      • 3-Methylcholanthrene 투여와 성장과정이 흰쥐 간조직 microsomal cytochrome P-450 효소계에 미치는 영향

        간조직 microsome내 약물대사 효소는 endoplasmic reticulum에 주로 존재하여 세포막과 결합되어 있고 여러 내인적(內因的) 물질과 외인적(外因的)물질 및 많은 다른 이물질 대사에 관여하는 전자전달계로서 작용한다. 또한 이 효소계에는 mixed function oxidase 또는 monooxygenase라 불리우는 중요한 효소군이 존재하며 이들 효소군에는 cytochrome P-450이라는 중요한 성분이 포함되어 있다. Cytochrome P-450은 이 효소계에서 기질과 산소의 결합장소이며 terminal oxidase로 작용한다. 본 실험에서는 3-Methylcholanthrene(3-MC)을 투여하였을 때와, 암컷과 숫컷의 신생, 미성숙 그리고 성숙 흰쥐에서 성별과 성장단계에 따른 micrsomal cytochrome P-450효소계의 활성도변화에 대하여 알고자 하였다. 암컷과 숫컷의 흰쥐 간조직에서 3-MC 투여와 성별 및 성장단계에 따른 cytochrome P-450, NADPH-cytochrome C reductase, cytochrome b_5, NADH-cytochrome C reductase, lipid peroxidation, p-nitroanisole-O-demethylase의 활성도 변화를 측정하여 다음과 같은 결론을 얻었다. 1. 3-MC를 흰쥐에 투여하였을 때 hepatic microsomal cytochrome P-450의 활성도가 모두 증가되었으며, 성장단계에 따른 활성도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 신생, 미성숙 흰쥐의 순서로 나타났고 숫컷의 경우에는 미성숙 흰쥐가 세단계중 높고 성숙, 신생 흰쥐의 순서로 나타났다. 2. 3-MC를 투여하였을 때 hepatic microsomal NADPH-cytochrome C reductase의 활성도는 모두 증가 되었으며 성장단계에 따른 활성도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 신생, 미성숙 흰쥐의 순서로 나타났고 숫컷의 경우에는 미성숙 흰쥐가 세단계중 높고 성숙, 신생 흰쥐의 순서로 나타났다. 3. 3-MC를 투여하였을 때 hepatic microsomal cytochrome b_5의 활성도는 암컷의 경우 미성숙 성숙 흰쥐에서, 숫컷의 경우 성숙 흰쥐에서 증가되었다. 성장단계에 따른 활성도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 신생, 미성숙 흰쥐의 순서로 나타났고 숫컷의 경우에는 미성숙 흰쥐가 세단계중 높고 성숙, 신생 흰쥐의 순서로 나타났다. 4.3-MC를 투여하였을 때 hepatic microsomal NADH-cytochrome C reductase의 활성도는 암컷의 경우 성숙 흰쥐에서, 숫컷의 경우 미성숙, 성숙 흰쥐에서, 증가되었다. 성장단계에 따른 활성도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 신생, 미성숙 흰쥐의 순서로 나타났고, 숫컷의 경우에는 성숙 흰쥐가 세단계중 높고 미성숙, 신생 흰쥐의 순서로 나타났다. 5. 3-MC를 투여하였을 때 hepatic microsomal lipid peroxide의 형성은 대부분 증가되었으며 성장단계에 따른 활성도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 미성숙, 신생 흰쥐의 순서로 나타났고 숫컷의 경우에는 성숙 흰쥐가 세단계중 높고 미성숙, 신생 흰쥐의 순서로 나타났다. 6. 3-MC를 투여하였을 때 hepatic microsomal p-nitroanisole-O-demethylase의 활성도는 모두 증가되었으며, 성장단계에 따른 활서도는 암컷의 경우에는 성숙 흰쥐가 세단계중 높고 신생, 미성숙 흰쥐의 순서로 나타났고 숫컷의 경우에는 미성숙 흰쥐가 세단계중 높고 신생, 성숙 흰쥐의 순서로 나타났다. The effects of 3-Methylcholanthrene (3-MC), sex, and developmental stage on the hepatic microsomal cytochrome P-450 enzyme system in rats were investigated. The result is as follows. 1. The contents of hepatic microsomal cytochrome P-450 were increased in 3-MC treated rats compared with control. In the three developmental stages of the female and male rats the contents of cytochrome P-450 were highest in the adult female rats followed by neonatal and immature male rats followed by adult and neonatal male rats. 2. The contents of hepatic microsomal NADPH-cytochrome C reductase were increased in 3-MC treated rats compared with control. In the three developmental stages of female and male rats the contents of NADPH-cytochrome C reductase were highest in the adult female rats followed by neonatal and immature female rats in that order and highest in the immature male rats followed by adult and neonatal male rats in that order. 3. The contents of hepatic microsomal cytochrome b_5 were increased in 3-MC treated immature female, adult female and male rats compared with control. In the three developmental stages of the female and male rats the contents of cytochrome b_5 were highest in the adult female rats followed by neonatal and immature female rats in that order and highest in the immature male rats followed by adult and neonatal male rats in that order. 4. The contents of hepatic microsomal NADH-cytochrome C reductase were increased in 3-MC treated adult female, immature male, and adult male rats compared with control. In the three developmental stages of NADH-cytochrome C reductase were highest in the adult female rats followed by neonatal and immature female rats in that order and highest in the adult male rats followed by immature and neonatal male rats in that order. 5. The formations of lipid peroxide were increased in 3-MC treated rats compared with control. In the three developmental stages of female and male rats the formations of lipid peroxide were highest in the adult female rats followed by the immature and neonatal female rats in that order and highest in the adult male rats followed by immature and neonatal male rats in that order. 6. The contents of hepatic microsomal p-nitroanisole-O-demethylase were increased in 3-MC treated rats compared with control. In the three developmental stages of the female and male rats the contents of p-nitroanisole-O-demethylase were highest in the adult female rats followed by the immature and neonatal female rats in that order and highest in the immature male rats followed by the neonatal and adult male rats in that order.

        Biodegradable retinal fixation devices obtain mechnical fixation of the retina with desirable chorioretinal scarring and with the potential for local, sustained release of antimetabolites and steroids to inhibit proliferative vitreoretinopathy. We manufactured a biodegradable retinal tack with barb that was designed in order to prevent intrusion from implantation of retinal tacks. This study was carried to evaluate the efficacy for retinal fixation and the capability for sustained release of drugs with a newly designed biodegradable retinal tack Biodegradable retinal tacks were made of polymers of glycolic acids and were designed with barbs in a shape to prevent the disinsertion. Biodegradale retinal tacks are divided into 3 parts, a conical portion that is inserted into the sclera, a cylinder portion that remains in the vitreous, and a neck portion between the pin and the cylinder. The tapered conical end was manufactured to allow easy insertion through the retina and choroid into the sclera. A cylinder portion was manufactured with a tapered angle that fixes firmly into the orifice of 19 gauge spinal needle. A neck portion, 0.4 mm in diameter, was designed to prevent disinsertion from following implantation of retinal tack. The applicator was a 19 gauge spinal needle and its orifice was prepared to 15°angle to accept the tapered cylinder portion of the retinal tack. The retinal tacks, secured in the needles, were passed through the formed vitreous and inserted into the retina, choroid, and sclera and were released by pushing the internal needle, usually within 2-3mm of the medullary ray of the posterior rabbit retina A retinal tack was placed in each of 8 pigmented rabbit eyes. Slit-lamp biomicroscopy, indirect ophthalmoscopy and fundus photography were performed periodically from 1 day to 8 weeks after surgery. Eight eyes were enucleated and studied by light microscopy at 8 weeks. Biomicroscopic evaluation of the animals revealed edemas adjacent to the retinal surfaces immediately after insertion of the biodegradable retinal tacks in all the animals. These edemas disappeared after 1 week. The first noticeable change in the size of retinal tacks was shown after 2weeks. The size of the retinal tacks gradually got smaller, decreasing to about one-half at 4 weeks and about one-third at 8 weeks. All retinal tacks remained in inserted places without any movement for an 8 week period. On light microscopy, epiretinal proliferations were seen to extend into the vitreous cavity. Cellular capsules that lined the inner aspect of the scleral defect caused by tack insertion were found. However the adjacent retina had a normal cytologic appearance and architecture in all specimens. We manufactured a biodegradable retinal tack that is designed to prevent intrusion from implantation of retinal tacks. All biodegradable retinal tacks reduce in size with time, but no retinal tacks extruded from the inserted place. The newly designed biodegradable retinal tack can be used for retinal fixation and may be used as a vehicle for the introduction of pharmacologic agents to prevent the cellular events that promote proliferative vitreoretinopathy.

        An improved retinal tack can be used to fix the retina in complicated retinal detachment and proliferative vitreoretinopathy. We used the retinal tack made by biodegradable polyglycolic acid, polylactic acid and copolymers. The tacks with polyglycolic acid were placed in the retina the right eyes of 10 rabbits as group 1, with polylactic-polyglycolic acid copolymers as group 2 in each left eyes. We examined the absorption rates and degree of intraocular inflammation at 1 day, 1, 2, 4, and 8 weeks and compared both eyes. The tacks were absorbed with time in both eyes, but more rapid in copolymers. And there was no inflammation in both eyes. Now, this study will be a base on the drug delivery system in intraocular pharmacological and surgical management.

        A new device, muscle clamping system was developed to facilitate exact quatifying technique A strabismus surgery, and reduce the risk of complications. The device is composed of a lower fixing body with three jaws and an upper supporting body. They are used to clip an extraocular muscle and fix it to the sclera with a single bite. Superior rectus recession on 16 rabbit eyes were performed with this new device. Conjunctival injection, muscle adhesion strength, and light microscopic findings were examined at 1, 2, 4 and 8 weeks postoperatively. The Conjunctival injection were minimal, adhesion power ranged from 420 to 600 gram gravity, which is sufficient in withstand the normal pull of human extraocular muscle. In microscopic exam, some Inflammatory cells and fibrosis were found. The new device was technically easy, fast, and accurate, so it may be useful in stabismus surgery.

        Molecular identification of subspecies of the sika deer, Cervus nippon, in deer farms from Northeastern China, North Korea, and Korea, based on cytochrome b sequences obtained from noninvasive samples

        To identify subspecies of the sika deer, Cervus nippon, and to select C. n. hortulorum individuals for restoration of this subspecies in Korea, we obtained cytochrome b partial sequences (375 bp) of sika deer (C. nippon) from deer farms in northeastern China, North Korea, mainland Korea, and Jeju Island. Based on noninvasive samples, these cytochrome b sequences were compared with the corresponding haplotypes of sika deer, which were obtained from GenBank. We identified five individuals of sika deer from deer farms in northeastern China and North Korea as C. n. hortulorum, and found that interbreeding between individuals belonging to different subspecies is common on deer farms in far-eastern Asia. We concluded that the five C. n. hortulorum individuals on deer farms from northeastern China and North Korea, detected in this study, cannot be considered as pure descendants of C. n. hortulorum in the wild.

        Taxonomic Status of Siberian Flying Squirrel from Korea(Pteromys volans aluco Thomas 1907)

        Sequences of mitochondrial DNA(mtDAN) cytochrome b gene(1,140 bp) and control region(803 bp) of Siberian flying squirrels from Korea(Pteromys volans aluco) and Mt. Changbai of northeast China(P. v. arsenjevi) were obtained to reexamine the taxonomic status of the Korean subspecies. In the cytochrome b gene, six haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjevi, and in control region, seven haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjeve. Furthermore, six haplotypes of cytochrome b gene of P. v. aluco from this study formed a clade with four haplotypes of P. v. arsenjevi in far-east Russia obtained from GenBank. We also investigated the research papers previously published that reported the length of tail vertebrae of P. volans, and found that the length was not sufficiently large as to be a key character of P. v. aluco. This result is not consistent with morphological description for its haplotype. Therefore, we conclude that P. v. aluco from Korea might possibly be a synonym of P. v. arsenjevi from north-east China and nearby Russia.

        Genetic Distinctness of the Korean Red-backed Vole (Myodes regulus) from Korea, Revealed by the Mitochondrial DNA Control Region

        To identify Korean red-backed voles (Myodes regulus) from Korea by mitochondrial DNA (mtDNA) sequencing, we obtained mtDNA control region sequences of 17 red-backed voles from Korea and northeast China, and these sequences were compared with the corresponding haplotypes of Myodes obtained from GenBank. We identified five red-backed voles from Mt. Changbai and Harbin as M. rufocanus and another three redbacked voles from Harbin as M. rutilus, respectively. Moreover, nine red-backed voles from Korea, showing the average nucleotide distance of 0.66% among nine haplotypes, were different from other species of Myodes, and the average distance between nine haplotypes of red-backed voles from Korea and seven haplotypes of M. rufocanus was 6.41%, whereas the average distance between nine haplotypes of red-backed voles from Korea and five haplotypes of M. rutilus was 14.8%. We identified the red-backed voles from Korea as M. regulus, and found that M. regulus is distinct in its mtDNA control region sequences as well, although we propose further analyses with additional specimens from East Asia using nuclear and mtDNA markers to confirm the distinctness of M. regulus.

        The present study summarizes a nonlinear analysis model developed to predict the load-deflection relationship of prestressed concrete beams with bond and/or unbonded tondons and ordinary bonded reinforcement. The stresses of bonded tendons and ordinary reinforcement are determined from the constitive equations of each material, equilibrium condition, and section compatiability condition at a specified loading step, while those of unbonded tendons are calculated based on the member compatiability condition instead of the section compatiability condition. The developed model uses an extensive iterative procedure to determine the neutral axis depth and stress of tendons at each element and each loading step. The effect of diagonal tensile crack on the distributions of moment and curvature is also identified using the truss mechanism for load transfer.
