      • 在來種 옥수수의 形態的 特性에 依한 分類

        This experiment was carried out to obtain genetic information for future corn breeding. The materials used for the study were obtained from the nation wide collection of Korean local corn lines. A total of 262 lines were used for the study of morphological characters and for the classification of lines. Results obtained are summarized as follows; 1. The days to flowering of lines ranged from 57 days to 87 days. Most lines had an average of 67 days of flowering days. 2. The number of tillers of lines showed a lot of variation among lines with 49.2% of coefficient of variation. 3. The coefficients of variation computed based on the phenotypic observation or measurement of each line were 36.1%, 27.2%, 20.0%, 16.4% and 16.3% for kernel weight per ear, 100 kernel weight, ear height, plant height and ear length, respectively. 4. Ear height, ear length, ear diameter, tiller number and days to flowering were highly and positively correlation with the plant height, Kernel size, ear size, and plant height were highly correlated with 100 kernel weight and kernel weight per ear. 5. The 262 corn lines were possibly classified into four major groups by the Euclidean distance. Group Ⅰ comprised 110 lines, group Ⅱ 74 lines, group Ⅲ 66 lines and group Ⅳ 12 lines, respectively. Group Ⅰ was characterized as having early maturity, medium plant height large kernel size and large ear size. Group Ⅱ had medium maturity, short plant height, medium kernel size and small ear size. Group Ⅲ had medium maturity, medium plant height, large kernel size and medium ear size. Group Ⅳ had late maturity, long plant height, small kernel size, small ear size and many tillering. 6. The plant height showed significant difference between group Ⅰ and Ⅱ, Ⅱ and Ⅲ, and Ⅱ and Ⅳ group. No statistical differences were observed between group Ⅲ and Ⅳ. The ear size of group Ⅰ was significantly different from those of group Ⅱ, Ⅲ and Ⅳ. Also difference of ear size between group Ⅱ and Ⅲ was significant. The kernel size, 100 kernel weight and kernel weight per ear were all significantly different among all groups classified. The row number was different between group Ⅰ and Ⅱ. The row number of lines in group Ⅳ was significantly different with group Ⅰ, Ⅱ, Ⅲ respectively. The number of tillers and flowering days of linfes in group Ⅳ were greatly different from those of group Ⅰ, Ⅱ and Ⅲ. 7. The corn lines collected from northwest plain regions and middle hilly regions in Korea had medium maturity, medium plant height, large ear and large kernels. The corn lines from middle eastern hilly regions had medium size of ear kernels. The corn lines from middle southern hilly regions had late maturity, small kernel size and many tillers. The corn lines from southwest plain areas had late maturity, long plant height and many tillers.

        Myometrial relaxation of mice via expression of two pore domain acid sensitive K<sup>+</sup> (TASK-2) channels

        Myometrial relaxation of mouse via expression of two-pore domain acid sensitive (TASK) channels was studied. In our previous report, we suggested that two-pore domain acid-sensing $K^+$ channels (TASK-2) might be one of the candidates for the regulation of uterine circular smooth muscles in mice. In this study, we tried to show the mechanisms of relaxation via TASK-2 channels in marine myometrium. Isometric contraction measurements and patch clamp technique were used to verify TASK conductance in murine myometrium. Western blot and immunehistochemical study under confocal microscopy were used to investigate molecular identity of TASK channel. In this study, we showed that TEA and 4-AP insensitive non-inactivating outward $K^+$ current (NIOK) may be responsible for the quiescence of murine pregnant longitudinal myometrium. The characteristics of NIOK coincided with two-pore domain acid-sensing $K^+$ channels (TASK-2). NIOK in the presence of $K^+$ channel blockers was inhibited further by TASK inhibitors such as quinidine, bupivacaine, lidocaine, and extracellular acidosis. Furthermore, oxytocin and estrogen inhibited NIOK in pregnant myometrium. When compared to non-pregnant myometrium, pregnant myometrium showed stronger inhibition of NIOK by quinidine and increased immunohistochemical expression of TASK-2. Finally, TASK-2 inhibitors induced strong myometrial contraction even in the presence of L-methionine, a known inhibitor of stretch-activated channels in the longitudinal myometrium of mouse. Activation of TASK-2 channels seems to play an essential role for relaxing uterus during pregnancy and it might be one of the alternatives for preventing preterm delivery.

        β-lactam계 항생물질 저항성을 지닌 Bacillus sp. J105 균주로부터 분비되는 베타 락탐 분해효소의 정제 및 특성

        Bacillus sp. J105 strain으로부터 유도된 β-lactamase는 ammonium sulfate 침전, 이온 교환 칼럼 크로마토그래피, 겔 여과 등의 과정을 거쳐 SDS-PAGE에서 단일 band로 정제하였다. 정제된 효소의 분자량은 31 kDa이었으며 등전점은 7.35이었다. 효소반응의 최적 pH와 온도는 각각 5와 40℃이었다. 정제 단백질의 총 아미노산 조성의 분석 결과, Gly과 Ala이 각각 14.1과 13.3 mole%로 가장 많은 아미노산 잔기를 차지하고 있었다. 정제 효소의 ampicillin을 기질로 하였을 때의 Km값은 1.33 mM이었고 Vmax값은 0.36 mM/㎖이었다. β-Lactamase, secreted from Bacillus sp. J105 strain was purified to a single band on SDS-PAGE by ammonium sulfate precipitation, ion exchange column chromatography and gel-filtration. The molecular weight of the purified enzyme was 31 kDa on SDS-PAGE and its isoelectric point was 7.35. Optimal pH and temperature for enzymatic reaction were 5 and 40℃, respectively. As a result of total amino acid composition analysis of the purified enzyme, Gly and Ala were occupied 14.1 and 13.3 mole %, respectively. Km and Vmax value of purified enzyme were 1.33 mM and 0.36 mM/㎖ using ampicillin as a substrate, respectively.

        Particle Swarm Optimization을 이용한 PET/CT와 CT영상의 정합

        영상정합 기술은 두 개 이상의 영상을 서로 맞추어, 각각의 영상이 가지고 있는 단점을 보완하여, 새로운 정보를 획득하게 하는 기술이다. 본 논문은 의료 영상간의 2D 영상 정합을 통해 환자의 점진적 병세파악에 도움을 주는 것을 목적으로 하고 있다. 서로 다른 시점과 장비로부터 얻어진 CT와 PET/CT영상을 정합하기 위하여 정확한 해부학적 정보를 제공하는 CT영상간의 정합을 먼저 수행하고 이를 통하여 얻어진 기하학적 정합파라미터들을 PET 영상에 적용하여, 독립 CT영상 위에 PET영상을 중첩하였다. 정합작업을 위해 먼저 각각의 CT영상에 대해 전처리 작업을 실시하였고, 영상의 변형은 affine 좌표변환을 이용하였다. 정합할 영상간의 유사도 평가를 위해 mutual information을 이용하였으며, 빠르고 정확한 정합을 위하여 최적화 알고 리듬인 particle swarm optimization 방법을 이용하였다. 이를 통해 실제 환자의 독립 CT와 PET/CT영상을 이용하여 실험하였고, PET/CT의 영상에서 확인할 수 있었던 병소에 대한 해부학적 위치 정보가 영상정합 과정을 통해 독립 CT 영상에서도 동일한 위치에 표시됨을 확인하였다. 제안된 알고리듬은 PET/CT 뿐만 아니라 향후 도입될 SPECT/CT, MRI/PET 등 다중영상기기와 기존의 독립 CT 영상기기와의 정합에도 폭넓게 사용될 것으로 기대된다. Image registration is a fundamental task in image processing used to match two or more images. It gives new information to the radiologists by matching images from different modalities. The objective of this study is to develop 2D image registration algorithm for PET/CT and CT images acquired by different systems at different times. We matched two CT images first (one from standalone CT and the other from PET/CT) that contain affluent anatomical information. Then, we geometrically transformed PET image according to the results of transformation parameters calculated by the previous step. We have used Affine transform to match the target and reference images. For the similarity measure, mutual information was explored. Use of particle swarm algorithm optimized the performance by finding the best matched parameter set within a reasonable amount of time. The results show good agreements of the images between PET/CT and CT. We expect the proposed algorithm can be used not only for PET/CT and CT image registration but also for different multi-modality imaging systems such as SPECT/CT, MRI/PET and so on.

        감송향(甘松香)이 HL-60 세포의 분화유도에 미치는 영향

        Objectives: To investigate PKC/MAPKs signaling pathways on the human promyelocytic leukemia HL-60 cell differentiation induced by Nardostachytis Rhizoma (N. Rhizoma). Methods : Differentiation of HL-60 cells was determined through expression of CD11b and CD14 surface antigens using flow cytometry and nitroblue tetrazolium (NBT) assay. PKC activity was measured by using a PKC Kinase Activity Assay kit (Non-Radioactive). Activation of ERK and protein levels of PKC soforms were determined by Western blot analysis. Results : Activation of extracellular signal-regulated kinase (ERK) increased time-dependently in differentiation of HL-60 cells induced by N. Rhizoma. Inhibitor of ERK (PD98059) significantly reduced NBT reduction activity induced by N. Rhizoma in HL-60 cells. In addition, N. Rhizoma increased protein level of PKC α, PKC βI and PKC βII isoforms, but the protein level of PKC γ was constant. PKC inhibitors (GF 109203X, Chelerythrine and H-7) inhibited HL-60 cell differentiation and the ERK activation induced by N. Rhizoma. Conclusions : N. Rhizoma induces granulocytic differentiation via a PKC/ERK signaling pathway in HL-60 cells

        재래종(在來種) 옥수수의 형태적(形態的) 특성(特性)에 의(依)한 분류(分類)

        새로운 유전(遺傳) 육종적(育種的) 재료(材料)를 얻고 탐색(探索)하기 위하여 수집(蒐集)한 재래종(在來種) 옥수수 262계통(系統)의 형태적(形態的) 특성분류(特性分類) 및 그 특성(特性)의 분포(分布), 상관(相關)둥을 요약(要約)하면 다음과 같다. 1. 각(各) 특성(特性)을 살펴 보면 개화소요일수(開花所要日數)에서 66일(日) 이하(以下)가 47%, 67~78일(日) 47%, 79일(日) 이상(以上)이 8%로 조(早) 중생(中生)에 고루 분포(分布)하였고 기타(其他) 특성(特性)들은 평균식(平均植)를 중심(中心)으로 양편에 고루 분포(分布)하였다. 2. 분얼수(分蘖數)는 변이계수(變異係數)가 49.2%로 특성(特性)들 중(中)에서 가장 큰 변이(變異)를 보였고 자수경(雌穗俓) 립중(粒重), 100립중(粒重), 착수고(着穗高), 초장(草長), 자수장(雌穗長) 등에서 각각(各各) 36.1, 27.2, 20.0, 16.3% 등으로 비교적 변이(變異)의 폭(幅)이 컸고 자수경(雌穗經), 입(粒)의 장(長), 폭(幅), 후(厚) 및 자수열수(雌穗列數), 개화소요일수(開化所要日數) 등에서 작은 변이(變異)를 보였다. 3. 특성(特性)들간에는 초장(草長)과 착수고(着穗高), 자수장(雌穗長), 자수경(雌穗俓), 분얼수(分蘖數), 개화소요일수(開花所要日數)와 높은 정(正)의 상관(相關)을 보였고, 100 립중(粒重), 자수당립(雌穗當粒)은 립(粒)의 장(長), 폭(幅), 후(厚) 및 초장(草長)과 높은 정(正)의 상관(相關)을 보였다. 4. 계통간(系統間)의 Euclidean distance에 의해 분류(分類)한 4개(個)의 군(群)은 I군(群)에 110개(個), II군(群)에 74개(個), III군(群)에 66개(個), IV군(群)에 12개(個) 계통(系統)이 속(屬)하였으며, 계통군(系統群) I은 조생(早生), 중간(中稈), 대립(大粒), 대형자수(大型雌穗)의 특징(特徵)을 계통군(系統群)II는 중생(中生), 중간(中稈), 중립(中粒), 소형자수(小型雌穗)의 특징(特徵)을, 계통군(系統群)III은 중생(中生), 중간(中稈), 대립(大粒), 중형자수(中型雌穗)의 특징(特徵)을 계통군(系統群)IV는 만생(晩生), 장간(長稈), 소형자수(小型雌穗), 소립(小粒), 다분얼(多分蘖)의 특징(特徵)을 각각(各各) 보였다. 5. 분류(分類)한 군별(群別) 평균초장(平均草長)은 I 군(群)과 II 군(群), II군(群)과 III, IV군(群) 사이에 유의차(有意差)가 인정(認定)되었고, 이삭의 크기에서는 IV군(群)과 II군(群), IV군(群)과 III군(群) 사이를 제외(除外)한 모든 군(群)사이에, 립(粒)의 크기는 II군(群)과 IV군(群) 사이를 제외한 모든 군(群)사이에 100 립중(粒重), 이삭당립중(當粒重)은 모든 군간(群間)에, 이삭열수(列數)는 I군(群)과 II군(群), IV군(群)과 I, II, III군(群) 사이에, 분얼수(分蘖數)와 개화일수(開花日數)는 IV군(群)과 I, II, II군(群) 사이에 각각(各各)그 유의차(有意差)가 인정(認定)되었다. 6. 강원도(江原道)와 제주도(濟州道)를 제외(除外)한 지역(地域)에서 수집(蒐集)한 옥수수를 지역별(地域別)로 나누었을 때 북서부평야지(北西部平野地)와 중부산악지(中部山岳地)의 옥수수는 중생(中生), 중간(中稈), 대형자수(大型雌穗), 대립(大粒)의 특징(特徵)을 중동부산악지(中東部山岳地)의 옥수수는 중생(中生), 중간(中稈), 중형자수(中型雌穗), 중립(中粒)의 특징(特徵)을 중서부평야지(中西部平野地)의 것은 중생(中生), 단간(短稈), 중형자수(中型雌穗), 중립(中粒)의 특징(特徵)을 남동부평야지(南東部平野地)의 것은 조생(早生), 단간(短稈), 소형자수(小型雌穗), 중립(中粒)의 특징(特徵)을 중남부산악지(中南部山岳地)의 것은 만생(晩生), 중간(中稈), 중 This experiment was carried out to obtain genetic information for future corn breeding.         Investigation of cell cycle arrest effects of actinomycin D at G1 phase using proteomic methods in B104-1-1 cells

        <P><B>Abstract</B></P><P>Actinomycin D was previously reported as an inhibitor of Shc/Grb2 interaction in B104-1-1 cells. Actinomycin D arrested the cell cycle at the G1 phase at 1nM, which is about 10 times lower than the inhibition of Shc/Grb2 interactions in B104-1-1 cells. To evaluate other mechanisms of actinomycin D affected suppression of tumors and cell growth, except inhibition of Shc/Grb2 interactions, we examined the proteomic expression profile by proteomic technology. We found up-regulation of MEKK3 and down-regulation of Hsp70 expression from proteomic analysis, which is a very interesting observation because MEKK3 is strongly related with G1 arrest of cell cycle and Hsp70 is also involved in cell cycle regulation. These results indicate that the anti-tumor effects of actinomycin D is due to synergic effects of various proteins regulated by the compound including inhibition of the Shc/Grb2 interaction and other signaling pathways in the cytoplasm. Here we provide a mechanism-based explanation for growth inhibition by actinomycin D using proteomic technology. Thus, this approach may be a potentially useful method to reveal new mechanisms of active compounds or drugs with unknown cellular function.</P>
