        Anti-inflammatory effect of hispidin on LPS induced macrophage inflammation through MAPK and JAK1/STAT3 signaling pathways

        Severe inflammatory reactions caused by macrophage activation can trigger a systemic immune response. In the present study, we observed the anti-inflammatory properties of hispidin on LPS induced RAW264.7 macrophage cells. Our results showed that hispidin treatment significantly reduced the production of cellular NO, IL-6 and reactive oxygen species (ROS) while has not inhibitory effect on TNF-α productions. Excitingly, hispidin treatment retains the phagocytosis ability of macrophages which enabling them to perform the function of removing foreign invaders. Signaling studies showed, hispidin treatment dramatic suppressed the LPS induced mitogen activated protein kinases (MAPK) and JAK/STAT activations. In conclusion, our findings suggest that hispidin may be a new therapeutic target for clinical treatment of macrophages-mediated inflammatory responses.

        The Role of Intestinal Microbiota and Mast Cell in a Rat Model of Visceral Hypersensitivity

        Background/Aims To explore the role of intestinal flora and mast cells in visceral hypersensitivity (VH). Methods The experimental animals were divided into 4 groups: control group, VH group, VH + VSL#3 group, and VH + ketotifen group. Stool samples were collected from each group (n = 3) for a further analysis using 16S ribosomal DNA gene sequence. Visceral sensitivity was evaluated by abdominal withdrawal reflex (AWR) score. Colon tissues of rats were obtained from each group. Mast cells were detected by toluidine blue staining. The degranulation of mast cells was assessed by transmission electron microscopy. Results VH rat model could successfully be induced by acetic acid enema combined with partial limb restraint method. Compared with rats in the control group, AWR score, number of mast cells, and degranulation of mast cells were increased in the VH rats, which could be reduced by administration of ketotifen or probiotic VSL#3. Clostridium sensu stricto 1 abundance was higher in the VH group compared to the control group, which could be restored by application of probiotic VSL#3. Conclusions Probiotic VSL#3 decreases visceral sensitivity in VH rats. The mechanism may be related to mast cell and intestinal flora. Change of Clostridium sensu stricto 1 abundance may be a basis for VH observed in irritable bowel syndrome and may be prevented by specific probiotic administration.         English Prepositional Passives from a Cognitive Grammar Perspective

        This paper aims to address the English prepositional passive from the perspective of cognitive grammar and argues that it resembles the prototype passive in several aspects, but syntactically and semantically it still deviates from the prototype. In the prepositional passive, the N/P2 places more constraints on affectedness, role prominence, or characterization, and the verb and the preposition function as an organic whole. In terms of transitivity and passivizability, the prepositional verb clause cannot be confined to objective analysis. Instead, the subjective construal of actual world can play a part. (i) Prepositions that describe a terminative path are more likely to be passivized than those describing an initiative path. (ii) The passivizibility of the preposition verb clause cannot be determined solely by the verbs and preposition but should be combined with the nominals, sometimes influenced by the selection of participants or setting, and the animacy and discreteness of the nominals.

      • Prototypicality and Markedness: A Study of [Subject+V1+V2ing] Passive Construction in English

        In an effort to answer ESL/EFL students’ questions about meanings, uses and functions of passive voice, this paper attempts to address two significant concepts-Prototypicality and Markedness-in studies of the categorization of the English passive voice system. In particular, it confines its discussion to [Subject+V1+V2ing] Passive Construction. On the basis of prototype theory and markedness theory, it is argued that category PASSIVE, starting from the prototype and unmarked form of central Passive ‘Be+Past participle’, extends in the chained way of concatenation to the adjacent near-prototype and unmarked form of Get-Passive, and expands in the way of radiation to peripheral, untypical and marked forms of [Subject+V1+V2ing] passive construction’ and the ‘V-able passive construction’. The idiosyncrasies and constraints of [Subject+V1+V2ing] passive construction in real context are discussed by using examples from corpus. Comparison and Contrast are also made among different types of passive construction and other related constructions in order to show that ‘[Subject+V1+V2ing] passive construction is one of the peripheral and marked members of passive voice system in English. Taking the Family resemblances as the operating principle, ‘[Subject+v1+v2ing] passive construction not only shares the common features with other passive constructions within the passive domain, but also shares the common properties with other constructions such as middle construction and progressive construction across domains. It also suggests that there exists vagueness among the various forms of passive constructions which are conducive to extend and expand the boundary of category PASSIVE and the perspective of construal and role alignment in cognition also play a significant part in categorization of passive voice and other related constructions.

        A Schema Category Theory-based Analysis of the Get+V-en Construction

        The purpose of this paper is to investigate English Get+V-en construction on the basis of the schema category theory proposed by Langacker(1987, 2000, 2009). Contrary to the passive or non-passive attribution of the Get+V-en construction, this study proposes that the Get+V-en construction is a fuzzy set that designates the gradient feature and its schema instantiates itself by abstracting the general features of the prototype and its extension. The passivization of the Get+V-en construction ranks in three layers, where the central Get-passive (agentful or agentless) serves as the prototype at the first layer. The psychological, reflexive, and reciprocal Get-passives are regarded as semi-Get-passives, functioning as instantiation one at the second layer. Both the adjectival and the formulaic Get-passives can be construed as pseudo Get-passives, representing instantiation two at the third level. The component get shows gradient features from lexical to copular, and V-en demonstrates a dynamic change being either verbal or adjectival in different variants of the Get+V-en construction. The schematic characterization of the central Get-passive can be summarized as follows: Get is a ‘semi-grammaticalized’ word that lies in between lexical verbs and genuine auxiliaries; The Get-passive is generally compatible with dynamic verbs with a causative meaning denoting a resultative state; It is subject-oriented and characteristically used in clauses involving adversity or benefit.

        A Cognitive Grammar Approach to the Active Construction in English

        As a grammatical category, the active construction in English shows many variations which puzzle EFL learners. Given this tendency to confuse, this paper aims to address the English active construction from the perspective of cognitive grammar. It is argued that there are prototype and non-prototype in the English active construction in terms of semantic and syntactic features. The prototypical active construction not only instantiates itself in simple, affirmative, declarative sentences with the resultative transitive verbs in the past form, but also profiles an action chain that involves two demarcated participants: the agent and patient. The agent, often coded as the subject, is in the topical position of the sentence and always exerts volitional influence on the patient, which is coded as the object and affected by the agent. In the non-prototypical active construction, the cognitive construal offers the better interpretations of interchangeable role archetypes in the subject and object position and selects different composite verbs invoked by viewing the event with the same conceptual content.

      • A Genetic Variant in MiR-146a Modifies Digestive System Cancer Risk: a Meta-analysis

        MicroRNAs (miRNAs) negatively regulate gene expression and act as tumor suppressors or oncogenes in oncogenesis. The association between a single nucleotide polymorphism (SNP) in miR-146a rs2910164 and susceptibility to digestive system cancers was inconsistent in previous studies. In this study, we conducted a literature search of PubMed to identify all relevant studies published before August 31, 2013. A total of 21 independent case-control studies were included in this updated meta-analysis with 9,558 cases and 10,614 controls. We found that the miR-146a rs2910164 polymorphism was significantly associated with decreased risk of digestive system cancers in an allele model (OR=0.90, 95%CI 0.87-0.94), homozygote model (OR=0.84, 95%CI 0.77-0.91), dominant model (OR=0.90, 95%CI 0.84-0.96), and recessive model (OR=0.85, 95%CI 0.79-0.91), while in a heterozygous model (OR = 0.99, 95% CI 0.89-1.11) the association showed marginal significance. Subgroup analysis by cancer site revealed decreased risk in colorectal cancer above allele model (OR=0.90, 95%CI 0.83-0.97) and homozygote model (OR=0.85, 95%CI 0.72-1.00). Similarly, decreased cancer risk was observed when compared with allele model (OR=0.87, 95%CI 0.81-0.93) and recessive model (OR=0.81, 95%CI 0.72-0.90) in gastric cancer. When stratified by ethnicity, genotyping methods and quality score, decreased cancer risks were also observed. This current meta-analysis indicated that miR-146a rs2910164 polymorphism may decrease the susceptibility to digestive system cancers, especially in Asian populations.
