        Tunable paclitaxel release carrier using diselenide-disulfide balance as regulator

        There are many reasons that lead to the failure of cancer chemotherapy, such as uncontrolled drugrelease, low drug utilization, and severe side effects. To overcome these obstacles, two kinds of thermaland redox-responsive copolymers with multiple diselenide/disulfide linkages, polyethylene glycol -altdiselenodipropionate/disulfhydryldipropionate-b-poly(N-isopropylacrylamide) (abbreviated as PEG-alt-DSeDP-b-PNIPAM and PEG-alt-DSDP-b-PNIPAM) were fabricated by alternative esterification and followingatom transfer radical polymerization. Afterward, these prepared copolymers were mixed in line withthe mass ratio of 8:0, 5:3, 3:5, 0:8 (denoted as S1, S2, S3, and S4, respectively), and self-assembled withpaclitaxel (PTX) to obtain PTX-loaded S1, S2, S3, and S4 nanomicellar assemblies, aiming to realize PTXtunable release using diselenide-disulfide balance as regulator. The chemical structures of these twocopolymers were characterized by gel permeation chromatography, indicating eight diselenide/disulfidelinkages and eight PEG units were contained in these copolymers. Moreover, the thermal-responsiveproperty was detected by UV–vis spectroscopy, meanwhile, the redox responsiveness was observed byTEM in the presence of 10 mM glutathione. We found that 76.90% of PTX was released from S1 nanomicelleswithin 23 h. In contrast, this percentage decreased to 64.53% for S4 nanomicelles even the incubationtime prolonged, indicating explosive and slow release behaviors of S1 and S4 nanomicelles,respectively. In addition, gradually decreased fluorescence intensity around the cellular nucleus wasoccurred from S1 to S4 orderly, which was consistent with cellular uptake and in vivo anti-tumor experiments. Taken together, this work not only provides a strategy for tunable PTX release, but also improveseffectiveness of PTX in cancer treatment.

      • Association Between HLA-DQ Genotypes and Haplotypes vs Helicobacter pylori Infection in an Indonesian Population

        Background: Helicobacter pylori is an important gastrointestinal pathogen related to the development of not only atrophic gastritis and peptic ulcer, but also gastric cancer. Human leukocyte antigens (HLA) may play particular roles in host immune responses to bacterial antigens. This study aimed to investigate the association between HLA-DQA1 and DQB1 genotypes and haplotypes vs H. pylori infection in an Indonesian population. Methods: We selected 294 healthy participants in Mataram, Lombok Island, Indonesia. H. pylori infection was determined by urea breath test (UBT). We analyzed HLA-DQA1 and DQB1 genotypes by PCR-RFLP and constructed haplotypes of HLA-DQA1 and DQB1 genes. Multiple comparisons were conducted according to the Bonferroni method. Results: The H. pylori infection rate was 11.2% in this Indonesian population. The DQB1*0401 genotype was noted to be associated with a high risk of H. pylori infection, compared with the DQB1*0301 genotype. None of the HLA-DQA1 or DQB1 haplotypes were related to the risk of H. pylori infection. Conclusions: The study suggests that HLADQB1 genes play important roles in H. pylori infection, but there was no statistically significant association between HLA-DQA1 or DQB1 haplotypes and H.pylori infection in our Lombok Indonesian population.

        Pure Ultraviolet Emission from ZnO Quantum Dots-Based/GaN Heterojunction Diodes by MgO Interlayer

        We demonstrate the fabrication and characterization of ZnO/GaNbasedheterojunction light-emitting diodes (LEDs) by using air-stableand solution-processable ZnO quantum dots (QDs) with a thin MgOinterlayer acting as an electron blocking layer (EBL). The ZnO QDs/MgO/p-GaN heterojunction can only display electroluminescence(EL) characteristic in reverse bias regime. Under sufficient reversebias, a fairly pure ultraviolet EL emission located at 370 nm derivingfrom near band edge of ZnO with a full width at half maximum(FWHM) of 8.3 nm had been obtained, while the deep-level emissionhad been almost totally suppressed. The EL origination andcorresponding carrier transport mechanisms were investigatedqualitatively in terms of photoluminescence (PL) results and energyband diagram.

        LncRNA linc01194 promotes the progress of endometrial carcinoma by up-regulating SOX2 through binding to IGF2BP1

        Objective: Endometrial carcinoma (EC) is one of the most common gynecological malignanttumors. Our study showed that long non-coding RNA (lncRNA) linc01194 plays an importantrole in EC. We explored the mechanism of lncRNA linc01194 in EC. Methods: The expression of lncRNA linc01194 was detected in The Cancer Genome Atlasdatabase and starBase database. The potential targeted protein of linc01194 was predictedthrough the starBase database. To determine the role of linc01194 in EC, we downregulatedor upregulated the level of linc01194 in EC cell lines and analyzed the cell behaviors and thechanges of its potential target proteins. Results: The expression of linc01194 in EC tissues is higher than that in normal endometrialtissues. The knockdown of linc01194 inhibited the cell proliferation, invasion and migrationand promoted the apoptosis of EC cells, while overexpression of linc01194 promoted cellproliferation, invasion and migration and inhibited the apoptosis of EC cells. The starBasedatabase revealed that linc01194 could bind to insulin-like growth factor 2 binding protein 1(IGF2BP1). Previous results showed that in EC, IGF2BP1 could promote the expression of sex-determining region Y-box 2 (SOX2) by promoting the stability of SOX2 mRNA. Our resultsshowed that linc01194 regulate the expression of IGF2BP1 and SOX2. Conclusion: Linc01194 can promote the expression of downstream protein SOX2 throughbinding to IGF2BP1, thus promoting the occurrence and development of EC.

        Effects of transport time and feeding type on weight loss, meat quality and behavior of broilers

        Objective: The purpose of this study is to determine the optimal time of transportation of floor-feed and scatter-feed broilers. Methods: Eighty healthy Arbor Acres (AA) broilers (21-day-old, 624.4 g, male, standard error = 6.65) were selected and randomly divided into two experimental groups (floor-feed and scatter-feed), then fed for three weeks. The experiment comprised a 2×4 factorial design with 2 feed patterns (floor-feed and scatter-feed) and 4 transport periods (2, 3, 4, and 5 h), and 4 replicates of 5 broilers (54-day-old, 2243 g, standard error = 46.65) was used to compare weight loss, meat quality and behavior index of different groups. Results: It appeared that drip loss, meat color and resting behavior of experimental broilers changed as length of transportation (p<0.05), however, weight loss and pH were not significantly transformed (p>0.05). Compared with floor-feed group, broilers in scatter-feed group had lower pH at 24 hours (3 h) and different behavioral indicators (p<0.05). Especially indicators after 3 h transportation, there were obvious differences between the two feeding modes in the behavior reaction of stress events before slaughter with different transport duration (p<0.05). The fluctuation of data on resting behavior with scatter-feed was significantly higher than that of floor-feed broilers. There was no interaction between transport time and different feeding methods for index tested of our experiment (p>0.05). Conclusion: Comprehensive analysis showed that the maximum transport duration of floor-feed and scatter-feed broilers should not exceed 3 h, and scatter-feed broilers were more likely prone to fear.
