Effects of ginsenoside on survival of human embryonic stem cell-derived cardiac cells after thawing

Human embryonic stem cells (hESCs) are promising cell source because of their unique self-renewal and pluripotency. Although hESC-derived cardiac cells are currently generated worldwide, cryopreservation of these cells is still limited due to low rate of post-thaw survival. Cryopreservation of hESC-derived cardiac cells is critical in that their long-term storage can accelerate their use in regenerative medicine. However, to date, there are few reports on efficient cryopreservation and post-thaw survival of hESC-derived cardiac cells. In this study, we evaluated the effects of ginsenoside, which is known to improve survival of rat embryonic cardiomyocytes against myocardial ischemia injury in diabetic rats (Wu et al., 2011), on the survival of hESC-derived cardiac cells after thawing. We induced differentiation into cardiac cells using our previously reported method (Kim et al., 2011). Differentiated, pre-beating stage cardiac cells were cryopreserved using either mass cryopreservation or vitrification. To evaluate the effects of ginsenoside (Re, Rb), we compared three sets: pre- and post-thaw treatment, pre- or post-thaw treatment only. The survival of post-thaw cardiac cells were evaluated using Trypan-blue and Annexin V staining. In addition, the three groups were treated with ROCK inhibitor Y-27632, and compared with non-treatment groups. The effect of ginsenoside was significant in post-thaw treatment group, i.e, thawed cells expressed cardiac specific genes and showed specific functionality such as spontaneous beating. Taken together, we demonstrated favorable effects of ginsenoside on the survival of hESC-derived cardiac cells after cryopreservation and thawing. These results suggest a possible application of well-known cardioprotectant ginsenoside in cell-based tissue engineering using hESC-derived cardiac cells.
태권도 호신술 트레이닝이 중년 여성의 뇌유래신경영양인자와 마이오카인 카텝신 B에 미치는 영향

목적 본 연구의 목적은 12주간의 태권도 호신술 트레이닝 프로그램이 중년 여성의 BDNF, IGF-1과 마이오카인 CTSB에 미치는 영향을 규명하는 것이다. 방법 중년 여성 20명을 대상으로 대조군(CG: 10명), 태권도 호신술 군(TSG: 10명)으로 무선 배정하였으며 태권도 호신술 트레이닝군은 1회 60분, 주 4회, 총 12주간 트레이닝 프로그램에 따라 운동을 실시하였다. 태권도 호신술 트레이닝 시작 전과 후에 각각 신체 조성, 기초체력검사와 혈액 채혈을 실시하였다. 결과 12주간 태권도 호신술 트레이닝에 따른 배근력, 팔굽혀펴기, 사이드스텝, 높이뛰기, 혈청 BDNF, IGF-1, CTSB에서 상호작용 효과가 나타났으며(P<.05), 이에 대한 주 효과 분석 결과 그룹 간 차이에서 사후 CG와 비교하여 TSG에서 유의하게 증가한 것으로 나타났다(P<.05). 또한 태권도 호신술 트레이닝에 따른 CTSB와 BDNF의 변화량은 유의한 정적 상관(r=0.881, P=0.001)이 나타났다. 결론 태권도 호신술 트레이닝은 BDNF, IGF-1와 마이오카인 CTSB를 증가시키는 데 효과적이었으며, 태권도 호신술 트레이닝 프로그램으로 인한 CTSB와 BDNF의 변화량은 유의한 상관관계가 나타나 CTSB의 증가에 따른 BDNF의 향상이 관계가 있음을 시사한다. 따라서 태권도 호신술 트레이닝은 노화의 진행 단계에 있는 중년 여성에서 기초체력을 증가시킬 뿐만 아니라 뇌기능과 관련된 마이오카인을 증가시킴으로써 신경영양인자의 발현을 향상시켜 인지 기능에 긍정적인 영향을 미치는 것으로 사료된다. Purpose The purpose of this study was to investigate the effects of 12-week taekwondo self-defense training program on BDNF, IGF-1 and myokine CTSB in middle-aged women. Method 20 middle-aged women over the age of 45 were randomly assigned to the control group(CG, n=10) and the taekwondo self-defense group(TPG, n=10). Taekwondo self-defense group exercised according to training programs for 60 minutes once, 4 times a week, for a total of 12 weeks. Body composition, fitness test and blood collection were performed before and after 12 weeks of taekwondo training, respectively. Results BDNF significantly increased after training in TSG compared to before training, and significantly increased in TSG compared to CG after training. IGF-1 significantly increased after training in TPG and TSG, and CTSB significantly increased after training in TSG only. In addition, there was a significant positive correlation between the delta values of serum CTSB and BDNF due to taekwondo training. Conclusion Taekwondo self-defense training was effective in increasing BDNF, IGF-1 and myokine CTSB. The delta values of CTSB and BDNF from the Taekwondo self-defense training program showed a significant correlation, suggesting that the increase of CTSB is associated with the improvement of BDNF. Therefore, Taekwondo self-defense training is thought to have a positive effect on cognitive function by enhancing the expression of neurotrophin by increasing the myokine related to brain function as well as increasing physical fitness in middle-aged women in the advanced stage of aging.

The study is analyzing real-world situations to analyze self-defense technology systems and functional effects. In particular, it emphasizes world Taekwondo and emphasizes real-world Taekwondo. This study analyzes the unique characteristics and efficiency of real-world Taekwondo through Taekwondo. The results of the investigation show that Taekwondo can improve its ability to defend against real-world threat scenarios through fast attack technology. In particular, successful attacks help prepare for defense and auxiliary attacks. It also emphasizes the functional value and practicality of real Taekwondo by analyzing attack techniques. This study verifies Taekwondo's self-defense capabilities. Continuous research and standardization are necessary to maximize the effectiveness against real-world threat situations. Moreover, this study suggests a future direction to improve Taekwondo's practicality.
Low Cost and High Performance Single Phase UPS Using a Single-Loop Robust Voltage Controller

Uninterruptible Power Supplies (UPSs) can be largely divided into the passive-standby, line-interactive and double-conversion methods. This paper proposes a double-conversion UPS with a low cost and high performance. This single phase UPS uses a single-loop robust voltage controller and 1-switch voltage doubler strategy PFC. The proposed UPS is composed of a single phase PFC, a half-bridge inverter, a battery charger and a battery discharger. Finally, the validity of proposed UPS was verified by various experimental tests.
Chitosan/poly(L-lactic acid) multilayered membrane for guided tissue regeneration

<P>A chitosan/poly(L-lactic acid) (PLLA) multilayered membrane was prepared for the purpose of guided tissue regeneration (GTR). The membrane was composed of the outer layers of chitosan mesh and the middle layer of nanoporous PLLA membrane. The outer membranes of chitosan fibrous mesh present a highly biocompatible and rough surface for ease of cell adherence. The PLLA membrane is intended to provide mechanical strength to the membrane, thereby preventing epithelial invasion. The PLLA membrane was applied as a local drug delivery carrier for growth factors. Regular pores in PLLA membrane were generated through phase inversion of polymer solution. The membrane retained its integrity during the degradation process in vitro (for up to 8 weeks), which is a requirement for providing enough space in vivo in the GTR procedure. Osteoblasts firmly attached to broad cytoplasmic extensions along with the microfibers of the membrane. The phenotypic expression of cultured osteoblasts on the membrane was measured by a reverse transcriptase-polymerase chain reaction. New bone had formed beneath the chitosan/PLLA membrane. These results suggested that the chitosan/PLLA membranes properly functioned as biocompatible and mechanically stable barriers for GTR. © 2008 Wiley Periodicals, Inc. J Biomed Mater Res, 2009</P>
