        Two Maternal Lineages Revealed by Mitochondrial DNA D-loop Sequences in Chinese Native Water Buffaloes (Bubalus bubalis)

        Little is known about the origin and genetic diversity of swamp buffaloes in China. To obtain more knowledge on genetics of the water buffalo in China, the complete mitochondrial D-loop sequences of 30 samples from 6 native types were investigated. The results revealed 12 mitochondrial haplotypes with 50 polymorphic sites. Among these polymorphic sites, there were 49 transitions and 1 transversion. The average nucleotide diversity and haplotype diversity estimated from mtDNA D-loop region in 6 Chinese water buffalo types were 0.00684 and 0.798, respectively, showing rather abundant mitochondrial genetic diversity. The Neighbor-Joining (NJ) tree of mtDNA of Chinese water buffaloes was constructed according to the 12 haplotypes. The NJ tree indicated two lineages being designated lineage A and lineage B, in which lineage A was predominant, and lineage B was at low frequency. The new lineage B was first discovered and defined in 6 Chinese water buffalo types. These results showed that two different maternal lineages were involved in the origin of domestic swamp buffaloes in China and the lineage B was probably an introgression from Southeast Asian buffaloes.

        Aerodynamic performance of a nonplanar multi-rotor air-craft in hover

        Experimental tests and numerical simulations were typically presented with a rotor spacing of 1.2 times the diameter to investigate the effect of disc angle (DA) on the hover performance of non-planar multi-rotor unmanned aerial vehicles (UAVs). Torque, thrust coefficient, power coefficient, and power loading measurements were performed to evaluate the performance of the vehicles with different rotor rational speeds and DAs. The hovering performance exhibits a maximum improvement of 10.3 % when DA = 50° compared with the traditional planar muti-rotor system. The flow fields were also simulated by unsteady Reynoldsaveraged Navier-Stokes solver. The downwash shifts, flow contractions, pressure difference enlargements, and wake inclinations were discovered and contributed to the interference reduction with the increasing DA. Based on these observations, the tilt-rotor configuration has a noticeable enhancement on the hovering performance of UAVs with proper DA. The insights gained can be applied as guidance for further UAV design.

        The present study was designed to isolate and characterize novel chemical constituents of the stem bark of Juglans mandshurica Maxim. (Juglandaceae).The chemical constituents were isolated and purified by various chromatographic techniques. The structures of the compounds were elucidated on the basis of spectral data (1D and 2D NMR, HR-ESI-MS, CD, UV, and IR) and by the comparisons of spectroscopic data with the reported values in the literatures. Two long chain polyunsaturated fatty acids (1 and 2) were obtained and identified as (S)-(8E, 10E)-12-hydroxy-7-oxo-8, 10-octadecadienoic acid (1) and (S)-(8E, 10E)-12-hydroxy-7-oxo-8,10-octadecadienoic acid methyl ester (2). To the best of our knowledge, this is the first report on the isolation and structural elucidation of the two new conjugated ketonic fatty acids from this genus.

        Simulation Research of Multi-morphological Microstructures Chaining Mechanism of Nano-magnetic Fluid

        For nano-magnetic fluid(NMF), the magnetic nano-particle(MNP) kinematic behavior in a magnetic field and the chain-like microstructure evolution are important to study the NMF properties at the microscopic level. However, the chain structure formation mechanism and the effects of multiple environmental factors on microstructural morphology remain unclear. In this paper, the interaction of a two-particle system, chain structure formation process, microstructure evolution and response time simulation of NMF is investigated by the discrete element simulation method. The results show that the magnetic dipole and repulsive forces dominate the chain structure formation and evolution under a uniform magnetic field. MNPs assemble into chain-like structures and various complex structures along the magnetic f ield direction. The volume fraction, magnetization intensity and particle size significantly affect the microstructure and the response time. The above study can obtain the specific morphology of the microstructure at different working conditions and broaden the application of NMF in practical engineering.

      • Smad4 Expression in Hepatocellular Carcinoma Differs by Hepatitis Status

        Aims: Primary hepatocellular carcinoma (HCC) is a common malignancy often related to hepatitis viral infection. Smad4 is known to mediate the TGF-${\beta}$ pathway to suppress tumorigenesis. However, the function of Smad4 in HCC is still controversial. In this study we compared levels of Smad4 in HCC tissues with or without hepatitis virus infection and adjacent normal-appearing liver. Methods: Samples from HCC patients were analyzed for Smad4 protein and mRNA expression by immunohistochemistry (IHC), RT-PCR and Western blotting. Results: We found that tumor tissues expressed less Smad4 mRNA and protein than the adjacent tissues. Most HCC tumor tissues were negative for Smad4 in IHC staining, while the majority of adjacent tissues were positively stained. Interestingly, protein levels were higher in HCC tissues with viral hepatitis than those without virus infection. Suppression of expression appeared closely related to HCC, so that Smad4 appears to function as a tumor suppressor gene (TSG). Conclusion: Patients with hepatitis viral infection, at higher risk for HCC, exhibited increased Smad4 protein expression suggesting hepatitis virus may modulate Smad4 expression, which is functionally distinct from its putative role as a TSG. Smad4 expression may thus be an applicable marker for diagnosis and/or a target to develop therapeutic agents for HCC.

      • An Updated Pooled Analysis of Glutathione S-transferase Genotype Polymorphisms and Risk of Adult Gliomas

        Objective: Glutathione S-transferases (GSTs) are multifunctional enzymes that play a crucial role in the detoxification of both the endogenous products of oxidative stress and exogenous carcinogens. Recent studies investigating the association between genetic polymorphisms in GSTs and the risk of adult brain tumors have reported conflicting results. The rationale of this pooled analysis was to determine whether the presence of a GST variant increases adult glioma susceptibility by combining data from multiple studies. Methods: In our meta-analysis, 12 studies were identified by a search of the MEDLINE, HIGHWIRE, SCIENCEDIRECT and EMBASE databases. Of those 12, 11 evaluated GSTM1, nine evaluated GSTT1 and seven evaluated GSTP1 Ile105Val. Between-study heterogeneity was assessed using ${\chi}^2$-based Q statistic and the $I^2$ statistic. Crude odds ratios (ORs) with corresponding 95% confidence intervals (CIs) were used to estimate the association between GSTM1, GSTT1 and GSTP1 polymorphisms and the risk of adult gliomas. Results: The quantitative synthesis showed no significant evidence to indicate an association exists between the presence of a GSTM1, GSTT1 or GSTP1 Ile105Val haplotype polymorphism and the risk of adult gliomas (OR, 1.008, 1.246, 1.061 respectively; 95% CI, 0.901-1.129, 0.963-1.611, 0.653-1.724 respectively). Conclusions: Overall, this study did not suggest any strong relationship between GST variants or related enzyme polymorphisms and an increased risk of adult gliomas. Some caveats include absence of specific raw information on ethnic groups or smoking history on glioma cases in published articles; therefore, well-designed studies with a clear stratified analysis on potential confounding factors are needed to confirm these results.

        Overexpression of a Phosphatidylinositol-Specific Phospholipase C Gene from Populus simonii ×  P. nigra Improves Salt Tolerance in Transgenic Tobacco

        Phosphatidylinositol-specific phospholipase C (PI-PLC) catalyzes the hydrolysis of phosphatidylinositol-4,5-bisphosphate into diacylglycerol and inositol 1,4,5-trisphosphate. It can play an essential role in plant stress response and signaling. However, the functions of PLCs remain unclear in wooden plants. This study carried out the bioinformatics analysis of the PLC gene family in poplar. The expression pattern analysis suggested the transient up-regulation of PsnPLC in salt stress conditions. The transgenic tobacco plants overexpressing PsnPLC were generated by the Agrobacterium-mediated leaf disc method. The transgenic lines showed a significant increase in plant height, SOD and POD activity, and proline content under salt stress. By contrast, transgenic lines showed a decreased malondialdehyde content under salt stress. Comparative transcriptome analysis indicated that the overexpression of PsnPLC mainly affects membrane-associated GO terms and signaling pathways, and the differential expression of salt-responsive genes may contribute to the enhanced salt tolerance. Overall, the results indicated that PsnPLC plays an essential role in modulating salt tolerance in transgenic tobacco.

        Two new (1 and 3) two known diarylhep-tanoids (2 and 4), along with two tetralones (5 and 6), one naphthoquinone (7), four phenylpropanoids (8-11), and phenol (12) were isolated from the leaves of Juglans mandshurica. Their structures were elucidated on the basis of spectral and chemical data. Compounds 2 and 10 are firstly isolated from this plant and 8 and 12 were isolated from the Juglans genus for the first time. Among these compounds, only 7 cxhibitcd moderate cytotoxicities against cultured Mgc-803, A549, K562, and Hela tumor cell lines with IC50 values of 25.90, 28.60, 39.06, 44.90 LLM, respectively.

        Renewable low-cost brassica rapa subsp. Extract for protection of Q235 steel in H2SO4 medium: Experimental and modeling studies

        The research aims to explore the corrosion protection performance and mechanism of the Brassica rapasubsp. extract (BRE) considered as a new environmental protection corrosion inhibitor. In this research,the inhibition performance of BRE toward Q235 steel in H2SO4 solution was evaluated through combiningpotentiodynamic polarization and electrochemical impedance. The results showed that the BRE moleculescould form a dense protective film on steel surface, effectively preventing from the attack of corrosiveions. Specifically, the inhibition efficiency brought by 200 mg/L of BRE at each temperature exceeded93 %. The results of Fourier transform infrared spectroscopy (FTIR) and UV–vis spectroscopy showed thatthe BRE molecules was well adsorbed on the steel surface. In the meantime, the results of N-Fe bond of Xrayphotoelectron spectroscopy (XPS) indicated that the nitrogen-atom in BRE molecules worked as theactive site to form multiple anchoring effects in Q235 steel. Calculations based on density functional theory(DFT) and molecular dynamics (MD) simulations have theoretically revealed the adsorption mechanismof BRE on the surface of Q235 steel at the molecular/atomic level.
