        Ameliorative effects of Dictyota dichotoma on hepatotoxicity induced by gibberellic acid in albino rats

        Gibberellic acid (GA3) is a natural plant growth regulator that is crucial for plant structural and functional development. We examined the alleviating capacity of brown algae (Dictyota dichotoma) on biochemical and molecular degenerative processes caused by sub-chronic exposure to gibberellic acid resulting in hepatic cell apoptosis. Adult male albino rats were divided into five equal groups: the first group received distilled water, the second group was treated with GA3, the third group was administered D. dichotoma extract suspended in 1% carboxymethylcellulose (CMC), the fourth group was administered both GA3 and D. dichotoma simultaneously, and the fifth group received 1% CMC orally, 5 days per week for a total of 50 days. The results indicated that GA3 induced a significant increase in liver function parameters based on serum levels of alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), and albumin, which indicate hepatotoxicity. A marked increase in malondialdehyde (MDA) levels and a marked decrease in reduced glutathione (GSH), glutathione-S-transferase (GST), and superoxide dismutase (SOD) were observed as a result of induction of lipid peroxidation and oxidative stress. Histopathology revealed severely degenerated hepatocytes including cytoplasmic vacuolations and many apoptotic cells with weak Bcl2 expression. Similarly, there was a significant up-regulation of gene and protein expression levels for the pro-apoptotic markers, Caspase-3 and Bax, and an increase in pro-inflammatory marker levels, tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) as well as C-reactive protein (CRP). The co-administration of D. dichotoma restored the disrupted biochemical, histopathological, molecular, and inflammatory changes resulting from GA3 toxicity. Our results confirm the antioxidant, anti-inflammatory, anti-apoptotic, and hepatoprotective potential of D. dichotoma.

        Automated condition assessment of concrete bridges with digital imaging

        The reliability of a Bridge management System depends on the quality of visual inspection and the reliable estimation of bridge condition rating. However, the current practices of visual inspection have been identified with several limitations, such as: they are time-consuming, provide incomplete information, and their reliance on inspectors' experience. To overcome such limitations, this paper presents an approach of automating the prediction of condition rating for bridges based on digital image analysis. The proposed methodology encompasses image acquisition, development of 3D visualization model, image processing, and condition rating model. Under this method, scaling defect in concrete bridge components is considered as a candidate defect and the guidelines in the Ontario Structure Inspection Manual (OSIM) have been adopted for developing and testing the proposed method. The automated algorithms for scaling depth prediction and mapping of condition ratings are based on training of back propagation neural networks. The result of developed models showed better prediction capability of condition rating over the existing methods such as, Naive Bayes Classifiers and Bagged Decision Tree.

        Protective effect of hydrogen sulfide against cold restraint stress-induced gastric mucosal injury in rats

        Hydrogen sulfide (H2S) is an endogenous gaseousmediator plays a potential role in modulating gastricinflammatory responses. However, its putative protectiverole remains to be defined. The present study aimed toevaluate role of the exogenously released and endogenouslysynthesized H2S in cold restraint stress (CRS)-induced oxidativegastric damage in rats. Rats were restrained, andmaintained at 4 C for 3 h. The H2S donor, sodium hydrosulfide(NaHS) (60 lmol/kg) was injected intraperitoneally(i.p.) before CRS. Our results revealed that NaHS pretreatmentsignificantly attenuated ulcer index, free and total acidoutput, and pepsin activity in gastric juice along withdecreased gastric mucosal carbonyl content and reactiveoxygen species production. This was accompanied byincreased gastric juice pH and mucin concentration inaddition to restoring the deficits in the gastric reduced glutathione,catalase as well as superoxide dismutase enzymeactivities. NaHS pretreatment markedly reduced the serumlevel of tumor necrosis factor (TNF-a) and myeloperoxidaseactivity compared to CRS-non-treated. Moreover, NaHSpreadministration significantly abrogated the inflammatoryand the deleterious responses of gastric mucosa in CRS. Theprotective effects of H2S were confirmed by gastric histopathologicalexamination. However, pretreatment withthe H2S-synthesizing enzyme, cystathionine-gamma-lyaseinhibitor, beta-cyano-L-alanine (50 mg/kg, i.p.) reversed thegastroprotection afforded by the endogenous H2S. Collectively, our results suggest that H2S can protect ratgastric mucosa against CRS-induced gastric ulcerationpossibly through mechanisms that involve anti-oxidant andanti-inflammatory actions alongside enhancement of gastricmucosal barrier and reduction in acid secretory parameters.

        Staphylococcus aureus derived hyaluronic acid and bacillus Calmette-Guérin purified proteins as immune enhancers to rabies vaccine and related immuno-histopathological alterations

        Purpose: One of the essential goals regarding the successful control of rabies infection is the development of a safe, effective, and inexpensive vaccine. the current study aimed to evaluate the inactivation potential of β-propiolactone (βPL), binary ethyleneimine (BEI), and hydrogen peroxide (H2O2). Materials and Methods: Estimating the inactivation kinetics of βPL, BEI, and H2O2 revealed that the tested inactivants could completely and irreversibly inactivate rabies virus within 2, 12, and 4 hours, respectively while maintaining its viral immunogenicity. The potency of βPL, BEI, and H2O2 inactivated vaccines was higher than the World Health Organization acceptance limit and were in the order of 3.75, 4.21, and 3.64 IU/mL, respectively. Monitoring the humoral and cellular immunity elicited post-immunization using Staphylococcus aureus derived hyaluronic acid (HA) and bacillus Calmette-Guérin purified protein derivative (PPD) adjuvanted rabies vaccine candidates were carried out using enzyme-linked immunosorbent assay. Results: Results demonstrated that both adjuvants could progressively enhance the release of anti-rabies total immunoglobulin G as well as the pro-inflammatory mediators (interferon-gamma and interleukin-5) relative to time. However, a higher immune response was developed in the case of HA adjuvanted rabies vaccine compared to PPD adjuvanted one. The harmful consequences of the tested adjuvants were considered via investigating the histopathological changes in the tissues of the immunized rats using hematoxylin and eosin stain. Lower adverse effects were observed post-vaccination with HA and PPD adjuvanted vaccines compared to that detected following administration of the currently used alum as standard adjuvant. Conclusion: Our findings suggested that HA and PPD could serve as a promising platform for the development of newly adjuvanted rabies vaccines with elevated immune enhancing potentials and lower risk of health hazards.
