        A nucleotide variant in promoter of the human CDH13 gene which affects its transcription activity is associated with colorectal cancer

        T-cadherin is frequently down regulated in various cancers, however, the underlying mechanisms responsible have yet to be elucidated. A genome wide association study of a cohort of Korean adults revealed that the T-cadherin rs3865188 single nucleotide polymorphism (SNP) was associated with serum Adiponectin levels and that its genotypic variants were correlated with risk for colorectal cancer (CRC). To test the function of rs12444338, a SNP tightly linked to the rs3865188 SNP, in T-cadherin transcriptional regulation in colorectal cancer, its effect on transcriptional activity and the capacity of binding activity attributable to allelic variation of the rs12444338 SNP was investigated. An electrophoretic-mobility-shift assay (EMSA) revealed a specific nucleoprotein complex unique to the T allele probe, which displayed lower promoter activity when compared to the G allele. Based on the results of the EMSA using mutant probes, the consensus sequence of the putative transcription factor binding site was determined. Additionally, candidates for putative binding factors to the T allele were also identified. Collectively, the study data suggested that the rs12444338 SNP was involved in transcriptional regulation of T-cadherin gene (CDH13) and that the differential binding of transcription factors at the rs12444338 SNP resulted in altered gene expression. These results elucidate, at least in part, the regulation of T-cadherin expression in CRC and provide further information regarding the effect of nucleotide variation in its promoter region.

      • Structural and functional study of ChuY from <i>Escherichia coli</i> strain CFT073

        <P><B>Abstract</B></P> <P>The uropathogenic <I>Escherichia coli</I> strain CFT073 contains multiple iron and heme transport systems, which facilitate infection of the host urinary tract. To elucidate the molecular and cellular function of ChuY, a hypothetical gene in the heme degradation/utilization pathway, we solved the crystal structure of ChuY at 2.4 Å resolution. ChuY has high structural homology with human biliverdin and flavin reductase. We confirmed that ChuY has flavin mononucleotide (FMN) reductase activity, using NAD(P)H as a cofactor, and shows porphyrin ring binding affinity. A <I>chuY</I> deletion-insertion strain showed reduced survival potential compared to wild-type and complemented strains in mammalian cells.         Design and Methodology for the Korean Observational and Escitalopram Treatment Studies of Depression in Acute Coronary Syndrome: K-DEPACS and EsDEPACS

        Depression is common after acute coronary syndrome (ACS), adversely affecting cardiac course and prognosis. There have been only a few evidence-based treatment options for depression in ACS. Accordingly, we planned the Korean Depression in ACS (K-DEPACS) study, which investigated depressive disorders in patients with ACS using a naturalistic prospective design, and the Escitalopram for DEPACS (EsDEPACS) trial, which assessed the efficacy and safety of escitalopram for treating major or minor depression in patients with ACS. Participants in the K-DEPACS study were consecutively recruited from patients with ACS who were recently hospitalized at Chonnam National University Hospital, Gwangju, South Korea. Diagnoses were confirmed by coronary angiography from 2005. Data on depressive and cardiovascular characteristics were obtained at 2 weeks, 3 months, 12 months, and every 6 months thereafter following the index ACS admission. The K-DEPACS participants who met the DSM-IV criteria for major or minor depressive disorder were randomly assigned to groups in the 24-week, double-blind, placebo-controlled EsDEPACS trial beginning in 2007. The outcome of treatments for depressive and other psychiatric symptoms, issues related to safety, including general adversity, and cardiovascular factors were assessed. The K-DEPACS study can significantly contribute to research on the complex relationships between depression and ACS. The results of the EsDEPACS trial provide an additional treatment option for clinicians treating these patients.

        Compact 트랙터용 기어타입 펌프는 경운, 이앙, 파종, 운반 등을 수행하기 위해 작업기에 필요한 동력을 전달해주는 핵심 유압부품이다. 트랙터 작업조건에 따라 기어타입 펌프의 회전속도를 가변시켜 사용하기 때문에 압력과 유량에 대한 성능 효율을 유지하는 것이 어렵다. 이에 본 연구에서는 Compact 트랙터용 기어타입 펌프의 작업 효율 향상 검증을 위해 성능 특성 분석을 진행하였다. 효율(기계, 용적, 전효율)시험은 농기계용 기어타입 펌프 신뢰성 평가기준에 따라 회전속도(600 ~ 2,400 rpm) 600 rpm, 작동압력(6 ~ 24 MPa) 6 MPa단위로 회전속도 대비 압력 16개 포인트에서 효율을 산출하였다. 효율시험 분석결과 1,800 rpm - 12 MPa, 18 MPa과 2,400 rpm - 18 MPa, 24 MPa에서 높은 효율을 보였으며, 회전속도 600 rpm, 1,200 rpm에서는 압력과 상관없이 낮은 효율을 보였다. 향후 본 연구 결과를 활용하여 Compact 트랙터용 기어타입 펌프의 효율 향상 연구에 활용할 예정이다.

        <P>The sigmaE-dependent stress response in bacterial cells is initiated by the DegS- and RseP-regulated intramembrane proteolysis of a membrane-spanning antisigma factor, RseA. RseB binds to RseA and inhibits its sequential cleavage, thereby functioning as a negative modulator of this response. In the crystal structure of the periplasmic domain of RseA bound to RseB, the DegS cleavage site of RseA is unstructured, however, its P1 residue is buried in the hydrophobic pocket of RseB, which suggests that RseB binding blocks the access of DegS to the cleavage site.</P>

        α-Glucosidase 저해제 생산 균주, Gluconobacter oxydans CK-2165의 특성 및 배양 최적화

        본 논문은 당뇨병 치료제로 알려진 Miglitol은 α-glucosidase 저해제로, 산업적으로 포도당과 에탄올아민으로부터 세 단계의 화학 및 생물전환 과정을 거쳐 합성되며. acetic acid bacteria에 속하는 Gluconobactor oxydans (G. oxydans)는 불완전 산화를 통해 1-deoxy-1-(2-hydroxyethylamino)-D-glucitol (P1)을 Miglitol의 전구체인 6-(2-hydroxyetyl) amino-6- deoxy-α-L-sorbofuranose (P2)로 생물 전환시키는 균주이다. 본 연구에서는 토양으로부터 스크리닝하여 선발된 고효율 생 물전환 박테리아인 CK-2165의 균주를 동정하고 최적의 발효조건을 탐색하고자 하였다. 16S rDNA 서열과 계통수 분석결 과 CK-2165는 G. oxydans에 속하는 미생물임을 결정하였으며, API 20E kits를 사용하여 선발된 균주의 탄소원 이용성을 실험한 결과 glucose, mannose, inositol, sorbitol, rhamnose, sucrose, melibiose, amygdalin, arabinose와 같은 탄소원에 대한 이용성을 가진 균주임을 확인하였다. 또한 산업적 생산을 위하여 배양 조건을 최적화하였고 균체를 이용하여 생물전환 반응에 중요한 요소들을 조사하였다. 생물전환 반응을 위해 사용되는 균체는 균 성장 단계 중 후기 정지기 (late-stationary phase)에 수확한 균체가 가장 높은 활성을 나타내었고 생물전환 반응에는 MgSO<sub>4</sub>가 필수적임을 확인하였다.

        Electronic structures of CO adsorbed Pt-skin surface on Pt–Co and Pt–Ni alloys

        By using angle resolved photoemission spectroscopy, we investigate the electronic structures of Pt-skin layer of Pt–Co and Pt–Ni alloys with CO molecules on the surface. Measured Fermi surface maps and band dispersions reflect the signatures of chemical bonding between Pt-skin layer and CO molecules. Furthermore, the degree of chemical bonding strength of CO molecules, estimated from the energy shift of the participating bands, is found to be reduced on both Pt bimetallic alloys. Our results show how the surface band structure of Pt bimetallic alloys is modified with molecular orbitals of CO molecules on the surface, revealing the important role of the electronic structure in the determination of chemical properties of bimetallic alloys.

      • Gene-gene interaction analysis identifies a new genetic risk factor for colorectal cancer

        <P><B>Background</B></P><P>Adiponectin levels have been shown to be associated with colorectal cancer (CRC). Furthermore, a newly identified adiponectin receptor, T-cadherin, has been associated with plasma adiponectin levels. Therefore, we investigated the potential for a genetic association between T-cadherin and CRC risk.</P><P><B>Result</B></P><P>We conducted a case–control study using the Korean Cancer Prevention study-II cohort, which is composed of 325 CRC patients and 977 normal individuals. Study results revealed that <I>rs3865188</I> in the 5’ flanking region of the T-cadherin gene (<I>CDH13</I>) was significantly associated with CRC (<I>p</I> = 0.0474). The odds ratio (OR) for the TT genotype as compared to the TA + AA genotype was 1.577 (<I>p</I> = 0.0144). In addition, the interaction between <I>CDH13</I> and the adiponectin gene (<I>APN</I>) for CRC risk was investigated using a logistic regression analysis. Among six <I>APN</I> single nucleotide polymorphisms (<I>rs182052</I>, <I>rs17366568</I>, <I>rs2241767</I>, <I>rs3821799</I>, <I>rs3774261</I>, and <I>rs6773957</I>)<I>,</I> an interaction with the <I>rs3865188</I> was found for four (<I>rs2241767</I>, <I>rs3821799</I>, <I>rs3774261</I>, and <I>rs6773957</I>). The group with combined genotypes of TT for <I>rs3865188</I> and GG for <I>rs377426</I> displayed the highest risk for CRC development as compared to those with the other genotype combinations. The OR for the TT/GG genotype as compared to the AA/AA genotype was 4.108 (<I>p</I> = 0.004). Furthermore, the plasma adiponectin level showed a correlation with the gene-gene interaction, and the group with the highest risk for CRC had the lowest adiponectin level (median, 4.8 μg/mL for the TT/GG genotype vs.7.835 μg/mL for the AA/AA genotype, <I>p</I> = 0.0017).</P><P><B>Conclusions</B></P><P>The present study identified a new genetic factor for CRC risk and an interaction between <I>CDH13</I> and <I>APN</I> in CRC risk.       • Long-term performance of anaerobic digestion for crop residues containing heavy metals and response of microbial communities

        <P><B>Abstract</B></P> <P>In order to investigate the long-term stability on the performance of the anaerobic digestion process, a laboratory-scale continuous stirred-tank reactor (CSTR) was operated for 1100 days with sunflower harvested in a heavy metal contaminated site. Changes of microbial communities during digestion were identified using pyrosequencing. According to the results, soluble heavy metal concentrations were lower than the reported inhibitory level and the reactor performance remained stable up to OLR of 2.0g-VS/L/day at HRT of 20days. Microbial communities commonly found in anaerobic digestion for cellulosic biomass were observed and stably established with respect to the substrate. Thus, the balance of microbial metabolism was maintained appropriately and anaerobic digestion seems to be feasible for disposal of heavy metal-containing crop residues from phytoremediation sites.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Stability of AD was evaluated for the treatment of heavy metal-containing crop residues. </LI> <LI> Changes of microbial communities during digestion were identified using pyrosequencing. </LI> <LI> AD process remained stable throughout the experimental periods without effects of heavy metals. </LI> <LI> Microbial communities did not suffer from heavy metals and conformed to the substrate. </LI> </UL> </P>
