        Radicicol Inhibits iNOS Expression in Cytokine-Stimulated Pancreatic Beta Cells

        Here, we show that radicicol, a fungal antibiotic, resulted in marked inhibition of inducible nitric oxide synthase (iNOS) transcription by the pancreatic beta cell line MIN6N8a in response to cytokine mixture (CM: TNF-${\alpha}$, IFN-${\gamma}$, and IL-$1{\beta}$). Treatment of MIN6N8a cells with radicicol inhibited CM-stimulated activation of NF-${\kappa}B$/Rel, which plays a critical role in iNOS transcription, in a dose-related manner. Nitrite production in the presence of PD98059, a specific inhibitor of the extracellular signal-regulated protein kinase-1 and 2 (ERK1/2) pathway, was dramatically diminished, suggesting that the ERK1/2 pathway is involved in CM-induced iNOS expression. In contrast, SB203580, a specific inhibitor of p38, had no effect on nitrite generation. Collectively, this series of experiments indicates that radicicol inhibits iNOS gene expression by blocking ERK1/2 signaling. Due to the critical role that NO release plays in mediating destruction of pancreatic beta cells, the inhibitory effects of radicicol on iNOS expression suggest that radicicol may represent a useful anti-diabetic activity.

        이매방 살풀이춤의 ‘여형화(女形化)’에 대한 논의

        우리는 이매방의 춤을 이야기할 때 ‘아름답다’, ‘교태적이다’, ‘농염하다’라고 표현한다. 이러한 수사(修辭)는 그의 춤이 다분히 여성적인 속성을 갖고 있기 때문에 붙여진 것이다. 본 연구는 이매방의 살풀이춤이 여형화의 양식체계로 구축되어 있음을 논의하였다. 춤에서 ‘여형화’란 남성이 여성의 모습으로 춤을 추는 형식, 혹은 양식이라고 할수 있다. 이매방의 춤은 다음과 같은 여형화의 특성을 보인다. 첫째, 이매방의 살풀이춤은 경극의 ‘화단’이나 가부키의 ‘게이세이’와 같이 화려하고 교태스런 배역 특성을 보인다. 둘째, 살풀이춤의 복식은 남성과 여성의 구분을 두고 있으되 상당히 장식적인 여성성을 지향하고 있다. 셋째, 이매방의 양성 지향적인 성 정체성은 탐미적인 미의식으로 확장되어 여형화 된 춤 양식 체계로 환원되었다. 넷째, 이매방 살풀이춤의 기법은 가공된 여성의 신체상을 기반으로 구축되었다. 즉, 응축된 근육의 밀도 높은 작용을 통해 신체를 ‘축소’하거나 ‘왜곡’하여 관능적이고 요염한 여성성을 신체상으로 구현하여 춤에 반영하였다. 이상으로 미루어 이매방 살풀이춤은 남성이 추는 여성의 춤, 혹은 기방춤을 전형 특성의 하나로 구축하고 있다고 하겠다. When we talk about Lee Mae-bang's Salpul-i chum, we use the expressions ‘beautiful’, ‘exciting’, and ‘congested’. This knowledge was attached because his dance has a feminine attribute. This study discussed that Lee Mae-bang's Salpul-i chum is constructed as Yeohyeonghwa's aquaculture system. First, Lee Mae-bang's Salpul-i chum exhibits gorgeous and cunning character, such as Peking Opera's 'Huā dàn' or Kabuki's 'Keisei'. In dance[chum], ‘Yeohyeonghwa’ can be said to be a form or style in which men dance in the form of women. Lee Mae-bang's dance shows the following characteristics of Yeohyeonghwa. Second, Salpul-i chum's costumes differentiate between men and women, but aim for a highly decorative femininity. Third, Lee Mae-bang's bisexual sexual identity has been stylized into a feminine attitude through aesthetic pursuit. Fourth, Lee Mae-bang Salpul-i chum's technique expresses the processed female body image. In other words, Salpul-i chum expresses the body of a sensual and fascinating woman by 'shrinking' or 'distorting' the body through the dense action of condensed muscles. In this regard, Lee Mae-bang is said to be constructing a male dance or gibangchum as one of the characteristics of Jeonhyeong[original form].

        Rapid HPLC determination of gastrodin in Gastrodiae Rhizoma

        Gastrodin is a major biologically active ingredient in members of the genus Gastrodia. For this reason, there are many reports on the quantification of gastrodin in Gastrodiae Rhizoma (GR) and in GR-containing herbal preparations. HPLC, HPLC-MS, and TLC are the major approaches for gastrodin quantification; however, they usually require complicated pre-treatment, lengthy analysis, and expensive instruments. Therefore, a rapid and reliable method for determining gastrodin in GR is necessary. Optimal HPLC separation was achieved using a Chromolith Performance RP-18e ($4.8{\times}100mm$, $5{\mu}m$) stationary phase. The optimal mobile phase was a mixture of water (A) and acetonitrile (B) with a gradient of 1 % B at 0-8 min, 20 % B at 8-10 min, 80-100 % B at 10-12 min, and 100 % B at 12-13 min, followed by equilibration with 1 % B for 2 min at a flow rate of 1.5 mL/min. The detection wavelength was UV220 nm. Gastrodin appeared within 4 min under the above conditions. The calibration curve of gastrodin showed good linearity in the 0.25-10.0 range ($r^2=0.9998$). The limit of detection ($0.13{\mu}g$), limit of quantification ($0.25{\mu}g$), and reproducibilities of area and retention time (0.04 and 3.24 %, respectively) were within acceptable ranges. In addition, the intra-day precision and accuracy of gastrodin were 0.74 and $100.63{\pm}0.04%$, respectively, while the inter-day precision and accuracy were 0.06 and $99.25{\pm}0.05%$, respectively. The range of mean gastrodin content in six GR samples, which were cultivated at different sites, was 0.37-0.79 %. This result may be a guideline for the quality control of GR and GR-containing medicinal preparations.

        Development of New Method for Antioxidant Capacity with ORP-pH System

        Many methods are used in the measurement of antioxidative capacity. These meth­ods require very complex procedures and pretreatment. Our suggestions for research will be simple and accurate methods for obtaining many kinds of samples, especially colored samples such as natural product extracts for measuring antioxidative capacities. For oxidation-reduction potential (ORP) system value, we examined the relationships between the ORP-pH system and the ORAC, FRAP methods. To evaluate ORP System value, we calculated the absolute slope/intercept from the linear regression of each standard material at different concentrations and ORP-pH system, and compared the correlations with ORAC and FRAP values.

        A Cluster-Based Energy-Efficient Routing Protocol without Location Information for Sensor Networks

        With the recent advances in Micro Electro Mechanical System (MEMS) technology, low cost and low power consumption wireless micro sensor nodes have become available. However, energy-efficient routing is one of the most important key technologies in wireless sensor networks as sensor nodes are highly energy-constrained. Therefore, many researchers have proposed routing protocols for sensor networks, especially cluster-based routing protocols, which have many advantages such as reduced control messages, bandwidth re-usability, and improved power control. Some protocols use information on the locations of sensor nodes to construct clusters efficiently. However, it is rare that all sensor nodes know their positions. In this article, we propose another cluster-based routing protocol for sensor networks. This protocol does not use information concerning the locations of sensor nodes, but uses the remaining energy of sensor networks and the desirable number of cluster heads according to the circumstances of the sensor networks. From performance simulation, we found that the proposed protocol shows better performance than the low-energy adaptive clustering hierarchy (LEACH).

      • Cooperative Activity of Subunits of Human Ferritin Heteropolymers in Escherichia coli

        We constructed a comparative expression system in order to produce recombinant human ferritin homo- and heteropolymers in Escherichia coli. Human ferritin H-(hfH) and L-chain (hfL) genes were expressed without amino acid changes under the control of a tac promoter. Ferritin heteropolymers of varying subunit composition were also produced by combining two different expression systems, a bicistronic expression system and a coplasmid expression system. As a result, recombinant H-chain ferritin and ferritin heteropolymers were catalytically active in forming iron core in vivo. In particular, the ferritin heteropolymer that is composed of 7% H-subunit and 93% L-subunit was capable of forming an iron core of the protein, while the L-chain ferritin homopolymer was inactive in vivo. This result indicates that the two H-subunits (i.e., 7% H-subunit content) are important to keep ferritin active in the cells. In addition, human ferritins were identified as the major iron binding proteins in the transformed cells. Also, the amount of iron bound to the recombinant ferritins was proportional to the H-subunit content in ferritin heteropolymers in vivo.

        Quantitative Analysis of Biogenic Amines in Raw and Processed Foods of Animal Origin on Korean Domestic Market

        This study was performed to examine the levels of biogenic amines (BAs) in foods of animal origin such as egg, ham, sausage, milk, cheese and yoghurt distributed on Korean domestic markets, and to compare the results with those of western countries. Egg yolk had more BAs than egg white. BAs detected in ham samples were the highest level in barbecued tender loin ham. Sausage samples had less BAs than ham samples. However, the delicatessen sausages had significantly higher levels of histamine than the other sausage samples (p<0.001). Cadaverine, spermidine and spermine were detected in market milks and their concentrations were very low compared with other samples. In Cheddar cheese, tyramine (44.46${\pm}$0.83 ${\mu}$g/g) was the major BA. The major BA of stirred yoghurt samples was histamine. These results suggest that BAs of the foods of animal origin distributed on Korean domestic markets were not much different from those of western countries and would not cause any harmful effect to consumers.

        c-Jun N-terminal Kinase Contributes to Norepinephrine-Induced Contraction Through Phosphorylation of Caldesmon in Rat Aortic Smooth Muscle

        Vascular smooth muscle contraction is mediated by activation of extracellular signal-regulated kinase (ERK) 1/2, an isoform of mitogen-activated protein kinase (MAPK). However, the role of stress-activated protein kinase/c-Jun N-terminal kinase (JNK) in vascular smooth muscle contraction has not been defined. We investigated the role of JNK in the contractile response to norepinephrine (NE) in rat aortic smooth muscle. NE evoked contraction in a dose-dependent manner, and this effect was inhibited by the JNK inhibitor SP600125. NE increased the phosphorylation of JNK, which was greater in aortic smooth muscle from hypertensive rats than from normotensive rats. NE-induced JNK phosphorylation was significantly inhibited by SP600125 and the conventional-type PKC (cPKC) inhibitor Go6976, but not by the Rho kinase inhibitor Y27632 or the phosphatidylinositol 3-kinase inhibitor LY294002. Thymeleatoxin, a selective activator of cPKC, increased JNK phosphorylation, which was inhibited by $G{\ddot{o}}6976$. SP600125 attenuated the phosphorylation of caldesmon, an actin-binding protein whose phosphorylation is increased by NE. These results show that JNK contributes to NE-mediated contraction through phosphorylation of caldesmon in rat aortic smooth muscle, and that this effect is regulated by the PKC pathway, especially cPKC.
