Osteoarthritis, characterized by articular cartilage degradation, is the leading cause of chronic disability in older adults. Studies have indicated that circular RNAs are crucial regulators of chondrocyte development and are involved in the progression of osteoarthritis. In this study, we investigated the function and mechanism of a circular RNA and its potential for osteoarthritis therapy. The expression levels of circCREBBP, screened by circular RNA sequencing during chondrogenic differentiation in adipose tissue-derived stem cells, and TGFβ2 were significantly increased in the cartilage of patients with osteoarthritis and IL-1β-induced chondrocytes. circCREBBP knockdown increased anabolism in the extracellular matrix and inhibited chondrocyte degeneration, whereas circCREBBP overexpression led to the opposite effects. Luciferase reporter assays, rescue experiments, RNA immunoprecipitation, and RNA pulldown assays confirmed that circCREBBP upregulated TGFβ2 expression by sponging miR-1208, resulting in significantly enhanced phosphorylation of Smad1/5 in chondrocytes. Moreover, intra-articular injection of adeno-associated virus-sh-circCrebbp alleviated osteoarthritis in a mouse model of destabilization of the medial meniscus. Our findings reveal a critical role for circCREBBP in the progression of osteoarthritis and provide a potential target for osteoarthritis therapy.
Obstacle-negotiation performance on challenging terrain for a parallel legwheeled robot

High obstacle-negotiation performance of quadruped robots for the challenging terrain is strongly demanded in some fields, including payload delivery and disaster relief. In this study, a parallel leg-wheeled robot with the high mobility performance on targeted types of rough terrain is proposed. In contrast to other obstacle-negotiating robots, the strength of this robot is that it has adequate over-obstacle capabilities with a horizontal body in terms of different obstacle shapes, which mainly derives from the multi-DOF (degree of freedom) flexible locomotion of legs and the independent actuated wheels as end effectors. Particularly for a steep terrain where a large inertia robot negotiates it with low efficiency and stability, the proposed step-over gait, combined with static gait and wheeled locomotion, improves the overobstacle efficiency and maintains pose stability. Finally, simulations and experiments verify the performance on negotiating the diverse obstacles with level body over challenging terrain.

We investigated whether cornin, an iridoid glycoside isolated from fruits of Verbena officinalis L., regulated angiogenesis and thereby improved functional outcomes after stroke and discovered a potential mechanism. The effects of cornin on proliferation of rat artery smooth muscle cell (RASMC) and signalling was investigated in vitro. Adult male rats were subjected to 1 h of middle cerebral artery occlusion (MCAO) and reperfusion and treated with or without 25 mg/kg of cornin, starting 24 h after ischemia and reperfusion, by continuous intravenous injection daily for 14 days. Neurological functional tests were performed and cerebral Evans blue extravasation was measured. Angiogenesis and angiogenic factor expressions were measured by immunohistochemistry and Western blotting, respectively. Cornin increased the proliferation of RASMC and enhanced the expression of Wnt5a, b-catenin, cyclin D1 and angiopoietin-1 (Ang1). Cornin treatment promoted angiogenesis in the ischemic brain core and improved functional outcomes after stroke. Cornin-treated MCAO rats showed significant increase in vascularization and expression of vascular endothelial growth factor and Ang1 and phosphorylation of Tie2 and Akt compared with vehicle-treated MCAO rats. The Ang1/ Tie2 axis and Wnt/b-catenin pathways appear to mediate cornin-induced angiogenesis.

Ethic and value theory of new contemporary Taoism, should be the development and inheritance of excellent tradition of ethical thoughts of ancient Taoism, and it also should be a new annotation of Taoism ethical value on actual requirements of current society. Only if contemporary new Taoism carries forward traditional Taoism thoughts of advocating simply and honestly, getting along with subordinates humbly, having few desires and selfishnesses, and combines Ethical and Confucianism thoughts together to build the Confucianism thoughts of humanity and courtesy on Taoism thought that to be honest and sincere, having few desires and selfishnesses, we can overcome and avoid the 'dissimilation' danger of Confucianism thoughts of humanity and courtesy.

Adopting the methods of literature and field research to systematically sort out the advanced experience of developed countries in international medical tourism, analyse the advantages and shortcomings of Hainan’s international medical tourism in the context of the construction of free trade port, so as to clarify the optimization path of Hainan’s international medical tourism. It is believed that: the ecological and policy advantages of Hainan’s international medical tourism are obvious, but the core level of medical technology and service mode still need to be improved and perfected, and the development process has a long way to go. In the face of the development dilemma, we should learn from the development experience of international medical tourism powerhouse, play the advantages of Hainan free trade port, strengthen the brand promotion and create top professional medical team, set up Chinese medicine health, reproductive assisted technology and other speciality specialties, the establishment of medical tourism referral system to speed up the cultivation of the local medical tourism industry composite talent training, and then gradually build a perfect Hainan international medical tourism system.
Development of stress-tolerant transgenic plants via RNA metabolism control

Environmental stresses including drought, extreme temperatures, and high salinity are major factors that severely limit crop productivity worldwide. To overcome yield loss due to these environmental stresses, a large number of researches have been conducted to understand how plants respond to and adapt these environmental stresses. Posttranscriptional regulation as well as transcriptional regulation of gene expression is recognized as a key regulatory process in plant stress responses, and these cellular processes are regulated by diverse RNA-binding proteins (RBPs). Over the last years, we have extensively investigated the functional roles of RBPs that harbor an RNA-recognition motif at the N-terminal half and a glycine-rich region at the C-terminal half (glycine-rich RNA-binding proteins, GRPs), zinc finger-containing GRP, and cold shock domain proteins (CSDPs) in Arabidopsis thaliana, rice (Oryza sativa), wheat (Triticum aestivum), and rapeseed (Brasicca napus) under stress conditions. Our comparative analysis demonstrated that certain family members display RNA chaperone function during stress adaptation process in monocotyledonous plants as well as in dicotyledonous plants. These findings point to the importance of the regulation of mRNA metabolism in plant response to environmental stresses and shed new light on the practical application of these RBPs to develop stress-tolerant transgenic crops.
