Development of Marker-Free Transgenic Potato Hairy Roots Using Agrobacterium rhizogenes

Agrobascterium rhizogenes KCTC2744 and A. tumefaciens LBA4404 harboring the binary vector pBIl21 were used to transform potato plants. Transformation efficiency was monitored by detecting the β-glucuronidase (GUS) transcripts and the enzyme activity. The GUS expression of potato hairy roots regenerated by A. rhizogenes-mediated transformation was found to be approximately two-fold higher than that by A. tumefaciens-mediated transformation. All hairy roots were induced by A. rhizogenes on Murashige and Skoog medium without antibiotics. The hairy roots were transformed to pBI121 and pCAMbar-by A. rhizogenes KCTC2744. The GUS expression patterns, including hairy roots regeneration and staining characteristics, were investigated. No significant differences were observed between the general transgenic hairy roots and the selectable marker-free transgenic hairy roots. The polymerase chain reaction amplification and Southern blots confirmed a stable integration of foreign genes into all of the marker-free transgenic hairy roots. This is the first genetic engineering approach to the transgenic potato hairy roots genome without the use of a selectable marker gene.
Effects of anti-mullerian hormone supplementation on cryopreserved and transplanted ovarian tissues

Ovarian tissue cryopreservation and transplantation causes follicle depletion. To overcome this problem, we investigate the effect of Anti-Müllerian hormone (AMH), a follicle recruitment control hormone, supplementation before and/or after mouse ovarian transplantation. A total of 120 5-week-aged BD F-1 female mice were used. The mice were randomly divided into four groups according to AMH doses (0, 5, 25, 125 μg/mL, respectively). AMH was injected intraperitoneally on every other day for a week before, after, or before and after transplantation of ovaries under kidney capsules was performed. One week after transplantation, follicular normality was evaluated by histological analysis and TUNEL assay. In Group A and C, morphologically intact follicle (G1) ratios of AMH treated groups showed no statistically significant difference. In Group B, G1 ratios of 25 and 125 μg/mL of AMH treated groups were higher than those of 5 μg/mL treated group, but there was no improvement in G1 ratio after AMH treatment. In every group, apoptotic follicle ratios did not show any trend according to AMH treatment. Proportions of primordial follicle were not significantly different according to AMH treatment in all groups. The result of the present study demonstrated that AMH treatment during on transplantation of cryopreserved ovaries has no significant effect on follicle survival and prevention of follicle depletion.

<P><B>Abstract</B></P><P>Beta amyloid (Aβ) is believed one of the major pathogens of Alzheimer’s disease (AD), and the reduction of Aβ is considered a primary therapeutic target. Immunization with Aβ can reduce Aβ burden and pathological features in transgenic AD model mice. Transgenic potato plants were made using genes encoding 5 tandem repeats of Aβ1–42 peptides with an ER retention signal. Amyloid precursor protein transgenic mice (Tg2576) fed with transgenic potato tubers with adjuvant showed a primary immune response and a partial reduction of Aβ burden in the brain. Thus, Aβ tandem repeats can be expressed in transgenic potato plants to form immunologically functional Aβ, and these potatoes has a potential to be used for the prevention and treatment of AD.</P>

Purpose: Axillary lymph node dissection (ALND) and sentinel lymph node biopsy (SLNB) are important for staging of patients with node-posi�tive breast cancer. However, these can be avoided in select micrometastatic diseases, preventing postoperative complications. The present study evaluated the ability of axillary lymph node maximum standardized uptake value (SUVmax) on positron emission tomography-comput�ed tomography (PET-CT) to predict axillary metastasis of breast cancer. Methods: The records of invasive breast cancer patients who underwent pretreatment (surgery and/or chemotherapy) PET-CT between Janu�ary 2006 and December 2014 were reviewed. ALNs were preoperatively evaluated by PET-CT. Lymph nodes were dissected by SLNB or ALND. SUVmax was measured in both the axillary lymph node and primary tumor. Student t-test and chi-square test were used to analyze sensitivi�ty and specificity. Receiver operating characteristic (ROC) and area under the ROC curve (AUC) analyses were performed. Results: SUV-tumor (SUV-T) and SUV-lymph node (SUV-LN) were significantly higher in the triple-negative breast cancer (TNBC) group than in other groups (SUV-T: 5.99, P<0.01; SUV-LN: 1.29, P=0.014). The sensitivity (0.881) and accuracy (0.804) for initial ALN staging were high�er in fine needle aspiration+PET-CT than in other methods. For PET-CT alone, the subtype with the highest sensitivity (0.870) and negative predictive value (0.917) was TNBC. The AUC for SUV-LN was greatest in TNBC (0.797). Conclusion: The characteristics of SUV-T and SUV-LN differed according to immunohistochemistry subtype. Compared to other subtypes, the true positivity of axillary metastasis on PET-CT was highest in TNBC. These findings could help tailor management for therapeutic and diag�nostic purposes.

Lobular endocervical glandular hyperplasia (LEGH) is a benign lesion characterized by non-invasive proliferation of endocervical glandular cells. Adenoma malignum, known as minimal deviation adenocarcinoma of the uterine cervix is an extremely well-differentiated mucinous adenocarcinoma. The objective of this study was to look for the characteristic findings in LEGH and the major consideration in the differential diagnosis with adenoma malignum. We carried out a retrospective analysis of cases treated at our hospital from 2002 to 2012. All cases of LEGH and adenama malignum presented profuse watery vaginal discharge, and transvaginal ultrasonography and MRI findings showed multicystic lesion. Pap smear was usually normal and conization was the main diagnostic method, however conization was not appropriate in cases of deep and highly located lesion. In these cases, we performed laparoscopic excisional biopsy or hysteroscopic biopsy with frozen section for differential diagnosis of adenoma malignum from LEGH. LEGH, LEGH with adenocarcinoma in situ, and adenoma malignum present similar clinical findings. Due to apparent difference in the treatment, an accurate preoperative differential diagnosis is necessary. Conization is the diagnostic method of choice. But, in cases of deep and highly located lesion or where conization could not include the exact lesion, further evaluation such as laparoscopic excisional biopsy or hysteroscopic biopsy with frozen section instead of conization only shoule be considered.
자궁경부의 원발성 Signet Ring Cell Adenocarcinoma

Signet ring cell adenocarcinoma of the cervix is most commonly considered to be metastatic in origin. We describe one case of primary signet ring cell adenocarcinoma of the cervix occuring in 49-year-old patient. The lesion was located at the posterior lip of the cervix and about 3 cm in size. This case was parametrium thickening in pelvic examination and positive for HPV type 18. CCRT (concurrent chemoradiotherapy) was done for clinical FIGO stage ⅡB adenocarcinoma of the cervix. The patient is alive for 6month since diagnosis and disappear lesion of the cervix. A upper gastroenteroscopy, colonoscopy, cystoscopy, intravenous pyelogram, abdominal pelvic CT, PET CT, mammogram were reported negative. Therefore we concluded this case for a primary cervical origin of signet ring cell adenocarcinoma of the uterine cervix.

백제금동대향로는 사비시대 백제를 대표하는 최고의 금속공예품이다. 그런데 이 향로의 출토장소가 陵寺라는 先王의 명복을 비는 추모사찰이며, 또 불교의 상징인 세 겹으로 된 연잎이 묘사되어 있음에도, 지금까지는 여기에서 묘사된 산을 박산이라는 신선사상과만 연결시켜서 이해했다. 그러나 본고는 백제금동대향로의 산형이 박산이 아닌 불교의 수미산일 개연성을 모색해 본 것이다. 이와 같은 주장근거는 다음과 같다. 첫째, 불교의 수미산과 박산이 중국불교 안에서, 상호간의 영향관계에 의해 형태적인 유사성을 확보하고 있다는 점. 둘째, 신선은 육체를 가진 존재이며 박산 역시 삶의 세계이므로, 사후라는 추모의 의미와는 맞지 않는다는 점. 셋째, 발견 장소가 능사이며 향로에 연잎이 묘사되어 있어, 불교적인 영향이 분명하게 확인된다는 점. 넷째, 백제를 대표하는 또 다른 유물인 옥충주자의 <수미산도>에 백제 금동대향로와 매우 유사한 모습이 표현되어 있다는 점. 다섯째, 수미산정인 도리천은 불교의 최고 이상군주인 전륜성왕이 죽은 뒤에 재탄생하게 되는 장소라는 점. 이와 같은 근거들에 입각하여, 본고에서는 백제금동대향로의 산형이 기존의 박산주장과는 달리 수미산으로도 볼 수 있다는 견해를 제시하였다. 박산설의 근거가 중국박산향로와의 형태적인 유사성만을 가지는 것에 비해, 수미산 주장은 형태와 내용적인 면에서 보다 높은 정합성이 확보된다. 또 이를 통해서 단순히 박산향로의 진일보가 아닌, 백제불교와 백제문화의 특징을 정립해 볼 수도 있다는 점에서 본고는 충분한 연구의의를 확보한다고 판단된다. Baegje Geumdong Daehyangro(金銅大香爐, great golden incense burner) is one of the typical metallic artifacts from Late Baegje. This incense burner was exhumed at Neungsa(陵寺) a temple to cherish and pray for the blessing of deceased former kings. And it has triple lotus petals a symbol of Buddhism. In spite of it, this burner has been considered only in connection with Sinseon(神仙) under the presupposition that the mountain depicted on it was Bagsan(博山). But, this paper seeks the possibility that the mountain form on Baegje Geumdong Daehyangro is not Bagsan but Mt. Sumeru of Buddhism. The grounds for such assertion are as follows: First, Mt. Sumeru of Buddhism and Bagsan have similar forms under the correlative influence in Chinese Buddhism. Second, as Sinseon has the flesh and Bagsan also exists in the world of life, they are not well appropriate for cherishing the deceased. Third, as it was found at Neungsa and the incense burner was decorated with lotus leaves, there must have been some Buddhist influence on it. Fourth, in < Sumisando(須彌山圖, painting of Mt. Sumeru) > of Ogchungjuja(玉蟲廚子) another typical artifact of Baegje, there is a very similar form as that on Baegje Geumdong Daehyangro. Fifth, Trayastrimsa(?利天) on the top of Mt. Sumeru is the place where the cakra-varti-rajan(轉輪聖王) the supreme ideal king of Buddhism is reborn after death. Based on above grounds, this paper suggests that the mountain form on Baegje Geumdong Daehyangro can be reckoned as Mt. Sumeru rather than Bagsan of conventional theories. While the Bagsan theory(博山說) is based on the fact that it is similar in the form as Chinese Bagsan Incense Burner(中國博山香爐), this Mt. Sumeru theory can have greater validity both in the form and the contents. In that we can find out the uniqueness of Baegje culture and Baegje Buddhism through it rather than the simple development of Bagsan burner, this study will have sufficient significance.
