18S-srRNA 분석에 의한 Acantbamoeba 한국 분리주들의 분류 검토

Acanthamoeba spp. spread in soil, ponds, air and swimming pool, cause a granulomatous amoebic meningitis and amoebic keratitis in human and experimental animals. Their classification had depended upon morphological characteristics and physiological conditions such as thermophilic status and generation times. However, because amoeba species showed the morphological and physiological diversity in isolates, many attempts have been made on identification of Acanthamoeba spp. Restriction fragments length polymorphism (RFLP) analysis of IBS-small subunit ribosomal RNA (srRNA) was an useful tool for classification among morphologically and genetically closely-related species. In this study, 18S-srDNAs of amoebae were amplified by PCR with primer encoding 18S-srRNA gene and digested with restriction endonudeases, and RFLP analysis was applied on classification of Acanthamoeba Korean isolates (YM-4, YM-5 and YM-7), comparing with reference amoebae, A. culbertsoni, A. polyphaga and A. royreba. Trophozoites of six Acanthamoeba spp. had typical acanthopoda, but did not show any morphological differences. Cyst of Acanthamoeba sp. YM-7 was similar to that of A. polyphaga which was designated as group Ⅱ Acanthamoeba. Morphologically other amoebae belonged to group Ⅲ Acanthamoeba. Digestion of 18S-srDNA with six enzymes, Dde Ⅰ, Hae Ⅲ, Hind Ⅲ, EcoR Ⅰ, Rsa Ⅰ and Sph Ⅰ, resulted in various DNA fragments. Using the method of Nei and U (1979) for RFLP analysis, genetic divergence was not observed between A culbertsoni and Acanthamoeba sp. YM-4. Between A. culbertsoni and Acanthamoeba sp. YM-5, genetic distance was 0.070, 0.364 between Acanthamoeba sp. YM-4 and A. polyphaga, and 0.277 between Acanthamoeba sp. YM-7 and A. polyphaga. In comparison with A. culbertsoni and A. polyphaga, Acanthamoeba sp. YM-7 showed genetic distance of 0.330 and 0.154, respectively. Thus, Acanthamoeba sp. YM-7 is a similar species to A. polyphaga. Acanthamoeba sp. YM-5 is a different species from A. polyphaga, but closely-related to A. culbertsoni. Acanthamoeba sp. YM-4 can be classified as a subspecies or a strain of A. culbertsoni.

We present the characteristics of the Seoul National University 4k Camera (SNUCAM) and report its performance on the 1.5m telescope at the Maidanak observatory in Uzbekistan. SNUCAM is a CCD camera with a pixel scale of 0.266" in $4096{\times}4096$ format, covering $18.1'{\times}18.1'$ field of view on the 1.5m. The camera is currently equipped with Bessell UBVRI, $H{\alpha}$, SDSS ugriz, and Y-band filters, allowing us to carry out a variety of scientific programs ranging from exoplanet studies to survey of quasars at high redshift. We examine properties of SNUCAM such as the bias level and its temporal variation, the dark current, the readout noise, the gain, the linearity, the fringe patterns, the amplifier bias, and the bad pixels. From our observations, we also constructed the master fringe frames in I-, z-, and Y-band. We outline some of the current scientific programs being carried out with SNUCAM, and demonstrate that SNUCAM on the 1.5m can deliver excellent images that reach to the $5-{\sigma}$ detection limits of R~25.5 mag and z~22.7 mag in 1 hour total integration.

In this study, we tried to get and make and analysis of the important informations to develop the appropriate teaching materials which are indispensable for arithmatic and science education in the primary schools of different areas in Kang Won Do. We classified four areas; cities, rural, fishing villages and mineralized ones. And we surveyed the present actural condition of teaching materials and its property. Many of the schools in each area have not enough teaching materials, because of the budgetary deficit and the shortage of time so that, in fact, pupils have been taught with no exploring activities but only a model experiment or recitation by the teachers. In conclusion, teaching materials which are suitable for the each actural situations should be developed and be come into wide use by the teachers. Especially, in the rural and fishing areas, it is necessary to be encouraged for using more teaching materials. Finally we should try to secure a firm and enduring plans for teaching materials.

It is crucial for the safety of the passengers to design the brakes of the vehicle appropriately. One must accurately estimate the temperature increase of the brake during the braking operation and provide sufficient cooling or heat loss to maintain the stress below the allowable limit of the braking system. In this study three models were used to obtain the temperature distribution during the braking process of an actual part was investigated and compared. A two-dimensional axi-symmetric model was sufficient for obtaining the temperature distribution; however, it was found to be unsatisfactory for analyzing the stresses. In the case of the tree-dimensional portion of the model the temperature distribution turned out to be similar to the actual temperature measurement. However, under repeated braking conditions the maximum equivalent stress turned out to be greater that the yield stress; thus, the current design requires research.

Stress of a caliper brake predesigned by a domestic company are analyzed by ABAQUS utilizing the finite element method. The analysis was perfomed in two parts-first the cylinder block and then the carrier block. The value of the von Mises equivalent stress at some portions of the break turned up to over the yield stress of cast iron. The critical area of the originally designed model was examined. It was found that the von Mises values stress values at the critical weak point of carrier block decreased considerably after it was redesigned.
