Serotonin-Norepinephrine Reuptake Inhibitors for Pain Control: Premise and Promise

<P>The precise mechanisms of pain perception and transmission in the central nervous system have not been fully elucidated. However, extensive data support a role for the monoamine neurotransmitters, serotonin and norepinephrine, in the modulation of pain. Experiments with animal models of pain indicate that noradrenergic interventions, and to a lesser extent serotonergic interventions, reduce pain-related behavior. This is supported by data from clinical trials in humans in which antidepressants have been shown to reduce pain and functional impairment in central and neuropathic pain conditions. These effects are particularly well-studied in trials with serotonin-norepinephrine reuptake inhibitors (SNRIs), which have provided a useful tool in the clinician’s arsenal, particularly considering the limitations of other classes of pain medications such as opioids, anti-inflammatories, and anticonvulsants (i.e., limited efficacy, safety and tolerability issues). Moreover, painful physical symptoms are frequently comorbid with major psychiatric disorders such as major depressive disorder and anxiety disorders. This paper reviewed and summarized the rationale and potential role of SNRIs for the control of pain including clinical and preclinical background. Currently evidence does not definitely support a role of the SNRIs, while limited data propose a putative promise of SNRIs in the treatment of pain related disorders including fibromyalgia and depressed patients with multiple somatic complaints. More researches are warranted to generalize currently available preliminary evidences.</P>
Toll-like Receptor 2 in Autoimmune Inflammation

TLR signaling is critical for broad scale immune recognition of pathogens and/or danger molecules. TLRs are particularly important for the activation and the maturation of cells comprising the innate immune response. In recent years it has become apparent that several different TLRs regulate the function of lymphocytes as well, albeit to a lesser degree compared to innate immunity. TLR2 heterodimerizes with either TLR1 or TLR6 to broadly recognize bacterial lipopeptides as well as several danger-associated molecular patterns. In general, TLR2 signaling promotes immune cell activation leading to tissue inflammation, which is advantageous for combating an infection. Conversely, inappropriate or dysfunctional TLR2 signaling leading to an overactive inflammatory response could be detrimental during sterile inflammation and autoimmune disease. This review will highlight and discuss recent research advances linking TLR2 engagement to autoimmune inflammation.
Automatic categorization of chloride migration into concrete modified with CFBC ash

The objective of this investigation was to develop rules for automatic categorization of concrete quality using selected artificial intelligence methods based on machine learning. The range of tested materials included concrete containing a new waste material - solid residue from coal combustion in fluidized bed boilers (CFBC fly ash) used as additive. The rapid chloride permeability test - Nordtest Method BUILD 492 method was used for determining chloride ions penetration in concrete. Performed experimental tests on obtained chloride migration provided data for learning and testing of rules discovered by machine learning techniques. It has been found that machine learning is a tool which can be applied to determine concrete durability. The rules generated by computer programs AQ21 and WEKA using J48 algorithm provided means for adequate categorization of plain concrete and concrete modified with CFBC fly ash as materials of good and acceptable resistance to chloride penetration.
A Comparative Measure of Decentralization for Southeast Asia

In this article we set out a fine-grained measure of the formal authority of intermediate subnational government for Indonesia, Malaysia, the Philippines, South Korea, and Thailand that is designed to be a flexible tool in the hands of researchers and policymakers. It improves on prior measures by providing annual estimates across ten dimensions of regional authority; it disaggregates to the level of the individual region; and it examines individual regional tiers, asymmetric regions, and regions with special arrangements. We use the measure and its elements to summarize six decades of regional governance in Southeast Asia and conclude by noting how the Regional Authority index could further the dialogue between theory and empirics in the study of decentralization and democratization.
