        Experimental study on seismic performance of steel reinforced concrete T-shaped columns

        This study investigates the seismic performance of steel reinforced concrete (SRC) T-shaped columns under low cyclic loading tests. Based on test results of ten half-scale column specimens, failure patterns, hysteretic behavior, skeleton curves, ultimate strength, ductility, stiffness degradation and energy dissipation capacity were analyzed. The main variables included loading angles, axial compression ratios and steel ratios. The test results show that the average values of the ductility factor and the equivalent viscous damping coefficient with respect to the failure of the columns were 5.23 and 0.373, respectively, reflecting good seismic performance. The ductility decreased and the initial stiffness increased as the axial compression ratio of the columns increased. The strength increased with increasing steel ratio, as expected. The columns displaced along the web had higher strength and initial stiffness, while the columns displaced along the flange had better ductility and energy dissipation capacity. Based on the test and analysis results, a formula is proposed to calculate the effective stiffness of SRC T-shaped columns.

        An innovative geometry control method for short-line match precast segmental bridges

        The occurrence of unexpected horizontal offset in the instrument or target will result in accumulated horizontal deviation in segment alignment with traditional short-line match method. A geometry control method, the four-point method, is developed for precast segmental bridges to avoid the influences of unexpected horizontal offset. The concept of the four-point method is elucidated. Furthermore, the detailed instruments and instructions are introduced. Finally, the four-point method is validated through a practical engineering application. According to the survey data, after short-line match precast construction, the vertical deviations on both sides vary between -5 mm and 5 mm in almost all segments, and the horizontal deviations vary between -4 mm and 4 mm in all segments. Without on-site adjustment, the maximum vertical and horizontal closure gaps are 12.3 and 26.1 mm, respectively. The four-point method is suggested to alleviate the issues associated with relatively poor soil conditions in casting yard.

        A missense variant in NCF1 is associated with susceptibility to multiple autoimmune diseases

        <P>Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disease with a strong genetic component characterized by autoantibody production and a type 1 interferon signatures. Here we report a missense variant (g.74779296G>A; p.Arg90His) in NCF1, encoding the p47phox subunit of the phagocyte NADPH oxidase (NOX2), as the putative underlying causal variant that drives a strong SLE-associated signal detected by the lmmunochip in the GTF2IRD1-GTF2I region at 7q11.23 with a complex genomic structure. We show that the p.Arg90His substitution, which is reported to cause reduced reactive oxygen species (ROS) production(2), predisposes to SLE (odds ratio (OR) = 3.47 in Asians (P-meta = 3.1 x 10(-104)), OR = 2.61 in European Americans, OR = 2.02 in African Americans) and other autoimmune diseases, including primary Sjogren's syndrome (OR = 2.45 in Chinese, OR = 2.35 in European Americans) and rheumatoid arthritis (OR = 1.65 in Koreans). Additionally, decreased and increased copy numbers of NCF1 predispose to and protect against SLE, respectively. Our data highlight the pathogenic role of reduced NOX2-derived ROS levels in autoimmune diseases.</P>

        Simultaneous Estimation of Spatial Frequency and Phase Based on an Improved Component Cross-Correlation Algorithm for Structured Illumination Microscopy

        Accurate estimation of spatial frequencies and phases for illumination patterns are essential to reconstructing super-resolution images in structured illumination microscopy (SIM). In this manuscript, we propose the improved component cross-correlation (ICC) algorithm, which is based on optimization of the cross-correlation values of the overlapping information between various spectral components. Compared to other algorithms for spatial-frequency and phase determination, the results calculated by the ICC algorithm are more accurate when the modulation depths of the illumination patterns are low. Moreover, the ICC algorithm is able to calculate the spatial frequencies and phases simultaneously. Simulation results indicate that even if the modulation depth is lower than 0.1, the ICC algorithm still estimates the parameters precisely; the images reconstructed by the ICC algorithm are much clearer than those reconstructed by other algorithms. In experiments, our home-built SIM system was used to image bovine pulmonary artery endothelial (BPAE) cells. Drawing support from the ICC algorithm, super-resolution images were reconstructed without artifacts.

        Polyacrylonitrile mesoporous composite membranes with high separation efficiency prepared by fast freeze-extraction process

        Polymer mesoporous membranes are widely employed in many membrane processes includingultrafiltration and dialysis, and become more crucial with increasing demand in chemical and lifefields. Here we report a highly-efficient polymer membrane with sub-5 nm pores and highflux frompolyacrylonitrile (PAN) nanoparticles produced from dilute PAN solution via fast freeze-extractionprocess. The25 nm-size PAN nanoparticles forming brad-like chains are dispersed in ethanol solutionand used to prepare the membrane byfiltration-assisted assembly method across a macroporoussupport. Effects of solvents and usage of PAN on membrane formation is studied in detail. The resultantmembranes comprise of the nanoparticles stacked on the support to form a mesoporous separation layerwith a controllable thickness and a cut-off of below 5 nm. They exhibit excellent separation performancesin ultrafiltration of protein and nanoparticle solutions. Typically, the membrane with 94.6% rejection of5 nm gold nanoparticles has a pure waterflux of 823 L m 1 h 1 bar 1 and perfect rejection for ferritinmolecules and 10 nm gold nanoparticles. The newly developed membranes should have wide applicationin chemical, environment and lifefields.

        Knockdown of lncRNA PVT1 Inhibits Vascular Smooth Muscle Cell Apoptosis and Extracellular Matrix Disruption in a Murine Abdominal Aortic Aneurysm Model

        This study was designed to determine the effects of the long non-coding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) on vascular smooth muscle cell (VSMC) apoptosis and extracellular matrix (ECM) disruption in a murine abdominal aortic aneurysm (AAA) model. After injection of PVT1-silencing lentiviruses, AAA was induced in Apolipoprotein E-deficient (ApoE?/?) male mice by angiotensin II (Ang II) infusion for four weeks. After Ang II infusion, mouse serum levels of pro-inflammatory cytokines were analysed, and aortic tissues were isolated for histological, RNA, and protein analysis. Our results also showed that PVT1 expression was significantly upregulated in abdominal aortic tissues from AAA patients compared with that in controls. Additionally, Ang II treatment significantly increased PVT1 expression, both in cultured mouse VSMCs and in AAA murine abdominal aortic tissues. Of note, the effects of Ang II in facilitating cell apoptosis, increasing matrix metalloproteinase (MMP)-2 and MMP-9, reducing tissue inhibitor of MMP (TIMP)-1, and promoting switching from the contractile to synthetic phenotype in cultured VSMCs were enhanced by overexpression of PVT1 but attenuated by knockdown of PVT1. Furthermore, knockdown of PVT1 reversed Ang II-induced AAA-associated alterations in mice, as evidenced by attenuation of aortic diameter dilation, marked adventitial thickening, loss of elastin in the aorta, enhanced aortic cell apoptosis, elevated MMP-2 and MMP-9, reduced TIMP-1, and increased proinflammatory cytokines. In conclusion, our findings demonstrate that knockdown of lncRNA PVT1 suppresses VSMC apoptosis, ECM disruption, and serum pro-inflammatory cytokines in a murine Ang II-induced AAA model.
