        FSCB phosphorylation in mouse spermatozoa capacitation

        It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation.         Background and Objectives: Mutation or inactivation of the p53 is important mechanism in a wide variety of human tumors. The purpose of this study is to investigate the relationship between expression amount of p53 protein and the grade of astrocytic gliomas. Materials and Methods: This study analyzes 40 paraffin-embedded astrocytic glioma, including 10 cases of low grade, 20 cases of high grade, and 10 cases of maligant glioma (glioblastoma multiforme) were used to analyze the status of abnormal accumulation of wild type of p53 protein. The evaluation of the immunostaining result was based on the percentage of positive neoplastic cells. Results: Average positive cell rate of p53 protein were 4.2%, 35.4% and 16.2% in low grade astrocytoma, high grade astrocytoma and glioblastoma multiforme, respectively. Conclusion: p53 gene mutations were correlated with histologic grade of glial tumors. High grade and glioblastoma multiforme showed higher positive cell rate than low grade astrocytoma. These results suggest that the high positive rates of p53 protein is related to the poor prognosis.

        IRS-2 Partially Compensates for the Insulin Signal Defects in IRS-1<sup>-/-</sup> Mice Mediated by miR-33

        Insulin signaling is coordinated by insulin receptor substrates (IRSs). Many insulin responses, especially for blood glucose metabolism, are mediated primarily through Irs-1 and Irs-2. Irs-1 knockout mice show growth retardation and insulin signaling defects, which can be compensated by other IRSs in vivo; however, the underlying mechanism is not clear. Here, we presented an Irs-1 truncated mutated mouse ($Irs-1^{-/-}$) with growth retardation and subcutaneous adipocyte atrophy. $Irs-1^{-/-}$ mice exhibited mild insulin resistance, as demonstrated by the insulin tolerance test. Phosphatidylinositol 3-kinase (PI3K) activity and phosphorylated Protein Kinase B (PKB/AKT) expression were elevated in liver, skeletal muscle, and subcutaneous adipocytes in Irs-1 deficiency. In addition, the expression of IRS-2 and its phosphorylated version were clearly elevated in liver and skeletal muscle. With miRNA microarray analysis, we found miR-33 was down-regulated in bone marrow stromal cells (BMSCs) of $Irs-1^{-/-}$ mice, while its target gene Irs-2 was up-regulated in vitro studies. In addition, miR-33 was down-regulated in the presence of Irs-1 and which was up-regulated in fasting status. What's more, miR-33 restored its expression in re-feeding status. Meanwhile, miR-33 levels decreased and Irs-2 levels increased in liver, skeletal muscle, and subcutaneous adipocytes of $Irs-1^{-/-}$ mice. In primary cultured liver cells transfected with an miR-33 inhibitor, the expression of IRS-2, PI3K, and phosphorylated-AKT (p-AKT) increased while the opposite results were observed in the presence of an miR-33 mimic. Therefore, decreased miR-33 levels can up-regulate IRS-2 expression, which appears to compensate for the defects of the insulin signaling pathway in Irs-1 deficient mice.

        Monosaccharide as a Central Scaffold Toward the Construction of Salicylate-Based Bidentate PTP1B Inhibitors via Click Chemistry

        The discovery of carbohydrate-based bioactive compounds has recently received considerable interest in the drug development. This paper stresses on the application of 1-methoxy-O-glucoside as the central scaffold, whereas salicylic pharmacophores were introduced with diverse spatial orientations probing into the structural preference of an enzymatic target, i.e. protein tyrosine phosphatase 1B (PTP1B). By employing regioselective protection and deprotection strategy, 2,6-, 3,4-, 4,6- and 2,3-di-O-propynyl 1-methoxy-O-glucosides were previously synthesized and then coupled with azido salicylate via click chemistry in forming the desired bidentate salicylic glucosides with high yields. The inhibitory assay of the obtained triazolyl derivatives leads to the identification of the 2,3-disubstituted salicylic 1-methoxy-O-glucoside as the structurally privileged PTP1B inhibitor among this bidentate compound series with micromole-ranged $IC_{50}$ value and reasonable selectivity over other homologous PTPs tested. In addition, docking simulation was conducted to propose a plausible binding mode of this authorized inhibitor with PTP1B. This research might furnish new insight toward the construction of structurally different bioactive compounds based on the monosaccharide scaffold.

        ICCD: Interactive Continuous Collision Detection between Deformable Models Using Connectivity-Based Culling

        <P>We present an interactive algorithm for continuous collision detection between deformable models. We introduce multiple techniques to improve the culling efficiency and the overall performance of continuous collision detection. First, we present a novel formulation for continuous normal cones and use these normal cones to efficiently cull large regions of the mesh as part of self-collision tests. Second, we introduce the concept of ldquoprocedural representative trianglesrdquo to remove all redundant elementary tests between nonadjacent triangles. Finally, we exploit the mesh connectivity and introduce the concept of ldquoorphan setsrdquo to eliminate redundant elementary tests between adjacent triangle primitives. In practice, we can reduce the number of elementary tests by two orders of magnitude. These culling techniques have been combined with bounding volume hierarchies and can result in one order of magnitude performance improvement as compared to prior collision detection algorithms for deformable models. We highlight the performance of our algorithm on several benchmarks, including cloth simulations, N-body simulations, and breaking objects.</P>

        Some remarks on problems of subset sum

Burr posed the following question: Determine conditions on integers sequence $B$ that imply either the existence or the non-existence of $A$ for which $P(A)$ is the set of all non-negative integers not in $B$. In this paper, we focus on some problems of subset sum related to Burr's question.

        Isolation of Dibutyl Phthalate-Degrading Bacteria and Its Coculture with Citrobacter freundii CD-9 to Degrade Fenvalerate

        Continued fenvalerate use has caused serious environmental pollution and requires large-scale remediation. Dibutyl phthalate (DBP) was discovered in fenvalerate metabolites degraded by Citrobacter freundii CD-9. Coculturing is an effective method for bioremediation, but few studies have analyzed the degradation pathways and potential mechanisms of cocultures. Here, a DBPdegrading strain (BDBP 071) was isolated from soil contaminated with pyrethroid pesticides (PPs) and identified as Stenotrophomonas acidaminiphila. The optimum conditions for DBP degradation were determined by response surface methodology (RSM) analysis to be 30.9 mg/l DBP concentration, pH 7.5, at a culture temperature of 37.2℃. Under the optimized conditions, approximately 88% of DBP was degraded within 48 h and five metabolites were detected. Coculturing C. freundii CD-9 and S. acidaminiphila BDBP 071 promoted fenvalerate degradation. When CD-9 was cultured for 16 h before adding BDBP 071, the strain inoculation ratio was 5:5 (v/v), fenvalerate concentration was 75.0 mg/l, fenvalerate was degraded to 84.37 ± 1.25%, and DBP level was reduced by 5.21 mg/l. In addition, 12 fenvalerate metabolites were identified and a pathway for fenvalerate degradation by the cocultured strains was proposed. These results provide theoretical data for further exploration of the mechanisms used by this coculture system to degrade fenvalerate and DBP, and also offer a promising method for effective bioremediation of PPs and their related metabolites in polluted environments.

        SOME REMARKS ON SUMSETS AND RESTRICTED SUMSETS

        Let A be a finite set of integers. For any integer $h{\geq}1$, let h-fold sumset hA be the set of all sums of h elements of A and let h-fold restricted sumset $h^{\wedge}A$ be the set of all sums of h distinct elements of A. In this paper, we give a survey of problems and results on sumsets and restricted sumsets of a finite integer set. In details, we give the best lower bound for the cardinality of restricted sumsets $2^{\wedge}A$ and $3^{\wedge}A$ and also discuss the cardinality of restricted sumset $h^{\wedge}A$.

        The genus Cephonodes from Xisha islands, Hainan Province, China, with description of a new species Cephonodes sanshaensis Deng & Huang, 2023 sp. nov. (Lepidoptera: Sphingidae)

        A new species, Cephonodes sanshaensis Deng & Huang, sp. nov. is described from Xisha islands nearSansha City, China. Photographs of the adults and their genitalia are provided. The new species is similarto C. hylas (Linnaeus, 1771) and C. picus (Cramer, 1777) but can be easily distinguished by characters in themale genitalia: the right lobe of the uncus is hook-shaped with distinctly acute apex, the left valva is longand narrow with a truncate apex, and the right valva is broad and knife-shaped. Molecular analysis basedon cytochrome c oxidase I gene barcode sequences is used to infer the phylogenetic position of the newspecies within the genus Cephonodes. An updated key and checklist to the worldwide species of thegenus Cephonodes are also provided.
