        밀어(Rhinogobius brunneus)의 산란행동 및 초기생활사

        본 연구는 밀어 (Rhinogobius brunneus)의 산란행동 및 초기생활사를 밝히기 위하여 4가지 수온조건 하에서 실험을 수행되었다. 본 실험을 위해 밀어는 금강 지류중의 하나인 갑천에서 성숙된 수컷과 암컷을 채집하였으며, 일정기간의 순치과정을 거친 후에 실험을 실시하였다. 본 실험 결과에 따르면, 성숙된 성어들의 구애행동 및 산란행동의 유형은 다음과 같이 요약된다. 밀어의 성숙된 수컷은 산란에 앞서 세력권을 형성한 후 자신의 세력권 안에 산란을 위한 둥지를 형성한 후, 성숙된 암컷을 유인 하여 둥지에 하부에 타원형 난을 단층으로 붙이는 부착 난 종으로 파악되었다. 수정된 알들은 수컷에 의해 보호 받는 종(Parental care species)임 이 관찰되었으며, 수정난의 평균 장경은 1.5±0.1 ㎜,단경은 0.7±0.1 ㎜였다. 실험실내 4개의 수온처리군에 따르면, 고온처리군 보다는 저온 처리군에서 부화가지 더 긴 시간이 소요되었으며, 높은 수온일수록 부화까지 소요되는 시간이 단축되는 것으로 나타났다. 또한, 18℃의 낮은 실험수온에서 부화된 자어는 약 3.6 ㎜의 전장을, 25℃ 이상의 높은 실험수온에서 부화된 자어는 3.1~3.2 ㎜의 전장을 보였다. 또한, 처리군에서 수온이 높을수록 빠른 성장을 보여 부화까지 소요되는 시간이 단축되는 결과를 보였으나, 부화 시 자어의 크기는 높은 수온 상태에서 보다는 낮은 수온상태에 서 부화된 개체가 큰 것으로 나타났다. 본 밀어에 대한 발생생태학적 기초연구는 수질오염 및 서식지 파괴에 의해 어종의 다양성이 빠르게 감소하는 상황에서 어류 개체군 보호 및 생태계보호에 기초자료를 제공할 것으로 사료된다. The objective of the study was to elucidate early life history of Rhinogobius brunneus, based on laboratory observations and field works, along with its courtship and spawning behaviors. The male and female fish matured were sampled for the lab experiments from Gap- Stream, which is one of the tributaries of Geum-River, Korea. The spawning behaviors and courtship of matured adults had a typical pattern; matured males of Rhinogobius brunneus built up spawning nests and had their own territory before the spawning, and the behavior then triggered the egg spawning of female with active courtship. The females attached elliptic egg mass as a single layer, on the stones below the spawning nest. After the spawning, the egg mass was guarded by males only until the hatching. The size of egg fertilized, measured by microscope was about 1.5±0.1 ㎜ in length and 0.7±0.1 ㎜ in the egg' s width. According to the lab experiments conducted under four temperature regime, the lapsed time for the hatching was shortened as temperatures increased in the treatments. On the contrary to the hatching time, the average length of newly hatched larvae showed a reverse relation with water temperature. The larvae, considered a final stage of the early life history was about 3.6 ㎜ in total length for water temperature of 18℃, whereas they were between 3.1 ㎜ and 3.2 ㎜ for over 25℃. This indicates that higher water temperature may decrease the growth rate in the early life history. These results may provide valuable information for the population conservation and ecosystem protection under accelerated water pollution and habitat degradations in Korea.

      • 결장직장암 환자의 임상적 고찰 : 조선대학교 부속병원의 10년 간의 결과

        Background and Objectives: The colorectal cancer rates 4th in the total cancer prevalence in Korea. The clinical patient demographics were considered with the incidence rate of the colon and rectal cancer. The clinicopathological factors were analyzed for the 5 year survival rate. Materials Methods: Of the 282 patients who were operated from Jan. 1, 1992 to Dec. 31, 2001 and diagnosed by pathological biopsy as colo-rectal cancer, 234 patients were followed up. The age, sex, location of the cancer, TNM stage, operation method, histopathological classification, metastatic region, complications after operation and the survival rate were investigated. Results: Of the 234 patients, male and female consisted of 120 cases (51.3%) and 114 cases (46.7%), with an average age of 61.6 years. The average follow up period was 52.3 months and the peri operative mortality was 5.6%. The site of the cancer was most frequent at the rectum with 127 cases (54.3%). The patients were classified according to the TNM stages with 38 cases of stage 1 (16.2%), 98 cases of II (41.9%), 72 cases of III (30.8%) and 26 cases of IV (11.1%). The 5 year survival rate in the age group older than 50 was 63.3 % and 73.5% in the group younger than 50. In regard to the operation type, the 5 year survival rate was 65.4% for elective operation and 65.8% for emergency operation. The 5 year survival rate for well differentiated, moderately differentiated, poorly differentiated cancer, and mucinous adenocarcinoma was 73.7%, 60.2%, 58.3%, and 92.6%, respectively. The 5 year survival rate of each of the TNM stage is 81.1%, 74.8%, 57.0%, and 11.5% for Classes I, II, III, and IV, respectively. Conclusion: The clinical factors were analyzed for the 5 year survival rate with no statistical significance for the age, sex, and location of the cancer. The TNM stage and histopathological differentiation were statistically significant. The colon cancer showed better prognosis compared to the rectal cancer.

        Insertion of reporter constructs into the mammalian genome leads to variable gene expression due to position effects at the site of integration. This random integration has limited the gene therapy of human genetic disorders by its undesirable effects. We report here the newly constructed plasmid vector(pIRES-neo-YJ) based on the concepts of homologous recombination and position-independent promoter enhancing of beta-globin matrix attachment region(Glb-MAR). chromosome 7 centromere-specific alpha satellite(alphoid) DNA sequence was cloned into pIRES-neo-YJ for homologous recombination of the cloned gene with the centromeric region of chromosome 7, which is genetically silent. Beta Glb-MAR sequence that allows high levels of transcription independent of the chromosomal site of integration was also inserted into pIRES-neo-YJ to ensure the stable and higher expression of the cloned genes. We expect that pIRES-neo-YJ would provide a valuable tool to eliminate random integration of cloned genes into the undesirable chromosomal region and their short-lived expression which often encounters during the construction of transgenic animals and human gene therapy.

        Insulation Coating of Fe–Si–Cr Soft Magnetic Powder by Selective Oxidation

        This study examines the insulation coating technology of Fe–Si–Cr powder via selective oxidation annealing, which oxidizeselements selectively by controlling the oxidation potential. The study calculated the oxidation driving force of Fe, Si, andCr, and conducted a thermodynamic analysis of oxidation and reduction conditions according to temperature and oxidationpotential. Based on the results, a selective oxidation annealing was performed in an atmosphere in which Fe is reduced andonly Si and Cr are selectively oxidized. The oxidation potential was controlled through the partial pressure ratio of hydrogenand water vapor. The XPS analysis results confirmed that a Si and Cr complex oxide layer formed on the powder surfaceafter the selective oxidation annealing. Afterward, withstanding voltages were analyzed to evaluate the insulation property. Then, the withstanding voltage of the powder applying the selective oxidation annealing increased significantly comparedto that of the initial powder. Further analysis showed that the powder annealed in an air atmosphere had a significantly lowersaturation magnetic flux density than the initial powder, while the powder applying the selective oxidation annealing hadonly a slightly reduced saturation magnetic flux density.

        Studies on Trichomonas vaginalis in 180 obstetics and gynecologic out-patient and 217 prosititues were carried out at Daejeon areas during August to September, 1988. All the vaginal specimens were cultured on las’s casein hydrolysate-serum at 37.5℃ for 10 days. The results are summarized as follows: 1. The prevalence of Trichomonas vaginalis was 13.9% in total examined group. 2. The prevalence of Trichomonas vaginalis was 7.8% in 180 obsterics and gynecologic out-patients and 18.9% in 217 prostitutes. 3. According to age group, the prevalence of obstetrics and gynecologic patients were 10.7%(0―29 age group in years), 9.1% (40―49 age group in years), 5.6% (30―39 age group in years) and 5.3% (over 50 age group in years). 4. According to age group, the prevalence of obstetrics and gynecologic patients were 25.3% (0―29 age group in years), 25.0% (40―49 age group in years) and 11.4% (30―39 age group in years). 5. The most affected age group of Trichomonas vaginalis was 20―29 age group in obstetics and gynecologic out-patients and prostitutes.

So, in study, we tested exhaust gas by various mode in light duty diesel engine. Therefore, we can know about NOx and smoke seriousness.

        환경친화적 의류제품에 대한 소비자의 특성분석

        The purpose of this study is to analyze the characteristics of the environment-friendly customers in Korean fashion market. The study is analyzed through questionary paper to the domestic female group of more than or equal to age of twenty living in Seoul. The collected questionary paper is analyzed with SPSS/PC program and the method of statistical analysis used for this study is frequency analysis, factor analysis, cluster analysis, t-test, and crosstab analysis. The result of analysis of the characteristics of the environment-friendly customers involves the following: Firstly, there were no significant difference in the demographic characteristics between the environment-friendly customer group and non-environment-friendly customer gruop. Secondly, environment-friendly customers had more interest in educational program about environmental problems, had seen and heard more environmental program through current news from TV, newspapers, and magazines, and had more interest in recycling or separated collecting of garbage, and been doing better recycling or separated collecting. These serial processes of research, have the meaning in confirming that segmented market for environment-friendly customers exists in the domestic fashion market, and these will not only set up the basis of research on the market of environment-friendly fashion product but also be able to contribute to propose future direction of research.

        NIHSS 활용을 통한 뇌졸중 환자 간호의 질향상 전략

        문제: 뇌졸중 환자를 돌보는 간호현장에서는 급성기 뇌졸중 환자의 증상악화에 대한 조기사정과 적절한 대처가 매우 중요한 문제이다. 그러나 임상현장에서는 이를 객관화하고 수치화하여 모든 의료진이 의사소통할 수 있는 유용한 도구를 사용하고 있지는 못하는 실정이다. 이러한 문제를 해결하기위해 NIHSS(National Institute of Health Stroke Scale)를 신경과 진료의사를 중심으로활용하고 있지만 아직 간호현장에서는보편적으로사용하지 않고있다. 이에 뇌졸중환자간호에 NIHSS를 활용하여 뇌졸중 증상악화의 조기사정과 빠른 대처로 궁극적으로는 뇌졸중환자 간호의 질을보다 향상시키기 위하여 본활동이 시도되었다. 목적: 첫째, 간호사의 NIHSS 측정 신뢰도의 현수준을 점검하고 이를향상시키기 위한 프로그램을 개발·적용한다. 둘째, 간호사가급성기 뇌졸중환자에게 NIHSS 측정을 통하여 뇌졸중악화를 얼마나 예측할 수 있는지 분석하고 이를 향상시킬수있는 방법을 모색한다. 셋째, 궁극적으로 급성기 뇌졸중 환자의 증상악화의 조기 발견과치료를위한의료진의 올바른 대처지침을 마련하여 이를 적용한다. 의료기관: 경기도에 소재한 대학병원의 뇌졸중 집중치료실 질 향상 활동: 첫째, 간호사의 NIHSS 측정 신뢰도 향상을 위해 「NIHSS 간호사 교육 프로그램」을 마련하였다.

        세라믹 브라켓의 제거 시 Er : YAG 레이저의 효능

        본 연구에서는 세라믹 브라켓 제거에 Er:YAG 레이저 조사가 도움이 되는지 알아보고, 브라켓 제거에 적합한 레이저 조사 방법을 연구하였으며, 또 이렇게 적용된 레이저가 치수와 법랑질에 손상을 주는지도 알아보았다. 총 190개의 치아, 단결정 세라믹 브라켓(MISO), 다결정 세라믹 브라켓(Transcend series 6000)과 KEY Laser3를 사용하였다. 실험군은 세라믹 브라켓의 종류(단결정, 다결정)와 레이저의 에너지(140, 300, 450, 600 mJ)에 따라 분류하였으며, 레이저를 브라켓당 두 곳에 1펄스씩 조사하고, 전단 강도를 측정하였다. 대조군은 레이저를 조사하지 않는 군으로 하였다. 레이저 조사에 의한 열 효과는 브라켓 하방 법랑질과 치수강에서 측정하였으며, 전단 강도 측정 후 치면에 남아있는 접착제의 양을 접착제 잔류 지수(adhesive remnant index)를 이용하여 평가하였다. 레이저 조사로 인한 접착제의 파괴 양상과 법랑질 표면 변화를 주사전자현미경으로 관찰하였다. 모든 세라믹 브라켓군에서 레이저 에너지가 증가 할수록 전단 강도는 유의하게 감소하였다. 또한, 브라켓 하방 법랑질에서 최대 온도 변화는 평균 3.78˚C 상승에 그쳤으며, 치수강에서 최대 온도 변화는 평균 0.9˚C 상승에 그쳤다. 주사전자현미경을 이용한 법랑질과 접착제 단면 관찰에서 접착제 표면이 레이저에 의해 붕괴되어 분화구 모양의 구덩이로 관찰되었으며, 일부 시편에서 약 10 - 30μm의 법랑질 손상이 발견되었다. Transbond XT로 부착된 단결정 도재 브라켓(MISO)의 디본딩에 Er:YAG 레이저를 이용할 경우, 300 - 450 mJ의 레이저 에너지를, 그리고 다결정 도재 브라켓(Transcend series 6000)의 경우는 450 mJ 정도의 에너지를 사용하는 것이 효과적이고 안전할 것으로 생각된다. Objective: The aim of this study was to find out whether Er:YAG laser can aid in debonding ceramic brackets, and to see what kind of method will be the most appropriate for debonding. Methods: One hundred and ninety teeth, monocrystalline brackets (MISO, HT, Ansan-Si, Korea), polycrystalline brackets (TranscendTM series 6000, 3M Untek, Monrovia, CA, USA) and the KEY Laser3 (KavoDental, Biberach, Germany) were used. Experimental groups were classified according to the type of ceramic brackets, and the amount of laser energy (0, 140, 300, 450, 600 mJ). After applying laser on the bracket at two points at 1 pulse each, the shear bond strength was measured. The effect of heat caused by laser was measured at the enamel beneath the bracket and pulp chamber. After measuring the shear bond strength, adhesive residue was evaluated and enamel surface was investigated using SEM. Results: All ceramic bracket groups showed a significant decrease in shear bond strength as the laser energy increased. The greatest average temperature change was 3.78˚C on the enamel beneath the bracket and 0.9˚C on the pulp chamber. Through SEM, crater shape holes caused by the laser was seen on the enamel and adhesive surfaces. Conclusions: If laser is applied on ceramic brackets for debonding, 300 - 450 mJ of laser energy will be safe and efficient for monocrystalline brackets (MISO), and about 450 mJ for polycrystalline brackets (Transcend series 6000).

      • 톡소포자충 검출에 있어서 DNA 분리방법 및 반응 조건에 따른 중합효소 연쇄반응의 민감도 비교

        To optimize the PCR for the diagnosis of toxoplasmosis, different DNA extraction method, PCR condition to conform the target DNA and Southern blot hybridization were carried out. The results were as follows. 1. On DNA extraction, G NOME DNA isolation kit method from the tachyzoites of Toxoplasma RH strain and split second DNA isolation kit method from the blood of Toxoplasma injected mouse were most effective on yield and purity of DNA products than any other methods. 2. PCR products were relatively dependent upon the variation of MgCl_2 concentration and pH in PCR buffer. 3. As the PCR results after serial dilution, G NOME DNA isolation method from tachyzoites showed the sensitivity to 0.05 fg and the split second DNA isolation kit method showed to 5 pg. In the result, the sensitivity of PCR was increased to 10-1000 times after adding CTAB in these two methods. 4. In two steps PCR, denaturation and annealing, after adding CTAB, G NOME DNA isolation kit method showed sensitivity to 5 pg and split second DNA isolation kit method to 5 ng at the annealing of 72℃ and the former showed the sensitivity to 50 fg and the latter showed the same sensitivity with it at 72℃, at the annealing of 65℃. 5. As the result of comparison to the sensitivity of target DNA with sequntial amplification method and Southern blot hybridization, the one was 100-1000 times more sensitive than that of the other.
