        Suppression of Matrix Metallopeptidase-3 Expression in Human Dermal Fibroblasts by Decursin from Angelica gigas Nakai Root Extracts Fermented with Jeju Lava Seawater

        목적: 최본 연구는 제주 용암해수로 숙성된 참당귀 추출물의 인간 진피섬유아세포 내 콜라겐분해효소 발현 감소 효과를 분석하고 그 효과에 대한 지표물질을 파악하는 것을 목적으로 진행되었다. 방법: 제주 용암해수를 이용해 저온숙성하여 추출한 참당귀 추 출물의 지표물질 확인을 위해 고성능 액체 크로마토그래피(HPLC) 분석을 진행하였다. 참당귀 뿌리 추출물 및 decursin의 인간 진 피섬유아세포 내 세포생존율을 측정하고 실시간 유전자증폭 정량(qRT-PCR) 분석을 통한 콜라겐분해효소 발현 변화를 분석하였 다. Decursin으로 진행한 결과와 참당귀 뿌리 추출물의 콜라겐분해효소 발현 감소 효과를 비교하여 그 상관관계를 파악하였다. 결 과: HPLC 분석 결과, 참당귀 추출물 내 주요한 지표물질로 decursin이 확인되었다. 인간 진피섬유아세포에서 참당귀 뿌리 추출물 과 decursin은 각각 200 μg/mL, 20 μM 미만의 농도에서 세포 생존율에 낮은 영향을 보였다. 또한, MMP3 발현에 대한 참당귀 뿌 리 추출물과 decursin의 감소 효과를 qRT-PCR을 통해 확인하였다. 결론: 본 연구결과를 통해 제주 용암해수로 숙성된 참당귀 뿌 리 추출물과 이 추출물로부터 순수분리된 지표물질 decursin의 콜라겐분해효소인 MMP3 발현 감소 효능을 확인하였으며, 이를 통 해 참당귀 뿌리 추출물 및 decursion은 주름개선용 기능성화장품 소재로 활용 가능함을 과학적으로 증명하였다. Purpose: This study aims to understand the role Angelica gigas (A. gigas ) Nakai root extract (AGNRE) fermented with Jeju lava seawater in collagenase suppression in human dermal fibroblasts, and identify the major active compound responsible for it suppressive effect. Methods: AGNREs were prepared by fermentation with Jeju lava water at low temperature and analyzed for identifying the major active compound in these fermented root extracts using high-performance liquid chromatography (HPLC). Water-soluble tetrazolium salt (WST-1) assay and real-time polymerase chain reaction (qRT-PCR) were performed to investigate the cell viability and expression level of collagenase gene. Comparative experiments were performed using AGNREs and decursin to confirm the downregulation of collagenase expression and analyze the correlation between them. Results: HPLC analysis revealed decursin to be the major active compound in AGNREs. Analysis of data obtained by WST-1 assays at concentration of 200 μg/mL for AGNREs and <20 μM for decursin did not show any cytotoxicity in human dermal fibroblasts. qRT-PCR analysis revealed that AGNREs as well as decursin downregulated the expression of matrix metalloproteinases-3 (MMP3 ) gene in human dermal fibroblasts. Conclusion: These analyses suggest that AGNREs fermented with Jeju lava water as well as decursin isolated from these extracts hold a great potential to be applied as functional anti-wrinkle agent in cosmetic.

      • The Mechanical Aspects of Formation and Application of PDMS Bilayers Rolled into a Cylindrical Structure

        <P>A polydimethylsiloxane (PDMS) film with its surface being oxidized by a plasma treatment or a UV-ozone (UVO) treatment, that is, a bilayer made of PDMS and its oxidized surface layer, is known to roll into a cylindrical structure upon exposure to the chloroform vapor due to the mismatch in the swelling ratio between PDMS and the oxidized layer by the chloroform vapor. Here we analyzed the formation of the rolled bilayer with the mechanical aspects: how the mismatch in the swelling ratio of the bilayer induces rolling of the bilayer, why any form of trigger that breaks the symmetry in the in-plane stress level is needed to roll the bilayer uniaxially, why the rolled bilayer does not unroll in the dry state when there is no more mismatch in the swelling ratio, and how the measured curvature of rolled bilayer matches well with the prediction by the theory. Moreover, for the use of the rolled bilayer as the channel of the microfluidic device, we examined whether the rolled bilayer deforms or unrolls by the flow of the aqueous solution that exerts the circumferential stress on the rolled bilayer.</P>

        다채널 멀티미디어 전송용 임베디드 Audio Video Bridging 플랫폼 설계 및 구현

        본 논문에서는 고품질 멀티미디어 데이터를 IEEE 802.1 Audio Video Bridging 기반의 네트워크를 통해 스마트 자동차와 스마트 홈 및 스마트 공연장에서 실시간으로 송수신할 수 있는 임베디드 플랫폼을 설계를 통해 정밀한 시간 동기화 표준인 IEEE 802.1AS와 QoS 보장을 위한 자원할당 표준인 IEEE 802.1Qat 프로토콜 분석을 통해 성능을 검증한다. 이를 위해 MRP(Multiple Registration Protocol), MMAP(Multicast Address Acquisition Protocol), IEEE 1722, 1722.1 등 AVB의 핵심 Layer-2 프로토콜을 리눅스 운영체제를 기반으로 구현한다. 아울러 상용 AVB 엔드포인트와 연동 실험을 통해 상호 호환성 및 멀티미디어 스트림의 실시간 송수신이 가능함을 보인다. In this paper, we designed an embedded audio video bridging (AVB) platform based on IEEE 802.1BA for real-time multimedia transmission in smart-car, smart-home, smart-theater, and then evaluated a performance of the implemented platform by analysis of IEEE 802.1AS (time synchronization protocol) and IEEE 802.1Qat (stream reservation protocol). Especially, the AVB Layer-2 protocol of MRP(Multiple Registration Protocol), MMAP(Multicast Address Acquisition Protocol), IEEE 1722, 1722.1 etc. was and implemented by linux based operating system. It is shown by interoperability tests with commercial products that the implemented platform transmits real-time multichannel AV data over AVB networks for Multichannel Multimedia Transmission.

        요오드화수은(Mercury Iodide, HgI2)은 높은 원자번호, 고밀도, 효율적인 밴드갭, 그리고 높은 비저항의 특성을 가지기 때문에 방사선 영상 검출기로 적용하기 위한 연구가 활발히 이루어져 왔다. 기존 연구 자료들에 의하면, HgI2 광도전체는 직접 변환 방식의 검출 물질로 높은 민감도 특성, 낮은 누설전류 등의 우수한 특성을 가지고 있으나 제작된 방사선센서는 재현성과 신뢰성에서 문제점을 가지고 있기 때문에 상용화에 제약을 가지고 있었다. 본 연구에서는 HgI2 필름의 제작 공정에 있어서 산화물을 첨가함으로써 제작된 영상 센서의 재현성과 신뢰성을 향상시키기 위한 연구가 수행되었다. 미세 사이즈의 산화물의 첨가는 변환 층의 충진율을 높여줄 뿐만 아니라 전하 전달 효율을 높여 주기 때문에 전하 트랩효율을 감소시킬 수 있다. 제작된 HgI2 필름의 형태적, 전기적 특성을 평가함으로써 최적의 제작조건과 산화물 첨가비를 최적화한 이후, 방사선 영상 센서를 제작하여 엑스선 영상을 획득함으로써 직접 변환 방식의 영상 센서로써의 가능성을 검증하였다. Mercury Iodide(HgI2) photoconductor has been widely studied for the application of radiation image sensor due to its inherent properties such as high atomic number, high density, sufficient band-gap energy, and high resistivity. Though HgI2 photoconductor has the excellent properties like high sensitivity and low leakage current, it is not commercialized due to the problem of reproducibility and stability. In this paper, the electrical properties of fabricated HgI2 film with mixing oxidation materials in HgI2 fabrication process were improved. The addition of fine-size oxidation materials enabled to increase filling ratio of conversion layer and reduce the charge trap efficiency. The fabrication process and the mixture ratio of oxidation materials were optimized by evaluating morphological and electrical properties of the fabricated HgI2 compound film. Finally, from the fabricated HgI2 compound image sensor, phantom images were acquired by incident x-ray energy and the applicability of direct conversion X-ray detector was verified.

      • 산화물을 첨가한 HgI₂ 엑스선 검출기의 특성 평가

        To improve the performance of HgI2 composite x-ray detector, we studied its structure that reduced charge trap and estimated electrical properties as incident x-ray energy. The composite detector exhibits lower performance in x-ray sensitivity, dark current, and reproducibility than single-crystal detector or poly-crystal detector, because dielectric constant of polymeric glue and grain size of semiconductor particles affected charge transport properties. Therefore, in order to improve the charge transport properties, metal oxide such as SiO2 or TiO2 were added to HgI2 composite x-ray detector and its characteristics were investigated. The effect of metal oxide in HgI2 composite was analysed by comparing the properties of metal oxide mixed HgI2 composite x-ray detector with those of simple HgI2 composite x-ray detector. The samples were divided into three kinds of films, HgI2/TiO2 composite x-ray detector, HgI2/SiO2 composite x-ray detector, and simple HgI2 composite x-ray detector. All samples were fabricated by sedimentation method, one kind of particle-in-binder (PIB) methods. And the each thickness of them was around 200um. To investigate electrical properties of the samples, dark current of samples was measured as a function of applied bias. As a result, when 1 V/um of the electric field was applied, 4.78 nA/cm2, 16.58 nA/cm2, and 7.10 nA/cm2 were measured for HgI2/SiO2 composite x-ray detector, HgI2/TiO2 composite x-ray detector, and simple HgI2 composite x-ray detector. And then x-ray sensitivity of samples was measured in RQA-5 condition (70kVp, 100mA, 0.03s, 21mmAl phantom). The result respectively showed 23.15 nC/mR/cm2, 19.25 nC/mR/cm2, and 24.25 nC/mR/cm2 of x-ray sensitivity for the each samples. Furthermore, reproducibility, aging effect, and signal lag of samples were measured. Consequentially, the all measured results like dark current, x-ray sensitivity, reproducibility, and signal lag showed the best properties in HgI2/SiO2 composite x-ray detector among three kinds of samples

        CNN(Convolution Neural Network)은 합성곱(Convolution)을 이용해서 시각적 이미지를 분석하는데 사용되는 인공지능 기술이다. 본 논문에서는 CNN을 이용한 실시간 심리 상담 서비스에 대해 논한다. 상담서비스에 심리학과 CNN을 접목시킴으로써 내담자의 사진을 심리학적 비언어 행동을 기반으로 분석하여 내담자의 예상 심리를 파악하고, 유의미한 상담 자료를 생성해 상담의 질을 향상시킬 수 있도록 한다.

        Physicochemical Properties of the Fermentation of Poly Hydroxy Acid (PHA) using the Lactobacillus plantarum J2K-27 Strain

        목적: 본 연구는 Lactobacillus plantarum J2K-27 (L. Plantarum J2K-27) 유산균을 발효한 폴리하이드록시산(PHA)의 변화를 분 석하고 그 효능을 통해 화장품 소재로서 가능성을 확인해보았다. 방법: L. Plantarum J2K-27 균주를 이용한 PHA의 발효에 의한 성분변화를 확인하고자 고성능 액체 크로마토그래피(HPLC) 분석을 진행하였다. 발효된 PHA의 항산화 효능 및 세포생존율을 측 정하고 히알루론산 및 프로콜라겐 type 1 생성량 측정을 통해 화장품 소재로 가능성을 확인하였다. 결과: HPLC 분석 결과, 발효된 PHA는 락토바이오닉산(lactobionic acid)과 젖산(lactic acid)의 성분을 생성하는 것을 확인할 수 있었다. 또한, 발효된 PHA는 농도 의존적으로 항산화 효과를 나타내었으며, 세포 독성이 없는 농도에서 프로콜라겐 및 히알루론산의 생성량이 농도 의존적으로 증가 하는 것이 확인 되었다. 결론: 본 연구 결과를 종합하여, L. Plantarum J2K-27 균주를 이용한 PHA는 lactobionic acid과 lactic acid 의 성분을 생성하면서 기능성 화장품 소재로서 가능성이 있음을 확인하였다 Purpose: This study aimed to investigate the characteristics of fermented poly hydroxyl acid (F-PHA) using the Lactobacillus plantarum J2K-27 strain, and identify the applicability of its biological compounds. Methods: F-PHA was analyzed using high-performance liquid chromatography (HPLC) to identify its active compounds. Furthermore, the radical scavenging activities were evaluated for F-PHA. The cell viability and expression of procollagen type 1 and hyaluronic acid were investigated. Results: Changes in F-PHA represented the increase of lactobionic acid and lactic acid after fermentation. F-PHA increased radical scavenging activity in a dosedependent manner. After fermentation, F-PHA was confirmed to increase the expression of procollagen type 1 synthesis and hyaluronic acid without cytotoxicity. Conclusion: These results confirmed that F-PHA shows generally higher activities functional cosmetic material.
