, and OVID. The focus was on Chinese studies about cuisine, nutritional characteristics, and health but drawing on literature all over the world. Then, nutritional characteristics and health effects of regional cuisines in China in these related literatures were analyzed and summarized in this paper. Results and conclusions: Different cuisines have their specific nutritional advantages and disadvantages. The health statuses of the residents are closely associated with the dietary structure and food composition in regional cuisines. Regional cuisines need to be reformed and innovated ceaselessly to promote national health. It is necessary to adopt a scientific cooking method, increase food consistency and variety, and develop healthy dishes. The awareness of the nutrition and health among chefs and residents should be enhanced. The safety and hygiene of the restaurants should be strengthened in order to adapt to the transformation of the dining mode and to further standardize the evaluation criterion of dish.
