β-Glucosidase 생산 유산균주의 분리와 이를 이용한 팽화홍삼 추출액의 생물전환

팽화홍삼에 존재하는 major ginsenoside를 minor ginsenoside로 생물전환하기 위하여 β-glucosidase 활성이 높은 유산균주를 탐색하였다. 된장으로부터 분리한 YLB 8 균주가 β-glucosidase 활성이 가장 높았으며 Leuconostoc mesenteroides로 동정되었다. L. mesenteroides YLB 8을 MRS 배지에서 배양하였을 경우 최대의 β-glucosidase 효소 활성을 나타내기 위한 최적 배양 조건은 glucose 첨가량 1%(w/v), 초기 pH 8.0, 배양온도 30oC, 배양시간 16시간이었다. L. mesenteroides YLB 8을 이용하여 팽화홍삼 추출액을 생물전환하였을 경우 최적 온도는 30oC었으며, 팽화홍삼 추출액의 농도는 12oBx이었다. 팽화홍삼 추출액내에 존재하는 major ginsenoside인 Rb1과 Rb2는 minor ginsenoside인 Rg3로 전환되었으며, Re는 Rg1으로 전환되었으며, 이 중 일부는 Rh1과 Rh2로 전환되었다. Lactic acid bacteria producing high levels of β-glucosidase were screened to convert the major ginsenosides into the minor ginsenosides in puffed red ginseng. A strain named YLB 8 isolated from doenjang showed the highest β- glucosidase activity and was identified as Leuconostoc mesenteroides. The optimum conditions for the maximum production of β-glucosidase, when L. mesenteroides YLB 8 was cultured in MRS medium, were as follows; glucose concentration of 1% (w/v), initial pH 8.0, culture temperature of 30oC, culture time of 16 h. When Puffed red ginseng extract was bioconverted using L. mesenteroides YLB 8, the optimum temperature and the concentration of Puffed red ginseng extract was 30oC and 12oBx, respectively. As a result of bioconversion, the major ginsenosides Rb1 and Rb2 were converted into minor ginsenoside Rg3, and Re was converted into Rg3 and successively into Rh1 and Rh2 in part.
SPECT Image Analysis Using Computational ROC Curve Based on Threshold Setup

We proposed the objective ROC analysis method based on the setting of threshold value for evaluation of single photon emission computed tomography (SPECT) image. This proposed ROC analysis method uses the quantification computational threshold value to each signal on the SPECT image. The SPECT images for this study were acquired by using Monte Carlo n-particle extended simulation code (MCNPX, Ver. 2.6.0, Los Alamos National Laboratory, USA). The basic SPECT detectors and specific water phantom were realized in the simulation, and we could get the simulation results by the simulation operation. We tried to analyze the reconstructed images using threshold value application based objective ROC method. We can get the accuracy information of reconstructed region in the image. This proposed ROC technique can be helpful when we have to evaluate the weak signal for the NM image. In this study, the proposed threshold value based computational ROC analysis method can provide better objectivity than the conventional ROC analysis method.

Since bovine leukosis caused considerable economic loss to the dairy industry, seroepidemiological survey on bovine leukosis was carried out for the dairy herds in Kyunggi province. 1. When compared the results of immunodifussion test with those of enzyme-linked immunosor-bent assay(ELISA) for 94 dairy herds sera, the relationship between the immunodifussion test and ELISA were showen high corresponding rate with sensitivity(97.5%) and specificity(92.6%). 2. In immunodiffusion test for bovine leukosis virus (BLV) antibody in 570 dairy cattle from 30 herds, mean positive rate for BLV antibody was 28.2 %. The positive rate by districts were 16.5% in central, 35.4% in east, 17.3% in west, 29.1% in south, 31.6% in north, 43.7% in northeast. 3. When the results of serological studies was analyzed by age groups, the number of positive was increased gradually with the advanced in age of herds. The highest positive rate was found in the age over 6 years. 4. Of 30 dairy herds examined, 5 herds(16.7%) have no reactions against BLV antigen while 15 herds (50%) showed the range of 1~5 positive cattle and 5 herds(16.7%), the rang of over 11 positive cattle.
Reference based simulation study of detector comparison for BNCT-SPECT imaging

To investigate the optimal detector material for prompt gamma imaging during boron neutron capture therapy, in this study, we evaluated the characteristic regarding radiation reaction of available detector materials using a Monte Carlo simulation. Sixteen detector materials used for radiation detection were investigated to assess their advantages and drawbacks. The estimations used previous experimental data to build the simulation codes. The energy resolution and detection efficiency of each material was investigated, and prompt gamma images during BNCT simulation were acquired using only the detectors that showed good performance in our preliminary data. From the simulation, we could evaluate the majority of detector materials in BNCT and also could acquire a prompt gamma image using the six high ranked-detector materials and lutetium yttrium oxyorthosilicate. We provide a strategy to select an optimal detector material for the prompt gamma imaging during BNCT with three conclusions.
Deficits in motor abilities for multi-finger force control in hemiparetic stroke survivors

<P>The ability to control redundant motor effectors is one of hallmarks in human motor control, and the topic has been studied extensively over several decades since the initial inquiries proposed by Nicholi Bernstein. However, our understanding of the influence of stroke on the control of redundant motor systems is very limited. This study aimed to investigate the effect of stroke-related constraints on multi-finger force control abilities in a visuomotor task. Impaired (IH) and less-impaired hands (LH) of 19 hemiparetic stroke survivors and 19 age-matched control subjects were examined. Each hand repeatedly produced isometric forces to match a target force of 5 N shown on a computer screen using all four fingers. The hierarchical variability decomposition (HVD) model was used to separate force-matching errors (motor performance) into task-relevant measures (accuracy, steadiness, and reproducibility). Task-irrelevant sources of variability in individual finger force profiles within and between trials (flexibility and multiformity) were also quantified. The IH in the stroke survivors showed deficits in motor performance attributed mainly to lower accuracy and reproducibility as compared to control hands (p < 0.05). The LH in stroke survivors showed lower reproducibility and both hands in stroke also had higher multiformity than the control hands (p < 0.05). The findings from our HVD model suggest that accuracy, reproducibility, and multiformity were mainly impaired during force-matching task in the stroke survivors. The specific motor deficits identified through the HVD model with the new conceptual framework may be considered as critical factors for scientific investigation on stroke and evidence-based rehabilitation of this population.</P>
