Free Radical Scavenging Activity of the Seed of Phaseolus calcaratus Roxburgh

The seed of Phaseolus calcaratus Roxburgh (PHCR) is traditionally used for anti-pyretic and anti-inflammatory effects. Although these effects are believed to be related to its antioxidant potential, little information is available for the mechanisms by which PHCR seed might scavenge free radicals or otherwise act as an antioxidant. In the present study, we purified some fractions from the ethanol extract of PHCR seed and evaluated each fraction's ability to scavenge free radicals generated by cell-free systems. We also identified active compound that is putatively responsible for free radical scavenging by analyzing NMR spectra. PHCR samples exhibited a concentration-dependent radical scavenging activity against hydroxyl radicals, superoxide anions, and DPPH radicals. Of the samples tested, a methanol-eluted sub-fraction from the PHCR extract, named FF4, scavenged these radicals more effectively than the other fractions. We identified catechin-7-O-b-D-glucopyranoside as the active compound responsible for free radical scavenging potential of FF4.

The seeds of Raphanus sativus L. (RSL) and Phaseolus calcaratus Roxburgh (PHCR), the root of Scutellaria baicalensis (SB), and the flower of Lonicera japonica (LJ) have been traditionally used as herbal medicines for anti-inflammation. Unlike the SB and LJ, little information is available for the scientific bases that show the anti-inflammatory mechanisms of RSL and PHCR. In this study, we prepared boiled water extracts from the medicines and determined their potentials in inhibiting nitric oxide (NO) production, cyclooxygenase-2 (COX-2) expression, and tumor necrosis factor (TNF)-a and interleukin (IL)-6 secretion in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. The effects of the medicines on serum IgE levels in ovalbumin (OVA)-administrated mice were also studied. The medicines inhibited production of TNF-a and IL-6, and COX-2 expression in LPS-stimulated macrophages. Especially, PHCR water extract showed more potent inhibition on TNF-a production than SB and LJ extracts, but RSL extract did not exert these effects. Similar to the cases of SB and LJ, PHCR extract prevented the phosphorylation of IkBa and c-Jun, and the activation of NF-kB-DNA binding. Further, oral supplementation of PHCR extract attenuated significantly serum levels of total and OVA-specific IgE in OVA-treated animals. These results suggest a possibility that PHCR water extract can be used for the treatment of inflammatory and allergic diseases.
An Improved Synchronization Acquisition Method of Fast Frequency Hopping Signal

The synchronization technology is one of the key technologies in fast frequency hopping communication system. The performance of synchronous loop will directly affect the whole performance of the fast frequency hopping communication system. In view of the deficiency of synchronization time and synchronization anti-interference in the existing synchronization methods, an improved synchronization acquisition method is put forward, that is double judgment circular correlation method. The test results show that the improved synchronization acquisition method can effectively improve synchronization speed, increase synchronization anti-interference performance and reduce the complexity of synchronization loop.

Catechin-7-O-β-D-glucopyranoside (CA-G) was previously isolated from red bean (the seed of Phaseolus calcaratus cv. Roxburgh). This study examined the ability of CA-G to scavenge reactive oxygen species generated by cell-free systems and to protect cells from oxidative stress caused by hydrogen peroxide (H_2O_2). The mechanism by which CA-G exerts its antioxidant and anti-apoptotic action on H_2O_2-exposed cells was also investigated. CA-G treatment prevented H_2O_2-mediated apoptosis and inhibited the formation of single stand breaks in DNA in H_2O_2-exposed BJAB cells. CA-G suppressed mitochondrial stress and caspase activation caused by H_2O_2. Mechanistic experiments revealed that the antioxidant mechanism of CA-G on H_2O_2-mediated oxidative damage was due to the direct scavenging of hydroxyl radicals and/or to the chelation of metal ions that were used to produce hydroxyl radicals from H_2O_2 via the Fenton reaction. Collectively,these findings suggest beneficial roles of CA-G or CA-Grich red bean on the protection from oxidative damage.
Red Bean Extract Reduces Inflammation and Increases Survival in a Murine Sepsis Model

This study used in vitro and in vivo experimental models to investigate the anti-inflammatory potential of the butanol fraction of red bean ethanol extract (BF-RBEE),which contains the biologically active molecule catechin-7-β-D-glucopyranoside. Treatment with BF-RBEE inhibited nitric oxide production in lipopolysaccharide (LPS)-stimulated macrophages through suppression of extracellular signalregulated kinase and IκBα activation. In an in vivo murine sepsis model, oral administration of BF-RBEE improved mouse survival. Specifically, the survival rate of mice injected with LPS was 0 and 40% in ICR and BALB/c mice, respectively, whereas the survival of mice co-treated with BF-RBEE was 100% in both mouse types. This increase in survival with the BF-RBEE administration was correlated with decreased tumor-necrosis factor-α and interferon-γ and increased interleukin-10. Oral administration of BF-RBEE also inhibited total and ovalbumin-specific IgE production in experimental mice. These results suggest the possible usefulness of red beans in the treatment of inflammatory diseases.
