        Correlation between pr1 and pr2 Gene Content and Virulence in Metarhizium anisopliae Strains

        Metarhizium anisopliae is a widely studied model to understand the virulence factors that participate in pathogenicity. Proteases such as subtilisin-like enzymes (Pr1) and trypsin-like enzymes (Pr2) are considered important factors for insect cuticle degradation. In four M. anisopliae strains (798, 6342, 6345, and 6347), the presence of pr1 and pr2 genes, as well as the enzymatic activity of these genes, was correlated with their virulence against two different insect pests. The 11 pr1 genes (A, B, C, D, E, F, G, H, I, J, and K) and pr2 gene were found in all strains. The activity of individual Pr1 and Pr2 proteases exhibited variation in time (24, 48, 72, and 96 h) and in the presence or absence of chitin as the inductor. The highest Pr1 enzymatic activity was shown by strain 798 at 48 h with chitin. The highest Pr2 enzymatic activity was exhibited by the 6342 and 6347 strains, both grown with chitin at 24 and 48 h, respectively. Highest mortality on S. exigua was caused by strain 6342 at 48 h, and strains 6342, 6345, and 6347 caused the highest mortality 7 days later. Mortality on Prosapia reached 30% without variation. The presence of subtilisin and trypsin genes and the activity of these proteases in M. anisopliae strains cannot be associated with the virulence against the two insect pests. Probably, subtilisin and trypsin enzyme production is not a vital factor for pathogenicity, but its contribution is important to the pathogenicity process.

        Color discrepancy of single-shade composites at different distances from the interface measured using cell phone images

        Objectives This study aimed to evaluate the impact of substrate color and interface distance on the color adjustment of 2 single-shade composites, Vittra APS Unique and Charisma Diamond One. Materials and Methods Dual disc-shaped specimens were created using Vittra APS Unique or Charisma Diamond One as the center composite, surrounded by shaded composites (A1 or A3). Color measurements were taken with a spectrophotometer against a gray background, recording the color coordinates in the CIELAB color space. Illumination with a light-correcting device and image acquisition using a polarizing filter-equipped cell phone were performed on specimens over the same background. Image processing software was used to measure the color coordinates in the center and periphery of the inner composite and in the outer composite. The color data were then converted to CIELAB coordinates and adjusted using data from the spectrophotometer. Color differences (ΔE00) between the center/periphery of single-shade and outer composites were calculated, along with color changes in single-shade composites caused by different outer composites. Color differences for the inner composites surrounded by A1 and A3 were also calculated. Data were analyzed using repeated-measures analysis of variance (α = 0.05). Results The results showed that color discrepancies were lowest near the interface and when the outer composite was whiter (A1). Additionally, Charisma Diamond One exhibited better color adjustment ability than Vittra APS Unique. Conclusions Color discrepancies between the investigated single-shade composites diminished towards the interface with the surrounding composite, particularly when the latter exhibited a lighter shade.         Command Shaping Control for Micro-milling Operations

        Micro milling requires both high speed and high accuracy in order to economically produce parts with features on the scale of 1 μm. Micro mills are smaller and more flexible than traditional large-scale machines. Therefore, vibration of the machine structure is a significant problem. Given that micro milling requires high positioning precision, even small vibrations in the controller dynamics are problematic. The small-scale operation has considerably lower tool/workpiece interaction forces than traditional-scale milling. These low cutting forces have minimal effect on the machine structural re-sponse. Therefore, the dominant dynamic factor in exciting vibration can be the machine tool motion, rather than the workpiece/tool interaction. Given this realization, properly shaping the motions of the micro mill is a promising approach to reduce vibration. This paper presents a nonlinear command-shaping technique to reduce the vibrations of a micro mill that can be implemented with a standard CNC controller. The robustness of this technique to modeling errors and disturbances is investigated. Theoretical proofs and experimental demonstrations of the command-shaping technique are presented. The improved performance from the command shaping enables higher throughput and improved accuracy of the micro mill.

        What Is the Ideal Core Number for Ultrasound-Guided Prostate Biopsy?

        Purpose: We evaluated the utility of 10-, 12-, and 16-core prostate biopsies for detectingprostate cancer (PCa) and correlated the results with prostate-specific antigen (PSA)levels, prostate volumes, Gleason scores, and detection rates of high-grade prostaticintraepithelial neoplasia (HGPIN) and atypical small acinar proliferation (ASAP). Materials and Methods: A prospective controlled study was conducted in 354 consecutivepatients with various indications for prostate biopsy. Sixteen-core biopsy specimenswere obtained from 351 patients. The first 10-core biopsy specimens were obtainedbilaterally from the base, middle third, apex, medial, and latero-lateral regions. Afterward, six additional punctures were performed bilaterally in the areas more lateralto the base, middle third, and apex regions, yielding a total of 16-core biopsyspecimens. The detection rate of carcinoma in the initial 10-core specimens was comparedwith that in the 12- and 16-core specimens. Results: No significant differences in the cancer detection rate were found between thethree biopsy protocols. PCa was found in 102 patients (29.06%) using the 10-core protocol,in 99 patients (28.21%) using the 12-core protocol, and in 107 patients (30.48%) usingthe 16-core protocol (p=0.798). The 10-, 12-, and 16-core protocols were comparedwith stratified PSA levels, stratified prostate volumes, Gleason scores, and detectionrates of HGPIN and ASAP; no significant differences were found. Conclusions: Cancer positivity with the 10-core protocol was not significantly differentfrom that with the 12- and 16-core protocols, which indicates that the 10-core protocolis acceptable for performing a first biopsy.

        Determination of kinetic parameters for the sisal residue pyrolysis through thermal analysis

        Determining the kinetics of biomass degradation by thermogravimetry is complex due to the existence ofnumerous parallel and consecutive reactions. The individual use of models available in the literature haslimitations regarding the applicability of the data due to high adjustment errors or lack of process information. Thus, a new procedure was proposed to determine the degradation kinetics of sisal residue, atheating rates of 20–80 C/min and inert atmosphere. First, the reaction order was determined by thefit model for each heating rate, resulting in approximate values. Then, the activation energy and thepre-exponential factor were determined by free models (Flynn-Wall-Ozawa, Kissinger-Akahira-Sunose,and Friedman). These models were validated from the experimental data of conversion and temperature,together with the calculated kinetic parameters. Friedman’s model was the best fit. Finally, the errorsobtained are compatible with those in the literature, indicating that this procedure can be used in kineticsinvolving biomass pyrolysis.

        Volcanic-hypabyssal rock geochemistry of a subduction-related marginal basin ophiolite: Southeast Bohol Ophiolite-Cansiwang M?ange Complex, Central Philippines

        The Early Cretaceous Southeast Bohol Ophiolite-Cansiwang Mlange Complex and the Alicia Schist form thebasement of southeastern Bohol Island in central Philippines.New geochemical data show that four discrete groups consti-tute the volcanic and associated hypabyssal rocks of the ophi-olite-mlange complex: boninitic rocks (BON), enriched andnormal mid-ocean ridge basalt-like rocks (E-MORB; N-MORB) and high-magnesian andesites (HMA). Of these fourgroups, the BON are the most depleted in REEs and with themost pronounced negative Nb anomalies. Both MORB-liketypes exhibit subduction-zone influence as reflected in theirslight negative Nb anomalies. Characteristically with flat andLREE-depleted patterns, the HMA samples apear to mimicN-MORB patterns but with lower REE concentrations. Thisgeochemical diversity is best explained by a suprasubductionzone environment of formation as is also evident from field geo-believed to have ben concurrent with the ophiolite’s emplace-ment by subduction-accretion along a forearc margin. This tec-tonic boundary was later jammed into inactivity with the entryof the Alicia Schist that most likely was an oceanic bathymetrichigh. The intercalation of both tufaceous materials and pelagicchert with the pillow basalts are consistent with a marginalbasin tectonic setting.

        Sedoanalgesia With Midazolam and Fentanyl Citrate Controls Probe Pain During Prostate Biopsy by Transrectal Ultrasound

        Purpose: To assess the pain intensity of patients administered midazolam and fentanylcitrate before undergoing transrectal ultrasound-guided prostate biopsy. Materials and Methods: This was a study in patients with different indications for prostatebiopsy in whom 5 mg of midazolam and 50 μg of fentanyl citrate was administeredintravenously 3 minutes before the procedure. After biopsy, pain was assessed by useof a visual analogue scale (VAS) in three stages: VAS 1, during probe introduction; VAS2, during needle penetration into prostate tissue; and VAS 3, in the weeks followingthe exam. Pain intensity at these different times was tested with stratification by age,race, education, prostate volume, rebiopsy, and anxiety before biopsy. Pain was rankedaccording to the following scores: 0 (no pain), 1–3 (mild pain), 4–7 (moderate pain), and8–10 (severe pain). Statistical analysis was performed by using Kruskal-Wallis andWilcoxon two-tailed tests with a significance of 5%. Results: Pain intensity was not influenced by any risk factors. The mean VAS 1 scorewas 1.95±1.98, the mean VAS 2 score was 2.73±2.55, and the mean VAS 3 score was0.3±0.9, showing greater pain at the time of needle penetration than in other situations(VAS 2>VAS 1>VAS 3, p=0.0013, p=0.0001, respectively). Seventy-five percent of patientsreported a VAS pain scale of less than 3.1 or mild pain. Conclusions: Intravenous sedation and analgesia with midazolam and fentanyl citrateis a good method for reducing pain caused by prostate biopsy, even during probeinsertion.

        One-Anastomosis and Roux-en-Y Gastric Bypass Promote Similar Weight Loss, Patient Satisfaction, Quality of Life, Inflammation Grade, and Cellular Damage in the Esophagus and Gastric Pouch in a Short-term Follow-up

        Background: One-anastomosis gastric bypass (OAGB) is a simpler procedure than Roux-en-Y gastric bypass (RYGB); however, biliary reflux can occur and impair outcomes. This study aimed to compare outcomes of OAGB and RYGB. Methods: Twenty patients with morbid obesity were randomized prospectively into two groups: OAGB (n=10) or RYGB (n=10). Quality of life (36-item short-form health survey [SF-36]), satisfaction (Visick scale), and body mass index (BMI) were evaluated before and 6 months after the operation. All patients underwent esophagogastroduodenoscopy with gastric and esophageal mucosal biopsies at 3 and 6 months after their operation. Results: The study found no significant difference in BMI before surgery (OAGB, 43.2 kg/m2; RYGB, 43.1 kg/m2; P=0.90) or at 6 months postoperative (OAGB, 32.1 kg/m2; RYGB, 31.8 kg/m2; P=0.91). There was no significant difference in improvement of quality of life (four SF-36 domains) or satisfaction (P=0.08) between groups at 6 months. There was no statistical difference between gastric (P=0.10) and esophageal (P=0.76) inflammation grade at three or 6 months between the two groups. Conclusion: OAGB and RYGB are equally effective in terms of weight loss, patient satisfaction, and quality of life improvement at 6 months after the procedures. Inflammation grade and cellular damage in the gastric pouch and in the esophagus were similar.

      • Lived Experiences during the Enhanced Community Quarantine : Students’ Perspectives through Value Judgment Lenses

        The spread of Corona virus (COVID-19) is pandemic. As the congress enacted the Bayanihan: to Heal as One Act, officially designated as Republic Act No. 11469, the president implemented the enhanced community quarantine (ECQ) in Luzon. Due to ECQ, there is the cessation of work, business, schooling and transportation; so people just stay at home to avoid contagion. It was in this contention that the researchers conducted this study to explore and navigate the thoughts and perspectives of the 15 graduating college students about their lived experiences during the ECQ. The only instrument employed in this research was the closed-fixed interview that covered both factual and meaning-generation level. There were no predetermined questions asked, in order to remain as open and adaptable as possible to the participants’ thoughts and perspectives on the enhanced community quarantine thru their value judgment lenses. The researchers prepared interview questions; participants were asked thru phone calls and skype. Responses were analyzed and categorized to extract their thoughts and perspectives. Conclusions were drawn and future research directions were offered.
