<P>Enhancing the proliferative capacity of mesenchymal stem cells (MSCs) is critical for increasing their therapeutic potential in a variety of diseases. We hypothesized that lentivirus-mediated overexpression of canine octamer-binding transcription factor 4 (OCT4) might influence the proliferation of canine adipose tissue-derived MSCs (cATMSCs). cOCT4-cATMSCs were generated by transducing cATMSCs with a cOCT4-lentiviral vector. Increased expression of cOCT4 was confirmed using RT-PCR and immunoblotting. Immunophenotypic characterization using flow cytometry indicated that the CD29, CD44, CD73, CD90, and CD105 surface markers were highly expressed by both cOCT4- and mock-transduced cATMSCs (mock-cATMSCs), whereas the CD31 and CD45 markers were absent. We performed the osteogenic differentiation assay to evaluate the effects of cOCT4 overexpression on the osteogenic differentiation potential of cATMSCs. The results showed that cOCT4-cATMSCs had a much higher potential for osteogenic differentiation than mock-cATMSCs. Next, the proliferative capacities of cOCT4- and mock-cATMSCs were evaluated using a WST-1 cell proliferation assay and trypan blue exclusion. cOCT4-cATMSCs showed a higher proliferative capacity than mock-cATMSCs. Cell cycle analysis indicated that overexpression of cOCT4 in cATMSCs induced an increase in the proportion of cells in S and G2/M phases. Consistent with this, immunoblot analysis showed that cyclin D1 expression was increased in cOCT4-cATMSCs. In conclusion, our results indicate that lentivirus-mediated overexpression of cOCT4 increased the proliferative capacity of cATMSCs. OCT4-mediated enhancement of cell proliferation may be a useful method for expanding MSC population rapidly without loss of stemness.</P>

<P><B>Purpose</B></P><P>The use of hepatitis B core antibody (HBcAb)-positive grafts is increasing, especially where hepatitis B is endemic. However, this remains controversial because of the risk of development of de novo HBV infection.</P><P><B>Methods</B></P><P>We collected information obtained between January 2000 and December 2012 and retrospectively analyzed data on 187 HBsAg-negative donors and recipients were analyzed retrospectively. <I>De novo</I> HBV infection was defined as development of HBsAg positivity with or without detection of HBV DNA.</P><P><B>Results</B></P><P>Forty patients (21.4%) received HBcAb-positive grafts. Survival rate did not differ by donor HBcAb status (P = 0.466). <I>De novo</I> HBV infection occurred in five patients (12.5%) who were not treated with anti-HBV prophylaxis, and was significantly more prevalent in hepatitis B surface antibody (HBsAb)- and HBcAb-negative than HBsAb- and HBcAb-positive recipients (50% <I>vs</I>. 4.2%, P = 0.049). All patients except one were treated with entecavir with/without antihepatitis B immunoglobulin and four were negative in terms of HBV DNA seroconversion. No patient died.</P><P><B>Conclusion</B></P><P>HBcAb-positive grafts are safe without survival difference. However, the risk of <I>de novo</I> hepatitis B virus infection was significantly increased in HBsAb- and HBcAb-negative recipients. All patients were successfully treated even after recurrence.</P>

This paper is to investigate the spalling resistance of high performance concrete with the contents of cellulose fiber. According to test results, the use of CL lead to decrease in fluidity. For compressive strength, the use of CL had no influence on compressive strength. For spalling properties, plain concrete showed a severe spalling failure. The use of CL protected from spalling of concrete, but most specimens had scale failure and partial destruction of specimens. This is due to the insufficient fiber length and diameter of CL fiber, which was unable to discharging the internal vapour pressure. For this reason, CL fiber can not be used to protect from spalling oh high performance concrete. Residual strength was observed to 5~7% of original strength, RC column after fire test showed 20% in plain concrete, 25% in CL fiber concrete.

<P>GC1113, a hybrid Fc-fused erythropoietin, is a novel erythropoiesis-stimulating agent that is expected to have an extended duration of action. The preclinical data showed that the hemoglobin increase lasted longer following GC1113 administration than it did following the administration of darbepoetin alfa (NESP?). This study aimed to investigate the pharmacodynamic and pharmacokinetic characteristics and tolerability profiles of GC1113 in humans after single intravenous or subcutaneous administration and to compare the results with those for darbepoetin alfa.</P>

In-place curing box for specimens is used to cure the compressive strength specimens for control in place concrete. The box is composed of insulating chamber maintaining 20±3℃ of temperature. in this paper, strength and temperature history of specimens cured at in place curing box are investigated to verify field applicability. According to test results, air temperature at measured time shows large temperature variation and below zero, whereas, inside temperature of in place curing box maintains within 20±3℃ due to temperature control function. For curing condition, temperature of specimens cured at outside shows large temperature deviation. specimens cured at in-place curing box is not affected by outer temperature.
