        Bioactive glasses and electrospun composites that release cobalt to stimulate the HIF pathway for wound healing applications

        Background: Bioactive glasses are traditionally associated with bonding to bone through a hydroxycarbonate apatite (HCA) surface layer but the release of active ions is more important for bone regeneration. They are now being used to deliver ions for soft tissue applications, particularly wound healing. Cobalt is known to simulate hypoxia and provoke angiogenesis. The aim here was to develop new bioactive glass compositions designed to be scaffold materials to locally deliver pro-angiogenic cobalt ions, at a controlled rate, without forming an HCA layer, for wound healing applications. Methods: New melt-derived bioactive glass compositions were designed that had the same network connectivity (mean number of bridging covalent bonds between silica tetrahedra), and therefore similar biodegradation rate, as the original 45S5 Bioglass. The amount of magnesium and cobalt in the glass was varied, with the aim of reducing or removing calcium and phosphate from the compositions. Electrospun poly(ε-caprolactone)/bioactive glass composites were also produced. Glasses were tested for ion release in dissolution studies and their influence on Hypoxia-Inducible Factor 1-alpha (HIF-1α) and expression of Vascular Endothelial Growth Factor (VEGF) from fibroblast cells was investigated. Results: Dissolution tests showed the silica rich layer differed depending on the amount of MgO in the glass, which influenced the delivery of cobalt. The electrospun composites delivered a more sustained ion release relative to glass particles alone. Exposing fibroblasts to conditioned media from these composites did not cause a detrimental effect on metabolic activity but glasses containing cobalt did stabilise HIF-1α and provoked a significantly higher expression of VEGF (not seen in Co-free controls). Conclusions: The composite fibres containing new bioactive glass compositions delivered cobalt ions at a sustained rate, which could be mediated by the magnesium content of the glass. The dissolution products stabilised HIF-1α and provoked a significantly higher expression of VEGF, suggesting the composites activated the HIF pathway to stimulate angiogenesis.

      • Improved Decision Tree Classification (IDT) Algorithm For Social Media Data

        In this paper we used classification algorithms on social networking. We are proposing, a new classification algorithm called the improved Decision Tree (IDT). Our model provides better classification accuracy than the existing systems for classifying the social network data. Here we examined the performance of some familiar classification algorithms regarding their accuracy with our proposed algorithm. We used Support Vector Machines, Naïve Bayes, k-Nearest Neighbors, decision tree in our research and performed analyses on social media dataset. Matlab is used for performing experiments. The result shows that the proposed algorithm achieves the best results with an accuracy of 84.66%.

        Developing an Integrated Acupuncture Protocol for Treating Medial Tibial Stress Syndrome: A Delphi Consensus Study

        The present study employs the Delphi method to devise a consensus-based protocol for utilizing integrated acupuncture in treating medial tibial stress syndrome (MTSS). Twenty acupuncture experts contributed opinions across six key themes, including diagnosis, acupuncture points, additional Traditional Oriental Medicine modalities, treatment rationale, treatment duration/frequency, and integration of yoga/naturopathic therapies. Consensus, defined as a 70% agreement or higher, was reached on all themes, reflecting a collective acknowledgment of the necessity for a holistic approach to MTSS management. The final protocol includes six diagnostic criteria, six acupuncture points, one additional modality, two Traditional Oriental Medicine therapies, four treatment rationales, and six yoga/naturopathic therapies. The present comprehensive protocol offers valuable guidance for healthcare professionals seeking an integrated approach to MTSS management.

        Microstructure and Properties of Fully Aliphatic Polyimide/Mesoporous Silica Hybrid Composites

        We report the effect of the amount of the mesoporous material, SBA-15, on the basic traits of fully aliphatic polyimides (API). For this purpose, water soluble, fully aliphatic poly(amic acid) triethyl amine salts ($PAA_{(s)}$) were prepared and mixed with various amounts of SBA-15. Fully aliphatic polyimide hybrid composites containing the SBA 15-type mesoporous silica were synthesized successfully from bicyclo [2.2.2] oct-7-ene-2,3,5,6-tetracarboxylic dianhydride and alicyclic diamine, 4,4'-methylene bis(2-methylcyclohexylamine). The structure of the hybrid composites was confirmed by IR spectroscopic analysis. Scanning electron microscopy revealed the morphology of the compounds. The hybrid composites exhibited good thermal stability, reasonable transparency, and a low dielectric constant.

        Comparative Study between Swamp Buffalo and Native Cattle in Feed Digestibility and Potential Transfer of Buffalo Rumen Digesta into Cattle

        Rumen ecology plays an important role in the fermentation process and in providing end-products for ruminants. These studies were carried out to investigate variations in rumen factors namely pH, $NH_3-N$ and microorganisms in cattle and swamp buffaloes. Furthermore, studies on diurnal patterns of rumen fermentation and the effect of rumen digesta transfer from buffalo to cattle was conducted. Based on these studies, diurnal fermentation patterns in both cattle and buffaloes were revealed. It was found that rumen NH3-N was a major limiting factor. Rumen digesta transfer from buffalo to cattle from buffalo to cattle was achievable. Monitoring rumen digesta for 14d after transfer showed an improved rumen ecology in cattle as compared to that of original cattle and buffalo. It is probable that buffalo rumen digesta could be transferred. However, further research should be undertaken in these regards in order to improve rumen ecology especially for buffalo-based rumen.

        Artificial diets determine fatty acid composition in edible Ruspolia differens (Orthoptera: Tettigoniidae)

        There are increasing interests in rearing edible insects in Africa, but information on how the feeds modify their fatty acids is largely lacking. In this work, the influence of artificial diets on the fatty acid contents and composition in the edible Ruspolia differens (Serville, 1838), in Uganda was assessed. R. differens was reared on the mixtures of six gradually diversified diets of two, three, four, six, eight and nine feeds. The diets were formulated from rice seed head, finger millet seed head, wheat bran, superfeed chicken egg booster, sorghum seed head, germinated finger millet, simsim cake, crushed dog biscuit pellet and shea butter. Fatty acid methyl esters were prepared using direct transesterification method, and analysed using gas chromatography. The contents of saturated, monounsaturated and polyunsaturated fatty acid differed significantly among the diets. The more diverse diets resulted in increased content of the polyunsaturated fatty acids. The n6:n3 ratio differed significantly among the diets and between the sexes, with R. differens fed on the four-feed diet having a higher n6:n3 ratio than those fed on other diets. Also, the fatty acid composition differed significantly among the diets, and diet diversification corresponded with the proportions of polyunsaturated fatty acids, especially linoleic acid. Overall, our results demonstrate that higher levels of essential fatty acids can be achieved by rearing R. differens on highly diversified diets. These findings are important in informing the design of future mass-rearing program for this edible insect.

      • Platelet Derived Growth Factor-B and Human Epidermal Growth Factor Receptor-2 Polymorphisms in Gall Bladder Cancer

        Gall bladder cancer (GBC) is a gastro-intestinal cancer with high prevalence among north Indian women. Platelet derived growth factor-B (PDGFB) and human epidermal growth factor receptor-2 (HER2) may play roles in the etiology of GBC through the inflammation-hyperplasia-dysplasia-carcinoma pathway. To study the association of PDGFB and HER2 polymorphisms with risk of GBC, 200 cases and 300 controls were considered. PDGFB +286A>G and +1135A>C polymorphisms were investigated with an amplification refractory mutation system and the HER2 $Ile^{655}Val$ polymorphism by restriction fragment length polymorphism. Significant risk associations for PDGFB +286 GG (OR=5.25) and PDGFB +1135 CC (OR=3.19) genotypes were observed for GBC. Gender wise stratification revealed susceptibility for recessive models of PDGFB +1135A>C (OR=3.00) and HER2 $Ile^{655}Val$ (OR=2.52) polymorphisms among female GBC cases. GBC cases with gall stones were predisposed to homozygous +286 GG and +1135 CC genotypes. Significant risk associations were found for ACIle (OR=1.48), GAVal (OR=1.70), GAIle (OR=2.00) haplotypes with GBC cases and GCIle haplotype with female GBC cases (OR=10.37, P=<0.0001). Pair-wise linkage disequilibrium revealed negative associations among variant alleles. On multi-dimensional reduction analysis, a three factor model revealed significant gene-gene interaction for PDGFB +286A>G, PDGFB +1135A>C and HER2 Ile165Val SNPs with GBC. Protein-protein interaction showed significant association of PDGFB and HER2 with the epidermal growth factor receptor signaling pathway.

        The fatty acid composition of edible grasshopper Ruspolia differens (Serville) (Orthoptera: Tettigoniidae) feeding on diversifying diets of host plants

        Ruspolia differens (Serville) (Orthoptera: Tettigoniidae) is a highly valued edible grasshopper species in Africa. However, the effects of plant diets on lipid content and fatty acid composition of R. differens are not well understood. We tested the effects of four diets on the total lipid content and fatty acid composition of R. differens. Sixth instar nymphs of R. differens were reared on one, and mixtures of two, three, and six natural plant inflorescences. Individuals collected from the field constituted a control treatment. We extracted lipids and analyzed the fatty acid methyl esters using gas chromatography–mass spectrometry. We analyzed if the total lipid content, body weight, and fatty acid composition differed among diets and between the sexes using two‐way ANOVAs and a PERMANOVA model, respectively. The total lipid content and weight of R. differens did not differ among the diets. The nine common fatty acids were palmitic (mean across treatments, 26%), oleic (22%), palmitoleic (18%), linoleic (13%), stearic (7%), myristic (6%), myristoleic (4%), α‐linolenic (2%) and arachidic acid (1%). The composition of fatty acids and the proportion of essential fatty acids significantly differed among the diets. The proportion of essential fatty acids was highest in the control treatment (21%) but low in less diversified (one to three feed) diets (12–13%). This study demonstrates that the fatty acid composition in R. differens can be influenced through diet. Thus, with dietary manipulations, using local plants in Africa, it is possible to produce R. differens with preferred high quality essential fatty acids for human consumption.

      • Risk of Cardiovascular and Cerebrovascular Events in Hepatitis C Patients Following Completion of Direct-Acting Antiviral Therapy: A Retrospective Cohort Study

        Aims: Chronic hepatitis C virus (HCV) infection has been associated with adverse cardiovascular and cerebrovascular outcomes. Viral eradication with direct-acting antiviral (DAA) therapy may decrease the risk of these events among HCV patients. We aimed to characterize the risk of cardiovascular and cerebrovascular events among HCV patients treated with DAA regimens compared to untreated patients in US claims data. Methods: 322,276 adults with chronic HCV were enrolled in the database between January 2006 and September 2015. We identified 8,342 HCV patients dispensed at least 12 weeks of DAA therapy (excluding boceprevir and telaprevir) and, for comparison, 76,423 untreated HCV patients who had follow-up time in the DAA era. Events were identified by diagnostic claims for acute and chronic ischemic heart disease, angina pectoris, cardiomyopathy, heart failure, subarachnoid hemorrhage, occlusion and stenosis of precerebral or cerebral arteries, cerebrovascular disease, cerebral atherosclerosis, intracerebral hemorrhage, and transient cerebral ischemia. Hazard ratios (HRs) estimating risk of incident cardiovascular and cerebrovascular events associated with completion of DAA therapy were calculated with adjustment for covariates using Cox proportional hazards methods. Results: HCV patients dispensed a full course of DAA therapy were more likely to be male, over 55 years-old, with baseline diagnoses of cirrhosis, diabetes, or hypertension, and on cardiovascular medications. After adjustment for covariates, there was a reduced risk of total cardiovascular and cerebrovascular events in patients completing DAA therapy compared to untreated patients (HR=0.86, 95% confidence interval [CI]: 0.74-0.99). Adjusted HRs were similar for cardiovascular disease (HR=0.90, 95%CI: 0.76-1.05) and cerebrovascular disease (HR=0.89, 95%CI: 0.74-1.08). Conclusions: In this real-world cohort, DAA therapy appeared to reduce the risk of cardiovascular and cerebrovascular events in HCV patients even within a short period following therapy. The benefits of curative DAA therapy in reducing extrahepatic complications of HCV may be even greater with longer follow-up.
