      • Detection of p53 Common Intron Polymorphisms in Patients with Gastritis Lesions from Iran

        Background: p53 alterations have been implicated in the development of many cancers, such as gastric cancer, but there is no evidence of p53 intron alterations in gastritis lesions. The aim of this study was to investigate the p53 intron alterations in gastritis along with p53 and mismatch repair protein expression and microsatellite status. Materials and Methods: PCR-sequencing was conducted for introns 2-7 on DNA extracted from 97 paired samples of gastritis lesions and normal adjacent tissue. Abnormal accumulation of p53 and mismatch repair proteins was investigated using immunohistochemistry. In addition, microsatellite status was evaluated with reference to five mononucleotide markers. Results: Gastritis cases included 41 males and 56 females in the age range of 15-83 years, 87.6% being H.pylori positive. IVS2+38, IVS3ins16 and IVS7+72 were the most polymorphic sites. Their minor allele frequency values were as follows: 0.38, 0.21 and 0.06, respectively. Samples with GG genotype at IVS2+38 and CT at IVS7+72 had no insertion. Moreover, most of the stable samples (91.9 %) had a G allele at IVS2+38. All of the samples were IHC negative for p53 protein, microsatellite stable and expressed mismatch repair proteins. p53 alterations were prominent in the H. Pylori+ group, but without statistical significance. Conclusions: According to our results, some p53 polymorphisms such as IVS2+38, IVS3ins16 and IVS7+72, because of their correlations together or with microsatellite status may contribute to gastritis development. However, so far effects on p53 expression and function remain unclear. Therefore, a comprehensive survey is needed to delineate their biological significance.

        Investigation of microwave absorbing properties for magnetic nanofiber of polystyrenee-polyvinylpyrrolidone

        Aligned magnetic blend of polystyreneepolyvinylpyrrolidone (PSePVP) nanofibers were prepared by this method. First, polystyreneepolyvinylpyrrolidone (PSePVP) blend solution in THF was synthesized. Then magnetic of PSePVPeFe3O4epolyethylene glycol (PEG) was prepared by masking method. Finally, magnetic nanofiber of PSePVPeFe3O4ePEG was prepared by electrospinning method, too. An electric potential difference of 25 kV was applied between the collector and a syringe tip, and the distance between the collector and the tip was 13 cm. Fe3O4 is exhibit various magnetic properties of which the complex permeability and the permittivity, in particular, are important in determining their high frequency characteristics. The magnetic oxide particles and nanofiber of nanometer size were characterized by TEM and SEM respectively. The thermal properties of nanofibers were determined by TGA and DSC. The magnetic characterization of the fibers was also performed by VSM and AFM techniques. On the other hand, nanofiber with diameters ranging from 30 to 40 nm, showing at room temperature, coercive field values of around 25 kV and saturation magnetization was 1.1 emu/g. Microwave reflection loss of the sample was tested at 8e12 GHz microwave frequencies and the results showed that magnetic nanofiber possessed the microwave absorbing properties.

        A Web-Based Global Educational Model for Training in Semen Analysis during the COVID-19 Pandemic

        Purpose: In response to the COVID-19 pandemic, the American Center for Reproductive Medicine (ACRM) transitioned its annual training in assisted reproductive technology (ART) from a hands-on, laboratory-based training course to a fully online training endorsed by the American College of Embryology. Here we describe our experience and assess the quality of an online training format based on participant outcomes for the first three modules of a planned series of online ART training. Materials and Methods: These modules included manual semen analysis, sperm morphology and ancillary semen tests (testing for leukocytospermia, sperm vitality, and anti-sperm antibody screening). The virtual format consisted of lecture presentations featuring laboratory protocols with corresponding video demonstrations of routine techniques and best practices. Practical scenarios, troubleshooting, and clinical interpretation of laboratory results were also discussed. At the end of each module, an optional multiple choice question test was held as a prerequisite to obtain certification on the topics presented. Course quality was assessed using participant responses collected via online surveys. Results: The digital delivery methods used were found to have largely or completely met the participants’ expectations for all questions (>85%). The majority (>87%) of the participants either strongly agreed or agreed that the course content was well-structured with appropriate depth, and that their overall expectations of the course had been met. Conclusions: This training format appears to be a realistic teaching option to freely share highly specialized expertise and technical knowledge with participants from anywhere in the world with varying levels of competency or experience.
