        PICOT Inhibits Cardiac Hypertrophy and Enhances Ventricular Function and Cardiomyocyte Contractility

        <P>Multiple signaling pathways involving protein kinase C (PKC) have been implicated in the development of cardiac hypertrophy. We observed that a putative PKC inhibitor, PICOT (PKC-Interacting Cousin Of Thioredoxin) was upregulated in response to hypertrophic stimuli both in vitro and in vivo. This suggested that PICOT may act as an endogenous negative feedback regulator of cardiac hypertrophy through its ability to inhibit PKC activity, which is elevated during cardiac hypertrophy. Adenovirus-mediated gene transfer of PICOT completely blocked the hypertrophic response of neonatal rat cardiomyocytes to enthothelin-1 and phenylephrine, as demonstrated by cell size, sarcomere rearrangement, atrial natriuretic factor expression, and rates of protein synthesis. Transgenic mice with cardiac-specific overexpression of PICOT showed that PICOT is a potent inhibitor of cardiac hypertrophy induced by pressure overload. In addition, PICOT overexpression dramatically increased the ventricular function and cardiomyocyte contractility as measured by ejection fraction and end-systolic pressure of transgenic hearts and peak shortening of isolated cardiomyocytes, respectively. Intracellular Ca(2+) handing analysis revealed that increases in myofilament Ca(2+) responsiveness, together with increased rate of sarcoplasmic reticulum Ca(2+) reuptake, are associated with the enhanced contractility in PICOT-overexpressing cardiomyocytes. The inhibition of cardiac remodeling by of PICOT with a concomitant increase in ventricular function and cardiomyocyte contractility suggests that PICOT may provide an efficient modality for treatment of cardiac hypertrophy and heart failure.</P>

        Transdermal delivery of Minoxidil using HA-PLGA nanoparticles for the treatment in alopecia

        Background: Alopecia has become a very common disease that many people around the world are suffered. Minoxidil (MXD) is the most well-known commercialized drug in its treatment. However, in the case of MXD administration, there are some problems with low efficiency of transdermal delivery and additional side effects. Method: MXD and Rhodamine B (Rho B) are encapsulated in poly(Lactide-co-Glycolide) grafted hyaluronate nanoparticles (HA-PLGA/MXD NPs, HA-PLGA/Rho B NPs) which is prepared with W/O/W solvent evaporation method. After then, the investigation is carried out to confirm the feasibility of NPs in alopecia treatment. Results: Both of HA-PLGA/MXD NPs and HA-PLGA/Rho B NPs are successfully prepared. In addition, it is confirmed that HA-PLGA NPs sufficiently delivered to cells without any significant cytotoxicity by cell viability, cellular uptake and skin permeation test. Conclusion: Taken together, HA-PLGA NPs as a transdermal delivery carrier to hair follicle cells can be exploited to develop the efficient and effective platform of transdermal drug delivery for the treatment of various diseases.

        Novel synthetic pathway for methyl 3-hydroxybutyrate from b-hydroxybutyric acid and methanol by enzymatic esterification

        M3HB (Methyl 3-hydroxybutyrate), known as an alternative energy source for cells with impaired metabolicfunction, has recently been reported to have therapeutic effects on Alzheimer’s disease and inhibitionof apoptosis. In this study, the enzymatic conversion of M3HB from methanol and betahydroxybutyricacid (BHB) by lipase-catalyzed esterification was performed for the first time. The reactionconditions for maximum M3HB conversion were as follows: Novozym 435, 15 g/L; BHB:methanolmolar ratio, 1:2; reaction temperature, 40 C; and organic solvent, chloroform. The lipase-based M3HBsynthesis achieved about 94.6% conversion through a step-by-step optimization, and is expected to contributeto future bioindustry applications.

        Generation of a Human Monoclonal Antibody to Cross-Reactive Material 197 (CRM<sub>197</sub>) and Development of a Sandwich ELISA for CRM<sub>197</sub> Conjugate Vaccines

        Cross-reactive material 197 (CRM<sub>197</sub>) is a non-toxic mutant of diphtheria toxin containing a single amino acid substitution of glycine 52 with glutamic acid. CRM<sub>197</sub> has been used as a carrier protein for poorly immunogenic polysaccharide antigens to improve immune responses. In this study, to develop a sandwich ELISA that can detect CRM<sub>197</sub> and CRM<sub>197</sub> conjugate vaccines, we generated a human anti-CRM<sub>197</sub> monoclonal antibody (mAb) 3F9 using a phage-displayed human synthetic Fab library and produced mouse anti-CRM197 polyclonal antibody. The affinity (K<sub>D</sub>) of 3F9 for CRM<sub>197</sub> was 3.55 nM, based on Bio-Layer interferometry, and it bound specifically to the B fragment of CRM<sub>197</sub>. The sandwich ELISA was carried out using 3F9 as a capture antibody and the mouse polyclonal antibody as a detection antibody. The detection limit of the sandwich ELISA was <1 ng/ml CRM<sub>197</sub>. In addition, the 3F9 antibody bound to the CRM<sub>197</sub>-polysaccharide conjugates tested in a dose-dependent manner. This ELISA system will be useful for the quantification and characterization of CRM<sub>197</sub> and CRM<sub>197</sub> conjugate vaccines. To our knowledge, this study is the first to generate a human monoclonal antibody against CRM197 and to develop a sandwich ELISA for CRM<sub>197</sub> conjugate vaccines.

        Correlation with Daily Life, Physical Activity, Pain, and Degree of Disability of Office Workers with Non-Specific Chronic Neck Pain

        Objective: The purpose of this study was to prepare evidence for the prevention and management of nonspecific chronic neck pain (NSCNP) by examining the correlation between activity of daily living and physical activities of office workers with NSCNP. Design: Crossed-sectional studyMethods: 89 patients with NSCNP were recruited for this study. But 2 subjects met the exclusion criteria and were dropped out, and 86 subjects finally participated. Numerical pain rating scale (NPRS) and neck disability index (NDI) were used to check the pain intensity and disability of patients with neck pain, respectively. For the activity of daily living, computer use time, sleep time, and driving time were used. To find out the physical activities of the subjects, International Physical Activity Questionnaires (IPAQ-SF) was used. Correlation analysis was performed to find out the correlation of each variable. Results: A clear positive correlation was established between computer use time and pain (p<0.05), and a clear positive correlation was established between computer use time and disability index (p<0.05). The correlation between NPRS and NDI and physical activity total time, high intensity activity score, moderate intensity activity score, and walking score were not statistically significant (p>0.05). Conclusions: In treating patients with NSCNP, it is necessary to reduce the computer usage time as a professional factor or to educate the proper posture. In addition, rather than emphasizing physically comprehensive physical activity, grafting therapeutic exercise directly related to neck pain could have a more positive effect on NSCNP patients.

        Social and cultural biases and constraints have prohibited women from equally benefiting from economic development, resulting in feminization of poverty. Development assistance will be effective only when it can tackle gender inequality and promote women's empowerment. It is in this respect that Korean Women's Development Institute undertook a two-year research to develop strategies for gender mainstreaming of Korea's development assistance. While OECD/DAC and major donors consider gender as a cross-cutting issue, it is a separate sector of its own in Korea's ODA. As a result, projects are women-targeted than gender-responsive and women-targeted projects are rather oriented to basic human needs such as health than gender equality and women's empowerment. According to a recently adopted three year strategy, gender is selected one of highly significant thematic areas and gaining a bigger support in Korea's development assistance. As Korea is fast growing as a donor country, it is required that the country develops a systematic development assistance principle and institution for gender equality and cultivates gender experts in the field. Gender-responsive M&E guidelines and tools along with project design mechanisms need to be developed to ensure the application of the principle. Gender training for staff, experts and volunteers is in great and urgent need as well. It is also required that the country prepare itself for OECD/DAC membership by educating itself with gender equality guidelines and gender marker. The country will truly benefit from the membership when it actively participates in policy networks such as GENDERNET.

        메타버스 활용 한국어교육에 대한 교사 인식과 교육적 적용 연구

        본 연구는 메타버스 활용 한국어교육에 대한 교사 인식과 교육적 적용 관련 견해를 종합적으로 검토하고 이를통해 한국어교육에의 메타버스 적용 가능성을 탐색하는 데 궁극적인 목적이 있다. 이를 위해 메타버스 체험 여부가 향후 메타버스의 교육적 활용에 대한 인식에 미치는 영향을 감안하여 메타버스에 대한 교사 인식을 확인해 보고자 하였다. 또한 메타버스를 직접 체험해 본 교사들을 대상으로 메타버스를 한국어교육에서 어떻게 활용할 수있을지 메타버스 활용 수업의 가치와 필요성, 메타버스 에 적합한 활동 유형 및 수업 구성 방안, 메타버스 활용수업에서 예상되는 어려움과 지원 방향의 관점에서 요구를 확인해 보고자 하였다. 연구 결과 메타버스를 체험해본 교사 집단이 메타버스에 대한 이해도가 높고 유용성에 대한 인식, 효능감 면에서 통계적으로도 유의했다. 그러나 효능감이 매우 낮게 나온 미체험 집단에서도 사용 의지가 높았으며 체험 집단과 차이를 보이지 않아 흥미로웠다. 한국어 교육 현장에서 메타버스의 장점을 잘 활용하려면 역할극과 시뮬레이션, 상호작용이 있는 소그룹 학습활동과 같은 말하기 활동에 적합함을 확인할 수 있었다. 그리고 교사들이 메타버스 활용 교육의 필요성을 느끼고있다는 점을 반영하여, 향후 교원 양성, 각종 재교육 프로그램의 교과 내용에는 물론, 교원 자격 인증의 조건인학습 필요 과목 목록에도 이러한 내용이 포함되어야 할 것이다. The ultimate purpose of this study is to comprehensively review teachers’ perceptions of metaverse -based Korean language education and views related to educational application, and to explore the possibility of metaverse application to Korean education through this. For this purpose, the instructor’s perception of metaverse-based education was studied, considering effect of their prior metaverse experience. In addition, educational application of metaverse was investigated from the perspective of the value and necessity of metaverse-based education, factors to consider when planning metaverse-based education, and expected difficulties and support directions of metaverse-based education. As a result of the study, the group of instructors who had experience with metaverse had higher understanding of the metaverse and were also statistically significant in terms of perceived usefulness and sense of efficacy. However, even in the unexperienced group, which showed a very low sense of efficacy, the behavioral intention to use was high, not showing a significant difference from the experienced group. In order to fully take advantages of metaverse in Korean language education, it was confirmed that speaking activities such as role play, simulation, and small group learning activities with interaction were the most effective. Furthermore, the necessity of metaverse-based education must be included in instructors’ training and re-training programs, as well as in the required courses for instructor qualification program.

        Remote cerebellar hemorrhage after a neurosurgical procedure: A report of three cases

        Remote cerebellar hemorrhage (RCH) is a rare complication of neurosurgical procedures and is characterized by a typical bleeding pattern defined as the “zebra sign.” Only few cases of RCH have been reported in the English literature, and its pathophysiology remains unclear. In this report, we present the cases of three patients with RCH after three different procedures: burr-hole trephination and chronic subdural hematoma evacuation of bilateral cerebral convexity with subsequent subdural drain insertion, lumbar drainage for cerebrospinal fluid divergence for thoracic endovascular aortic repair, and combined bypass surgery for moyamoya disease.
