      • 곰팡이에서 분리한 Helicobacter pylori 항균물질의 분리 및 구조규명

        Helicobacter pylori is a Gram-negative bacterium which causes chronic gastritis and is associated with gastric ulcer, duodenal ulcer and gastric carcinoma. In the process of screening of antibacterial activities against H. pylori from soil microorgani는, fungus No. 60686 was isolated. After fementaion of No. 60686, the antibacterial compound was isolated, purified and followed by extraction of mycelium with orgainic solvents, acetone and ethyl acetate, through silica gel chromatography, LH-20 gel chromatography and HPLC. As a result of the structural analyses of the compound by IR, ^1H- and ^13C-NMR, FAB/Mass spectrophotomer, the compound having the antimicrobial activity was idenfitied as chaetoglobosin A (C_32H_36N_2O_5), a cytochalasan derivative. The antimicrobial activity of chaetoglobosin A was tested against Gram-positive and negative bacteria by paper disk method. Among the test strains of 9 Gram-positive bacteria and 18 Gram-negative bacteria containing 4 H. pylori strains, the growth of 4 H. pylori strains and 3 S. aureus strains (SG 511, 285 and 503) was only inhibited by chaetoglobosin A. Also it was shown that its growth inhibition against H. pylori strains was stronger than that against S. aureus strains at the treatment of the same concentration. Therefore it was concluded that chaetoglobosin A has a specific growth inhibition against H. pylori of the tested bacteria.

        Software development of large scale program such as Operating System is lengthy and costly process. In order to reduce cost, time and risk, there is currently general acceptance of the need for Software Reuse System during the whole development cycles. In this paper, the problem of existing reuse system methodology is analyzed. Also using this method Economy Statistics Program Reuse System(ESPRS) which supports all phase of software life cycles is designed. It appears that this ESPRS reduces considerably communication error of requirement specification between system-analyst and end-user.

        The simulation of target-scattered echo with the moving sonar platform and target in 3-dimensional ocean environment is essential to validate and evaluate the performance of a sonar system. This paper presents the improved target signal simulation on the basis of the highlight(HL) model and its realtime algorithm. In order to simulate the scattering highlight, the highlight is represented as a directional scatterer. The realtime generation algorithm of the target signal is realized by use of DSP chip, TMS320C40, where the 40 channels are equally separated to form a parallel processing task in 4 processors. The presented realtime-version of target signal simulation can be used as a target signal simulator in the development of ACM(Acoustic Counter Measure) and advanced sonar signal processing techniques.

      • 홍모기 (Culex pipiens pallens)에 있어서 trypsin의 분리 및 특성에 관한 연구

        Culex pipiens 모기에 있어서 흡혈 후 중장내에서 생성되는 단백질 분해효소인 trypsin의 특성에 대하여 조사하였다. 중장내의 trypsin의 활성을 조사한 결과 적정 pH는 8.1로 나타났다. 무기이온의 영향을 조사하였을 때 칼슘이온의 경우 50mM 농도에서 활성을 가장 높게 증가시켰으며, 마그네슘이온의 경우는 활성도를 미세하게 증가하거나 영향을 나타내지 못하였고, 아연이온에 의하여는 그 활성이 저해되었다. 흡혈 후 trypsin 활성은 20시간후부터 증가하기 시작하여 30시간에 최대의 활성을 나타내었고 그 후 감소하였다. Anion-exchange chromatography를 이용하여 흡혈 후 30시간 지난 중장내의 trypsin을 부분 분리한 결과 trypsin 활성을 나타내는 2개의 peak(PⅠ, PⅡ)가 존재하는 것으로 나타났다. 흡혈 후 24시간된 모기의 중장에서는 2종류의 단백질 peak가 나타났으나 PⅠ의 부위에서만 활성을 나타내었다. 흡혈 후 30시간이 지난 중장에서 활성이 나타나는 PⅡ는 흡혈 후 24시간에서 30시간 사이에 PⅡ의 해당하는 zymogen이 활성화됨으로써 나타나는 것으로 추정된다. 이들의 전기영동결과 C. pipiens는 7가지의 동이원소를 가지고 있었으며 그 중 4번∼7번째가 흡혈 후 30시간된 중장의 PⅡ에서 활성이 나타났다. Differential centrifugation 방법에 의한 trypsin의 중장내 분포를 조사한 결과 trypsin이 가용성 단백질인 것으로 밝혀졌으며 이 사실에서 microvilli로부터 중장의 강 내부로 분비되어 소화작용을 일으키는 것으로 추측된다. 이런 실험의 결과는 trypsin이 microvillar endoperitropic membrane을 통과하는 70,000 dalton 이하의 분자량을 갖음을 간접적으로 증명하고 있다. Trypsin, protease, in the midgut after bled meal from the mosquito, Culex pipiens pallens female has been examined. In the crude homogenate, the trypsin showed optimum activity at pH 8.1. The activity was stimulated by Ca^2+ at 50mM, slightly or no affected by Mg^2+, inhibited by Zn^2+. The trypsin activity in the adult females began to rise at. 20 hours after blood meal and reached at maximal level at 30 hours and started to declease. The result of anion -exchange(DEAE-cellulose) chromatography, trypsin has been found two active peaks(PⅠ, PⅡ) at the 30 hour female after blood meal. But at the 24 hour female after blood meal had two protein peaks, while only peak(PI) showed trypsin activity. The PII activity, showed in the midgut at 30 hour after blood meal, was supposed activation of zymogen PII during 24 hour to 30 hour. The result of electrophoresis, C. pipiens had seven isozymes and the 4-7th isozyme activity were appeared at PII fraction of 30hr midgut after blood meal. The distribution of trypsin in the midgut was investigated by differential centrifugation method. The result suggested that trypsin was secreted from microvilli to luminal space of midgut and the digestion was processed in the luminal space and the molecular weight was lower than 70,000 delton.

      • 지주막하 출혈을 동반한 용혈성 요독 증후군 1례

        Hemolytic uremic syndrome is characterized by Coombs-negative microangiopathic hemolytic anemia, thrombocytopenia, and acute renal failure in infant and children. At present, E. coli 0157 : H7 and other verotoxin-producing E. coli(VTEC) seem to be a major cause of this illness. The majority of patients have a prodrome of bloody diarrhea, and acute renal failure develops several days to weeks later. Although an optimal treatment remains unknown, the general management of hypertention and renal failure with early and frequent dialysis is the mainatay of treatment, and intravenous administration of immunoglobulin G is like to be effective. We experienced a case composed of bloody diarrhea, seizures, coma, and anuria in 1 year old girl who was diagnosed as HUS and recovered renal function completely with slight impairment of cognitive ability and behavioral function. We presented this case with a review of literatures.

      • 경쟁심리가 급성 냉통증 반응에 미치는 영향

        One of the most common symptoms in dental clinic is pain. The pain behavior is very variable because it is subjective and influenced by many factors such as sex, age, race, religion, etc. The purpose of this study is to evaluate effect of the competition on pain threshold and pain tolerance. A sample consisted of 55 individuals (24 males, 31 females) ranged from 23 to 32 years old. In the first session, the subjects were requested to place their each nondominant and dominant hand in ice water(0-2℃) and to raise the other hand when they feel the first pain and to leave it there until the pain reached an intolerable level. After 1 week, subjects were fully informed the purpose of this study which sex can endure the pain longer. We compared the first and the second session of the cold pressor pain threshold and tolerance time. The obtained Results were as follows ; 1. Cold pain threshold was not significantly different between sex and hand group. 2. In male's dominant hand group, cold pain tolerance time was significantly longer after competition. 3. In the dominant hand group, cold pain tolerance time of the male was significantly longer than females after competition. This study showed that competition can change the cold pain tolerance.

      • 신설아파트 열병합발전 도입에 대한 예비 타당성 분석기법 개발

        The object of this study is the development of a Method which is enable to review the preliminary feasibility for co-generation system in new apartment buildings. In Korea co-generation systems have been installed in most of large industrial plants and commercial buildings which consume a large quantity of electric and heat energy, for energy saving and cutting products cost, under positive governmental supports. However for apartment buildings which consume quite a large electric and heat energy, are still remained in conventional energy supply system, and are not popular to utilize useful co-generation system. One of the major reason for these is the lack of clear and easy justification tool. In this circumstance, this study can provide a tool to verify the feasibility of co-generation in apartment buildings with this handy tools for planners and designers beforehand.

      • Thermoplasma acidophilum의 20S Proteasome에 의한 ATPase-활성적 단백질분해에 관한 연구

        The eukaryotic 26S proteasome is an ATP/ubiquitin-dependent proteolytic complex consisting of the 20S core particle and 19S ATPase complex. However, because of its complexity and unstable properties, this study was carried out to present more simple and stable model for the ATP-activated proteolytic complex in prokaryotes which can take the place of the eukaryotic 26S proteasome. For this purpose, recombinant Thermoplasma 20S proteasome (T20S) and Methanococcus MS4, a sequence homolog of one ATPase subunit in the 19S ATPase complex, were successfully isolated from Escherichia coli (E. coli). The α and β subunits of T20S expressed in E. coli could assemble for themselves, and showed the peptide-hydrolyzing activity. Whereas both T20S and R20S (the 20S complex from rabbit skeletal muscle) had the highest peptidase activity against Suc-LLVY-AMC, a good substrate for chymotrypsin-like peptidase activity, the specific activity of T20S was slightly lower than that of R20S. In addition, several reagents such as KCI, SDS, and ovalbumin were shown to have different effects on the peptidase activities between T20S and R20S. When the ATPase activity of the purified MS4 were assayed , the Km for ATP was about 0.5 mM, and casein could stimulate the activity more than 2 fold without the change in Km. This result implicates that protein-activated ATPase may induce the conformational change of casein, and therefore suggests that MS4 ATPase may activate the proteolytic activity of the 20S proteasome via accelerating the recognition and translocation of the protein substrates.
