        Porcine cytomegalovirus (PCMV) causes piglet deaths, runting, inclusion body rhinitis, respiratory and reproductive disorders in swine herd, resulting in the significant economic losses in the swine industry. Recently, in the field of pig-to-human xenotransplantation, PCMV has been indicated as a potential xenogeneic pathogen with the risk for infection transmitted from the xenograft donor to the recipient. In this study, PCMV DNA polymerase gene was detected in the sera and nasal swabs from 30 heads of the weaned piglets affected with various wasting diseases and 10 heads of normal healthy piglets using polymerase chain reaction (PCR), and the diagnostic efficacy of PCR was evaluated. The prevalence of the PCR-positive by specimens from the diseased piglets were 83.3% in sera and 33.3% in nasal swabs. In the normal healthy group, the PCR-positive rates of sera and nasal swabs were 40% and 30%, respectively. Ten piglets (25.0%) out of 40 showed the simultaneously positive results in both of sera and nasal swabs. According to this result, it was found that the serum specimen was the most appropriate sample for detection of PCMV by PCR. The nucleotide sequence homology of DNA polymerase gene of PCMV KS1 as compared with the reference strain of PCMV (GenBank accesson no.AF027217) was 99.0%.

      • Effect of the spirobiindane group in sulfonated poly(arylene ether sulfone) copolymer as electrode binder for polymer electrolyte membrane fuel cells

        <P><B>Abstract</B></P> <P>Electrode properties determine the membrane-electrode assembly (MEA) performance of fuel cells and are influenced by the materials and processing. A high-performance electrode requires good adhesion between the membrane and catalyst, excellent gas permeability, and good ion conduction. Here, sulfonated poly(arylene ether sulfone) (SPAES) containing a bulky spirobiindane (Spiro) group is proposed as a new hydrocarbon electrode binder for MEAs. The effect of the Spiro group on MEA performance was compared to that of the common biphenylsulfone. The structural differences between the two binders affected the mass transport region of the current-voltage polarization (related to gas permeation). The spiro-polyethersulfone (PES) had 115 times higher O<SUB>2</SUB> permeability (30 Barrer) than that of PES. From <SUP>1</SUP>H NMR spectra, the degree of sulfonation (DS) of each SPAES was confirmed, where the proton conductivity of Spiro 30, Spiro 20, and BPS 30 were 28.2, 10.1, and 26.9mScm<SUP>−1</SUP>, respectively. In addition, the DS of the electrode binder contributed to the Ohmic region of the i–V curve. The Ohmic resistance of these hydrocarbon binders was similar to that of commercial Nafion binder (0.13–0.15Ωcm<SUP>2</SUP>). However, the charge transfer resistance of our binders was higher than that of Nafion due to insufficient gas permeability and low proton conductivity.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Long-term follow-up of a severely traumatized leg treated with ipsilateral fracture-united fibular transfer in a patient with amputation of the contralateral leg: a case report

        Extensive bone loss associated with severe vascular injury remains a challenge for lower extremity reconstruction. The fibular free flap has been utilized for many decades to reconstruct long-segment tibial defects. We present an unusual scenario of unilateral weight-bearing, wherein we salvaged the sole lower extremity by transfer of the fractured ipsilateral fibula and a bipedicled skin flap. A 38-year-old man sustained a severe crush injury in the right leg with loss of circulation. His left lower leg had a soft tissue defect measuring 20×15 cm with an exposed comminuted fracture and a 17-cm tibial defect, along with a segmental fracture of the fibula. Subsequently, we reconstructed the tibial defect by transferring a 17-cm-long section of the ipsilateral fibula. We covered the soft tissue defect with a bipedicled skin flap. The patient eventually began to ambulate independently after surgery.

        Coxsackie and Adenovirus Receptor Binding Ablation Reduces Adenovirus Liver Tropism and Toxicity

        <P>Human adenovirus-based vectors have emerged as a new promising vehicle for in vivo gene transfer-mediated therapy. However, the full potential of this methodology has not been fully realized because of the nonspecific tissue distribution of adenoviral vectors. Adenovirus infection is initiated by forming a complex between the fiber protein and a ubiquitously expressed host cell membrane protein, coxsackie B virus and adenovirus receptor (CAR). Therefore, ablating the adenovirus vector's ability to bind to the CAR is the first step in redirecting adenoviral tropism. To ablate CAR binding, we mutated the Bbeta sheet of the fiber knob, generating CAR-binding ablated replication-incompetent (dl-K420A-Z) and replication-competent (YKLK420A) adenoviral vectors. The in vitro transduction efficiency of dl-K420A-Z was significantly reduced in comparison to dl-LacZ carrying the wild-type fiber in CAR-positive cells but not in CAR-negative cells, suggesting that the mutation introduced in the Bbeta sheet of the fiber knob could disable the CAR-dependent transduction pathway. The in vivo transduction was also dramatically reduced in the liver and other organs for mice treated with dl-K420A-Z, compared with a cognate control vector, dl-LacZ. Concomitant with this attenuated gene transfer efficiency in vivo was a substantial reduction in the amount of general toxicity observed in the YKL-K420A-treated mice. Diminished toxicity was surmised from quantitative measurement of serum level of enzymes for liver and kidney function, hematologic chemistries, histopathology, and differences in lethality. Significant decrease in serum transaminases (alanine transferase [ALT] and aspartate transferase [AST]) was observed in mice treated with YKL-K420A. In addition, the lethality was lower in the YKLK420A- treated groups compared to the YKL-1-treated groups at all doses examined. Furthermore, the hepatopathologic analysis revealed that YKL-1 induced focal zonal necrosis and hepatocyte degeneration, while YKL-K420A induced mild spotty necrosis. In summary, this decreased vector tropism of CAR-binding ablated adenoviruses in normal tissues may increase the amount of virus available for infecting tumor cells and thus increase the antitumor efficacy with fewer unwanted side effects.</P>

        Genomic characteristics of triple negative apocrine carcinoma: a comparison to triple negative breast cancer

        Apocrine carcinoma is a rare breast cancer subtype. As such, the genomic characteristics of apocrine carcinoma with triple negative immunohistochemical results (TNAC), which has been treated as triple negative breast cancer (TNBC), have not been revealed. In this study, we evaluated the genomic characteristics of TNAC compared to TNBC with low Ki-67 (LK-TNBC). In the genetic analysis of 73 TNACs and 32 LK-TNBCs, the most frequently mutated driver gene in TNAC was TP53 (16/56, 28.6%), followed by PIK3CA (9/56, 16.1%), ZNF717 (8/56, 14.3%), and PIK3R1 (6/56, 10.71%). Mutational signature analysis showed enrichment of defective DNA mismatch repair (MMR)-related signatures (SBS6 and SBS21) and the SBS5 signature in TNAC, whereas an APOBEC activity-associated mutational signature (SBS13) was more prominent in LK-TNBC (Student’s t test, p < 0.05). In intrinsic subtyping, 38.4% of TNACs were classified as luminal A, 27.4% as luminal B, 26.0% as HER2-enriched (HER2-E), 2.7% as basal, and 5.5% as normal-like. The basal subtype was the most dominant subtype (43.8%) in LK-TNBC (p < 0.001), followed by luminal B (21.9%), HER2-E (21.9%), and luminal A (12.5%). In the survival analysis, TNAC had a five-year disease-free survival (DFS) rate of 92.2% compared to 59.1% for LK-TNBC (P = 0.001) and a five-year overall survival (OS) rate of 95.3% compared to 74.6% for LK-TNBC (P = 0.0099). TNAC has different genetic characteristics and better survival outcomes than LK-TNBC. In particular, normal-like and luminal A subtypes in TNAC have much better DFS and OS than other intrinsic subtypes. Our findings are expected to impact medical practice for patients diagnosed with TNAC.

        HVDC Overhaul 및 풍력발전을 고려한 제주계통 전력품질 분석

        제주의 전력계동은 HVDC로 육지계통과 연계되어 있으며, 2곳의 풍력발전단지가 연계되어 있다. 향후 대용량의 풍력발전이 연계된 상황에서 HVDC Overhaul을 시행하거나 어떤 사고로 HVDC가 끊어진다면 제주계통을 정상적으로 유지하는 것이 어려울 수도 있다. 이에 본 논문은 Overhaul 및 풍력발전이 제주계동에 어떤 영향을 미치는지를 분석하기 위하여 지난 5월에 이루어진 Overhaul 기간에 변전소 2곳 및 풍력발전단지 2곳을 선정하여 전력품질을 측정하고 분석하였다. 또한 풍력발전량에 따른 인근 D/L의 전력품질을 측정함으로써 용력발전이 전력계동에 미치는 영향을 분석하였고 그 영향정도가 명확하지 않은 부분에 대해서는 동계 데이터를 이용하여 분석하였다. Overhaul 기간에는 주파수가 매우 불안정 하였으며, 전압왜형률이 규정치를 초과하기도 하였다. 풍력발전량에 비례한 고조파 전류가 계동으로 유입되었으며, 발전량이 3,000[㎾] 이상일 때 인근 D/L의 전압파형이 발전량에 비례하여 왜곡되는 것을 확인할 수 있었다. Power system of Jeju is interconnected to the mainland using HVDC and that is also interconnected two wind farms. Control of Jeju power system will be difficult if HVDC is disconnected or HVDC is overhauled under large scale wind farms interconnected. We measured and analysed the power quality of two substation and two wind farms to assess that wind farms and HVDC overhaul have an effect on Jeju system or not during the HVDC overhaul. We found that the frequency of Jeju system is very unstable and the voltage distortion excess the limitation. Harmonic currents of wind farms flew into Jeju system in proportion to the power of wind turbine. And the voltage of nearby distribution line was distorted by power of wind turbine when the amount is more that 3,000[㎾].

        정금나무(Vaccinium oldhami) 열매의 항산화 효과

        천연물들은 현재까지 합성 항산화제들을 대체하기 위하여 많은 연구가 이뤄지고 있다. 본 연구에서는 천연 항산화제로서 화장품과 바이오산업에 적용하기 위해 정금나무 열매 70% 아세톤 추출물의 항산화능을 측정하였다. 정금나무 열매의 항산화능을 확인하기 위하여 폴리페놀의 함량을 측정, 전자공여능, ABTS·?, nitric oxide, hydrogen peroxide들의 소거 측정을 분석하여 다음과 같은 결과를 얻었다. 폴리페놀의 총 함량은 55.972 ㎎ TAE/g로 정량 되었다. 전자공여능 실험에서는 농도 의존적으로 DPPH radical 소거능이 증가 하였으며 500 ㎍/ml에서 98% 이상의 활성을 나타내어 양성 대조군인 BHA의 활성을 초과하는 것을 알 수 있었다. ABTS·?에 대한 소거활성은 500 ㎍/ml에서 66.7%의 결과로 나타났고, nitric oxide radical 소거능은 100 ㎍/ml에서 57%의 활성을 나타내었다. 또한 정금나무 열매 70% 아세톤 추출물의 hydrogen peroxide의 소거활성은 50 ㎍/ml에서 정금나무 열매 추출물은 70% 이상의 활성을 나타났으며, BHA 보다 높은 활성을 나타내었다. 천연물의 산업화 가능성에 있어 가장 중요한 것은 재료의 영속적인 수급과 적은 농도를 첨가하여도 높은 효과를 나타내는지의 여부이다. 매년 수확이 가능한 정금나무 열매를 이용한 본 연구의 결과에서 열매의 항산화능 확인 및 화장품 등 바이오산업에 천연항산화제 원료로서의 적용 가능성을 확인할 수 있었다. Natural compounds have been studied to substitute synthetic antioxidants. In this study, the anti-oxidant activity of 70% acetone extracts from the Vaccinium oldhami fruit was investigated for utilization as ingredients for the cosmetic and bio-industries. Anti-oxidant activity was determined by determining total polyphenolic content, electron donating ability, nitric oxide (NO) radical scavenging activity, ABTS·? cation radical scavenging activity and hydrogen peroxide scavenging activity. The polyphenolic content of 70% acetone extracts of the Vaccinium oldhami fruit was 55.972 ㎎ TAE/g. In electron donating activity, 70% acetone extracts of the Vaccinium oldhami fruit showed an effect of 93.9%, which was similar to BHA effect at a concentration of 500 ㎍/ml. In the NO radical scavenging ability, 70% acetone extracts of the Vaccinium oldhami fruit showed 60% at 500 ㎍/ml. ABTS·? cation radical scavenging activity of the Vaccinium oldhami fruit at a concentration of 1000 ㎍/ml was 75.7%. Also, hydrogen peroxide scavenging activity of 70% acetone extracts showed 80.8% at 100 ㎍/ml, whichwas higher than BHA. In the natural compound market, the most important factors are the ability to obtain high effects of a material in low concentrations and a long-lasting supply. The Vaccinium oldhami fruit can be harvested every year - this fulfills one of the requirements. From these results, we can confirm that the Vaccinium oldhami fruit has anti-oxidant abilities and has potential as a natural anti-oxidant agent to be utilized in the cosmeceutical and bio-industries.

        The Development and Validation of the Korean Strength Scale for Youth

        Objectives The purpose of this study was to develop a Korean Strength Scale for Youth and to examine its validity and reliability. Methods The Korean Strength Scale for Youth is based on a self-reporting, 100 item questionnaire that measures 25 strengths. Each scale item was given a 0–5 rating on a 6-point scale. In order to test the scale s validity and reliability, data were collected from 422 youths (aged 12–18 years). The strength measures included in our analyses were those contained within the Korean Strength Scale for Youth, the Positive Affect and Negative Affect Schedule, the Korean Well-Being Scale for Adolescents, and the Short Five Factor Personality Scale for Children. Results The results of exploratory factor analysis of the Korean Strength Scale for Youth scale-measures indicated the importance of four factors. The Korean Strength Scale for Youth was shown to have acceptable psychometric properties, including good internal consistency, test-retest reliability, and high convergent correlations. Conclusion The Korean Strength Scale for Youth is a reliable and valid instrument that can be used to assess strengths in adolescents
