        A 17-year-women developed mixed warm and cold antibody type autoimmune hemolytic anemia associated with systemic lupus erythematosus. The patients was admitted to our hospital for syncope. Jaundice was observed. Urinalysis revealed protein and hemoglobin. Her hemoglobin was 4.6g/dl and reticulocytes were 10%. Total bilirubin was elevated, while haptoglobin and complement were abnormally reduced. ANA and mitochondrial Ab were detected. The direct and indirect Coombs test were positive. The direct antiglobulin test was positive ; IgG and C3d were detected on the red cell surface. The cold agglutinin(CA) titer was 128. The patients was initially treated with high dose steroid therapy. However the patients did not responded to high dose steroid therapy. We treated the patients with plasmapheresis and synchronized cyclophosphamide therapy. Clinical remission of hematologic complications was achieved in this patients. We report a case of mixed type autoimmune hemolytic anemia with SLE that responded plasma pheresis and synchronized cyclophosphamide therapy.

        Quantitative comparison of mRNA expression of glucosyltransferase(GTF) between xylitol-resistant(X^(R)) and xylitol-sensitive(X^(s)) mutans streptococci

        Since the long-term exposure of mutans streptococci to xylitol is known to select for xylitol-resistant(X^(R)) natural mutants, the occurrence and survival of such X^(R) strains were performed in batch culture methods. The aim of the study was to compare the differentiation and quantification of mRNA expression of the gtf genes of X^(R) and X^(S) mutans streptococci. Using a real-time reverse-transcription polymerase chain reaction, the expression of each gtf was determined. In X^(R) strains, the relative levels of transcription of gtfB and gtfC were decreased while that of gtfD was increased, suggesting the presence of independent promoters. It also suggested that mutation related to production of glucosyltransferase occurred under the exposure of xylitol could explain the caries-preventive mechanisms of xylitol. 자일리톨에 장기간 노출된 mutans streptococci는 자일리톨에 내성이 발현되어 자일리톨 내성균주가 생성된다고 알려져 있다. 본 연구의 목적은 mutans streptococci에서 자일리톨 내성균주와 감성균주의 gtf 유전자 발현량을 각각의 유전자별로 정량적으로 분석하고 비교하는 것이다. 실시간 역전사 중합효소연쇄반응법을 이용하여 각각의 gtf 발현을 조사한 결과 gtfD는 증가한 반면. gtfB와 gtfC는 감소하였는데 이는 각 유전자의 독립된 조절기전이 존재함을 보여주는 것이다. 또한 자일리톨에 노출된 mutans streptococci에서의 glucosyltransferase와 연관된 유전자변형이 자일리톨의 치아우식증 예방효과의 작용기전 중 하나임을 알 수 있었다.

      • 等電點 電氣泳動에 의한 배추와 양배추 育成系統에서의 自家不和合性 關聯 glycoproteins의 比較分析

        等電點 電氣泳動을 이용하여 배추의 몇 개의 5-allele 造成과 그들의 F₁에서 自家不和合性의 발현과 관련된 glycoprotein의 band 樣相을 비교 분석한 결과는 다음과 같다. 1. 배추의 內婚系와 이들 간의 에서 柱頭內 glycoprotein의 band pattern을 分析한 결과 兩親의 band pattern이 F₁으로 그대로 遺傳되므로 써 명확한 同定 및 區分이 가능하였다. 2. 공시된 배추의 계통 중에서 8개의 等電點을 발견하였는데 대부분의 系統에서 pl 7.0 또는 8.6에서 major band를 보이고 있었다. 따라서 major band만으로는 S-allele genotype의 명확한 동정이 불가능한 경우도 상당히 있었다. 3. 상기 bands (pI 6.8 and 7.7)에서의 glycoprotein의 分子量은 pI 7.0에서 49,000-50,000이었고 pI 8.6에서 59,000으로 推定되었다. 4. 양배추는 배추에 비해서 glycoprotein의 major bands와 minor bands수가 많고 매우 다양하게 나타나므로써 S-allele 동정이 배추보다 매우 용이할 것임을 보여 주었다. Band patterns of glycoproteins associated with the expression of self-incompatibility were examined in the stigmatic extracts of self-incompatible Chinese cabbage inbred lines and the hybrids. Clear relationship in the band pattern could be confirmed in the parents and their hybrid, indicating the inheritance of S-alleles to the progeny. Total of 8 isoelectric points were indentified in the inbreds, with the major bands at pI 7.0 and 8.6. Thereforne, in some genotypes, identification of cultivars or S-allele genotypes were impossible with the major bands only. The molecular weights of the glycoproteins were estimated to be 49,000-50,000 for pI 7.0 and 59,000 for pI 8.6 bands. The stigmatic extracts from cabbages showed numerous major and minor bands so that identification of S-allele genotype will be much easier as compared to chinese cabbage.

      • Diborane(B_2H_6) 가스의 합성에 관한 연구

        A reaction between NaBH_4 and BCl_3 gas was carried out to find a new process for the synthesis of B_2H_6 gas. The optimum conditions for the synthesis of B_2H_6 gas were as follows : first step reaction temperature : 15∼25℃, BCl_3 mole ratio to NaBH_4 : 0.14, second step reaction temperature : 50℃, BCl_3 mole ratio to NaBH_4 : 0.19, amount of triglyme to 5g of NaBH_4 : 80㎖. Under above conditions, conversion and yield of B_2H_6 gas were 88.90% and 85.91% respectively.

      • 인체지방기질세포를 통한 척수 손상에 대한 신경학적 회복

        중추신경계 질환의 회복은 손실된 세포재생, 손상된 수초대체, 기능적 재연결을 담당하는 척추중추신경계의 제한적 기능으로 인해 저해된다. 중추신경계 질환의 치료방법으로 줄기 세포 이식은 기능적 결함을 감소시킬 수 있는 중요연구 과제로 부상하고 있다. 본 연구에서는 척수 손상된 백서에 인체지방기질세포의 이식을 통한 효과를 검증하는데 목적을 둔다. 척수 손상은 흉수신경 9번째와 10번째에서 혈관용 클립을 통해 유발되었으며 척수 손상후 7일후에 지방기질세포의 이식을 시행하였다. 척수 손상된 동물 모델에 CM-Dil로 염색된 지방기질세포를 이식하게 되면 척수 손상부위에 지방기질세포들이 채워질 것으로 추측된다. 또한 지방 기질세포로 이식한 동물에서 행동측정지침인 BBB 측정값이 높으며 척수 손상 정도가 조직학적으로 감소될 경우 이식한 지방기질세포들이 신경세포나 성상세포로는 분화되지는 않을 경우로 간주될 것이다. 예상한 결과들은 지방으로부터 분리된 기질세포가 신경자극인자들을 분비하거나 손상부위의 조직과 직접적으로 접촉함으로서 기능적, 행동적 향상에 기여한다고 볼 수 있다. 본 연구 결과는 척수 손상 치료에 지방기질세포의 임상적 응용 가능성을 제안한다. 핵심용어 : 중추신경계, 척수 손상, 인체지방기질세포

      • Cyclin-dependent kinase(CDK)와 세포주기(cell cycle)조절을 통한 새로운 항암제의 개발 현황

        The cell-division cycle is a tightly controlled process that is regulated by the cyclin/CDK family of protein kinase complexes. Stringent control of this process is essential to ensure that DNA synthesis and subsequent mitotic division are accurately and coordinately executed. There in now strong evidence that CDKs, their regulators, and substrates are the targets of genetic alteration in many human cancers. As a result of this, the CDKs have been targeted for drug discovery and a number of small molecule inhibitors of CDKs have been identified. Our attempt here is to illustrate the potential for development of therapeutics to treat human cancers by interfering with cell-cycle progression. Because of the central role that they play in advancing the division cycle, CDKs have been targeted for drug discovery and a number of small molecule compounds have now been identified as CDK inhibitors. These strategies and other targets of intervention within the cell cycle are discussed in our review.

      • LiCoO₂의 합성 공정변수에 따른 리튬이차전지 정극의 전기화학적 특성 연구

        리튬이차전지용 정극활물질 LiCoO₂를 소성법 을 이용하여 합성공정변수를 달리하여 합성하고 합성공정변수가 이들의 물리화학적인 재료특성 및 전기화학적인 특성에 미치는 영향을 조사하였다. 출발 물질로 LiOH · H₂O와 Co₃O₄를 사용하였으며, 750℃, 6시간 이상의 조건에서 결정의 LiCoO₂가 합성되기 시작하였으며, 반응온도가 높아지고 반응시간이 길어질수록 LiCoO₂의 결정성이 증가하였다. 여러 합성 조건 중 가장 우수한 전기화학적 특성을 보이는 조건은 반응옹도 750℃ 반응시간 24시간이었다. 이 조건에서 합성한 LiCoO₂로 Li/1M LiCIO₄ in PC + DME/LiCoO₂의 cell을 구성한 후 0.25mAh/㎠며의 전류밀도로 충 · 방전 하였을 경우 초기 방전 용량은 130mAh/g이였으며, 10cycle 후 방전 용량은 130mA h/g에서 109mAh/g으로 16%의 용량감소가 일어났다. LiCoO₂, the active material of positive electrode in lithium secondary battery, was synthesized by calcination with LiOH · H₂O and C0₃O₄. And relationships between synthesis condition and physical and electrochemical characteristics of LiCoO2 were investigated. Synthesis condition of 750℃, 6hr, LiCoO₄ crystal began to be formed, and the crystallinity of LiCoO₂ increased with reaction temperature and reaction time. LiCoO₂ synthesized by calcination at 750℃ during 24hr showed best electrochemical characteristics. The initial discharge capacity of LiCoO₂ synthesized under this condition was 130mAh/g, and after 10 cycles, the discharge capacity decreased to 109mAh/g.

        자동차 관련직종 근로자에서 상지 근골격계 증상 호소율과 관련요인

        Objectives : The purpose of this study was to determine the prevalence and related factors of upper limb musculoskeletal symptoms among workers in automobile related jobs. Methods : Two-hundred and twenty-one workers volunteered in this study consisting of 112 seat installers and 109 auto mechanics. A questionnaire was administered to the workers consisting of questions on demographics, work type and duration, sleep hours, health habits, and standardized descriptions of NIOSH on musculoskeletal symptoms. Results : Complaint rates of neck and upper extremity musculoskeletal symptoms by anatomical site were as follows: shoulder, 52.0%; neck, 47.1%; wrist and hand 39.4%; elbow, 26.2% and; arm, 24.4%. Significantly higher prevalence of shoulder musculoskeletal symptom was found in middle school than high school graduate, married than unmarried, less than 6 hours sleep than over 6 hours sleep and, seat installer than auto mechanic group. A significantly higher prevalence of arm, elbow, hand and wrist musculoskeletal symptoms was disclosed in the less than 6 hours sleep than over 6 hours sleep group and the seat installer than the auto mechanic group. Multiple logistic regression analysis identified education, sleep hours and, job type as the main affecting factors for shoulder musculoskeletal symptoms. Sleep hours and job type were the main affecting factors for wrist and hand musculoskeletal symptoms. Sleep hours were the main affecting factors for arm and elbow musculoskeletal symptoms. Conclusions : The prevalence of neck and upper limb musculoskeletal symptoms was high in workers who did routine tasks at confined places like seat install-line work. It is recommended that workers performing simple and routine tasks be provided adequate sleep time since lack of sleep was the main affecting factor for most upper limb musculoskeletal symptoms in automobile factory related jobs workers.

      • Vancomycin B 의 불순물 연구

        Vancomycin B obtained by fermentation includes its analogs. Even though the amounts of the analogs are very little because of careful purification steps, those can give rise to toxicities. Therefore, impurities contained in vancomycin B produced by fermentation should be identified. A pyridine buffer was applied for separation of impurities, and good NMR spectra were collected except more downfield shifts than 7 ppm. Four impurities included in vancomycin B were identified.
