        녹차 및 결명자 추출물의 교정용 브라켓과 치면 사이의 경계부에서 분리된 mutans streptococci에 대한 항균작용

        본 연구는 치아우식증에 관련된 Mutans streptococci 표준균주 및 임상분리 균주의 성장억제를 유도할 수 있는 천연 생약추출물을 검색하기 위하여 실시되었다. 녹차추출물은 CHMC-2032를 사용하였으며, 결명자 추출물은 50% 에탄올을 이용하여 얻었다. 이들 추출물의 Mutans streptococci 표준균주 및 교정환자들에서 브라켓과 치면 사이의 경계부에서 분리된 각각 10 균주씩의 S. mutans 및 S. sobrinus에 대한 최소성장억제농도를 액체배지 희석법으로 구하였다. 그 결과 CHMC-2032의 S. mutans 및 S. sobrinus의 표준 균주, S. sobrinus의 대부분 임상분리 균주에 대한 최소성장억제농도는 5 mg/ml이었다. 그러나 결명자 추출물에 의한 S. mutans 및 S. sobrinus에 대한 세균 성장억제 효과는 미미하였다. 본 연구 결과 치아우식증의 예방적 측면에서 결명자차보다는 녹차를 마시는 것이 유리하며, CHMC-2032를 이용하여 구강양치용액을 제조하여 사용할 경우 교정환자를 포함하여 대부분의 사람에게서 치아우식증 예방 효과가 있으리고 추정된다. Mutans streptococci is the major causative factor in dental caries. Especially, orthodontic patients with fixed appliance are a risk group for dental caries. Because fixed appliances attached on teeth may change the environment of dental plaque, the enamel decalcification or dental caries around the bracket and band is a major side effect of orthodontic treatment. The aim of this study was to search plant extracts that have antimicrobial effect on mutans streptococci. Seed-extract of Casio tora were prepared with ethanol and CHMC-2032, the leaf-extracts from Camellia sinensis extract, was obtained extract, 2 type strains and 20 clinical isolates of mutans streptococci isolated from the interface between orthodontic brackets and tooth surfaces in the orthodontic patients were used in this study. The minimal inhibitory concentration of CHMC-2032 was 5 mg/ml on the S. mutans KCTC 3065, S. sobrinus KCTC 3088, and 8 clinical isolates of S. sobrinus. However, there was no antibacterial effect of seed-extract of C. tora on mutans streptococci. These data suggest that green tea may be more effective than the tea prepared from C. tora in the prevention of enamel decalcification or dental caries around brackets.

        I have carried out a variance analysis on four hundred seventy four University students living in Chollabook-Do area to compare and analyse the characteristics between Sasang Constitutionand health related physical fitness. This survey is to provide basic data needed for excercise prescription of the collegians hereafter. The result of said survey shows below: 1. Muscular strength muscular endurance and cardiorespiratory endurance are not much effected by Sasang Constitution. 2. But flexibility and body at ratio are considerably varied according to each of Sasang Constitution. In conclusion, it is recommended to write excercise prescription toward college students under the consideration that the flexibility end body fat ratio are quite related to each of Sasang Constitution.

        The hydraulic excavator has been a popular research object for automation because of its multi-workings and economic efficiency. When it works crane tasks, most of disasters happen. The objective of this paper is to design each components and to construct boom, arm, bucket circuit. These models modeled with AMESim show us change of variables and behavior of excavator. Simulation model will be used for simulator of excavator.

      • 綠茶추출물에 의한 치매 치료 및 예방에 관한 연구

        Alzheimer's disease (AD) is characterized by amyloid deposition and associated loss of neunons in brain regions involved in learning and memory processes. Several causes of evidence support that the congnitive disturbance is closed associated with the deficit of cerebral acetylcholine neurotransmission, and the effect of carboxyl terminal 105 amino acid fragment (CT105) of the amyloid precursor protein (APP) on the gene expression of proinflammatory cytokines. We tested it on the scopolamine-induced amnesia model of the ICR mouse using the Moms water maze, with repeated orally administration of 1st Green-Tea extract (200 mg/kg) and 2nd Green-Tea extract (200 mg/kg). The Green-Tea prevents impairment of learning and memory and neuronal loss in mouse models of cognitive disturbance and it demonstrated selectivity for inhibition of acetylcholinesterase (AChE). Furthermore, the repeated administration of Green-Tea, CTlO5-induced alzheimer's mouse model showed central cholinergic activity by ameliorates learning and memory impairment, and isolation of CD14 microglia showed significantly decreases intracellular release of the proinflammatory cytokines tumor necrosis factor- a, interleukin-18 and reactive oxygen species (ROS). Because of its composite profile, oral therapeutic index and a prophylactic, Green-Tea is considered the better therapeutic candidate for the treatment of Alzheimer's disease.

      • 五加皮 (Acanthopanacis cortex)抽出物을 첨가한 발효김치의 급여가 생쥐의 免疫作用에 미치는 影響

        This experimental study was carried out to evaluate the effects of Kimchi intake of Acanthopanacis cortex extract (APCE) supplementation on cytokine-induction and immune response in mice. To study in experiments using male Balb/c mice fed Kimchi and Kimchi of APCE supplementation (addition of 2% of total Kimchi weight) containing fed experimental diet during 2 weeks. Experimental mice were fed control diet or diet containing freeze-dried Kimchi at the level of 5%(w/w) or 5% freeze-dried Kimchi with 2% APCE supplementation. The main ingredient of Kimchi was Korean cabbage and fermentation was carried out at 4C for three weeks. Freeze-dried 2% APCE supplementation was added to Kimchi at the begining of fermentation. In order to investigate the effect of Kimchi intake of APCE supplementation (5%Kimchi-2%APCE), the following was performed; body weight, food intake, hematological parameter, serum level of mouse interleukin-4 (mIL-4) and mouse interferon- y (mIFN- y ), and the percentage of CD3+/CD4+, CD3+/CD8+, B220+ in splenic cells. The results of final body weight, and food diet intake of two Kimchi groups were lower than those of the control group (not supplemented experimental diet). The hematology change obtained from the level of WBC (white blood cell) and platelet were not affected by feeding different dietary regiments, but the level of RBC (red blood cells) HB (hemoglobin), and spleen weight of two Kimchi groups were increased significantly than those of the control group. The serum level of IL-4 and IFN- Y of two Kimchi groups were increased significantly than those of the control group, also enhanced the percentages of the CD3+/CD4+ and CD3+/CD8+ bv 5% freeze-dried Kimchi. and 5%Kimchi-2%APCE group were 43.9 and 65.2%, and 96.0 and 208% than those of the control group, respectively. From these results, it can be concluded that Kirnchi itself has an immuno-stimulatory effect and Kimchi contaning 2% APCE supplementation has the more pronounced effect in vivo system.

      • 액체크로마토그래피/질량분석법을 이용한 랫트 혈장내의 Surfactin C의 정량법에 관한 연구

        Surfactin isolated from Bacillus subtilis has a strong surface tension-lowering activity, together with antiviral, antitumor, fibrinolytic and hypocholesterolemic activities. LC/MS is a simple, rapid and effective technique for the determination of surfactin C in rat serum. The separation of surfactin was achieved on reverse phase column. The mobile phase consisted of 0.1% trifluoracetic acid in water (A) and acetonitrile (B). As a result of analysis of blank rat serum samples, matrix interference was not shown. The values of precision and accuracy were satisfied w the guideline. A newly developed LC/MS method for the determination of surfactin C was successfully applied in its pharmacokinetic analysis.

        Acinetobacter sp. BE-254에 의한 유화제의 생산

        Bioemulsifier를 생산하는 미생물을 토양으로부터 screening하였다. 그 중에서 유화활성과 유화안정성에서 가장 우수한 BE-254 균주를 순수분리하여 동정한 결과, Acinetobacter속으로 판명되었다. Bioemulsifier 생산을 위한 최적배지 조성은 n-hexadecane 4%, NaNO_3 0.2%, KH_2PO_4 0.05%, MgSO_4·7H_2O 0.05%, CaCl_2 0.01%, yeast extract 0.01%이였으며, 최적온도와 pH는 각각 30℃와 7.0이였다. 이러한 조건에서 500㎖용 shaking flask에 최적배지 50㎖를 넣어 배양했을 경우 대수증식기 말기인 5일째 균의 증식과 유화활성이 가장 높게 나타남에 따라 Acinetobacter sp. BE-254에 의한 bioemulsifier의 생산은 균의 생육과 밀접한 관련이 있는 것으로 판단되었다. 유화기질로서 hydrocarbon류, edible oil류, 그리고 petroleum fraction등에 작용시켰을 경우 이들 물질에 대해서 비교적 높은 활성을 나타내어 유화제 로서의 우수성을 시사 하였다. The strain producing bioemulsifier was isolated from soil samples. The isolated strain was identified as the genus Acinetobacter through its morphological, cultural and physiological characteristics. The highest emulsification activity and stability by Acinetobacter sp. BE-254 was observed after 5 days of cultivation in the culture medium containing n-hexadecane 4%, NaNO_3 0.2%, KH_2PO_4 0.01%, MgSO_4·7H_2O 0.01%, CaCl_2 0.01% and yeast extract 0.01%. The optimum pH and temperature for bioemulsifier production were pH 7.0 and 30℃, respectively. Furthermore the most of bioemulsifier was produced during the exponential growth phase, and this suggested that the bioemulsifier production was growth-associated. The bioemulsifier showed good emulsification activity on various emulsifying substrates such as hydrocarbons, edible oils, and petroeum fractions.

        충전가능 콤포짓트 레진의 마찰력 및 마모량 비교

        When using posterior composite resins, wear resistance is still a problem. Recently condensable composite resins which have proper condensability like dental amalgam and increased wear resistance have been introduced. The purpose of this study was to compare the wear resistance of newly developed condensable composite resins with those of universal composite resins by measuring the amount of wear and frictional force with a friction tester (Friction Player, FPR-2000, Rhesca, Japan). Two condensable composite resins (Surefil; Dentsply, Synergy Compact; Coltene) and two universal composite resins (Z100; 3M, Spectrum; Dentsply) were studied. The surface of 3 min(D)×8 mm(L) specimens was polished with wet 600 grit SiC paper. The specimens of non-aging group were stored in 37 ℃, 75 % ethanol for 7 days. After fixing SiC paper on the turn table of friction tester, 500 g load was applied to the specimen. With the linear velocity of 0.1257 cm/sec, the wear procedure was done for 10 minutes in distilled water, which resulted in 1.27 m wear distance. Amount of wear was measured by microscopic measuring device of a micro hardness tester (Shimadzu, Japan) in 0.1㎛ scale, and friction force was calculated from the frictional force-time graph. Regardless of the aging, there was no significant difference in wear resistance between condensable composite resins and universal composite resins (p>0.05). In every test groups, there was some variation in frictional forces initially, however there was no significant change depending on time (p>0.05). There was a weak correlation between wear amounts and frictional force.

        광도와 온도가 가시광선 중합형 치과용 레진의 중합에 주는 영향

        본 연구에서는 2종의 다이메타크릴레이트 치과용 레진 (urethane dimethacylate, UDMA와 bisphenol A glycerolate dimethacylate, Bis-GMA)이 광중합 반응하는 과정에서 변화되는 물성을 동역학분석법 (dynamic mechanical analysis, DMA)으로 관찰하였다. 광중합에 사용한 가시광선 (λ=400-500 nm)의 광도는 20 mW/㎠에서 70, 120, 300 및 600 mW/㎠로 변화시켜 그에 따른 저장탄성율 (storage modulus, G')과 손실탄성율 (loss modulus, G")의 변화를 측정하였고, 측정온도도 25 ℃에서 37, 50 및 60 ℃로 변화시켜 온도가 중합과정에 주는 영향을 관찰하였다. 광조사 시간은 모든 시편에 대하여 측정 시작 직후 30초로 일정하게 하였다. 각 조건에서 측정한 G' 과 tan δ (=G"/G') 결과를 이용하여 겔화 (gelation)와 유리화 (vitrification) 전이시점 (transition point)을 결정하였으며, 이를 이용하여 TTIT (Time-Temperature-Intensity-Transformation) 중합 도표를 작성하였다. 이 중합도표를 보면 광도 변화가 온도 변화보다 중합반응에 더 민감하게 영향을 주었으며, 특히 광도는 겔화보다 유리화에 훨씬 크게 영향을 주는 것을 알 수 있었다.
