        Brown Guinea Pig와 Hairless Mouse를 이용한 광노화 모델에서 LGNC-5의 경구반복투여에 의한 피부주름발생 및 색소침착억제 효과

        비타민과 불포화지방산, 그리고 폴리페놀의 복합처방인 LGNC-5 (LG생활건강 기술연구원 개발)가 건강기능식품의 개별인증을 받을 수 있는지의 여부를 확인하기 위해 hairless mice와 brown guinea pig 모델을 이용해 자외선에 의한 주름생성 및 색소침착 억제력 평가를 실시하여 다음과 같은 결괴를 얻었다. 1. Hairless mice를 대상으로 실시한 주름억제실험에서 육안관찰과 피부주형의 영상분석결과, LGNC-5군은 농도의존적인 주름발생 억제효과를 보였으며, 고용량군(x60)인 LGNC-5H군의 경우 10주후 정상군과 유사한 수준의 양호한 피부 상태를 보였다. Hematoxylin & Eosin stain 에서도 LGNC-5H군에서는 UV 대조군에 비해 표피 두께와 과각화가 현저하게 줄어드는 것으로 조사되어 우수한 주름발생억제 효과를 확인하였다. 2. Brown guinea pig을 대상으로 실시한 색소침착억제력 실험에서 육안관찰과 색차계를 이용한 측정결과, LGNC-5H군에서는 설험개시 7일 후부터 UV 대조군에 비해 색소침착이 억제되기 시작해, 설험 종료시까지 색소침착억제효과를 보였다(p<0.05). Fontana-masson silver stain 에서도 LGNC-5H군에서는 UV 대조군에 비해 melanin 침챡량이 현저하게 감소된 것으로 조사되어 LGNC-5H의 색소침착억제효과를 확인하였다. 3. 10주간의 주름억제실험과 4주간의 색소침착 억제실험에 사용한 모든 설험동물에서 사망 혹은 시험물질 투여로 인한 특이한 임상증상이 관찰되지 않았다. 시험물질 투여 기간 중 사료와 음수 소비량 역시 유의한 변화를 관찰할 수 없었으며, 대조군과 실험군 모두 정상적인 체중 증가를 나타내었다. 이상의 실험결과를 종합하면 LGNC-5은 안전하고 경구반복투여 시 UV조사로 인해 hairless mice에서 발생되는 주름과 brown guinea pig에서 생성되는 색소침착을 효과적으로 억제시키는 처방인 것으로 평가되었다. Chronic exposure of UV radiation in sunlight results in a number of biological responses, including erythema, sunburn cell formation, immune suppression, photoaging, and skin cancer. In this study, we investigated the effects of LGNC-5 (LG Nutricosmetics-5, the mixture of vitamins, unsaturated fatty acid and polyphenol) on UV-induced skin damage. Oral administration of LGNC-5 for 10 weeks resulted in inhibition of UVB-induced wrinkle formation in SKH-1 hairless mice skin compared to UV control group as a dose dependent manner. LGNC-5 treatment also inhibited UV radiation-induced skin pigmentation in brown guinea pigs examined by colorimeter and Fontana-masson silver staining. These results mean that LGNC-5 as a dietary supplement could be useful to attenuate solar UVB-induced melanogenesis and premature skin aging.

        낙동강수계의 유기염소계 농약류 잔류특성과 위해성 평가

        Five organic chlorinated pesticides (OCPs) were monitored for the water samples in the Nakdong river basin. Samples were collected from the 33 representative points and the 5 industrial complex discharge points over 6 sampling events. Heptachlor and trans-heptachlor epoxide were not detected for all samples while dieldrin was detected once at the first sampling event. The measured concentrations of hexachlorobenzene and cis-heptachlor epoxide for the representative sites ranged from N.D to 0.0057 ng/L and from N.D to 0.078 ng/L, respectively. The levels of hexachlorobenzene and cis-heptachlor epoxide were N.D to 0.173 ng/L and N.D to 0.134 ng/L, respectively. It seems like that risk at the monitored level of organic chlorinated pesticide was not serious. However, the maximum concentration of cis-heptachlor epoxide exceeded the water quality criteria (WQC) of U.S EPA. The average concentrations of organic chlorinated pesticides for the industrial complex discharge points were relatively higher than those for the representative sites of the Nakdong river, however, the risk by industrial OCP discharge seems also not serious.

        Subcutaneous Toxicity Evaluation of a Combination Vaccine against Hantaan and Puumala Viruses in Rats for 4 Weeks

        Hantaan (HTNV) and Puumala (PUUV) viruses cause hemorrhagic fever with renal syndrome in human. In the present study, the repeated dose toxicity of the HTNV-PUUV combination vaccine was evaluated in Sprague-Dawley rats. Animals were injected subcutaneously for 28 days with dosages of 0, 0.017, 0.17 and 1.7 dose/kg body weight per day, respectively. No any significant changes of body weight, food and water consumptions were shown. There were no death and clinical findings during the experimental period. In both male and female rats, there were not significant changes in hematological and serum biochemical analysis, urinalysis, and ophthalmoscopic and histopathological examination. These results indicate that the HTNV-PUUV combination vaccine may have no toxic effects and no observed adverse effect level (NOAEL) may be over 1.7 dose/kg/day at subcutaneous route in rats.

        당종류와 최종가열온도가 다른 결착제로 제조한 찰현미 쌀엿강정의 물리화학적, 관능적 특성 변화

        Yetgangjeong is a traditional Korean rice cookie which used rice syrup (Chocheong) to bind fried rice grains. Brown waxy rice and binders prepared using different sugar types and end heating temperatures were assessed to improve the textural properties and functionality and to promote global consumption. Binder was made by mixing different ratios of starch syrup, rice syrup, honey, sugar, and trehalose, and the binder mixture was heated up to a temperature of $100^{\circ}C$ or $105^{\circ}C$. Specific electrical conductivity of binder containing ST (starch syrup and trehalose) showed the lowest values (9.51 and 9.77), and binders containing the rice syrup showed increased values for specific electrical conductivity. The fructose content was higher in the binder mixture containing C (starch syrup and sugar) than in the binder mixture containing ST, but it did not affect the end temperature. Viscosity of the binder was 123.90-175.20 cP and the binder showed higher viscosity at the end heating temperature (EHT) of $100^{\circ}C$ compared at EHT of $105^{\circ}C$. The fracturability of brown rice yetgangjeong prepared using different sugar types was higher at EHT of $100^{\circ}C$ than at that of $105^{\circ}C$ and it was reduced with an increase in the rice syrup content. However, the hardness of yetgangjeong was lower at $100^{\circ}C$, unlike fracturability. With respect to sensory evaluation, its moistness and stickiness decreased, but the crunchiness increased with addition of trehalose instead of sucrose. The overall eating quality score of yetgangjeong was higher at EHT of $105^{\circ}C$ than at that of $100^{\circ}C$ in every experiment and the highest score was found for SHT (starch syrup, honey and trehalose) with 20% rice syrup at EHT of $105^{\circ}C$ (20%RSHT105 (starch syrup, honey, and trehalose containing 20% rice syrup)).

        Co-infection of Aspergillus spp. with Erysipelothrix rhusiopathiae in a red-crowned crane: a case report

        This case describes outbreaks of acute aspergillosis in a red-crowned crane. A six-month-old, male, crane had showed clinical signs (i.e. anorexia, performance loss, ruffled feathers and drooped wings and open mouth breathing, etc.) before death. In necropsy examination, spherical to oval nodules disseminated from the respiratory tract to other organs. Those nodules were formed predominantly in air sacs, lung, peritoneum, serosa of esophagus and trachea. The nodules varied in size from 1 mm to over 1cm and the color was white to yellow. Microscopically, most of lung architecture were replaced by multiple foci which were characterized by well demarcated eosinophilic and karyorrhetic debris and surrounded by numerous Inflammatory cell. Most within necrotic center of the nodules, large numbers of fungal hyphae were present. Microbiology result indicated fungal growths on sabroud dextrose agar and bacterial growths on blood agar. Bacteria identified as E. rhusiopathiae using MALDI-TOF (microflex, BRUKER, USA) and fungi identified as A. fumigatus, A. terreus by sequencing the ITS1 and ITS4 regions. To confirm the route of infection, we checked the existence of the same pathogens in cohabitant (i.e. mother crane). The young age and weakened immunity (i.e. bacterial infection, etc.) causes fatal aspergillosis in birds.

        <P>Background</P><P>Naturally occurring antioxidants were used to regulate the skin damage caused by ultraviolet (UV) radiation because several antioxidants have demonstrated that they can inhibit wrinkle formation through prevention of matrix metalloproteinases (MMPs) and/or increase of collagen synthesis.</P><P>Objective</P><P>We examined the effect of oral administration of the antioxidant mixture of vitamin C, vitamin E, pycnogenol, and evening primrose oil on UVB-induced wrinkle formation. In addition, we investigated the possible molecular mechanism of photoprotection against UVB through inhibition of collagen-degrading MMP activity or through enhancement of procollagen synthesis in mouse dorsal skin.</P><P>Methods</P><P>Female SKH-1 hairless mice were orally administrated the antioxidant mixture (test group) or vehicle (control group) for 10 weeks with UVB irradiation three times a week. The intensity of irradiation was gradually increased from 30 to 180 mJ/cm<SUP>2</SUP>. Microtopographic and histological assessment of the dorsal skins was carried out at the end of 10 weeks to evaluate wrinkle formation. Western blot analysis and EMSA were also carried out to investigate the changes in the balance of collagen synthesis and collagen degradation.</P><P>Results</P><P>Our antioxidant mixture significantly reduced UVB-induced wrinkle formation, accompanied by significant reduction of epidermal thickness, and UVB-induced hyperplasia, acanthosis, and hyperkeratosis. This antioxidant mixture significantly prevented the UVB-induced expressions of MMPs, mitogen-activated protein (MAP) kinase, and activation of activator protein (AP)-1 transcriptional factor in addition to enhanced type I procollagen and transforming growth factor-β2 (TGF-β2) expression.</P><P>Conclusion</P><P>Oral administration of the antioxidant mixture significantly inhibited wrinkle formation caused by chronic UVB irradiation through significant inhibition of UVB-induced MMP activity accompanied by enhancement of collagen synthesis.</P>
