        The purpose of this study is to develop a transurethral liquid suppository containing prostaglandin E_(1)(PGE_(1))-loaded microemulsion, which undergoes a phase transition to gels at body temperature. The effects of oils, ethanol as solvent and HCI as pH-controlling agent on the physicochemical properties of liquid suppositories composed of poloxamer P 407. P 188 and carbopol was investigated. The stability of PGE_(1) and release of PGE_(1) from liquid suppository were evaluated. Oils such as Neobee and soybean oil significantly decreased the gelation temperature but increased the gel strength of liquid suppository due to their strongly binding with the components of liquid suppository base. However, ethanol slightly did the opposite. The pH of liquid suppositories hardly affected the gelation temperature and gel strength due to addition of very small HCI(0.005-0.01%). A liquid suppository [PGE_(1)/P 407/P 188/carbopol/Neobee/ethanol/HCI(0.4/14/14/0.4/7/2/0.005%)], which had the gelation temperature (34.2±0.6℃) and gel strength (31.35±4.37 sec) suitable for liquid sup-pository system, was easily administered and not leak out from the body. About 60% of PGE_(1) was released out within 2h from this formulation. It was shown that the release of PGE_(1) was proportional to the square root of time, indicating that PGE_(1) might be released from the suppository by Fickian diffusion. It was stable at 4℃ for at least 2months. The results suggest that transurethral liquid suppository containing prostaglandin E_(1)-loaded microemulsion is thought to be a con-venient, safe and effective dosage form for PGE_(1). However, it should be further developed as a more convenient and stable dosage form for PGE_(1).

        본 논문에서는 fiber optical field test 에서의 통신과 파이버설치, 시험, 유지/보수 및 손실측정에 사용되는 광전화기를 설계 하고자 한다. 본 장비는 음성을 광신호로 변조하여 파이버로 내보내어 설치자/시험자/수리자 등이 상대방과 통신할 수 있도록 한다. 이번에 설계한 광전화기는 FM 변/복조 방식을 이용한 아날로그 양방향 광전화기로써, 마이크로폰을 통해 입력되는 음성신호를 광신호로 변환하고 이를 다시 FM변조한 후 신호를 파이버로 전송한다. 그리고 Photo Diode를 통해 수신된 광신호는 FM 복조기를 통해 아날로그 신호로 복조한 후 적절한 이득으로 취하여 스피커로 전송되고 전송된 신호는 스피커를 통해 사람이 들을 수 있는 소리로 바뀌게 된다. 단일 파이버(Single Optical Fiber)를 통한 쌍방향 동시 통화는 13㎛의 LED를 사용하여 FM 변조 방식을 채용하므로 가능하였다.         Studies on carbon fiber epoxy composite is being conducted broadly in industries because of its specific stiffness, specific strength and good fatigue resistance. Because carbon fiber epoxy composite has high vibration characteristics, if existing metal material is alternated by carbon fiber epoxy composite, structure will be low weight, satisfy 1st bending natural frequency and have high fatigue resistance. In this study, research on to manufacture hybrid propeller-shaft using carbon fiber epoxy composite instead of current steel propeller shaft of rear wheel drive vehicle. For this, change of 1st bending natural frequency by the number of layers of carbon fiber epoxy composite on the outer surface of aluminum shaft was observed. Compared the change of 1st bending natural frequency by FEA with computer simulation and vibration test. Through this, the amount of increase in 1st bending natural frequency of hybrid shaft by the increase in number of layers of carbon fiber epoxy composite and high vibration characteristics of hybrid shaft was acknowledged.

        Development of a model for animal health monitoring system in Gyeongnam I. Design, data and frequencies of selected dairy cattle diseases

        A national animal health monitoring system(NAHMS) in Gyeongnam area was started from 1997 to develop statistically valid data for use in estimating disease frequencies in dairy cattle, and the associated costs. The objectives of this study were to : (1) discribe what was done to implement and maintain the system in Gyeongnam ; (2) present selected disease frequencies ; (3) discuss the epidemiological consideration of what was done and implications for the results obtained. Veterinary Medical Officers(VMOs-professors and graduate students from Gyeongsang National University, Faculty of Gyeongnam Livestock Promotion Institute, and Clinic veterinarians) served as data collectors. After training on current disease and management problems of dairy cattle, interview techniques, sampling methods, and data collection instruments, the VMOs participated in selection of the sample herds and data gathering. Forty of 167 dairy herds were selected randomly and the VMOs visited farms once a month for 12 months to collect data about management, disease, inventory, production, preventive treatment, financial and any other relevant data. Strict data quality control devices were used. Specific feed-back was developed for the producers and data collectors. Of the three age groups studied, cows had the greatest number of disease problems. The six disorders found most frequently were (from the highest to the lowest) breeding problems, clinical mastitis, birth problems, gastrointestinal problems. metabolic problems, and lameness. In young stock, respiratory, multiple system, breeding problems, and gastrointestinal problems were pre dominant, whereas in calves, gastrointestinal, respiratory, and integumental problems were major.

        Molecular Cloning and Characterization of a Large Subunit of Salmonella typhimurium Glutamate Synthase (GOGAT) Gene in Escherichia coli

        Two pathways of ammonium assimilation and glutamate biosynthesis have been identified in microorganisms. One pathway involves the NADP-linked glutamate dehydrogenase, which catalyzes the amination of 2-oxoglutarate to form glutamate. An alternative pathway involves the combined activities of glutamine synthetase, which aminates glutamate to form glutamine, and glutamate synthase, which transfers the amide group of glutamine to 2-oxoglutarate to yield two molecules of glutamate. We have cloned the large subunit of the glutamate synthase (GOGAT) from Salmonella typhimurium by screening the expression of GOGAT and complementing the gene in E. coli GOGAT large subunit-deficient mutants. Three positive clones (named pUC19C12, pUC19C13 and pUC19C15) contained identical Sau3AI fragments, as determined by restriction mapping and Southern hybridization, and expressed GOGAT efficiently and constitutively using its own promoter in the heterologous host. The coding region expressed in Escherichia coli was about 170 kDa on SDS-PAGE. This gene spans 4,732 bases, contains an open reading frame of 4,458 nucleotides, and encodes a mature protein of 1,486 amino acid residues (Mr =166,208). The EMN-binding domain of GOGAT contains 12 glycine residues, and the 3Fe-4S cluster has 3 cysteine residues. The comparison of the translated amino acid sequence of the Salmonella GOGAT with sequences from other bacteria such as Escherichia coli, Salmonella enterica, Shigella flexneri, Yersinia pestis, Vibrio vulnificus and Pseudomonas aeruginosa shows sequence identity between 87 and 95%.

        Effect of 2,3,4,4',5-pentachlorobiphenyl (PCB 114) on Morphological Changes in the Testis of the Rats

        This study was performed to examine the effect of 2,3,4,4',5-pentachlorobiphenyl (PCB 114) on morphological changes in the testes of rats. PCB 114 (20 mg/kg/week) in corn oil was intraperitoneally injected into adult rats for 2, 5, and 8 weeks, with injections carried out weekly. The total body weights and testicular weights were measured at 3, 6, and 9 weeks during PCB treatment, respectively. The morphological changes in the rat testes were then analyzed by light microscopy (LM) and transmission electron microscopy (TEM). The results showed that PCB 114 caused no significant change in the body weight but caused a marked increase in the testicular weights at 6 weeks. The LM results proved that the size of their seminiferous tubules slightly increased by PCB treatment, and cytoplasmic vacuolation and damaged nuclear membrane of spermatogenic cells were evident at 6 weeks in the electon microscopical examination. These results suggest that the reproductive of the adult male rats is effective to PCB 114, and that may induce degenerative changes in testicular parenchyma of adult male rats.

        <P>Skeletal muscle is an important organ in our body and a dynamic composite of proteins. Citrus aurantium L. has been widely used in oriental medicine in Eastern Asia for a long time. It contains over 100 bioactive compounds and flavonoids that regulate the inflammatory response and tumorigenesis, through various mechanisms. In the present study, we investigated changes in the protein pattern using two-dimensional electrophoresis (2-DE) and matrix assisted laser desorption ionization time of flight mass spectroscopy (MALDI-TOF/MS) to assess the anti-inflammatory effect of flavonoids isolated from Korean C. aurantium L. in lipopolysaccharide (LPS)-induced L6 cells. L6 skeletal muscle cells were pretreated with flavonoids for 1 h and stimulated with LPS for 24 h. Proteins from the L6 cells of the control, LPS treated and flavonoid treated groups were extracted and resolved by 2-DE using pH 4-7 IPG strips loaded with 150 μg of protein. Forty-one differentially expressed protein spots were identified (more than two-fold was considered significant, p < 0.05), and 18 were detected by MALDI-TOF/MS. These results suggest that proteomics can be used to identify changes in the expression of marker proteins and the anti-inflammatory effect of flavonoids isolated from Korean C. aurantium L.</P>

        Reconstruction of Medial Orbital Wall Fractures without Subperiosteal Dissection: The "Push-Out" Technique

        Background Various surgical methods for repairing medial orbital wall fractures have been introduced. The conventional technique requires total separation of the displaced orbital bones from the orbital soft tissues. However, subperiosteal dissection around the fracture can cause additional damage. The aim of the present study is to introduce a method of reconstructing medial orbital wall fractures without subperiosteal dissection named the "push-out" technique. Methods Six patients with post-traumatic enophthalmos resulting from an old medial orbital wall fracture and 10 patients with an acute medial orbital wall fracture were included. All were treated with the push-out technique. Postoperative computed tomography (CT) was performed to assess the correct positioning of the implants. The Hertel scale and a comparison between preoperative and postoperative orbital volume were used to assess the surgical results. Results Restoration of the normal orbital cavity shape was confirmed by examining the postoperative CT scans. In the old fracture group, the median orbital volume of the fractured side was $29.22cm^3$ preoperatively, and significantly improved postoperatively to a value of $25.13cm^3$. In the acute fracture group, the median orbital volume of the fractured side was $28.73cm^3$ preoperatively, and significantly improved postoperatively to a value of $24.90cm^3$. Differences on the Hertel scale also improved, from 2.13 mm preoperatively to 0.25 mm postoperatively in the old fracture group and from 1.67 mm preoperatively to 0.33 mm postoperatively in the acute fracture group. Conclusions The push-out technique can be considered a good alternative choice for old medial orbital wall fractures with posttraumatic enophthalmos, acute medial orbital wall fractures including large fractured bone segments, and single-hinged greenstick fractures.

        Anti-oxidant and anti-inflammatory effects of Fraxinus rhynchophylla on lipopolysaccharide (LPS)-induced murine Raw 264.7 cells

        Fraxinus rhynchophylla (Oleaceae), a deciduous tree, is known to have properties that include anti-inflammatory, convergence, febricide, antiblenophthalmia, antidiarrhea, antileukorrhea, and so forth. In addition, it has been used for traditional herbal medicine in East Asian countries, including Korea. In this study, we investigated the antioxidant and anti-inflammatory effects of Fraxinus rhynchophylla ethanol extract (FRE) in lipopolysaccharide (LPS)-induced murine macrophage Raw 264.7 cells with FRE pretreatment. We performed DPPH-assay, Western blot, and Reverse Transcription-Polymerase Chain Reaction (RT-PCR). FRE showed 85% free radical scavenging activity at concentrations of 80 μg/ml. Results of this study also showed that FRE down-regulates Cox-2 and iNOS expression in mRNA and protein level. In conclusion, crude ethanol extract of Fraxinus rhynchophylla exhibited antioxidant and anti-inflammatory activities, and it may potentially provide a valuable source of natural herbal agent to inhibit inflammation.
