In this study, an attempt was made to clarify whether Ginseng enhances secretion of catecholamines in the isolated perfused adrenal glands and to investigate its mechanism of action, making use of total saponin of Ginseng(GTS). GTS(200㎍) produced an increased secretion of catecholamines(CA) significantly in the isolated perfused rabbit adrenal gland. Secretion of CA induced by GTS was inhibited by atropine-treatment and secretion evoked by acetylcholine(Ach) also blocked. Physostigmine -perfusion potentiated the CA secretory effect of GTS. Adenosine lowered CA secretion evoked by GTS, but rather potentiated the effect on secretion induced by Ach. Ouabain reduced the secretory responses evoked by either procedures (Ach or GTS). Mecamylamine, while blocking the secretory effects of Ach perfusion, did not affect the secretion of CA induced by GTS. Perfusion with Ca-free Krebs containing EGTA (5mM) for 30 min totally blocked Ach-induced secretory effects, but did not secretory responses evoked by GTS. These experimental results suggest that GTS increases secretion of CA in the isolated perfused rabbit adrenal glands via direct action on chromaffin cells with partly mediation of muscarinic receptors.
