        FTIR을 이용한 브라질 Itabira 지역 에메랄드의 분광학적 특성 연구

        현재 에메랄드의 중요한 산지로 자리 잡아가고 있는 Itabira 지역 에메랄드의 분광학적 특성을 연구하기 위하여 에메랄드를 C-축에 수직, 수평인 방향으로 각각 절단하였으며, H2O 관련 범위의 좀 더 세부적인 피크 관찰을 위하여 KBr로 압축한 pellet시료를 만들어 적외선 분광분석(FTIR) 실험을 수행 하였다. H2O 관련 범위에서 5,271cm-1, 1,638cm-1 피크가 나타나는 것으로 보아 본 시료는 Type-Ⅱ에 해당함을 관찰할 수 있었으며, H2O-Na-H2O 배열 형태를 가지는 Type-Ⅱa에 해당하는 피크가 아주 강하게 나타나고 있다. H2O 관련 범위를 제외한 특정 범위에서는 CO2, Cl 피크 등이 관찰 되었으며, 적외선 분광분석법을 이용하여 타입의 분류뿐만 아니라 상대적인 피크 강도 비교를 통하여 Na2O 함량 또한 예측 가능한 것으로 사료된다. Emerald deposit located on Itabira, Brazil is one of the major one in the world. We applied three different analytical approaches on Itabira emerald samples, (1) perpendicular to the c-axis, (2) parallel to the c-axis, (3) emerald pallet mixed with KBr, using Fourier Transform Infrared (FTIR) spectroscopy, to understand and compare spectroscopic characteristics of three Itabira emerald samples. Occurrence of 5,271 cm-1 and 1,638 cm-1 absorption peaks within H2O-related range indicates that the samples belong to Type-II emerald. These emerald samples also display strong absorption peak generated from Type-IIa emerald preserving H2O-Na-H2O sequence. CO2- and Cl-related absorption peaks observed within specific range except for H2O-related range. These observations and results suggest that FTIR analysis can be used for not only classification of emerald types, but also prediction of Na2O content within the emerald through comparison of relative peak intensity.

        민간건강보험에서 정신과 질환에 대한 보험비 지급 관련 약관 현황과 문제

        Objectives : Recently, Korean psychiatrists have noticed the complaints from probands and family members that private health insurance companies do not pay for most psychiatric disorders. Furthermore, Probands cannot even apply for insurance because of their medical record of psychiatric disorders. Authors investigated and reviewed contracts of Korean private insurance companies to find reasons for banning psychiatric disorders from insurance policies. Methods : Authors reviewed more than 800 contracts from 48 insurance companies. Results : Among all the psychiatric diagnoses, few of them-dementia and some other organic mental disorders-are guaranteed to be paid from insurance companies. Less then 10 contracts say they pay for psychiatric illnesses. Most insurance companies have contracts prohibiting F codes ; however, there are not enough reasons in these contracts. Conclusions : In the private health insurance system, Psychiatric illnesses have almost no rooms. It is very urgent to add space for patients with psychiatric illnesses and psychiatrists in the insurance policies.

      • A pilot study of differential brain activation to suicidal means and DNA methylation of CACNA1C gene in suicidal attempt patients

        <P><B>Abstract</B></P> <P>Suicide is a serious public health problem. In this study, we investigated functional brain changes to novel visual stimuli of suicidal means, DNA methylation status, and the relationship between the two markers. 14 suicidal attempt patients (SAs) and 22 healthy controls were included. Pictures of facial expressions and suicidal means were shown to subjects during fMRI scanning. 11 CpG sites within transcription factor binding site of CACNA1C gene were selected. In knives (K) vs. natural landscape (NL) condition, left middle frontal gyrus (Brodmann's area, BA 6 and 46) and left inferior frontal gyrus (BA 9) were shown to be significantly higher brain activation in the SAs than the controls (p<0.001). DNA methylation percentages of CpG site 4 (p=0.005) and 6 (p=0.037) were found to be related to the SAs. In the SAs, methylation degree of site 4 and site 6 was positively correlated with signal intensity of K vs. NL condition in left thalamus. The degree of site 4 was positively correlated with signal intensity in left middle and inferior frontal gyri in SAs. The possibility that these findings might be involved in the neurobiology of suicidal behavior is suggested.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Functional brain change and DNA methylation were investigated in suicidal attempters. </LI> <LI> Distinct brain activation and DNA methylation were found in suicidal attempters. </LI> <LI> The possibility of a neurobiological evidence for suicide is suggested. </LI> </UL> </P>

      • A bioorthogonal approach for imaging the binding between Dasatinib and its target proteins inside living cells

        <P>Herein, we present a simple readout of the binding between a chemical drug and its target proteins in the cytoplasm by using a two-step bioorthogonal labeling method combined with spatially-localized expression of proteins. Dasatinib was modified with transcyclooctene (TCO), and its cytoplasmic target kinases were expressed in intracellular compartments, such as endosomes and F-actins. After bioorthogonal labeling, the colocalization between Dasatinib and its target proteins was observed in intracellular compartments.</P>

      • 제대혈의 적혈구 제거 방법에 관한 연구 : 제대혈 은행 설립을 위한 기초 연구 Basic Study for the Establishment of Cord Blood Bank

        배경:제대혈에서 적혈구를 제거하는 것은 제대혈 은행의 효율적인 운영 및 제대혈이식시 ABO 부적합 수혈, DMSO 독성 등의 부작용을 해결하기 위한 이상적인 방법이다. 저자들은 동일한 제대혈로 여러가지 적혈구 제거법을 동시에 시행하여, 이들에서 CD34양성 세포의 수득률 및 조혈세포 집락형성능을 비교하여 제대혈 은행 설립의 기반 기술을 확립하고자 하였다. 방법:제왕절개 직후 헤파린 처리된 용기에 제대혈을 채취하였다. 제대혈은 5개의 용기에 분산하여 실온에서 보관하였고 12시간 이내에 적혈구를 분리하였다. 적혈구 제거 방법으로 Ficoll 비중 분리법, gelatin 침전법, 적혈구 용혈법, starch 침전법을 각각의 제대혈에서 시행하였다. 전혈과 적혈구가 제거된 제대혈에서의 세포 생존율, 적혈구 제거율과 단핵구 및 CD34양성 세포의 수득률, 조혈세포 집락형성능을 비교 분석하였다. 결과:18례에서의 평균 제대혈 채취양은 81.3 mL(범위 42~128 mL)이었다. 서로 다른 적혈구 제거법들간에 세포 생존률과 적혈구 제거율은 차이가 없었다. 단핵구 수득률은 3% gelatin침전법(47.7±14.5%)이 다른 적혈구 제거법보다 유의하게 높았다. 3% gelatin침전법은 Ficoll비중 분리법과 Starch침전법에 비해 CD34 양성 세포수가 통계적으로 유의하게 높았다(36.1±5.4×10³/mL, 15.0±3.5×10³/mL, 20.7±4.2×10³/mL). 또한 CFC의 집락수도 3% gelatin침전법에서 Ficoll 비중 분리법과 적혈구 용혈법에 비해 우수한 결과를 보였다(52.0±7.1×10²/mL, 32.8±4.7×10²/mL, 36.8±5.7×10²/mL). 그러나 제대혈 전혈의 CD34 양성 세포수(65.7±11.2×10³/mL) 및 CFC의 집락수(117.8±15.8×10²/mL)를 적혈구 제거법을 이용한 실험군과 비교하였을 때 전혈군에서 높은 것을 알 수 있었다. 결론:3% gelatin 침전법은 다른 적혈구 제거법과 비교해 볼 때 더 우수한 단핵구, CD34 양성 세포 수득률 및 조혈세포 집락형성능을 보였다. 그러나 다른 보고들과는 달리 3% gelatin 침전법은 CD34양성 세포 및 CFC 집락수의 절대값이 제대혈 전혈과 비교하여 의미있게 낮았다. 이러한 결과는 시약의 종류와 실험방법의 차이에 기인하는 것으로 사료되며, 최상의 세포 수득률을 얻기 위하여 적혈구 분리 방법의 표준화를 위한 연구가 진행되어야 할 것으로 생각된다. Background:Efficient volume reduction is important to make a cost-effective umbilical cord blood (UCB) banking system. The aim of this study was to find a method of red cell depletion of UCB without major losses of the hematopoietic progenitor-CD34+ cells. Methods:Eighteen cord blood samples were collected in heparinized bottle immediately after Cesarean section. Five aliquots of each cord blood were stored at room temperature and processed within 12 hours. We used 4 different RBC depletion methods (Ficoll density gradient separation, gelatin sedimentation, red cell lysis, starch sedimentation) and compared the results of viability, RBC reduction rate, mononuclear cell (MNC) recovery rate, CD34+ cells and colony forming unit (CFC).Results: The mean collected cord blood volume was 81.3 mL (range 42~128 mL). There were no differences of viability and RBC reduction rate among groups. MNC recovery rate were significantly higher in gelatin group (47.7±14.5%) than other RBC depletion methods. Gelatin group had significantly higher CD34+ cell count than Ficoll and starch group (36.1±5.4×10³/mL, 15.0±3.5×10³/mL, 20.7±4.2×10³/mL) and higher CFC count than Ficoll and RBC lysis group (52.0±7.1×10²/mL, 32.8±4.7×10²/mL, 36.8±5.7×10²/mL). But the results of CD34+ cell count (65.7±11.2×10³/mL) and CFC count (117.8±15.8×10²/mL) of untreated control group are significantly higher than those of all other treated groups. Conclusion:Umbilical cord blood processed with 3% gelatin sedimentation has higher recovery rate of MNC, CD34+, cell and CFC than other groups. Unlike other reports, the absolute numbers of CD34+ cell and CFC count of 3% gelatin sedimentation method are significantly lower than those of untreated control group. Different laboratory processes with gelatin could be one cause of these differences, a study for standardized laboratory methods should be established to obtain higher recovery rate of hematopoietic cells after RBC depletion.

        Hydrophobically modified glycol chitosan nanoparticles as carriers for paclitaxel

        <P><B>Abstract</B></P><P>Self-assembled nanoparticles based on hydrophobically modified glycol chitosan (HGC) were prepared as a carrier for paclitaxel. HGC conjugates were prepared by chemically linking 5β-cholanic acid to glycol chitosan chains using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide chemistry. In phosphate-buffered saline (PBS; pH 7.4), the synthesized HGC conjugates formed nano-sized particles with a diameter of 200 nm and exhibited high thermodynamic stability as reflected by their low critical aggregation concentration (0.03 mg/ml). Paclitaxel was efficiently loaded into HGC nanoparticles up to 10 wt.% using a dialysis method. The paclitaxel-loaded HGC (PTX-HGC) nanoparticles were 400 nm in diameter and were stable in PBS for 10 days. These PTX-HGC nanoparticles also showed sustained release of the incorporated of paclitaxel (80% of the loaded dose was released in 8 days at 37 °C in PBS). Owing to sustained release, the PTX-HGC nanoparticles were less cytotoxic to B16F10 melanoma cells than free paclitaxel formulated in Cremophor EL. Injection of PTX-HGC nanoparticles into the tail vein of tumor-bearing mice prevented increases in tumor volume for 8 days. Finally, PTX was less toxic to the tumor-bearing mice when formulated in HGC nanoparticles than when formulated with Cremophor EL.</P>

        Expression of human β‐defensin‐2 in the prostate

        <P>We found the expression of human beta-defensin-2 (HBD-2) in the prostate for the first time and LPS, a gram negative bacterial component, upregulated HBD-2 in prostate epithelial cells. We are looking for other antimicrobial peptides expressed in the prostate besides human beta-defensin-2. Also, we are studying the relationship between antimicrobial peptides and the development or progression of prostate diseases. OBJECTIVE To investigate the expression and regulation of human beta-defensin-2 (HBD-2) in the prostate. PATIENTS AND METHODS Normal human prostate epithelial cell line (RWPE-1), human prostate cancer cell lines (DU-145, PC-3), and paraffin-embedded prostate tissue from patients with benign prostatic hyperplasia (BPH) were analysed by RT-PCR and immunohistochemical staining. HBD-2 expression was also analysed by RT-PCR and ELISA in RWPE-1 cells treated with lipopolysaccharide (LPS). Nuclear factor-kappa B (NF-kappa B) activation was assessed by I kappa B alpha immunoblotting and electrophoretic mobility shift assay (EMSA). RESULTS BPH tissue and all of the tested prostate cell lines other than PC-3 constitutively express HBD-2 mRNA. HBD-2 protein was strongly detected in prostate gland tissue surrounded by inflammatory cells including macrophages. Exposure to LPS induced HBD-2 upregulation and NF-kappa B activation, as assessed by I kappa B alpha phosphorylation and degradation in RWPE-1 cells. Bay11-7082, an NF-kappa B inhibitor prevented LPS-induced HBD-2 production in RWPE-1 cells. CONCLUSIONS Prostate epithelial cells may constitutively express HBD-2, and its expression was upregulated by LPS. Our data indicate that HBD-2 may be an important immunomodulatory factor in prostate function. Expression of HBD-2 in normal prostates and the potential role of HBD-2 in prostatitis and BPH should be addressed in the future.</P>
