<P><B>Abstract</B></P> <P><B>Introduction</B></P> <P>The detection of <I>BRCA1/2</I> mutations is important because PARP1 inhibitors are approved for germline and/or somatic <I>BRCA</I>-mutated advanced ovarian cancer. Next-generation sequencing (NGS) is increasingly used in clinical practice for <I>BRCA1</I>/2 mutations. The purpose of this study was to consider several conditions of NGS <I>BRCA1/2</I> assay applicable to clinical laboratory tests, in particular for using formalin fixed paraffin embedded (FFPE) ovarian tissues.</P> <P><B>Materials and methods</B></P> <P>We selected 64 ovarian cancer patients and performed Oncomine™ BRCA assay using FFPE tissue. Effect of FFPE sample quality was analyzed by NGS quality parameters including deamination metric. Somatic variants were selected by removing germline variants of peripheral blood and interpreted as pathogenic, variants of unknown significance, and false positive.</P> <P><B>Results</B></P> <P>We found a positive relationship between the number of variants over the deamination metric and FFPE age (<I>P</I> < 0.001) with a cutoff values of approximately 0.7 and 60 months, respectively. When comparing NGS results with Sanger sequencing, NGS misreported 3 of 15 variants using default parameters which were corrected after changing parameters. We detected somatic variants in eight patients and classified them into pathogenic (n = 3), VUS (n = 3) and false positive (n = 2).</P> <P><B>Conclusions</B></P> <P>This study is important for improving <I>BRCA1/2</I> mutation detection capabilities of NGS analytical pipelines and strategy to overcome their limitations using FFPE tissue in ovarian cancer patients.</P>

<P>A sticker-type system with hybrid integration of CMOS IC and organic optical sensors is proposed to monitor photoplethysmogram (PPG) signals. To solve problems with the previous solely organic sensor-based works, CMOS IC is implemented in 180 nm technology under 5 V/1.5 V dual power supply. The silver-wire printed planar-fashionable circuit board (P-FCB) is used to connect the CMOS IC with organic sensors. The proposed hybrid system has the five following key features: 1) Power-efficient structure of organic sensor; 2) Integrated analog front-end and digital processor; 3) Degradation compensation scheme; 4) Large parasitic elements optimized design; and 5) Motion artifact rejection scheme. The stickertype PPG monitoring system has mass of only 2g, including the batteries, and consumes only 233 mu W to operate. The PPG signal could be acquired from various body parts (finger, wrist, and neck). The peripheral oxygen saturation level (SpO(2)) extraction results are verified by comparison with a commercial sensor device.</P>

<P>A small form-factor, lightweight wireless capsule endoscope (WCE) system with body channel communication (BCC) transceiver ICs is proposed for VGA-resolution image transferring and capsule localization. The transceiver ICs are composed of capsule chip and receiver chip, implemented in a 65-nm CMOS process. In the capsule side, the proposed system provides 360° image capturing through four-camera integration to reduce the miss rate. Also, a dual-band pulse-shaping BCC transmitter is proposed to enable low-power (<1 mW), high-speed (80 Mb/s) image transmission. In the receiver side, contact attenuation compensated-received signal strength indicator (CAC-RSSI) is proposed to increase the capsule localization accuracy. The external system is composed of eight-receiver nodes that allow signal demodulation and localization of the capsule. To reduce transceiver payload and system power consumption, image encoder is implemented in the capsule. The transceiver IC is integrated into the capsule system and verified with body mimicking phantom. The proposed system satisfies compatibility with medical grade diagnosis by operating longer than 8 h with 4 fps and 12 h with 2 fps with conventional two silver oxide 55-mA · h coin batteries. The tested localization accuracy shows less than sub-centimeter range.</P>

A uterocutaneous fistula is rarely reported clinical condition after uterine procedures. Many diagnostic and management strategies are being suggested. In this case report, uterocutaneous fistula after pelviscopic myomectomy was diagnosed simply with hystero-salpingo contrast sonography and managed by surgical tract excision without hysterectomy and uterine wall dehiscence repair combined with medical treatment using gonadotropin-releasing hormone agonist succeeded to preserve fertility in young woman.

<P>A wearable electrical impedance tomography (EIT) system is proposed for the portable real-time 3-D lung ventilation monitoring. It consists of two types of SoCs, active electrode (AE)-SoC and Hub-SoC, mounted on wearable belts. The 48-channel AE-SoCs are integrated on flexible printed circuit board belt, and Hub-SoC is integrated in the hub module which performs data gathering and wireless communication between an external imaging device. To get high accuracy under the variation of conductivity, the dual-mode current stimulator provides the optimal frequency for time difference-EIT and frequency difference-EIT with simultaneous 4 k-128 kHz impedance sensing. A wide dynamic range instruments amplifier is proposed to provide 94 dB of wide dynamic range impedance sensing. In addition, the 48-channel AE system with the dedicated communication and calibration is implemented to achieve 1.4-m Omega sensitivity of impedance difference in the in vivo environment. The AE-/Hub-SoCs occupy 3.2 and 1.3 mm2in 65-nm CMOS technology and consume 124 mu W and 1.1 mW with 1.2 V supply, respectively. As a result, EIT images are reconstructed with 90% of accuracy, and up to 10 frames/s real-time 3-D lung images are successfully displayed.</P>

<P><B>Abstract</B></P> <P>A series of Fe-30 wt%Mn-10.5 wt%Al-1.1 wt%C steels with Mo addition from 0 to 5 wt% were prepared to investigate the effect of Mo on the microstructure and tensile deformation behavior of austenitic lightweight steel. When the Mo content was below 4 wt%, the microstructure of solution-treated samples consisted of austenite and κ–carbide, while Mo-enriched M<SUB>6</SUB>C and M<SUB>23</SUB>C<SUB>6</SUB> carbides were additionally precipitated in samples containing Mo more than 4 wt% during a solution treatment at 1050 °C. These carbides inhibited austenite grain growth during the solution treatment, resulting in significant grain refinement in the samples containing more that 4 wt% of Mo. Tensile test results showed that the yield strength gradually decreased with an increase in the Mo content up to 3 wt% due to the suppression of κ–carbide precipitation, whereas it significantly increased when the Mo content exceeded 4 wt% due to grain refinement and precipitation strengthening caused by Mo-enriched carbides. During the tensile deformation, the strain hardening rates of all alloys increased and then the deformation mode subsequently changed with an increase in the Mo content from shearband-induced plasticity (SIP) to microband-induced plasticity (MBIP). Finally, the change in the κ-carbide precipitation behavior upon the addition of Mo and its effect on the deformation behavior were carefully analyzed and discussed through nanoindentation experiments, first-principles calculations and atom probe tomography analyses.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Strawberry (Fragaria ${\times}$ ananassa) is a globally-cultivated and popular fruit crop, prized for its flavor and nutritional value. Sweetness, a key determinant of fruit quality, depends on the sugar composition and concentration. We selected eight strawberry cultivars based on the fruit soluble solids content to represent high and low sugar content groups. The average soluble solid content was $13.6^{\circ}Brix$ (Okmae, Geumsil, Aram, and Maehyang) and $2.9^{\circ}Brix$ (Missionary, Camino Real, Portola, and Gilgyung53), for the high and low sugar content groups, respectively. Sucrose was the main sugar in the cultivars with high sugar content, whereas fructose was the main component in the low sugar content cultivars. Fruit starch concentration ranged from $3.247{\pm}0.056$ to $3.850{\pm}0.055g/100g$, with a 12% higher concentration in the high sugar content cultivars. Additionally, we identified 41 sugar metabolism-related genes in Fragaria ${\times}$ ananassa and analyzed the relationship between their transcripts and the sugar accumulation in fruit. FaGPT1, FaTMT1, FaHXK1, FaPHS1, FaINVA-3, and FacxINV2-1 were highly expressed in the high sugar content cultivars, while FapGlcT, FaTMT2-1, FaPHS2-1, FaSUSY1-1, and FaSUSY1-2 were highly expressed in the low sugar content cultivars. In general, a greater number of genes encoding sugar transporters or involved in sugar synthesis were highly expressed in the high sugar content cultivars. Contrarily, genes involved in sugar degradation were preferentially transcribed in the low sugar content cultivars. Although gene expression was not perfectly proportional to sugar content or concentration, our analysis of the genes involved in sugar metabolism and accumulation in strawberries provides a framework for further studies and for the subsequent engineering of sugar metabolism to enhance fruit quality.
JYNNEOS vaccine safety monitoring in the Republic of Korea, 2022: a cross-sectional study

Objectives: With the recent global mpox outbreak, the JYNNEOS vaccine (Modified Vaccinia Ankara-Bavarian Nordic) was developed as a third-generation smallpox vaccine and initially favored for mpox immunization. Vaccine-associated side effects contribute to vaccine hesitancy. Consequently, tracking adverse events post-immunization is crucial for safety management. This study used data from the national active vaccine safety surveillance conducted in Korea from August 25 to November 24, 2022 to detect potential safety signals and adverse events. Methods: Data on health conditions following vaccination were gathered from web-based surveys and reported via active surveillance through the Immunization Registry Information System. This follow-up system functioned via a text message link, surveying adverse events and health conditions beginning on the second day post-vaccination. Information about specific adverse events, including both local and systemic reactions, was collected. Results: The study included 86 healthcare workers who had received at least 1 dose of the JYNNEOS vaccine. Among the respondents, 79.1% reported experiencing at least 1 adverse event, with the majority being local reactions at the injection site. The incidence of adverse events was higher following the first dose (67.9%) than after the second dose (34.4%). The most frequently reported adverse event for both doses was mild pain at the injection site. Conclusion: The study provides crucial information on the safety of the JYNNEOS vaccine, demonstrating that most adverse events were manageable and predominantly localized to the injection site. Nonetheless, additional research is needed on the safety of various vaccine administration techniques and the vaccine’s effects on broader demographics.

Purpose: On the basis of evidence, we aimed to reevaluate the necessity of the empirical proton pump inhibitor (PPI) trial for children with suspected gastroesophageal reflux disease (GERD). Methods: We analyzed the frequency of GERD in 85 school-age children with gastroesophageal reflux (GER) symptoms, who received 24-hour esophageal pH monitoring and/or upper endoscopy. According to the reflux index (RI), the children were classified into normal (RI <5%), intermediate (5%≤ RI <10%), or abnormal (RI ≥10%) groups. Results: Fifty six were female and 29 were male. Their mean age was 12.6±0.5 (±standard deviation) years (range: 6.8–18.6). The RI analysis showed that the normal group included 76 patients (89.4%), the intermediate group included 6 patients (7.1%), and the abnormal group included 3 patients (3.5%). The DeMeester score was 5.93±4.65, 14.68±7.86 and 40.37±12.96 for the normal, intermediate and abnormal group, respectively (p=0.001). The longest reflux time was 5.56±6.00 minutes, 9.53±7.84 minutes, and 19.46±8.35 minutes in the normal, intermediate, and abnormal group, respectively (p=0.031). Endoscopic findings showed reflux esophagitis in 7 patients. On the basis of the Los Angeles Classification of Esophagitis, 5 of these patients were included in group A, 1 patient, in group B and 1 patient, in group C. Conclusion: The incidence of GERD was very low in school-age children with GER symptoms. Therefore, injudicious diagnostic PPI trials would be postponed until the actual prevalence of GERD is verified in future prospective studies.
