        Gelatin methacryloyl (GelMA) loaded with concentrated hypoxic pretreated adipose-derived mesenchymal stem cells(ADSCs) conditioned medium promotes wound healing and vascular regeneration in aged skin

        Aging skin is characterized by a disturbed structure and lack of blood supply, which makes it difficult to heal once injured. ADSCs secrete large amounts of cytokines, which promote wound healing and vascular regeneration through paracrine secretion, and the number of cytokines can be elevated by hypoxic pretreating. However, the components of ADSCs are difficult to retain in wounds. Gelatin methacrylate (GelMA) is a photopolymerizable hydrogel synthesized from gelatin and has recently emerged as a potentially attractive material for tissue engineering applications. GelMA loaded with concentrated hypoxic pretreated ADSCs conditioned medium could provide a new method of treating wounds in aged skin.Primary ADSCs were isolated from human adipose tissue and characterized by flow cytometry and differentiation test. ADSCs in passages 4-6 were pretreated in the hypoxic and normoxic environments to collect conditioned medium, the conditioned medium was then concentrated to prepare concentrated ADSCs conditioned medium(cADSC-CM)(the one collected from ADSCs under hypoxia was called hypo-CM ,and the one from normoxia was called nor-CM). The concentration of cytokines was detected. After treated with cADSC-CM, the abilities of proliferation, migration, and tube formation of human umbilical vascular endothelial cells (HUVECs) were assayed, and Akt/mTOR and MAPK signal pathway was detected using western blotting. GelMA+hypo-CM hydrogel was prepared, and a comprehensive evaluation of morphology, protein release efficiency, degradation rate, mechanical properties, and rheology properties were performed. Full-thickness skin wounds were created on the backs of 20-month-old mice. After surgery, GelMA, GelMA+F12, GelMA+hypo-CM, and GelMA+nor-CM were applied to the wound surface respectively. H&E, Masson, and immunohistochemistry staining were performed, and a laser Doppler perfusion imager was used to evaluate the blood perfusion. The student’s t-test was used for analysis between two groups and a one-way analysis of variance (ANOVA) was used for analysis among multi groups.Our results revealed that 1) wounds in aged skin healed more slowly than that in young skin and exhibited poorer perfusion; 2) hypoxic pretreated ADSCs secreted more cytokines including VEGF by activating HIF1α; 3) hypo-CM promoted proliferation and migration of HUVECs through VEGF/Akt/mTOR and MAPK signal pathway; 4) GelMA-hypoCM accelerated wound healing and angiogenesis in aged skin in vivo.GelMA loaded with concentrated hypoxic pretreated adipose-derived mesenchymal stem cells conditioned medium could accelerate wound healing in aged skin by promoting angiogenesis.

        Effects of temperature on the local fracture toughness behavior of Chinese SA508-III welded joint

        The structural integrity of welded joints in the reactor pressure vessel (RPV) is directly related to the safety of nuclear power plants. The RPV is made from SA508-III steel in a pressurized water reactor. In this study, we investigated the effects of temperature on the tensile and fracture toughness properties of Chinese SA508-III welded joint in different sampling areas in order to provide reference data for structural integrity assessments of RPVs. The specimens used in tensile and fracture toughness tests were fabricated from the base metal (BM), weld metal (WM), and the heat-affected zone (HAZ) in the welded joint. The representative testing temperatures included the ambient temperature (20 C), upper shelf temperature (100 C), and service temperature (320 C). The results showed that temperature greatly affected the fracture toughness (JIC) values for the SA508-III welded joint. The JIC values for BM and HAZ both decreased remarkably from 20 C to 320 C. The fracture morphologies showed that the BM and HAZ in the welded joint exhibited fully ductile fracture at 20 C, whereas partial cleavage fracture was mixed in ductile fracture mode at 100 C and 320 C. The WM exhibited the ductile and cleavage fracture mixed mode at various temperatures, and the JIC values showed slight changes

        Preparation of Co<sub>3</sub>O<sub>4</sub>/NF Anode for Lithium-ion Batteries

        Due to its characteristics of light weight, high energy density, good safety, long service life, no memory effect, and environmental friendliness, lithium-ion batteries (LIBs) are widely used in various portable electronic products. The capacity and performance of LIBs largely depend on the performance of electrode materials. Therefore, the development of better positive and negative materials is the focus of current research. The application of metal organic framework materials (MOFs) derivatives in energy storage has attracted much attention and research. Using MOFs as precursors, porous metal oxides and porous carbon materials with controllable structure can be obtained. In this paper, rod-shaped Co-MOF-74 was grown on Ni Foam (NF) by hydrothermal method, and then Co-MOF-74/NF precursor was heat-treated to obtain rodshaped Co<sub>3</sub>O<sub>4</sub>/NF. Ni Foam was skeleton structured, which effectively relieved. The change of internal stress changes and destroys the structural volume of the electrode material and reduces the capacity attenuation. Co<sub>3</sub>O<sub>4</sub>/NF composite material has a specific discharge capacity of up to 1858 mA h/g for the first time, and a reversible capacity of up to 902.4 mA h/g at a current density of 200 mA/g, and has excellent rate and impedance performance. The synthesis strategy reported in this article opens the way to design high-performance electrodes for energy storage and electrochemical catalysis.

        Self-supervised feature matched virtual try-on

        Virtual try-on is a technology that enables users to preview the effect of wearing a target garment without wearing the actual garment. However, existing image-based virtual try-on methods often require additional human parsing or segmentation operations to generate intermediate representations required for garment deformation and texture fusion. These operations not only increase the computational complexity and memory consumption, but also limit the real-time and portability of virtual try-on. Additionally, inaccurate parsing results can lead to misleading final generated images. To overcome these challenges, we propose a self-supervised feature matched virtual try-on network, which can directly generate high-quality try-on results from human body images and target clothing images without any additional input. Specifically, we design an optical flow warp module, which focuses on the optical flow changes between the person image and the clothing image to achieve accurate clothing alignment and deformation. Furthermore, a feature refine warp module is designed to enhance the features of the extracted optical flow information and the original character segmentation and analysis operations, reducing the influence of background clutter features on the content, and ensuring that the wrinkles and deformation of the replacement clothes are close to the original clothes. The feature match module is developed to calculate the feature matching loss of the converted clothing and the generated results of the teacher network and the student network, and the corresponding knowledge is distilled and passed to the student network to assist in self-supervised training. We conduct experiments on the VITON dataset and show that our model can generate high quality and high resolution, and our proposed method outperforms the state-of-the-art virtual try-on methods both qualitatively and quantitatively.

        Effect of drying temperature on the sugars, organic acids, limonoids, phenolics, and antioxidant capacities of lemon slices

        Changes in contents of sugars, organic acids, limonoids, phenolics contents, and antioxidant capacities of lemon slices dried at different temperatures were evaluated. Air drying (AD) promoted losses of sugars, citric acid, ascorbic acid, extractable phenolics (EPs), and nonextractable phenolics (NEPs), while it introduced an increase in limonoids. Phenolics of lemon were mainly presented in their extractable form. Hesperidin and eriocitrin were the main EPs; protocatechuic acid and poncirin were the predominant NEPs. The decrease in extractable phenolic acid, EP, and NEP content in lemon is lower at low drying temperatures, while the increase in non-extractable phenolic acid content is higher at high drying temperatures. The antioxidant capacity of EP was higher than that of NEP. Phenolics contributed to antioxidant capacities of lemon slices, and flavonoids were the main contributors among phenolics. Considering limonoids contents and the high levels of EP, NEP, and antioxidant capacities, AD at 60 C could be an appreciate treatment for dehydrating lemon slices.

        L-lysine and L-arginine inhibit the oxidation of lipids and proteins of emulsion sausage by chelating iron ion and scavenging radical

        Objective: To evaluate the effects of L-lysine (Lys)/L-arginine (Arg) on lipid and protein oxidation of emulsion sausage during storage and its possible mechanism. Methods: Four samples were prepared based on the presence or absence of additional sodium isoascorbate, Lys, or Arg: sample A (control), sample B (0.05 g of sodium isoascorbate), sample C (0.4 g of Lys), and sample D (0.4 g of Arg). Peroxide value (POV), thiobarbituric reactive substances (TBARS), protein carbonyls and thiols were measured. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical-scavenging, ferrous ion-chelating ability were also measured. Results: Compared with the control, the sample treated with sodium isoascorbate, Lys or Arg had significantly lower POV during the initial 20 days, TBARS during the initial 15 days. Protein carbonyls were significantly lower compared Sample B, C, and D with A during the later storage (10 to 25 days); basically, protein thiols became lower during storage when the samples were treated with sodium isoascorbate, Lys, or Arg. Both Lys and Arg had weak reducing power but strong ferrous ion-chelating activity and DPPH radical- and hydroxyl radical-scavenging activity. Conclusion: Both Lys and Arg effectively inhibited the oxidation of lipids and proteins in emulsion sausage by scavenging free radicals and chelating ferrous ions. The results obtained may be favorable for the prevention of lipid and protein oxidation during processing and storage of meat products.

        Patient preferences and shared decision making for the traditional Chinese medicine treatment of lung cancer: A discrete choice experiment study

        Background: Traditional Chinese Medicine (TCM), integrating patient preferences into decision-making process, has been widely used in the multimodality therapy of lung cancer. This study aimed to estimate patient preferences for treatment and shared decision-making (SDM) modes concerning inpatient TCM treatment of lung cancer in Shanghai in order to provide a basis for clinical decision-making process on TCM therapy for lung cancer. Methods: This study was conducted among patients (n = 347) from nine tertiary hospitals in Shanghai by discrete-choice experiment (DCE) survey and Shared Decision-Making Questionnaire-patient version (SDM-Q-9) survey. The DCE was developed with the inclusion of the most relevant attributes at appropriate levels for the TCM treatment of lung cancer. The empirical data analyses of patients were performed using mixed logit models. Additionally, subgroup analysis was conducted. Results: In total, 347 respondents completed the questionnaire. All attributes’ coefficients were statistically significant for patients’ preferences. Patients showed strong preferences for increasing disease control rate, relieving nausea and vomiting, reducing the risk of side effects, and were concerned about monthly out-of-pocket costs. Subgroup analysis indicated that patients with a lower SDM-Q-9 score and those who were satisfied with medical services emphasized more importance of higher disease control rate. Furthermore, most of the patients (90.20%) self-reported a high willingness to use SDM during the decision-making process. Conclusion: In Shanghai, patients mainly preferred SDM and considered disease control rate as the most essential attribute in the TCM treatment of lung cancer. The study findings could underscore the importance of considering patients’ preferences and promote SDM.

        Transcriptomic analysis reveals the molecular mechanism of apoptosis induced by Muscovy duck reovirus

        This study was to investigate the molecular mechanism underlying the apoptosis induced by Muscovy duck reovirus (MDRV) through a transcriptomic analysis. Muscovy ducklings were infected with MDRV strain YB and the apoptotic cells in their livers were examined with terminal-deoxynucleotidyl-transferase-mediated nick end labeling and flow cytometry. Genes differentially expressed in the livers of the MDRV-infected ducklings were screened by comparing them with those of uninfected ducklings and were analyzed with a transcriptomic method to illuminate the mechanism of MDRV infection. The results showed that MDRV infection strongly induced apoptotic cells in liver. Significant pathway enrichment determined by a Kyoto Encyclopedia of Genes and Genomes analysis showed that MDRV activated the death receptor family signaling pathway (Fas, TNFR1), the interleukin receptor signaling pathway (IL1, IL3), the phosphatidylinositol 3-kinase signaling pathway, NF-ҝB signaling pathway and calcium ions signaling pathway to induce apoptosis. This was verified by SYBR-Green-based fluorescence quantitative PCR. In conclusion, MDRV induce apoptosis by activation multi signaling pathways.
