        Modified heat of hydration and strength models for concrete containing fly ash and slag

        This paper describes the development of modified heat of hydration and maturity-strength models for concrete containing fly ash and slag. The modified models are developed based on laboratory and literature test results, which include different types of cement, fly ash, and slag. The new models consider cement type, water-to-cementitious material ratio (w/cm), mineral admixture, air content, and curing conditions. The results show that the modified models well predict heat evolution and compressive strength development of concrete made with different cementitious materials. Using the newly developed models, the sensitivity analysis was also performed to study the effect of each parameter on the hydration and strength development. The results illustrate that comparing with other parameters studied, w/cm, air content, fly ash, and slag replacement level have more significantly influence on concrete strength at both early and later age.

        A numerical study on the damage of projectile impact on concrete targets

        This paper presents the numerical simulation of the rigid 12.6 mm diameter kinetic energy ogive-nosed projectile impact on plain and fiber reinforced concrete (FRC) targets with compressive strengths from 45 to 235 MPa, using a three-dimensional finite element code LS-DYNA. A combined dynamic constitutive model, describing the compressive and tensile damage of concrete, is implemented. A modified Johnson_Holmquist_Cook (MJHC) constitutive relationship and damage model are incorporated to simulate the concrete behavior under compression. A tensile damage model is added to the MJHC model to analyze the dynamic fracture behavior of concrete in tension, due to blast loading. As a consequence, the impact damage in targets made of plain and fiber reinforced concrete with same matrix material under same impact velocities (650 m/s) are obtained. Moreover, the damage distribution of concrete after penetration is procured to compare with the experimental results. Numerical simulations provide a reasonable prediction on concrete damage in both compression and tension.

      • Monitoring of bridge overlay using shrinkage-modified high performance concrete based on strain and moisture evolution

        High performance concrete (HPC) has been extensively used in thin overlay for repair purpose due to its excellent strength and durability. This paper presents an experiment, where the sensor-instrumented HPC overlays have been followed by dynamic strain and moisture content monitoring for 1 year, under normal traffic. The vibrating wire and soil moisture sensors were embedded in overlay before construction. Four given HPC mixes (2 original mixes and their shrinkage-modified mixes) were used for overlays to contrast the strain and moisture results. A calibration method to accurately measure the moisture content for a given concrete mixture using soil moisture sensor was established. The monitoring results indicated that the modified mixes performed much better than the original mixes in shrinkage cracking control. Weather condition and concrete maturity at early age greatly affected the strain in concrete. The strain in HPC overlay was primarily in longitudinal direction, leading to transverse cracks. Additionally, the most moisture loss in concrete occurred at early age. Its rate was very dependent on weather. After one year, cracking survey was carried out by vision to verify the strain direction and no cracks observed in shrinkage modified mixes.

        Therapeutic inhibition of SGK1 suppresses colorectal cancer

        Colorectal cancer (CRC) is one of the leading causes of death worldwide. Thus, the development of new therapeutic targets for CRC treatment is urgently needed. SGK1 is involved in various cellular activities, and its dysregulation can result in multiple cancers. However, little is known about its roles and associated molecular mechanisms in CRC. In present study, we found that SGK1 was highly expressed in tumor tissues compared with peri-tumor samples from CRC patients. In vitro experiments revealed that SGK1 overexpression promoted colonic tumor cell proliferation and migration and inhibited cell apoptosis induced by 5-fluorouracil (5-FU), while SGK1 shRNA and inhibitors showed the inverse effects. Using CRC xenograft mice models, we demonstrated that knockdown or therapeutic inhibition of SGK1 repressed tumor cell proliferation and tumor growth. Moreover, SGK1 inhibitors increased p27 expression and promoted p27 nuclear accumulation in colorectal cancer cells, and p27 siRNAs could attenuate the repression of CRC cell proliferation induced by SGK1 inhibitors.         Toremifene, an Alternative Adjuvant Endocrine Therapy, Is Better than Tamoxifen in Breast Cancer Patients with CYP2D6*10 Mutant Genotypes

        Purpose Tamoxifen showed individual differences in efficacy under different CYP2D6*10 genotypes. Our study evaluated the prognosis of tamoxifen or toremifene in hormone receptor (HR)–positive breast cancer patients under different genotypes. Materials and Methods CYP2D6*10 genotypes of HR-positive breast cancer patients were determined by Sanger sequencing, and all the patients were divided into tamoxifen group or toremifene group. Results A total of 268 patients with HR-positive breast cancer were studied. The median follow-up time was 72.0 months (range, 5.0 to 88.0 months). Of these, 88 (32.9%), 114 (42.5%), and 66 (24.6%) patients had C/C, C/T, and T/T genotypes, respectively. Among patients who received tamoxifen (n=176), the 5-year disease-free survival (DFS) rate in patients with C/C and C/T genotype was better than that in patients with T/T genotype, and the difference was statistically significant (p < 0.001 and p=0.030, respectively). In patients receiving toremifene, CYP2D6*10 genotype was not significantly associated with DFS (p=0.325). Regardless of genotypes, the 5-year DFS rate was higher in patients treated with toremifene than in patients with tamoxifen (91.3% vs. 80.0%, p=0.011). Compared with tamoxifen, toremifene remained an independent prognostic marker of DFS in multivariate analysis (hazard ratio, 0.422; p=0.021). For all the 180 patients with CYP2D6*10 C/T and T/T genotypes, the 5-year DFS rate was significantly higher in the toremifene group than in the tamoxifen group (90.8% vs. 70.1%, p=0.003). Conclusion Toremifene may be an alternative adjuvant endocrine therapy for patients with CYP2D6*10 mutant genotypes.
