Intrinsic expression of viperin regulates thermogenesis in adipose tissues

<P><B>Significance</B></P><P>The expression of numerous interferon (IFN)-inducible proteins is triggered by IFN or stimuli that activate IFN signaling pathways in innate immune responses. However, many of these proteins are expressed without stimulation in specific tissues, suggesting that their intrinsic expression may have novel functions. We show here that viperin, an IFN-inducible protein, regulates fatty acid β-oxidation–mediated thermogenesis in adipose tissues. Viperin deficiency up-regulates the expression of thermogenesis- and fatty acid β-oxidation–related genes, which in turn increases heat production, reduces fat mass, improves high-fat diet–induced glucose tolerance, and enhances cold tolerance. Thus, the intrinsic expression of viperin plays a novel, pivotal role in thermoregulation.</P><P>Viperin is an interferon (IFN)-inducible multifunctional protein. Recent evidence from high-throughput analyses indicates that most IFN-inducible proteins, including viperin, are intrinsically expressed in specific tissues; however, the respective intrinsic functions are unknown. Here we show that the intrinsic expression of viperin regulates adipose tissue thermogenesis, which is known to counter metabolic disease and contribute to the febrile response to pathogen invasion. Viperin knockout mice exhibit increased heat production, resulting in a reduction of fat mass, improvement of high-fat diet (HFD)-induced glucose tolerance, and enhancement of cold tolerance. These thermogenic phenotypes are attributed to an adipocyte-autonomous mechanism that regulates fatty acid β-oxidation. Under an HFD, viperin expression is increased, and its function is enhanced. Our findings reveal the intrinsic function of viperin as a novel mechanism regulating thermogenesis in adipose tissues, suggesting that viperin represents a molecular target for thermoregulation in clinical contexts.</P>
Dextran/PLGA Double-Layered Microspheres for Sustained Release of Doxorubicin

Biodegradable polymer microspheres have been extensively developed as drug carriers that alleviate burst release and control drug release profiles. In this study, we report a simple method to develop double-layered microspheres for controlled and sustained drug release. Double layered microspheres were prepared using PLGA and natural hydrophilic polymer dextran by a simple method based on an emulsion-solvent evaporation method. Microspheres had a double layered structure with dextran layer in the core and PLGA on the surface due to phase separation and encapsulated doxorubicin as a model drug in their core. The molecular weight and concentration of PLGA dominated the structure, in vitro degradation and drug release profiles of microspheres. Dextran/PLGA double- layered microspheres had a diameter about 200 m and alleviated initial burst release. Drug release profiles from dextran/PLGA double-layered microspheres can becontrolled by manipulating the thickness of shell of the microspheres. We anticipate great potential of dextran/PLGA double-layered microspheres for controlled and sustained drug delivery, given their simple formulation and the control of drug release profiles.
USE OF ITEM RESPONSE THEORY IN MARKETING RESEARCH

There are two purposes of this paper. One is to present a brief introduction to item response theory in conjunction with marketing research. The other is to present a review of current use of item response theory in representative marketing research journals. Several item response theory relevant papers were recently published in various marketing research journals. Because models under item response theory from simple to complex were used without any systematic introduction in marketing research, this paper briefly presents main concepts in item response theory. An encyclopaedia entry (Kim, 2015) and two graduate-level textbooks (Baker & Kim, 2004, 2017) are mainly referred and used for the first purpose. A content analysis was done for the second purpose with 28 item response theory relevant articles on marketing research journals. Articles are sorted based on the classification framework by Thissen and Steinberg (1986). Many articles reviewed relied on some type of unidimensional dichotomous item response theory models. Articles published recently within 10 years that used item response theory models were more complicated both mathematically and statistically than other previously published articles in marketing research journals. The taxonomic tabulations in this study should aid marketing researchers who are planning their continuous training in item response theory, and faculty who design or teach courses on marketing research methods for advanced undergraduate and graduate students.

There are two methods to treating onychomycosis. They are oral therapy and topical therapy. In this study we investigated the topical therapy. Penetration enhancer is necessary for effective topical dose of nail. KOH, NaOH and NaBH4 that have a potentiality as a penetration enhancer were dissolved in distilled water at 3% w/v concentration. And then nails are treated with penetration enhancers. The nail swelling rate(%) is calculated by using the nail weight of before and after experiment. The nails treated with penetration enhancer are fully dried and then we observed the surface of nails by using SEM and AFM. The three potential penetration enhancers showed high nail swelling rate and the surface of nails was more lifted than non-treating group or control group. So three penetration enhancers as enhancing the drug penetration are excellent.

Tramadol HCl(TH)-loaded poly(L-lactide-co-glycolide)(PLGA) microspheres were prepared by O/O solvent evaporation method for sustained release. We investigated the release behavior according to PLGA molecular weight and concentration. TH-loaded PLGA microspheres were characterized on the surface and cross-section morphology by SEM. TH-loaded PLGA microspheres had smooth surfaces and various pores in internal structure. As the PLGA molecular weight and PLGA concentration increased, the release rate of TH decreased. The behaviors of degradation was decreased according to increased PLGA molecular weight. These results showed that the release behaviors can be controlled by various of molecular weight and concentration of PLGA.
High Malaria Prevalence among Schoolchildren on Kome Island, Tanzania

In order to determine the status of malaria among schoolchildren on Kome Island (Lake Victoria), near Mwanza, Tanzania, a total of 244 schoolchildren in 10 primary schools were subjected to a blood survey using the fingerprick method. The subjected schoolchildren were 123 boys and 121 girls who were 6-8 years of age. Only 1 blood smear was prepared for each child. The overall prevalence of malaria was 38.1% (93 positives), and sex difference was not remarkable. However, the positive rate was the highest in Izindabo Primary School (51.4%) followed by Isenyi Primary School (48.3%) and Bugoro Primary School (46.7%). The lowest prevalence was found in Muungano Primary School (16.7%) and Nyamiswi Primary School (16.7%). These differences were highly correlated with the location of the school on the Island; those located in the peripheral area revealed higher prevalences while those located in the central area showed lower prevalences. Plasmodium falciparum was the predominant species (38.1%; 93/244), with a small proportion of them mixed-infected with Plasmodium vivax (1.6%; 4/244). The results revealed that malaria is highly prevalent among primary schoolchildren on Kome Island, Tanzania, and there is an urgent need to control malaria in this area.
