Lipids, which along with carbohydrates and proteins are among the most important nutrients for the living organism, have a variety of biological functions that can be applied widely in biomedicine. A fatty acid, the most fundamental biological lipid, may be classified by length of its aliphatic chain, and the short-, medium-, and long-chain fatty acids and each have distinct biological activities with therapeutic relevance. For example, short-chain fatty acids have immune regulatory activities and could be useful against autoimmune disease; medium-chain fatty acids generate ketogenic metabolites and may be used to control seizure; and some metabolites oxidized from long-chain fatty acids could be used to treat metabolic disorders. Glycerolipids play important roles in pathological environments, such as those of cancers or metabolic disorders, and thus are regarded as a potential therapeutic target. Phospholipids represent the main building unit of the plasma membrane of cells, and play key roles in cellular signaling. Due to their physical properties, glycerophospholipids are frequently used as pharmaceutical ingredients, in addition to being potential novel drug targets for treating disease. Sphingolipids, which comprise another component of the plasma membrane, have their own distinct biological functions and have been investigated in nanotechnological applications such as drug delivery systems. Saccharolipids, which are derived from bacteria, have endotoxin effects that stimulate the immune system. Chemically modified saccharolipids might be useful for cancer immunotherapy or as vaccine adjuvants. This review will address the important biological function of several key lipids and offer critical insights into their potential therapeutic applications.

<P>In this paper, we present BMQ-Processor, a high-performance border-crossing event (BCE) detection framework for large-scale monitoring applications. We first characterize a new query semantics, namely, border monitoring query (BMQ), which is useful for BCE detection in many monitoring applications. It monitors the values of data streams and reports them only when data streams cross the borders of its range. We then propose BMQ-Processor to efficiently handle a large number of BMQs over a high volume of data streams. BMQ-Processor efficiently processes BMQs in a shared and incremental manner. It develops and operates over a novel stateful query index, achieving a high level of scalability over continuous data updates. Also, it utilizes the locality embedded in data streams and greatly accelerates successive BMQ evaluations. We present data structures and algorithms to support 1D as well as multidimensional BMQs. We show that the semantics of border monitoring can be extended toward more advanced ones and build region transition monitoring as a sample case. Lastly, we demonstrate excellent processing performance and low storage cost of BMQ-Processor through extensive analysis and experiments.</P>

<P>A strong promoter increases transcription of the genes of interest and enhances the production of various valuable substances. For a hyperthermophilic archaeon Thermococcus onnurineus NA1, which can produce H-2 from carbon monoxide oxidation, we searched for a novel endogenous strong promoter by transcriptome analysis using high-throughput RNA sequencing. Based on the relative transcript abundance, we selected one promoter to encode a hypothetical gene, of which homologs were found only in several Thermococcales strains. This promoter, P (TN0510) , was introduced into the front of CO-responsible hydrogenase gene cluster encoding a carbon monoxide dehydrogenase (CODH), a hydrogenase, and a Na+/H+ antiporter. In the resulting mutant strain, KS0510, transcription and translation level of the gene cluster increased by 4- to 14-folds and 1.5- to 1.9-folds, respectively, in comparison with those of the wild-type strain. Additionally, H-2 production rate of KS0510 mutant was 4.8-fold higher than that of the wild-type strain. The P (TN0510) was identified to be much stronger than the well-known two strong promoters, gdh and slp promoters from Thermococcus strains, through RT-qPCR and Western blotting analyses and kinetics of H-2 production. In this study, we demonstrated that the RNA-seq approach is a good strategy to mine novel strong promoters of use to a Thermococcus strain when developed as a biotechnologically promising strain to produce valuable products such as enzymes and metabolites through metabolic engineering.</P>

<P><B>Abstract</B></P><P>Bacterial DNA has a variety of immunostimulatory activities, such as the activation of B cells and natural killer cells, the induction of interferon-γ, and the induction of Th1-type immune responses. In contrast, mammalian DNA does not have these activities. To evaluate the genomic DNA sequences of <I>Mycobacterium bovis</I> that have immunostimulatory activity, we used a computer to analyze the <I>M. bovis</I> genome and we designed a series of synthetic, 20 base length, phosphodiester backbone oligodeoxynucleotides (ODNs) that contain CpG motifs (MB-ODNs). We screened the immunostimulatory MB-ODNs that induce the activation of the NF-κB-responsive IL-8 promoter in RAW 264.7 cells. Our experimental analyses demonstrate that the potent CpG DNA in the <I>M. bovis</I> genome has functional effects as a Th1-responsive adjuvant, and that it activates the transcription factor NF-κB. Moreover, we found that both the CpG motifs and the context of the sequence surrounding the CpG motif are important for the immunostimulatory activities. The identification of the potent immunostimulatory DNA sequence in a native bacterial genome may give insights to the optimal sequence for well-controlled immune responses.</P>
